Applicability of QbD-assisted Analytical Method for Simultaneous Detection of Tetrahydrocurcumin and Folic Acid in Developed Nanostructured

The present research explored the usefulness of (5929IN008, application number 202211045937), a combination of tetrahydrocurcumin and folic acid-loaded nanostructured lipidic carriers. Objectives:: To develop and validate a QbD-assisted method for simultaneous analysis of tetrahydrocurcumin (THC) and folic acid (FA). Applicability of the above method to determine total drug content (TDC) and entrapment efficiency (EE) of nanostructured lipid carriers (NLCs) loaded THC and FA. Methods:: A high-performance liquid chromatographic (HPLC) method was developed, optimized and validated using Box-Behnken design for improved method performance. Chromatographic separation was conducted on a Supelco 250 x 4.6 mm (5 μm) column with optimized mobile phase composition containing tetrahydrofuran: citric acid buffer pH 3.5 (50:50) at a flow rate of 0.4 mL.min-1 and diode array detection between 210 and 360 nm. Results:: The method developed in a concentration range of 1 to 100 μg.mL-1 was found to be linear (R2 0.999, p≤0.001), accurate (99.10-101.70%) and precise with high recovery values in intra and inter-day results. The system adaptability and robustness evaluation revealed that the percent recovery ranged from 96.90 to 102.80%, and the percent relative standard deviation (%RSD) values were less than 2%. Moreover, the method was further applied for the determination of TDC (86±6% and 96±8%) and drug EE (81±21% and 73±13%) for THC and FA, respectively. Conclusion:: The investigation indicated the applicability of the developed and validated method for the estimation of THC and FA in the developed nanostructured lipidic carriers
Wilderness Medical Society Clinical Practice Guidelines for the Treatment of Acute Pain in Austere Environments: 2024 Update
The Wilderness Medical Society (WMS) convened an expert panel to develop evidence-based guidelines for the management of pain in austere environments. Recommendations are graded based on the quality of supporting evidence as defined by criteria put forth by the American College of Chest Physicians. This is an update of the 2014 version of the “WMS Practice Guidelines for the Treatment of Acute Pain in Remote Environments” published in Wilderness & Environmental Medicine 2014; 25:41–49
Vulnerability Revisited: Leaving No One Behind in Research
This open-access book discusses vulnerability and the protection-inclusion dilemma of including those who suffer from serious poverty, severe stigma, and structural violence in research. Co-written with representatives from indigenous peoples in South Africa and sex workers in Nairobi, the authors come down firmly on the side of inclusion. In the spirit of leaving no one behind in research, the team experimented with data collection methods that prioritize research participant needs over researcher needs. This involved foregoing the collection of personal data and community researchers being involved in all stages of the research. In the process, the term ‘vulnerability’ was illuminated across significant language barriers as it was defined by indigenous peoples and sex workers themselves. The book describes a potential alternative to exclusion from research that moves away from traditional research methods. By ensuring that the research is led by vulnerable groups for vulnerable groups, it offers an approach that fosters trust and collaboration with benefits for the community researchers, the wider community as well as research academics
Creative approaches to landscape research: Using multisensory and multispecies research perspectives with marginalised groups
The research project discussed here used artist-led workshops to enable people from marginalised groups to explore and communicate their relationships with nature and landscapes. Aligning with multisensory and multispecies methodologies, five workshops took place in England’s north-west, in which participants from marginalised and excluded communities co-produced creative works representing their sensorial responses to those environments. Drawing on participants’ experiences of these workshops, and the resulting creative works, the affordances of location-specific creative activities as a means of facilitating connection with local landscapes are examined, as are the opportunities that creative methodologies offer in enabling marginalised and disadvantaged groups to engage in debates about nature and landscape use. Participants benefitted greatly from the project, which enabled small steps towards involving marginalised groups in debates about nature and landscape use. However, challenges remain for decision-making and a more equal distribution of power amongst humans and other species whose interests may be overlooked
Entrepreneurship education methods and venture creation intention: evidence from a developing country context
The purpose of this paper is to explore the use and impact of entrepreneurship education methods on students' entrepreneurial intentions. The study used a survey design to sample 555 primary data from undergraduate students in one public and one private university in Ghana. We analysed the data using structural equation modelling, exploratory and confirmatory factor analyses. The results of the structural equation modelling show that active methods of education were more effective in influencing students' entrepreneurial intention, although passive methods of teaching were dominant among most educators. The paper also reveals the influence of gender and personal characteristics on students' venture creation intentions. The findings, recommendations, and limitations are discussed below
Navigating the Post-Pandemic Normal: Learning from the Experiences of Cyprus-Based Female Researchers during the COVID-19 Pandemic
The article addresses the new normal for female researchers in the post-pandemic era by utilising information collected from the pandemic period, when disruption to ‘business as usual’ occurred. This information can inform a new normal that is as efficient, resilient, and ethical as possible. The research employs a case study methodology, with qualitative data collection and analysis approaches. To understand the circumstances faced by researchers in Cyprus during the pandemic, it was necessary to approach several scientists from different research areas and try to understand their experiences through conversations. The set of notes compiled from the completion of the informal conversations were analysed using thematic analysis. Three themes were identified: (a) misperceptions of flexibility in researchers’ schedules and workload, (b) lack of flexibility in adjusting expected outcomes or timelines, and (c) inability to compartmentalise personal and professional life. To better contextualise these findings, the study additionally explored survey results published by the European Commission on relevant topics, enabling a critical juxtaposition of the European perspective to the Cyprus-based findings of these pandemic-induced challenges. The study highlights significant misconceptions and overlooked dynamics regarding research conducted in times of crisis and offers avenues for ensuring research quality in analogous future scenarios
Real-time Monitoring of 3D Printing Using Between-layer Structural Similarity (BLSS)
This research introduces an innovative method specifically designed for comprehensive error detection in the 3D printing process. The Between-layer Structural Similarity (BLSS) technique gauges the similarity between displacement maps, which are generated using the Structural Similarity Index Measure (SSIM). This allows for the differentiation between consecutive layers in both simulated and actual prints. Utilising the Fast Fourier Transform (FFT), the displacement maps can be converted to the frequency domain and assessed for similarity. The proposed approach involves three key processing stages: print simulation and processing, print capture and processing, and BLSS-based structural analysis and evaluation. This methodology not only reduces material wastage but also lays the foundation for automated systems capable of halting or terminating live printing processes upon error detection. Experimental results reveal that printing defects occur below 97.5 % of the similarity value, establishing this threshold as appropriate. This underscores the effectiveness of the system in error detection and its potential for real-time quality control in 3D printing
The positional demands of explosive actions in elite soccer: Comparison of English Premier League and French Ligue 1
The aims of the present study were to: (i) quantify accelerations and decelerations of soccer players during match-play across two consecutive seasons from the English Premier League (EPL) and Ligue 1 (L1); and (ii) compare any positional differences between the two leagues. Fifty-eight male professional soccer players were monitored during all league matches (n = 144) across seasons 2020/21 and 2021/22. The absolute number of accelerations (> +3 m/s ) and decelerations (< -3 m/s ) and accelerations and decelerations per minute were examined. The relative number of accelerations and decelerations across all positions was higher with moderate effect sizes in the EPL when compared to L1 (p < 0.001, for both). Significant differences were observed in accelerations and decelerations across all playing positions (p < 0.001 and p = 0.001 respectively, with moderate to very large effect sizes), except for centre forwards (CF) (accelerations p = 0.40; ES = 0.16; decelerations p = 0.97; ES = 0.01). This study provides valuable insights into the positional acceleration and deceleration differences in the EPL and L1, which should be considered in match running performance evaluations. While confirming higher accelerations and decelerations in the EPL, the unique case of CF challenges current evidence, emphasising the need for a more granular understanding of the positional demands of explosive actions incorporating accelerations and decelerations in elite soccer.
