    Small molecules and heat treatments reverse vernalization via epigenetic modification in Arabidopsis

    Monocarpic plants flower only once and then produce seeds. Many monocarpic plants require a cold treatment known as vernalization before they flower. This requirement delays flowering until the plant senses warm temperatures in the spring. Exposure to high temperatures following vernalization causes devernalization, which cancels the vernalized state, inhibiting flowering and promoting vegetative growth. In this study, we screened over 16,000 chemical compounds and identified five small molecules (devernalizers; DVRs) that induce devernalization in Arabidopsis thaliana at room temperature without requiring a high-temperature treatment. Treatment with DVRs reactivated the expression of FLOWERING LOCUS C (FLC), a master repressor of flowering, by reducing the deposition of repressive histone modifications, thereby delaying flowering time. Three of the DVRs identified shared two structures: a hydantoin-like region and a spiro-like carbon. Treatment with DVR06, which has a simple chemical structure containing these domains, delayed flowering time and reduced the deposition of repressive histone modifications at FLC. RNA-seq and ChIP-seq analyses revealed both shared and specific transcriptomic and epigenetic effects between DVR06- and heat-induced devernalization. Overall, our extensive chemical screening indicated that hydantoin and spiro are key chemical signatures that reduce repressive histone modifications and promote devernalization in plants.journal articl

    Cross-Correlation Enhancement with GNSS-Derived SPM-Delay Using Azimuth Angle Filtering Towards Dust and Sandstorm Detection in Japan

    This letter proposes the application of the precise point positioning (PPP) technique used in the GNSS for detecting the dust and sandstorm (DSS) in the atmosphere. Many of the papers have reported that the GNSS-derived zenith total delay (ZTD) is affected to aerosols and sandstorms, however, directivity for arrival angles of sandstorms is not considered. Since the suspended particulate matter (SPM) including the yellow sand and PM2.5 usually comes from the west side from Japan, azimuthal angle filtering is introduced in the process of estimation of the ZTD. The enhancement in the cross-correlation between the proposed SPM-induced delay and measured time series of the ground SPM has shown.journal articl

    Large Language Models Can be Good Medical Annotators: A Case Study of Drug Change Detection in Japanese EHRs

    In this study, we combined automatically generated labels from large language models (LLMs) with a small number of manual annotations to classify adverse event–related treatment discontinuations in Japanese EHRs. By fine-tuning JMedRoBERTa and T5 on 6,156 LLM-labeled records and 200 manually labeled samples and then evaluating on a 100-record test set, T5 achieved a precision of 0.83, albeit with a recall of only 0.25. We noted that when training solely on the 200 human-labeled samples (that contained significantly few positive cases), the model failed to detect any adverse events, making a reliable measurement of precision or recall infeasible (that is, N/A). This underscores the potential of large-scale LLM-driven labeling as well as the need to improve recall and label quality in practical clinical scenarios.conference pape

    Model Predictive Control for a Soft Robotic Finger with Stochastic Behavior based on Fokker-Planck Equation

    The inherent flexibility of soft robots offers numerous advantages, such as enhanced adaptability and improved safety. However, this flexibility can also introduce challenges regarding highly uncertain and nonlinear motion. These challenges become particularly problematic when using open-loop control methods, which lack a feedback mechanism and are commonly employed in soft robot control. Though one potential solution is model-based control, typical deterministic models struggle with uncertainty as mentioned above. The idea is to use the Fokker-Planck Equation (FPE), a master equation of a stochastic process, to control not the state of soft robots but the probabilistic distribution. In this study, we propose and implement a stochastic-based control strategy, termed FPE-based Model Predictive Control (FPE-MPC), for a soft robotic finger. Two numerical simulation case studies examine the performance and characteristics of this control method, revealing its efficacy in managing the uncertainty inherent in soft robotic systems.conference pape
