Multiple charged amino acids of Plasmodium falciparum SURFIN4.1 N-terminal region are important for efficient export to the red blood cell
Plasmodium falciparum, an obligate intracellular protozoan parasite which causes the severe form of human malaria,
exports numerous proteins to the infected red blood cell that are important for its survival and of severe pathological effect to its host. These proteins and their export mechanisms are candidates for drug
and vaccine development, and among them is the Plasmodium SURFIN family of proteins. Previously we showed that the N-terminal region along with the sequence surrounding the transmembrane domain of SURFIN 4.1 is essential for its export to Maurer's clefts in the red blood cell cytoplasm. We proposed that this region is recognized by a machinery responsible for protein translocation across the parasitophorous vacuole membrane surrounding the parasite.
To understand the export mechanism further, we utilized a fluorescent protein-tagged mini-SURFIN 4.1 consisting of the minimum essential components for export. Alanine scanning of all charged amino acids
within the N-terminal region revealed that replacement of 3 glutamic acid and 2 lysine residues significantly impairs the export efficiency of this protein across the parasitophorous vacuole membrane. In addition,N-terminally Myc-tagged mini-SURFIN 4.1 and mini-SURFIN 4.2 with similar architectures were detected with anti-Myc antibody at Maurer's clefts, indicating that elements required for export to Maurer's clefts are conserved between SURFIN 4.1 and SURFIN 4.2 , and that N-terminal sequences of these SURFIN members are not cleaved during export. Our results implicate a conserved nature of SURFIN export
to the red blood cell, particularly an important role of multiple glutamic acid and lysine residues in the SURFIN N-terminal region
BODY COOLING EFFECTS OF IMMERSION IN HIGH-CONCENTRATION ARTIFICIAL CARBONIC ACID WATER
This study examines the body cooling effects of immersion in high-concentration artificial carbonic acid water. The subjects’ core (tympanic) temperature was increased by a pedaling ergometer. They then immersed their lower legs in carbonic acid water or tap water. The decrease in tympanic temperature was observed. The tympanic temperature significantly decreased more during immersion in stirred 25 / 30°C CO2 water than in tap water. The results suggest that immersing the lower legs in stirred 25 / 30°C carbonic acid water can suppress skin vascular constriction and reduce core temperature elevated by heat stress / exercise more effectively than stirred tap water of the same water temperature
肝細胞癌に対するTACE時の動脈損傷:損傷のリスク因子と残存狭窄の予測因子について
Purpose: To investigate clinical features and results of follow-up in patients with arterial injury during transcatheter arterial chemoembolization (TACE) for hepatocellular carcinoma. Methods: From 2005 to 2015, 2219 TACE procedures were performed in 906 patients in our hospital. Iatrogenic arterial injury occurred during 38 TACE procedures (sessions) in 35 patients (24 men, 11 women; mean age 71.8 years). The incidence of arterial injury was 1.7%. We evaluated the characteristics of arterial injury, and evaluated the risk factors for incomplete recanalization of the injured artery at follow-up angiography conducted after 1–11 months (mean 102.5 days).
Results: Iatrogenic arterial injury was caused by the microcatheter in 34 of 38 cases (86.8%). There were 15 cases (39.5%) in which the replaced hepatic artery or the extrahepatic artery was the parasitic supply. Extravasation occurred in five cases. In 36 of 38 cases, follow-up angiography was performed. We divided 36 cases into two groups: complete reopening (n = 24) and non-complete reopening (n = 12). The two groups were compared regarding the factors associated with incomplete recanalization of the injured artery at follow-up. Injury length >3 cm (p = 0.0002) and proximal arterial injury (proximal to the segmental artery; p = 0.03) were significant risk factors for non-complete reopening of the injured artery. Conclusion:
Iatrogenic arterial injury frequently occurred in the extrahepatic artery or replaced hepatic artery.
