T6SS4 is heterogeneously expressed in Yersinia pseudotuberculosis and is a target for transcriptional and post-transcriptional regulation
No full textThe type VI secretion system (T6SS) is a complex secretion system encoded by many Gram-negative bacteria to translocate effector proteins directly into target cells. Due to its high complexity and energy-intensive firing process, regulation of the T6SS is tightly controlled in many organisms. 'Y. pseudotuberculosis' encodes four complete T6SS clusters but lacks genes implicated in T6SS gene regulation in other microorganisms, indicating a distinct control mechanism. Here, we could show that the T6SS4 of 'Y. pseudotuberculosis' is heterogeneously expressed within a population, which is determined by the transcriptional T6SS4 activator RovC. Moreover, the T6SS4 and RovC are embedded in a complex and global regulatory network, including the global post-transcriptional regulator CsrA, the 'Yersinia' modulator A (YmoA), the global protease Lon, and RNases (PNP and RNase III). Post-transcriptional processing of the T6SS4 polycistron and different transcript stability within the operon also achieve a higher regulatory complexity. In summary, our work provides new insights into the sophisticated and complex regulatory network of the T6SS4 of 'Y. pseudotuberculosis', which clearly differs from regulation in other organisms
Academic Career Pathways in German Medicine: Current Structures and International Comparisons
No full textIntroduction: Academic qualification processes in medicine are an essential component of excellent research and teaching. In Germany, these qualification paths are characterized by heterogeneity in the regulations both between the federal states and between the faculties. This lack of uniformity in Germany stands in contrast to internationally established models with standardized and transparent career paths. In view of growing international mobility and increasing global competition, the need to reform academic careers in medicine in Germany is increasingly coming into focus. Methods: On the basis of a systematic analysis, higher education laws, doctoral, habilitation and appointment regulations of German medical faculties were examined for defined target criteria. This was followed by a literature search in Pubmed and Google Scholar databases as well as freely accessible documents on the qualification systems of other international universities. Results: The evaluation reveals considerable differences in the requirements, regulations and transparency of academic qualification levels in Germany. There is a lack of standardization processes in German regulations, particularly in the area of habilitation and extraordinary professorship. In an international comparison, there are clear differences between countries both in the existence of qualification levels and the qualification path to the qualification level. However, many countries have structured and transparent qualification levels with underlying tenure-track models or performance-oriented criteria. Conclusion: The results of the academic qualification structures in medicine show a need for reform of the German academic system in an international comparison. The introduction of transparent and structured career paths, based on systems already established in other countries (PhD model/tenure track system), would contribute to harmonization and the promotion of international comparability and mobilit

No full textIm Juni 2011 wurde die wissenschaftliche Kommission Hochschulmanagement im Verband der Hochschullehrer für Betriebswirtschaft e. V. (VHB) zum Zeitschriftenranking dieses Verbandes,
VHB-JOURQUAL, befragt. Die Ergebnisse zeigen, dass eine große Mehrheit der Mitglieder und Interessenten der Kommission zumindest im Detail kritisch zu diesem Ranking eingestellt ist, wobei es von einer beachtlichen Minderheit grundsätzlich abgelehnt wird. Dabei gibt es zwei unterschiedliche Motive für die Kritik. Die einen empfinden deutschsprachige BWL-Zeitschriften als zu schlecht beurteilt, die anderen eher als zu gut und befürworten internationale Rankings. Es werden fünf konkrete Empfehlungen formuliert.The Section of Academic Management of the German Academic Association for Business Research (VHB) has been asked about the journal ranking of this association, VHBJOURQUAL, in June 2011. The results of this survey show that a large majority of members and interested persons of the section are critical about this ranking or at least some details of it, while a considerable minority rejects it fundamentally. However, there are two different motives for this criticism. One side perceives that German journals of business administration are ranked too low, whereas the other side thinks that these journals are ranked too high and advocates international rankings. Five concrete recommendations are formulated
Corporate Governance und Arbeitnehmermacht im Aufsichtsrat:Eine angewandt spieltheoretische Analyse
No full textThe discussion about employee representation on supervisory boards has received much attention from scholars and politicians around the world. We provide new insights to this ongoing debate by employing power indices from game theory to examine the 'real' power of employees on boards and its effect on firm performance. Based on unique panel data of the largest listed companies in Germany, we find an inversely U-shaped relationship between labour power and Tobin's Q with a value-maximising labour power of approximately 43 %. Our results are robust to different game theoretical calculations of labour power, as well as various econometric models.Die Diskussion über Arbeitnehmermitbestimmung in Aufsichtsräten hat erhebliche Aufmerksamkeit von Wissenschaftlern und Politiker in vielen Ländern erfahren. Wir tragen neue Einsichten zu dieser fortdauernden Debatte bei, indem wir spieltheoretische Machtindices verwenden, um die Macht von Beschäftigten und deren Beitrag zum Unternehmenserfolg zu bestimmen. Mit originären Paneldaten der größten deutschen gelisteten Unternehmen finden wir eine umgekehrt U-förmige Beziehung zwischen Arbeitnehmermacht und Tobin's Q, wobei die wertmaximierende Arbeitnehmermacht bei rund 43 % liegt. Unsere Ergebnisse sind robust sowohl hinsichtlich unterschiedlicher spieltheoretischer Berechnungen der Arbeitnehmermacht als auch verschiedener ökonometrischer Modellierungen
Perfectly Secure Oblivious Algorithms for Geometric Problems
No full textBeim wahlfreien Zugriff auf einzelne Speicherzellen hängen die Speicherzugriffsmuster von Programmen von deren Eingaben oder Parametern ab. Darunter können auch sensible Daten wie beispielsweise Passwörter oder Krankenakten fallen. Wenn ein Angreifer Kenntnis des Speicherzugriffsmusters erlangt, dann können auch diese Informationen offenbart werden. Data-Oblivious Algorithmen und Datenstrukturen vermeiden dieses Datenschutzrisiko, indem sie Seitenkanäle im Zusammenhang mit dem Speicherzugriffsmuster beweisbar schließen. In dieser Arbeit betrachten wir Data-Oblivious Algorithmen für mehrere geometrische Probleme: Wir nutzen Eigenschaften der Probleme, um effiziente Data-Oblivious Algorithmen zu entwerfen, und wir erweitern unsere Ansätze zu allgemeineren Strukturen und Techniken. Dabei entwickeln wir Oblivious Datenstrukturen als Werkzeug für unsere Algorithmen, die auch für sich allein genommen von Interesse sind

No full textCurrently, background, qualification and training of German top politicians are widely discussed by scholars and media. This paper provides new insights to this discussion by analysing significant differences between the chancellors, vice chancellors as well as ministers of the inner and residual cabinets of the German federal governments between 1949 and 2009 with respect to their socio-demographic backgrounds and educational, economic and political human capital. Applications of different statistical methods reveal that the ministers of the inner cabinet have the most advantageous social background and the best education. Vice chancellors score highest with regard to their economic human capital, measured here by board seats before their current offices. The average tenure in the federal government as well as the expertise in the actual headed department is highest for chancellors.Aktuell werden die Herkunft, Qualifikation und Bildung deutscher Spitzenpolitiker rege von

Hier schafft die Fibel Klarheit: Sie zeigt, dass meist Abbildungen legal ohne kostenpflichtige Lizenzen genutzt werden können. Sie beleuchtet die urheberrechtlichen Rahmenbedingungen, insbesondere die Gemeinfreiheit von Werken und ihrer Reproduktionen sowie das im wissenschaftlichen Kontext so wichtige Zitatrecht. Auch wird auf die Rechte der Eigentümer der abgebildeten Artefakte sowie persönlichkeits- und datenschutzrechtliche Bestimmungen eingegangen, die ebenfalls der Nutzung meist nicht im Wege stehen. This primer provides clarity by demonstrating that images can usually be used legally without costly licenses. It highlights the copyright framework, particularly the public domain status of works and their reproductions, as well as the right to quote, which is so important in an academic context. It also addresses the rights of the owners of the artifacts depicted, as well as privacy and data protection regulations, which in most cases do not prevent use either. It explains the conditions under which the use of images is permitted and why publishers’ demands for licensing of images are often not based on legal necessities
Ascorbate mitigates oxidative stress and hemin cytotoxicity in heme oxygenase-1 deficiency
No full textHeme oxygenase-1 (HO-1) deficiency manifests with severe metabolic abnormalities, yet the underlying mechanisms remain incompletely understood. We investigated metabolic adaptations in HO-1-deficient cells using patient-derived lymphoblastoid cells (LCL) and HEK293T knockout cells. Both cell types exhibited concordant metabolic signatures, characterized by a substantial reduction in intracellular ascorbic acid levels, despite compensatory upregulation of sodium-dependent vitamin C transporter 2 (SVCT2) and downregulation of GLUT1, compared to wild-type cells. Ultrastructural examination revealed abnormal mitochondrial morphology in both models. Both cell types showed higher baseline hydrogen peroxide levels compared to wild-type cells. Treatment with 2-phospho-L-ascorbic acid (AA2P) restored cell viability in both models upon hemin-induced stress, with protection requiring functional SVCT2-mediated uptake. AA2P supplementation attenuated the elevated H2O2 levels without altering glutathione redox homeostasis as measured by GSH/GSSG ratios. These findings illuminate ascorbic acid metabolism as a critical node in HO-1 deficiency pathophysiology and suggest ascorbic acid supplementation as a potential therapeutic strategy for this rare disorder
