Purification and Characterization of Aldehyde Dehydrogenase 2 (ALDH2)
In 2023, alcohol use disorders (AUDs) affected 28.9 million individuals in the United States, including adolescents. Among them, approximately 757,000 fell between the ages of 12-17. Aldehyde dehydrogenase 2 (ALDH2) is a mitochondrial enzyme which plays a critical role in alcohol metabolism, responsible for oxidizing the toxic byproduct of acetaldehyde into acetate. The oxidation of ethanol is essential in preventing the accumulation of acetaldehyde, which can lead to adverse physiological effects, including nausea, alcohol-induced liver damage and increased cancer susceptibility

Relationships In-the-making in Virtual Learning Communities: Theorizing Relationships in Critical Multicultural Education
This paper explores the complex, discursive constructions of relationships in online learning communities in the pursuit of equity and diversity. Using Braidotti’s (2019) posthuman theory and diverse intellectual traditions, the authors contemplate how subjectivities and relationality complicate discourse on relationships and relationalities, which are critical elements in multiculturalism, in an online learning community. Post-qualitative inquiry (PQI) is utilized to consider these complexities. The authors interrogate the entangled relationships in the online setting to go beyond mainstream research and identify effectiveness in online learning. Using images, metaphors, and ethnodramatic writing, the authors theorize the discursive meanings of relationships by interpreting vignettes of lived experience to extend the current challenge of sustaining the spirit of diversity and equity in education for all. The authors suggest exploring the complexities of subjectivities and implementing innovative research methodologies to imagine new terms and praxis in critical multicultural education
I\u27m Not Having Fun Anymore : A Qualitative Analysis of Gender and Harassment in Bars and Clubs
Sexual harassment, while often spoken about in hushed tones or ignored altogether, happens frequently and often loudly. 66 percent of women who have experienced sexual harassment report that the harassment took place in a public space, such as work, school, or on the street (Kearl, 2018). Harassment is not discreet, but rather normalized, and those who experience it are expected to brush it off or take it in their stride. However, such harassment and subsequent dehumanization has a deteriorating effect on the mental well-being of those affected (Graham et. al., 2016). This study aims to hone in on the effect frequent harassment has on women in the public space of bars and clubs, and how such harassment affects their mental health and their preparation for going, as well how they protect themselves and others from harassment or assault in these spaces
Voices of Chinese American Emerging Adults: Complex Racial Identities, Family Dynamics, and Mental Health Experiences
Although Asian Americans are the fastest growing racial minority group in the United States, they remain significantly underrepresented in mental health research. Furthermore, research on Asian American mental health often aggregates the experiences of different Asian ethnic groups, creating a limited understanding of how distinct ethnic groups may differ in their mental health experiences. This interview study with Chinese American emerging adults ages 18–25 from varied family contexts (single-race, multiracial, and adoptive households) examines how members of this population make sense of the relationships between their identity, family, and mental health. This thesis posits that Chinese Americans\u27 nuanced experiences—particularly around identity and familial relationships—contribute to both positive and negative experiences with mental health. The findings from this project contribute to our understanding of Chinese American emerging adult identities, the influence of cultural values related to family and social status, and the mental health experiences of this population. The findings from this thesis may be beneficial for developing future projects focusing on distinct Asian ethnic groups and for supporting practitioners who work with the Chinese American population
Systematic Review Examining How Black Youth in the United States Cope with Stressful and Traumatic Experiences
Coping is an important behavioral skillset that youth use to emotionally regulate after experiencing stress or trauma. Research has repeatedly highlighted the stressors that Black youth experience, often finding that Black youth face higher levels of stress and trauma than their White peers. However, there has been a lack of research in understanding how Black youth cope with these higher levels of stress. This systematic review includes 72 studies (N = 16,205 participants) examining the frequency and effectiveness of coping strategies used by Black youth in response to a trauma or stressor, along with the associations between utilized coping strategies and various functional outcomes. Although previous reviews categorized coping strategies into domains, factors, and specific strategies, this framework does not accurately reflect the complex coping methods Black youth develop in response to the complex challenges stemming from capitalism and anti-Black systemic oppression. Findings indicate that, broadly, Black youth employ adaptive coping strategies; however, they also utilize complex, situation-specific strategies that best fit their unique experiences, with results showing adaptive and maladaptive strategies being associated with a mix of positive and negative outcomes. Finally, we critique the limitations of current coping research involving Black youth and suggest a research agenda for future studies to develop interventions that genuinely reflect and support their needs
Experiences and Perceptions of Gaslighting in the Workplace
The topic of gaslighting has gained increasing traction over the last several years. People have begun researching it in real-world settings such as relationships, politics, and the workplace. Furthermore, researchers have looked at the effects it has on the victim. However, there has been limited research on how one might react to an instance of gaslighting. This study examines how the victim’s race and the perpetrator’s position of power influence third parties’ recommendations on how to handle a scenario of workplace gaslighting. Psychology 101 students (N = 154) were randomly assigned to a 2 (race of victim: white vs black) x 2 (power position of perpetrator: co-worker vs supervisor) factorial design where they were asked to read a scenario depicting gaslighting in the workplace and report how likely they would recommend the five next steps: direct confrontation, report to HR, increase likeability, discuss with coworker, and avoid. Power had a significant main effect on direct confrontation, such that individuals were more likely to recommend direct confrontation for a coworker than a supervisor. There were no main effects of race or any interactions for any of the recommendations
Ultramodern Racism: A Critical Discourse Analysis of the Impact of Anti-Crt Legislation on Us History Curriculums
In the wake of Donald Trump’s failed 2020 bid for a second presidential term, there was a surge in anti-CRT legislation targeting how issues of race and racism could be taught in public school curricula. While proponents claim that anti-CRT laws will protect students of all races from discrimination, curriculums have historically been used as a tool by those with symbolic power to legitimize white supremacy and perpetuate anti-Black and color-evasive racism. Two states--Florida and Arkansas--recently enacted anti-CRT legislation in addition to Black history mandates. While legislators in these two states claim that their goal is to expand the teaching of African American history and equip all students to address issues of race and racism, the resulting US History state standards suggest otherwise. This offers a unique opportunity for critical discourse analysis to understand what is being taught in these two states and for what purposes. In the following case study, I will outline the methods and theoretical frameworks I used to answer the questions “What do the legally constructed narratives in Florida and Arkansas’s secondary US history curriculums say about Black people’s lived experiences from 1619 to the present? How does this curriculum intentionally inflict symbolic violence on students by perpetuating anti-Black and/or color-evasive racism?
Analysis of Antibody Affinity Maturation and Thermal Stability of Proteins Using Photophysical Approaches
Engineered antibodies (Abs) have found applications in disparate fields such as enzyme catalysis and medicinal therapeutics because of their exquisite binding affinity and specificity. To engineer antibodies for these purposes, a firm understanding of the underlying mechanisms that control Ab affinity and specificity is needed, so these properties can be tightly controlled and adapted to the application at hand. My research characterized the binding site of the anti-fluorescein Ab 4-4-20 using an array of biophysical techniques and gives insight into the binding site interactions that give rise to both affinity and specificity. We utilized Fab fragments of the anti-fluorescein Ab 4-4-20 as a model system to investigate the binding site of mature Abs. Site-directed mutagenesis was used to insert eight point-mutations into the Ab binding site at key amino acid residues that directly interacted with fluorescein in the binding pocket. The inherent fluorescence of the fluorescein antigen was utilized in a label-free fluorescence titration used to measure the dissociation constant Kd of fluorescein-Ab binding. Binding specificity was also assessed using the same technique but with a chlorine-substituted fluorescein analog. Binding was further assessed by fluorescence lifetime measurements of Ab-bound fluorescein using time-correlated single photon counting (TCSPC). Finally, the structural stability of the point mutants was determined using thermal melt curves with SYPRO® Orange (SyO) as a reporter dye. Additionally, we are interested in comparing the binding behavior of mature and germline Abs. We are optimizing the papain digestion of IgG Abs to FAB fragments which we will use with our inherent fluorescence titration technique to characterize the binding of germline and mature Ab pairs that also bind to fluorescein. SyO is a zwitterionic dye that is used for protein gel staining, thermal melt assays of proteins, and as a marker for misfolded proteins. However, while widely utilized, much of SyO’s photophysics remains unexplored. We studied the effect of pH on the photophysical properties of SyO in aqueous solution and found two well-defined transitions in the 0 to 10 pH range between three SyO species with distinct absorption and fluorescence properties. The aggregation behavior of the three pH-dependent species of SyO were characterized by their absorbance and fluorescence spectra. The aggregation behavior of SyO at neutral pH and aqueous solution was found to be essential to the utility of SyO as a reporter dye in protein assays. Furthermore, SyO-based assays need to be optimized for each individual protein system and this optimization process can be challenging because the mechanisms by which SyO fluorescence is increased through protein interactions are poorly understood. We characterized the steady-state and time-correlated fluorescence of SyO interacting with hen egg-white lysozyme (HEL), bovine alpha-lactalbumin (aLac), saccharomyces cerevisiae invertase (Inv), and bovine serum albumin (BSA) at a range of temperatures to understand the temperature-dependent mechanisms that lead to the enhancement of SyO fluorescence when it interacts with the model proteins
