Growth factor dependent regulation of p70S6 kinase: Evaluation of domain specific signals sensitive to rapamycin
Phosphorylation dependent regulation of S6K1 has been proposed in accordance with
the strategy of activation adapted by AGC family kinases. Accordingly three
phosphorylations at the turn motif, HM and AL are considered critical for S6K1
regulation , while the turn motif phosphorylation is thought to be constitutive ,the burden of the regulation is more or less entirely shared by the phosphorylations turn
over at HM and AL of the enzyme. Although both phosphorylations are equally
important without a question their individual roles in contributing to activation and
mediating rapamycin sensitivity remains controversial. Phosphorylations of the
enzyme at its HM brought about by rapamycin sensitive mTOR kinase is thought to
prime the enzyme for PDK1 dependent phosphorylations at AL, in accordance with
the prevalent dogma of sequential phosphorylations. Accordingly inactivation of
mTOR by rapamycin resulting in loss of HM phosphorylation and resultant loss of
phosphorylation at the AL is the model put forward to explain inhibition by
rapamycin. The attribution of mTOR as the kinase responsible to engender HM
phosphorylation, is however, based on scanty evidence carried forward simply on the
basis of the ability of mTOR to bind rapamycin. In fact S6K1 mutants supposedly
incapacitated for TOR kinase recruitment continue to be phosphorylated at the HM to
suggest that effects attributed to TOS motif deletion may simply be conformational
and not attributable to the loss of TOR kinase input at the HM. Further the
implication of kinases other than TOR in mediating HM phosphorylation compound
the ambiguity associated with TOR kinase involvement in mediating rapamycin
inhibition. This study therefore, attempts to understand as to how the dynamics of the
two phosphorylations in general and TOR mediated phosphorylation in particular
relates to rapamycin mediated inhibition of S6K1.
We present evidence that baculovirus mediated expression of S6K1 in insect cells
results in the failure of the enzyme to engender both HM and AL phosphorylations.
The enzyme surprisingly continues to be inhibited by rapamycin. Evidence indicating
redundant role of phosphorylation in general and TOR mediated phosphorylation in
particular for S6K1 inhibition by rapamycin challenge the prevailing dogma.We also
provide evidence about the interdependent turnover of the two phosphorylations such
as to exhibit all or none characteristics. The evidence that both AL and HM kinases
are completely insensitive to rapamycin, suggests that their co-ordinate loss to
rapamycin cannot be mechanistic and therefore has to be consequential. We finally
show that the loss of the two phosphorylations is also associated with inhibition of
S6K1 by agents like bupivacaine and lidocaine that target signalling pathways other
than TOR, to strongly suggest that the loss of HM and AL phosphorylations is a
general feature of S6K1 inhibition not necessarily governed by TOR inactivation

Ranked Set Sampling and Its Application in Quality Control
One of the keys to any Statistical inference is that the data involved be obtained via some formal mechanism that enables the experimenter to make valid judgments on the question(s) of interest. One of the most common mechanisms for obtaining such data is that of a simple random sample. Other more structured sampling designs, such as stratified sampling or probability sampling, are also available to help make sure that the obtained data collection provides a good representation of the population of interest. Any such additional structure of this type revolves around how the sample data themselves should be collected in order to provide an informative image of the larger population. With any of these approaches, once the sample items have been chosen the desired measurement(s) is collected from each of the selected items
Economics of Fisheries in Jammu and Kashmir: A Case Study of District Anantnag
Fishery is an important sector in most of the countries of the world from the
viewpoint of income and employment generation. The experience in these
countries indicates that the growth of the fishing sector stimulates the
development and employment in related industries which contribute significantly to
the total economic growth of these countries. Fisheries play an important role in
augmenting food supply and rising nutritional levels of the population. Besides being
an article of protein rich food, fishery also provides a number of by-products such as
fish manure, fish leather, fish glue and isinglass. The aquatic resources of the world
are varied, extensive and rich in potential. They can be broadly divided in to two
groups- marine and inland which in turn can be categorised as capture fisheries and
culture fisheries. With an extensive coastal line extending along the mainland, with a
fairly wide continental-shelf and slope high sea beyond, the world has rich marine
resources. The inshore waters forming the marginal sea together with the coastal
inlets constitute an environment of high productivity, offering great scope for culture
of a wide variety of marine organisms. Similarly the inland fishery resource
comprises of two types of waters namely the fresh water and the brackish, the former
including river systems, an extensive network of irrigation canals, reservoirs, lakes,
tanks, ponds etc. and the latter, the sprawling estuaries at the confluence of the river
system with the sea, a large number of lagoons, brackish water lakes, impoundments
and the vast areas of mangrove swamps containing tidal waters of fluctuating salinity
State and Islam in Turkmenistan (1991-2001)
Islam in Central Asia in general, and Turkmenistan in particular has to confront different stages in the process of its development. Earlier during the Soviets Islam as a religion was marginalized in so far its role and influence on common masses was concerned. After independence, it was expected, Islam with its whole system will re-emerge on the scenario of Turkmenistan. Niyazov, the president of the country initially proved all-liberal by providing administrative support regarding the teaching of Islamic principles at school, establishing Mosques at large scale in the region, however being a former communist leader, with the passage of time he pursued the footsteps of his predecessors.In order to have a meaningful discourse on the subject, interdisciplinary approach has been followed taking insight from history, geography, cultural anthropology, economics, political science and philosophy. The comparative methods were also employed for the understanding of different issues related to the subject. Since the study is based on theoretical and historical study as such all sources of information, books, official records, journals, periodicals, newspapers etc. were taken into account. Although enough of literature on the nature of State and Islam in Turkmenistan is not available, an attempt has been made to evaluate the available data with an objective analysis. Hardly any compact research work has been carried out on the history of Islam in Turkmenistan both in Soviet and in Post Soviet period, even the little studies related to Turkmenistan cover only disjointed and limited areas touching mainly general information. The present study therefore aims to fill up the gaps and present an objective analysis of the State and Islam in Turkmenistan (1991-2001)
