A 3D Co-culture model of endometrial carcinoma cells with peripheral blood immune cells (PBMCs) to study the activity of immune checkpoint inhibitors
The 3D co-culture model we describe was developed to study the potency of immune checkpoint inhibitors. In the assay, the first step is to provide 3D cell growth of endometrial carcinoma RL95-2 cells on a hydrogel, LifeGel®, and then to co-culture cancer cells with PBMCs isolated from healthy blood donors. After 5 days of the co-culture, in the presence of anti-PD-1/PD-L1 or anti-VISTA immune checkpoint inhibitors, the cancer cells’ viability is measured using a colorimetric assay and fluorescence readout.
The key features of the presented model are (1) a direct 3D co-culture model that involves cancer cells and immune cells represented by PBMCs, (2) the use of LifeGel® hydrogel, where cancer cells form spheroids on the surface, allowing immune cells and larger molecules, such as monoclonal antibodies, to penetrate the cancerous structures, and (3) the possible reactivation of immune cells by immune checkpoint inhibitors may be observed via killing/destroying effects on cancer structures
Ciało żydowskie w polskiej prozie modernistycznej w latach 1918-1939 wobec tradycji literatury XIX wieku
Artykuł analizuje sposób, w jaki polska proza modernistyczna lat 1918-1939 przejmuje i przetwarza repertuar konwencji oraz strategii reprezentowania żydowskiego ciała wypracowanych przed rokiem 1918. Autorzy modernistyczni odwołują się bowiem zarówno do wzorów przedstawiania ciała żydowskiego jako inkarnacji mitycznej Obcości, egzotycznej orientalności i rasowej semickości oraz rozmaitych ideologicznych wariantów tych obrazów, jak i do tradycyjnych schematów narracyjnych właściwych różnym obiegom, w tym do wzorców romansowych i sensacyjnych obiegu popularnego. Wyróżnione zostały dwie zasadnicze strategie. Pierwsza z nich, związana z modernistycznym relacjonizmem, opiera się na ujawnianiu problematyczności dotychczasowych reprezentacji. Jej głównym środkiem jest akcentowanie zmiennego charakteru ich konstrukcji zależnego od przyjętego punktu widzenia. Druga strategia - konstruktywistyczna - ma charakter prowokacyjnej interwencji i polega na potraktowaniu literackich konwencji, obrazów należących do społecznego imaginarium i ideologicznych dyskursów jako kulturowego zasobu, który podlega parodystycznemu użyciu. Efektem obu okazuje się z jednej strony budowanie dystansu wobec sposobu posługiwania się artystycznymi kliszami i społecznymi stereotypami, z drugiej zaś odsłanianie kulturowej genezy form rozumienia i odzwierciedlania rzeczywistości. W tym sensie oglądanie i czytanie żydowskiego ciała w literaturze międzywojennej wpisuje się również w podejmowaną przez modernizm rewizję XIX-wiecznego modelu świata.The article analyses how Polish modernist prose from 1918 to 1939 adopts and transforms the repertoire of conventions and strategies for representing the Jewish body developed before 1918. Modernist writers refer both to nineteenth century patterns of representing the Jewish body, understood as an incarnation of the mythical Other, defined as oriental or, if coded racially, as Semitic, and to nineteenth-century narrative schemes, mostly from romantic or sensational novels. Two basic strategies have been identified. The first attempted to expose the problematic nature of earlier representations by showing that their construction was changeable and dependent on the point of view of individual writers. The second saw these literary conventions and images as a cultural resource which needed to be deconstructed. These two strategies established a distance from how artistic clichés and social stereotypes were previously used and uncovered the cultural genesis of systems which sought to understand and reflect reality. In this sense, the new ways of observing and reading the Jewish body in the literature of the interwar period were an essential part of the modernist revision of the world-view of the nineteenth century

Integrated lab-in-syringe/lab-on-valve flow system for the synthesis of EDTA-capped silver nanoparticles and determination of creatinine
A fully integrated lab-in-syringe/lab-on-valve flow system was developed for the synthesis of EDTA-capped silver nanoparticles and their subsequent application in the spectrophotometric determination of creatinine. This work presents the first use of a lab-in-syringe system for nanoparticle synthesis and the first fully integrated flow system that unites both synthesis and analysis steps within a single system. The method is based on the aggregation of silver nanoparticles induced by creatinine in an alkaline medium, with detection performed by spectrophotometric measurement of the resulting colour change. The method was successfully applied to the determination of creatinine in human serum samples. Analytical validation of the method confirmed a limit of detection (LOD) of 0.04 mg/L and a limit of quantification (LOQ) of 0.14 mg/L. A linear range between 0.14 and 3.00 mg/L was obtained, with a relative error (RE) of 6.7 % and a coefficient of variation (CV, n=8) of 8.0 %, confirming satisfactory accuracy
