Molecular and functional characterization of ANKRD1, a candidate gene for total anomalous pulmonary venous return.
Total Anomalous Pulmonary Venous Return (TAPVR, OMIM %106700) is a rare congenital heart defect in which the pulmonary veins fail to enter the left atrium and drain instead into the right atrium or one of its venous tributaries. Previously, a TAPVR patient bearing a t(10;21) de novo balanced translocation was found in our laboratory and the ANKRD1/CARP gene, mapping on chromosome 10 about 130 Kb proximally to the breakpoint, was defined as a good TAPVR candidate gene. Significantly, we found highly increased ANKRD1 expression levels in lymphoblastoid cell lines derived from both the translocation-bearing proband and a second independent sporadic TAPVR patient, suggesting that disruption of the normal ANKRD1 expression pattern is associated with TAPVR. Moreover, an ANKRD1 non-conservative missense mutation was found in a third sporadic TAPVR patient. Such mutation was subsequently found to confer an increased stability to the ANKRD1 protein in vitro and to affect its function as a repressor of cardiac-specific gene expression. The aim of this doctoral project was to characterize at the molecular and functional level the ANKRD1 gene, in order to provide new evidences for its involvement in TAPVR pathogenesis. First, ANKRD1 function as a negative regulator of cardiac genes was further investigated, and its role as a repressor of Nkx2.5 and Pitx2cmediated activation of the cardiac-specific ANF promoter was demonstrated. Moreover, in situ hybridization assays revealed that the expression pattern of the murine Ankrd1 gene strongly overlaps with that of both Nkx2.5 and Pitx2 genes, supporting both the occurrence of a functional interaction between these genes and a role for ANKRD1 in TAPVR pathogenesis. Co-immunoprecipitation assays added a further support for this hypothesis, by showing a physical interaction between the ANKRD1 and Nkx2.5 proteins. The intracellular pathways involved in ANKRD1 turnover were also investigated. ANKRD1 was shown to behave as a short-lived protein in vivo, with the PEST motif playing a crucial role in its turnover rate. Moreover, intracellular degradation of ANKRD1 was found to be carried out mainly by the ubiquitin proteasome system, through several independent degron elements. These data defined for the first time both the intracellular pathway responsible for ANKRD1 degradation and the functional relevance of the PEST motif in this process. Finally, ANKRD1-overexpressing transgenic mice were generated in the attempt to develop an animal model for TAPVR. Several independent transgenic lines are currently under investigation in order to better define the role of ANKRD1 in heart development

Le spettrometrie di massa hanno, negli ultimi anni, aumentato la loro diffusione passando da tecniche avanzate di ricerca a tecniche di “largo consumo” grazie all’enorme avanzamento tecnologico e ai nuovi materiali utilizzati che ne hanno incrementato l’affidabilità e l’applicabilità, accanto ad una migliorata facilità d’uso. Tra gli analizzatori di massa disponibili commercialmente, l’analizzatore quadrupolare offre senza alcun dubbio una grande flessibilità di utilizzo accoppiata a un costo macchina e a dei costi di mantenimento contenuti.
Nel corso dei tre anni di studio del dottorato di ricerca sono stati sviluppati temi di ricerca riguardanti le metodologie analitiche in spettrometria di massa focalizzandosi principalmente sulla risoluzione di problematiche tecnico-strumentali. Le tematiche affrontate sono volutamente molto differenti e hanno riguardato non solo l’aspetto strettamente tecnico-analitico, ma hanno spaziato verso lo sviluppo di sequenze analitiche complete. Tali sequenze comprendono anche l’attenzione verso il campione, verso la definizione dei protocolli di analisi robusti e verso il
trattamento statistico dei dati. Questo processo è stato sviluppato per ottimizzare tutti i parametri legati all’analisi in grado di migliorare l’affidabilità del dato finale e di ottenere la massima informazione in esso celato.
Il primo filone di ricerca del quale mi sono occupato ha riguardato lo sviluppo e l’utilizzo di una metodica completa per la caratterizzazione geochimica di mineralizzazioni cuprifere per lo studio della provenienza di reperti archeologici dell’età del bronzo. La metodologia comprende il trattamento del campione in solido, la digestione acida dello stesso, l’analisi quantitativa delle componenti in traccia, la misura delle abbondanze isotopiche e dei loro rapporti per rame e piombo e il trattamento statistico multivariato dei dati attraverso. Tutte la metodologia ha utilizzato come strumento unico di analisi un ICP-QMS. L’analizzatore quadrupolare si dimostra estremamente valido per l’analisi quali-quantitativa di un gran numero elementi e su un gran numero di campioni in maniera veloce e accurata, ma non trova applicazioni di qualità nel campo dell’analisi isotopica che trova negli strumenti MC-ICP-SFMS la scelta ideale. Le ragioni di questa limitazione sono da ricercare innanzitutto nei forti limiti strumentali, quali l’instabilità del segnale, il mass bias, l’effetto del dead time del rivelatore e, sopra tutte, la non contemporanea rivelazione degli isotopi. Gli ultimi avanzamenti tecnologici hanno tuttavia smussato molto le limitazioni strumentali rendendo più competitivo questo tipo di analizzatore. Ci si è quindi focalizzati nello studio di tutti i parametri che influenzano la precisione e l’accuratezza dela misura ottenibile con un ICP-QMS per giungere ad un miglioramento generale delle prestazioni strumentali. È stata sviluppata una metodologia di bracketing modificata che migliora la produttività delle metodologie già esistenti andando a migliorare anche la qualità analitica del dato. In particolare con il protocollo sviluppato è stato possibile raggiungere un RSD pari a 0.018‰ sui rapporti isotopici 63Cu/65Cu che sono circa tre volte migliori di quelli riportati in letteratura.
Il secondo filone di ricerca ha riguardato la realizzazione di due reattori a scansione per condurre test catalitici su sistemi planari nano strutturati. La grande disponibilità di metodologie di sintesi controllata di sistemi collettivi nano strutturati ha fatto nascere negli ultimi anni l’esigenza di sviluppare sistemi di caratterizzazione per l’attività catalitica degli stessi. I nuovi reattori devono essere adatti a campioni planari in cui l’area superficiale è molto ridotta così come i tempi di contatto. La realizzazione dell’accoppiamento originale tra il reattore e un sistema di analisi quadrupolare commerciale si è sviluppato in due step attraverso due prototipi.
Il primo prototipo permette di fatto una scansione passo-passo ed è stato utilizzato per lo studio del dimensionamento della camera di reazione, dei flussi gassosi da utilizzare e per individuare le criticità strumentali che l’interfaccia reattore-analizzatore ha generato. Sulla base dei dati raccolti, è stato successivamente disegnato e realizzato il vero prototipo a scansione lineare continua, in grado di testare le proprietà catalitiche di una superficie planare di 28 mm2 di area minima. Il micro reattore è stato realizzato presso la nostra officina meccanica e verrà assemblato e testato nei mesi di dicembre-gennaio. Accanto allo sviluppo strumentale sono state affrontate le problematiche legate alla metodologia di testing delle prestazioni strumentali attraverso la sintesi mirata di campioni ad hoc. I catalizzatori planari utilizzati in questa fase sono stati prodotti presso i laboratori del centro CIMaINa di milano dal gruppo del prof. Paolo Milani. Lo studio di nuovi catalizzatori planari è stata anche affrontata durante un periodo di ricerca all’estero presso la “CHALMERS Tekniska Högskola” di Göteborg (Sverige) sotto la supervisione del Prof. Bengt Kasemo. Presso il CHALMERS ho potuto approfondire varie tecniche di sintesi e caratterizzazione per materiali nano strutturati e ho sintetizzato alcuni campioni planari caratterizzati da una struttura a “nanodiscs array” tramite metodologie litografiche colloidali. Tali catalizzatori sono stati sintetizzati per testare le prestazioni del secondo prototipo di reattore sviluppato.
Presso il CHALMERS ho anche seguito il design e l’assemblaggio di un micro-reattore accoppiatto a un analizzatore di massa quadrupolare per testare l’attività catalitica foto-stimolata di titania modificata. Il reattore è stato assemblato passo-passo andando a risolvere i problemi strumentali di interfaccia del sistema a ultra alto vuoto e il reattore a pressione atmosferica. Il reattore è stato testato con successo. Accanto al lavoro tecnico-pratico è stato sviluppato anche il protocollo di analisi che comprende la preparazione del campione, la sequenza analitica e il trattamento dei dati.

Infrared cameras for small telescopes: from requirements to device.
This thesis work concerns the study, the procurement and the commissioning of small infrared cameras for imaging with small sized ground-based telescopes.
Observations at infrared wavelengths are of great importance in astronomy. One of the most known advantages concerns the observation of the distant universe. Due to red-shift, distant objects have their spectral features moved toward higher wavelengths. If one wants to compare the restspectrum of a nearby galaxy in the V band (500-600 nanometers) with a galaxy at z=3, he will have to observe the latter between 2000 and 2400 nanometers, i.e. in the infrared region. Furthermore if the Lyman break (912 Ångstroms) is redshifted to the upper limit of the visible region (z~7-8), the object is detectable only in the infrared.
Further advantage of the infrared band with respect to the visible is the reduced absorption of the radiation by the interstellar medium. Indeed the absorption in the classical V band is about one order of magnitude greater than the absorption in the K band. Hence any observation of sources in dusty areas such as galactic centers, clouds, etc. benefits of being carried out in infrared instead of isible.
There is another class of objects the study of which that benefits from infrared observations. Every astronomical source for which the thermal blackbody emission is relevant, if cold, will be bright in the infrared band. These objects are typically brown dwarfs, planets, protostars, low mass stars.
During the past few decades, infrared astronomy has become a major field in astronomy due to the rapid advances in infrared detector technology. One of the most important breakthroughs coincided with the introduction of new detectors imported from other applications (mainly military), like the Rockwell Hawaii (1024x1024 pixels, pitch 18.5 μm) with a pixel size comparable to that of existing visible detectors. The Hawaii allowed, with the known differences in detector handling and reading, to collect images and spectra of the same kind and quality of images and spectra in the Visible wavelength domain.
The cost of these detectors and their extreme cryogenic requirements (77 K operating temperature instead of 150-200 K of a typical CCD) limited their application to large instruments for large (VLT) or dedicated (UKIRT) telescopes. Progresses in cryo-vacuum technologies, such as Stirling cryo-coolers and cooling-flow cooling systems, as well as progresses in detector technology leading to a higher reliability and a reduction of cost, allowed to think about Infrared imaging systems for small telescopes. This timely coincided with the new scientific requirement for very fast pointing IR imaging devices for the follow-up of fast transient sources such as Gamma Ray Bursts (GRBs) afterglows.
Fast pointing robotic telescopes for transient detections have decades of tradition in astronomy. However the first successful attempt to extend their coverage to the IR band is REM (Rapid Eye Mount) installed in 2003 at La Silla Observatory and currently in operation. REM was conceived as the ground-based complement of the satellite SWIFT, the optical monitor of which (UVOT) is limited at 650 nm toward the red, hence missing most of the potential high redshift GRBs detected by the Gamma Ray detector BAT.
Well beyond its original purposes REM is today serving the Italian Community as a long-term monitoring facility for variable stars, AGNs, Supernovae and many other types of programs that need IR observations. Moreover it invalidated the equation "IR equal to large telescope" and opened to a number of projects such as BOOTES-IR and AMICA-IRAIT that are described in details in this thesis.
The new frontier of infrared astronomy is in Antarctica. This continent offers features and advantages unique on the Earth. Bases like DOME-C show promising atmospheric transparency, wider and newer transmission bands, lower background emission, reduced water vapour atmospheric content, large isoplanatic angle, promising seeing and the possibility to observe for very long periods like the entire winter night of six months. AMICA-IRAIT is an Italian project to install a pathfinder telescope and infrared camera at DOME-C.
Nowadays thinking of IR imaging at small robotic telescopes is no longer a taboo. Pathfinders had their way and the pioneering technology is consolidating. I had the chance to be part of this consolidation process through the work done in three projects: INCA (INfrared CAmera), BIRCAM (Bootes InfraRed CAMera) and AMICA (Antarctic Multi-band Infrared Camera).
INCA is the study and the development of a fully functional prototype of an infrared camera implementing innovative materials and solutions.
BIRCAM is an astronomical infrared camera followed up end-to-end by gOlem group (INAF Osservatorio Astronomico di Brera-Merate, Italy), installed on the Spanish BOOTES-IR telescope (Observatorio de Sierra Nevada, Granada, Spain) and is designed for the Gamma Ray Burst afterglow observations.

Processi intramolecolari catalizzati da palladio per la sintesi di sistemi eterociclici.
Les réactions catalysées par le palladium occupent un rôle déterminant en chimie organique, puisqu’ils permettent de promouvoir la formation de liaisons carbone-carbone et carbone-hétéroatome lors de la préparation de composés naturels, de polymères et de produits d'intérêt pharmaceutique. En effet, le palladium présente une faible toxicité, ses composés sont relativement peu sensibles à l’oxygène et à l’humidité et tolèrent en outre beaucoup de groupes fonctionnels. Les méthodes synthétiques faisant intervenir le palladium procèdent souvent en conditions douces et fournissent des rendements élevés, avec d’excellentes chimio-, régio- et stéréosélectivité.
Dans cette thèse de doctorat, divers objectifs ont été atteints grâce à des réactions catalysées au palladium, qui a permis de construire des liaisons C-C et C-N de façon intramoléculaire. Une série de systèmes hétérocycliques azotés a ainsi été obtenue, ces derniers possédant une activité biologique et pharmacologique potentielle.
Quelques aspects des résultats obtenus doivent être soulignés.
Le premier concerne la réaction de Heck. Le mécanisme de cette réaction est connu depuis longtemps, même si des exemples d’isolation de complexe -alkylpalladium ont été rarement reportés dans la littérature. Le travail effectué dans la première partie de la thèse, pour obtenir de tétrahydro-spiroisoquinoléines, a permis pour la première fois d’isoler et de caractériser systématiquement une gamme assez large de complexes -alkylpalladium, intermédiaires de la réaction de Heck Ceux-ci se sont ensuite révélés des catalyseurs efficaces dans des réactions pallado-catalysées, telles que les réactions de Heck, de Suzuki et de Stille.
Une synthèse alternative de l'acide kaïnique a ensuite été effectuée. Il fait parti de la famille des kaïnoïdes, une importante classe d'aminoacides naturels nonprotéinogéniques.
L’acide kaïnique présente un noyau pyrrolidine, deux fonctions acide carboxylique, un fragment isopropylidène et 3 centres stéréogènes consécutifs en positions 2, 3 et 4 du cycle. Le palladium permet la formation d’une liaison C-C lors de l’étape clé de formation du système pyrrolidinone Par la suite, une méthode pallado-catalysée permettant d’accéder à des structures pyperazinoniques a été mise au point, simplifiant considérablement les méthodes de synthèse existantes dans la littérature.
Enfin, la faisabilité de réactions de couplage oxydant d'alcènes avec des hétérocycles électro-enrichis a été étudiée. Après optimisation des conditions réactionnelles, ces dernières ont été appliquées à une vaste gamme de composés. Le comportement des substrats indoliques s’est révélé particulièrement intéressant, ces derniers montrant un réarrangement de Plancher totalement sélectif
