An antibacterial peptide derived from urechis unicinctus, mutant and application thereof
本发明公开了一种天然抗菌肽及其在抗菌方面的应用。本发明的抗菌肽来源于单环刺螠。通过检索单环刺螠基因数据库,筛选分析获得抗菌肽成熟肽序列,其氨基酸序列如序列表中SEQ ID NO : 1所示。通过对比分析,该天然抗菌肽与目前已知所有抗菌肽的氨基酸序列存在明显差异,属于一种新型抗菌肽。抗菌实验结果表明,本发明的抗菌肽对革兰氏阳性细菌、革兰氏阴性细菌和真菌具有极强的抗菌活性,抗菌作用广谱高效,可用于制备抗菌、抑制细菌生长的药物、防腐剂、兽药、动物饲料以及化妆品

Discovery of a Promising CBP/p300 Degrader XYD129 for the Treatment of Acute Myeloid Leukemia
The epigenetic target CREB (cyclic-AMP responsive element binding protein) binding protein (CBP) and its homologue p300 were promising therapeutic targets for the treatment of acute myeloid leukemia (AML). Herein, we report the design, synthesis, and evaluation of a class of CBP/p300 PROTAC degraders based on our previously reported highly potent and selective CBP/p300 inhibitor 5. Among the compounds synthesized, 11c (XYD129) demonstrated high potency and formed a ternary complex between CBP/p300 and CRBN (AlphaScreen). The compound effectively degraded CBP/p300 proteins and exhibited greater inhibition of growth in acute leukemia cell lines compared to its parent compound 5. Furthermore, 11c demonstrated significant inhibition of tumor growth in a MOLM-16 xenograft model (TGI = 60%) at tolerated dose schedules. Our findings suggest that 11c is a promising lead compound for the treatment of AML
Synthesis of 12-Connected Three-Dimensional Covalent Organic Framework with lnj Topology
The structural exploration of three-dimensional covalent organic frameworks (3D COFs) is of great significance to the development of COF materials. Different from structurally diverse MOFs, which have a variety of connectivity (3-24), now the valency of 3D COFs is limited to only 4, 6, and 8. Therefore, the exploration of organic building blocks with higher connectivity is a necessary path to broaden the scope of 3D COF structures. Herein, for the first time, we have designed and synthesized a 12-connected triptycene-based precursor (triptycene-12-CHO) with 12 symmetrical distributions of aldehyde groups, which is also the highest valency reported until now. Based on this unique 12-connected structure, we have successfully prepared a novel 3D COF with lnj topology (termed 3D-lnj-COF). The as-synthesized 3D COF exhibits honeycomb main pores and permanent porosity with a Brunauer-Emmett-Teller surface area of 1159.6 m(2) g(-1). This work not only provides a strategy for synthesizing precursors with a high connectivity but also provides inspiration for enriching the variety of 3D COFs
Improvement of the Antioxidant and Antitumor Activities of Benzimidazole-Chitosan Quaternary Ammonium Salt on Drug Delivery Nanogels
The present study focused on the design and preparation of acid-responsive benzimidazole-chitosan quaternary ammonium salt (BIMIXHAC) nanogels for a controlled, slow-release of Doxorubicin HCl (DOX.HCl). The BIMIXHAC was crosslinked with sodium tripolyphosphate (TPP) using the ion crosslinking method. The method resulted in nanogels with low polydispersity index, small particle size, and positive zeta potential values, indicating the good stability of the nanogels. Compared to hydroxypropyl trimethyl ammonium chloride chitosan-Doxorubicin HCl-sodium tripolyphosphate (HACC-D-TPP) nanogel, the benzimidazole-chitosan quaternary ammonium salt-Doxorubicin HCl-sodium tripolyphosphate (BIMIXHAC-D-TPP) nanogel show higher drug encapsulation efficiency and loading capacity (BIMIXHAC-D-TPP 93.17 +/- 0.27% and 31.17 +/- 0.09%), with acid-responsive release profiles and accelerated release in vitro. The hydroxypropyl trimethyl ammonium chloride chitosan-sodium tripolyphosphate (HACC-TPP), and benzimidazole-chitosan quaternary ammonium salt-sodium tripolyphosphate (BIMIXHAC-TPP) nanogels demonstrated favorable antioxidant capability. The assay of cell viability, measured by the MTT assay, revealed that nanogels led to a significant reduction in the cell viability of two cancer cells: the human lung adenocarcinoma epithelial cell line (A549) and the human breast cancer cell line (MCF-7). Furthermore, the BIMIXHAC-D-TPP nanogel was 2.96 times less toxic than DOX.HCl to the mouse fibroblast cell line (L929). It was indicated that the BIMIXHAC-based nanogel with enhanced antioxidant and antitumor activities and acidic-responsive release could serve as a potential nanocarrier
Coastline Monitoring and Prediction Based on Long-Term Remote Sensing Data-A Case Study of the Eastern Coast of Laizhou Bay, China
Monitoring shoreline movements is essential for understanding the impact of anthropogenic activities and climate change on the coastal zone dynamics. The use of remote sensing allows for large-scale spatial and temporal studies to better comprehend current trends. This study used Landsat 5 (TM), Landsat 8 (OLI), and Sentinel-2 (MSI) remote sensing images, together with the Otsu algorithm, marching squares algorithm, and tidal correction algorithm, to extract and correct the coastline positions of the east coast of Laizhou Bay in China from 1984 to 2022. The results indicate that 89.63% of the extracted shoreline segments have an error less than 30 m compared to the manually drawn coastline. The total length of the coastline increased from 166.90 km to 364.20 km, throughout the observation period, with a length change intensity (LCI) of 3.11% due to the development of coastal protection and engineering structures for human activities. The anthropization led to a decrease in the natural coastline from 83.33% to 13.89% and a continuous increase in the diversity and human use of the coastline. In particular, the index of coastline diversity (ICTD) and the index of coastline utilization degree (ICUD) increased from 0.39 to 0.79, and from 153.30 to 390.37, respectively. Over 70% of the sandy beaches experienced erosional processes. The shoreline erosion calculated using the end point rate (EPR) and the linear regression rate (LRR) is 79.54% and 85.58%, respectively. The fractal dimension of the coastline shows an increasing trend and is positively correlated with human activities. Coastline changes are primarily attributed to interventions such as land reclamation, aquaculture development, and port construction resulting in the creation of 10,000.20 hectares of new coastal areas. Finally, the use of Kalman filtering for the first time made it possible to predict that approximately 84.58% of the sandy coastline will be eroded to varying degrees by 2032. The research results can provide valuable reference for the scientific planning and rational utilization of resources on the eastern coast of Laizhou Bay
Activity Pattern and Habitat Use of Shorebirds in an Artificial Wetland Complex: A Case Study of Breeding Pied Avocet in the Yellow River Delta, China
With the loss of substantial natural wetlands in coastal zones, artificial wetlands provide alternative habitats for many shorebirds. Scientific management of artificial wetlands used by shorebirds plays an important role in maintaining the stability of shorebird population. Satellite tracking technique can obtain high-precision location information of individuals day and night, providing a good technical support for the study of quantitative relationship between waterfowls and their habitats. In this study, satellite tracking method, Remote Sensing (RS) and Geographic Information System (GIS) technology were used to analyze the activity pattern and habitat utilization characteristics of Pied Avocet during breeding period in an artificial wetland complex in the Yellow River Delta (YRD), China. The results showed that the breeding Pied Avocets had a small range of activity, with a total core and main home range of 33.10 km(2) and 216.30 km(2), respectively. This species tended to forage in the pond and salt pan during the day and night, respectively, with an unfixed staying time in the breeding ground. The distance between breeding ground and feeding ground was less than 6 km. It is emphasized that in addition to improving the conditions of the remaining natural habitats, effective managing artificial habitats is a priority for shorebird conservation. This research could provide reference for the management of artificial wetlands in coastal zones and supply technique support for the protection of shorebirds and their habitats, and alleviate human-bird conflicts and sustainable development of coastal zones
Lateral flow chromatography strip system for rapid fluorescence determination of phycocyanin in water samples
Phycocyanin (PC) is of great significance to biomedicine and water environmental safety. Hence, it is indispensable to develop facile and rapid method for PC determination. In this investigation, a system containing lateral flow chromatography (LFC) strip (which was deposited with molecularly imprinted polymer (MIP) capped CdTe quantum dots (QDs) based mesoporous structured coated silica nanoparticles, SiO2@QDs@ms-MIP NPs) and miniaturized fluorimeter was first fabricated. In detail, a two-step strategy was utilized for preparation of SiO2@QDs@ms-MIP NPs, which consisted of modification of CdTe QDs onto the silica NPs first, and synthesis of mesoporous imprinting shell by using PC as template molecule and cetyltrimethylammonium bromide (CTAB) as surfactant. After that, novel fluorescence NPs possessing specific recognition and sensitivity toward PC in seawater and lake water were acquired. The resulting fluorescent sensing system exhibited outstanding performances, which included excellent sensitivity (4.5 nmol/L), satisfactory specificity (imprinting factor, 2.31), appropriate linearity range (0.01-5 mu mol/L), good recovery (96.0-101.7 %), excellent stability (relative standard deviation, RSD<1.1 %), wonderful reproducibility (RSD<1.1 %), and excellent anti-interference ability. The results of the fluorescent sensing system were superior to those of the commonly used ultraviolet (UV) method. The proposed strategy showed great potential for fast (<10 min) and convenient fluorescence detection of PC in real samples
Recent advances and challenges in monitoring chromium ions using fluorescent probes
Chromium is among the most strategic and critical transition metal elements and has extensive applications in both industrial and biological contexts. The most stable oxidation states of chromium are trivalent chromium (Cr (III)) and hexavalent chromium (Cr(VI)). Cr(III) is recognized as an effective trace nutrient, whereas Cr(VI) in its highest oxidation state poses a toxic threat to human health due to its potent oxidizing capacities. To mitigate the risk of poisoning, efficient detection methods have been developed to meet testing requirements. Comparing with traditional methods, colorimetric and fluorescent techniques have emerged as simple, time-efficient, highly selective and sensitive, cost-effective and particularly well-suited for biological applications. Furthermore, these methods excel in distinguishing between trivalent and hexavalent chromium, even in low-level concentrations and complex matrices. Since the scarcity of fluorescent probes for chromium, most existing reviews merely touch upon this topic without providing comprehensive coverage. Therefore, this review aims to consolidate information on small molecular fluorescent probes and fluorescent materials serve as fluorescent probes for monitoring Cr2+, Cr3+, Cr2O72- and CrO42- in environmental settings and living cells. We anticipate that this review will promote the development of novel fluorescent probes for chromium detection, facilitating their applications in chemical, biological and medical domains
