    Genetic diversity and population structure of Capitulum mitella (Cirripedia: Pedunculata) in China inferred from mitochondrial DNA sequences

    Capitulum mitella is an economically important species in China; however, information on the population structure of this species is limited. In this study, the phylogeographical pattern, population genetic profile, and demographic history of C. mitella were investigated using mitochondrial DNA (mtDNA) cytochrome c oxidase subunit I (COI). A 599-bp segment of this subunit was sequenced from 198 individuals sampled in eight localities along the Chinese coast. High haplotype diversity (0.656-0.788) and low nucleotide diversity (0.0016-0.00244) were observed. Analyses of molecular variance (AMOVA) and F-ST statistics suggested the absence of a significant population structure. Neutrality tests and mismatch distributions both indicated a recent population expansion. The current phylogeographic structure of C. mitella may be attributed to past population expansion and long-distance pelagic larval dispersal facilitated by present-day ocean currents.     Identification of the Bcl-2 family protein gene BOK from orange-spotted grouper (Epinephelus coioides) involved in SGIV infection

    Apoptosis plays vital roles in many physiological process and immune response. BOK is one of the central regulators in apoptosis. In this study, a new BOX homolog (Ec-BOK) was cloned and characterized from Orange-spotted grouper, Epinephelus coioides. Ec-BOK encoded a 210 amino acid peptides which shared 97% identity to Stegastes partitus BOK protein, contained four BH domains and one transmembrane region. Ec-BOK widely expressed in all analyzed tissues with the higher expressions in kidney and spleen. Its expression level was up-regulated after SGIV infection in vitro. Further analysis revealed that over expression of Ec-BOK inhibited viral genes transcriptions and virus replication in fish cell. Our findings suggested that Ec-BOK might play a role in the immune response against virus. (C) 2016 Elsevier Ltd. All rights reserved

    Prolonged post-rift magmatism on highly extended crust of divergent continental margins (Baiyun Sag, South China Sea)

    Three-dimensional (3D) seismic, borehole and geochemical data reveal a prolonged phase of post-rift magmatism on highly extended crust of the Baiyun Sag, South China Sea. Two volcanic complexes are identified and described in the context of continental rifting and diachronous continental breakup of the South China Sea. Biostratigraphic data from exploration wells BY7-1 and BY2, complemented by K-Ar datings from core samples, confirm that magmatic activity in the Baiyun Sag occurred in two main stages: (1) a first episode at the base of the Miocene (23.8 Ma); and (2) a second episode occurring at the end of the Early Miocene (17.6 Ma). The relative location of volcanic complexes in the Baiyun Sag, and their stratigraphic position, reveals prolonged magmatism inboard of the ocean-continent transition zone during continental breakup. We suggest that magmatism in the Baiyun Sag reflects progressive continental breakup in the South China Sea, with the last volcanic episode marking the end of a breakup sequence representing the early post-rift tectonic events associated with the continental breakup process. Seismic and borehole data from this breakup sequence records diachronous magma emplacement and complex changes in depositional environments during continental breakup. (C) 2016 The Authors.     Crustal structure across the post-spreading magmatic ridge of the East Sub-basin in the South China Sea: Tectonic significance

    The 140-km wide last phase of opening of the South China Sea (SCS) corresponds to a N145 degrees direction of spreading with rift features identified on swath bathymetric data trending N055 degrees (Sibuet et al., 2016). These N055 degrees seafloor spreading features of the East Sub-basin are cut across by a post-spreading volcanic ridge oriented approximately E-W in its western part (Zhenbei-Huangyan seamounts chain). The knowledge of the deep crustal structure beneath this volcanic ridge is essential to elucidate not only the formation and tectonic evolution of the SCS, but also the mechanism of emplacement of the post-spreading magmatism. We use air-gun shots recorded by ocean bottom seismometers to image the deep crustal structure along the N-S oriented G8G0 seismic profile, which is perpendicular to the Zhenbei-Huangyan seamounts chain but located in between the Zhenbei and Huangyan seamounts, where topographic changes are minimum. The velocity structure presents obvious lateral variations. The crust north and south of the Zhenbei-Huangyan seamounts chain is ca. 4-6 km in thickness and velocities are largely comparable with those of normal oceanic crust of Atlantic type. To the south, the Jixiang seamount with a 7.2-km thick crust, seems to be a tiny post-spreading volcanic seamount intruded along the former extinct spreading ridge axis. In the central part, a 1.5-km thick low velocity zone (3.3-3.7 km/s) in the uppermost crust is explained by the presence of extrusive rocks intercalated with thin sedimentary layers as those drilled at IODP Site U1431. Both the Jixiang seamount and the Zhenbei-Huangyan seamounts chain started to form by the intrusion of decompressive melt resulting from the N-S post-spreading phase of extension and intruded through the already formed oceanic crust.     Association between Arctic autumn sea ice concentration and early winter precipitation in China

    Associations between autumn Arctic sea ice concentration (SIC) and early winter precipitation in China are studied using singular value decomposition analysis. The results show that a reduced SIC almost everywhere in the Arctic Ocean, except the northern Greenland Sea and Canadian Basin, are accompanied by dry conditions over central China, extending northeast from the Tibetan Plateau toward the Japan Sea, the Bohai Sea and the Yellow Sea, and wet conditions over South China and North China. Atmospheric circulation anomalies associated with SIC variability show two wave-train structures, which are persistent from autumn to winter, leading to the identified relationship between autumn Arctic SIC and early winter precipitation in China. Given that the decline in autumn SIC in the Arctic Ocean is expected to continue as the climate warms, this relationship provides a possible long-term outlook for early winter precipitation in China

    A tumour necrosis factor receptor-like protein encoded by Singapore grouper iridovirus modulates cell proliferation, apoptosis and viral replication

    It has been demonstrated that tumour necrosis factor receptor (TNFR) homologues encoded by viruses are usually involved in virus immune evasion by regulating the host immune response or mediating apoptotic cell death. Here, a novel TNFR-like protein encoded by Singapore grouper iridovirus (SGIV VP51) was cloned and characterized. Amino acid analysis showed that VP51 contained three cysteine-rich domains (CRDs) and a transmembrane domain at its C terminus. The expression of VP51 in vitro enhanced cell proliferation, and affected cell cycle progression via altering the G1/S transition. Furthermore, VP51 overexpression improved cell viability during SGIV infection via inhibiting virus-induced apoptosis, evidenced by the reduction of apoptotic bodies and the decrease of caspase-3 activation. In addition, overexpression of VP51 increased viral titre and the expression of viral structural protein gene MCP and cell proliferation promoting gene ICP-18. In contrast, the expression of the viral apoptosis inducing gene, LITAF, was significantly decreased. Although all three CRDs were essential for the action of VP51, CRD2 and CRD3 exerted more crucial roles on virus-induced apoptosis, viral gene transcription and virus production, while CRD1 was more crucial for cell proliferation. Together, SGIV TNFR-like products not only affected cell cycle progression and enhanced cell growth by increasing the expression of the virus encoded cell proliferation gene, but also inhibited virus-induced apoptotic cell death by decreasing the expression of the viral apoptosis inducing gene. Our results provided new insights into understanding the underlying mechanism by which iridovirus regulated the apoptotic pathway to complete its life cycle

    Comparison of Bioavailability and Biotransformation of Inorganic and Organic Arsenic to Two Marine Fish

    Dietary uptake could be the primary route of arsenic (As) bioaccumulation in marine fish, but the bioavailability of inorganic and organic As remains elusive. In this study, we investigated the trophic transfer and bioavailability of As in herbivorous rabbitfish Siganus fuscescens and carnivorous seabass Lateolabrax japonicus. Rabbitfish were fed with one artificial diet or three macroalgae, whereas seabass were fed with one artificial diet, one polychaete, or two bivalves for 28 days. The six spiked fresh prey diets contained different proportions of inorganic As [As(III) and As(V)] and organic As compounds [methylarsenate (MMA), dimethylarsenate (DMA), and arsenobetaine (AsB)], and the spiked artificial diet mainly contained As(III) or As(V). We demonstrated that the trophic transfer factors (TTF) of As in both fish were negatively correlated with the concentrations of inorganic As in the diets, while there was no relationship between TTF and the AsB concentrations in the diets. Positive correlation was observed between the accumulated As concentrations and the AsB concentrations in both fish, suggesting that organic As compounds (AsB) were more trophically available than inorganic As. Furthermore, the biotransformation ability of seabass was higher than that in rabbitfish, which resulted in higher As accumulation in seabass than in rabbitfish. Our study demonstrated that different prey with different inorganic/organic As proportions resulted in diverse bioaccumulation of total As in different marine fish

    PfIRR Interacts with HrIGF-I and Activates the MAP-kinase and PI3-kinase Signaling Pathways to Regulate Glycogen Metabolism in Pinctada fucata

    The insulin-induced mitogen-activated protein kinase (MAPK) and phosphatidylinositol 3-kinase (PI3K) pathways are major intracellular signaling modules and conserved among eukaryotes that are known to regulate diverse cellular processes. However, they have not been investigated in the mollusk species Pinctada fucata. Here, we demonstrate that insulin-related peptide receptor of P. fucata (pfIRR) interacts with human recombinant insulin-like growth factor I (hrIGF-I), and stimulates the MAPK and PI3K signaling pathways in P. fucata oocytes. We also show that inhibition of pfIRR by the inhibitor PQ401 significantly attenuates the basal and hrIGF-I-induced phosphorylation of MAPK and PI3K/Akt at amino acid residues threonine 308 and serine 473. Furthermore, our experiments show that there is cross-talk between the MAPK and PI3K/Akt pathways, in which MAPK kinase positively regulates the PI3K pathway, and PI3K positively regulates the MAPK cascade. Intramuscular injection of hrIGF-I stimulates the PI3K and MAPK pathways to increase the expression of pfirr, protein phosphatase 1, glucokinase, and the phosphorylation of glycogen synthase, decreases the mRNA expression of glycogen synthase kinase-3 beta, decreases glucose levels in hemocytes, and increases glycogen levels in digestive glands. These results suggest that the MAPK and PI3K pathways in P. fucata transmit the hrIGF-I signal to regulate glycogen metabolism

    Composition and Distribution of Planktonic Ciliates in the Southern South China Sea During Late Summer: Comparison Between Surface and 75 m Deep Layer

    Ciliates are very important components in most marine ecosystem. They are trophic link between the microbial food web and grazing food chain. In this study, ciliates were collected from 11 sites in the southern South China Sea (SCS) during August 25 to September 28, 2011. Their composition and distribution at the surface and 75 m deep depth of the ocean were studied. A total of 30 species belonging to 22 genera were identified, and 22 species of 15 genera were Tintinnids. Eutintinnus fraknoii and E. stramentus were the most common species. The other dominants were strombidiids ciliates including Strombidium conicum and S. globosaneum, which were followed by the tide form, Mesodinium pulex. Ciliates abundance ranged from 46 ind L-1 to 368 ind L-1 in the open sites, 46-368 ind L-1 at surface and 73-198 ind L-1 at 75 m deep layer. In the Yongshu reef, ciliates abundance ranged from 167 ind L-1 to 365 ind L-1 in the water column, similar to that in Sanya coral reef waters. Ciliates composition showed obvious difference between surface and 75 m deep layer at station S2 (P < 0.05), while no similar result was observed at other sites. At 75 m deep layer, salinity was negatively related to mixed layer depth (P < 0.05), but positively to chlorophyll a concentration (P < 0.05), indicating that the change of vertical mixing in water column influenced vertical distribution of ciliates in the southern SCS

    The complete mitochondrial genome sequence of Cynoglossus abbreviatus (Pleuronectiformes: Cynoglossidae) with control region translocation and tRNA-Gln gene inversion

    Cynoglossus abbreviatus (Cynoglossidae, Soleoidei) is characterized by a bilaterally asymmetrical with both eyes on the left side. In this study, the complete mitogenome of this tongue sole has been reported for the first time. The gene order in C. abbreviatus mitogenome possesses a novel rearrangement like other tonguefish. The tRNA-Gln gene moves from the light strand to the heavy strand, accompanied by tRNA-Ile gene shuffling, leaving a large non-coding region (88 bp) between these two tRNAs. Additionally, the control region translocates to the place between ND1 and tRNA-Gln genes. The total length is 16,417 bp, with 30.9%, 29.5%, 24.9% and 14.7% for A, T, C and G, respectively (60.4% for AT content). These molecular data will provide useful information about the mechanism of gene reorganization in Cynoglossidae mitogenome and further phylogenetic study on Pleuronectiformes
