MATHEMATISCHE BEHANDLUNG VON SINGLE-CELL-PROTEIN-PROZESSEN
Hansenula polymorpha fed-batch cultivations were carried out on ethanol-substrate in
a cocurrent 275 cm high bubble column loop bioreactor. The studied single-cell-protein-(
SCP-) processes are time- and spacedependent.
The mathematical description leads to a set of coupled nonlinear partial differential
equations (distributed parameter system). The model equations contain X-, S-, and 0,-
balances in the liquid phases of column and loop as well as 0,-, No- (inert) and C0, -
balances in the gas phase of the reactor. Nonstationary model equations have been
solved and unknow parameters, volumetric mass transfer coefficient ky +a, 0,- and Syield
coefficients, Yyyo and Yyys> 0,- and S-saturation constants, Ko and Ko» have
been identified -by fast hybrid simulation methods. Some identified parameters depend
strongly on the process conditions, classified into unlimited growth, oxygen transferand
substrate-limited growth.
The time dependence of parameters during the dynamical processes needs adaptive control
strategies. Based on excellent agreement of experiments and mathematical model
on-line control algorithmen (OLFO=Open Loop Feedback Optimal Control) have been developed
for optimal control of batch processes in tower reactors

Zusammenfassung
In den folgenden einführenden Worten wird auf verschiedene Einfachverfahren,
die mit Erfolg in Gebrauch sind, hingewiesen. Sehr unterschiedliche
Gebiete, in denen Einfachverfahren verwendet werden, werden dargestellt.
Mikrobielle Proteingewinnung ist aus Einfachtechnologien entstanden.
Wenn heute fiir die mikrobielle Proteingewinnung im wesentlichen z.T.
sehr komplizierte Technologien angewendet werden sollen, so hat dies
seinen Ursprung darin, daß man seit 1962 bis etwa 1973/74 fest beabsichtigt
hatte, mikrobielle Proteine im wesentlichen aus Alkanen und
in den letzten Jahren besonders aus Methanol oder Methan zu gewinnen.
Inzwischen sind die konventionellen Kohlenstoffquellen wieder interessant
geworden und stehen z.T. auch in ausreichender Menge zur Verfügung.
Es sind dies besonders stärkehaltige Produkte, Melasse, Soja
und verschiedene andere Kohlenstoffquellen. Auch die co,-Fixierung
zur mikrobiellen Proteinbildung wird weiterhin bearbeitet. Cellulose
als Kohlenstoffquelle ist gegenwärtig an vielen Forschungsstätten der
Welt in Bearbeitung, ihre Anwendung ist aber noch keinesfalls gesichert.
Die genannten Kohlenstoffquellen sind außer der Cellulose zur Verwendung
in Einfachverfahren geeignet.
Die vergangenen Ölkrisen haben es bewirkt, daß Alkane als Kohlenstoffquelle
für die Züchtung von Mikroorganismen zur SCP-Gewinnung nur noch
in solchen Ländern angewandt werden, in denen sie in ausreichender
Menge zur Verfügung stehen. Hierzu gehören besonders die Ostblockländer.
Einfachverfahren sind nicht nur billig, sondern sie lassen sich auch
in Entwicklungsländern verhältnismäßig leicht einführen, da sie von
der einheimischen Bevölkerung auch bewältigt werden können.
Das Prinzip der Einfachtechnologien liegt darin, durch Auswahl des
Substrates, der Mikroorganismen und der Technologie ökologische Bedingungen
zu schaffen, die eine Entwicklung der zur Biomasse eingesetzten
Mikroorganismen begünstigen. Die Entwicklung anderer Mikroorganismen
wird dabei stark unterdrückt, aber nicht vollständig ausgeschlossen.
Dadurch muß der gesamte Aufwand zur Führung des Verfahrens
unter Ausschluß von Fremdkeimen nicht gemacht werden: Bei Einfachverfahren
hat also die Ökologie der Mikroorganismen eine ganz wesentliche
Bedeutung. Durch unsere vermehrten Kenntnisse der ökologischen Bedingungen
bei der Züchtung von Mikroorganismen könnten wir gegenwärtig
wieder vor einer neuen Entwicklung von Einfachverfahren stehen.
Aus all diesen Gründen ist es sicherlich richtig, vorhandene und bewährte
Einfachverfahren zur SCP-Herstellung einmal durchzusehen, um zu
klären, ob hier ein Potential für eine billige mikrobielle SCP-Gewinnung
existiert.
Im folgenden sollen einige Gruppen von Einfachverfahren vorgestellt
werden. Keinesfalls sind sämtliche hier vorgestellten Verfahrensgruppen
zur Erzeugung von SCP geeignet
MIKROBIELLE PROTEINGEWINNUNG - EINFUHRUNG
Nach den Produktionsverfahren und Einfachtechnologien wird in diesem
dritten Teil die mikrobielle Proteingewinnung behandelt. Im ersten Vortrag
von Teuber und Moebus wird über Arbeiten mit Molke als Substrat
und Prozeßwasser für die Produktion von SCP berichtet. In der ersten
Stufe wird die Lactose der Molke in Milchsäure vergoren und in der zweiten
Stufe mit enzymatisch abgebauter Stärke aus Weizenmehl als Substrat
in der umgesetzten Molke, Saccharomyces cerevisiae in fed batch Betrieb
kultiviert. Die Zusammensetzung der Biomasse wird näher untersucht
MOLKE ALS SUBSTRAT UND PROZESSWASSER FUR DIE PRODUKTION VON SCP
The aim of our study was to develop a technique allowing the production of yeast
protein from agricultural substrates by avoiding waste water. The product should be
of high biological quality and harmless in terms of toxicity. Using Saccharomyces
cerevisiae acidified whey, to which enzymatically hydrolysed wheat flour was added,
was fermented in a 20 m’ plant in a two step procedure:
1. Conversion of the lactose in whey into a mixture of lactic acid/ammonium-lactate
by lactic acid fermentation.
2. Fermentation of the suspension at high cell densities in the fermenter by deep
jet aeration.
Using a semi-continuous feeding system the RNS content in the yeast solids fell from
4,07 to 2,37 % within 21 h. In technical experiments carried out simultaneously on
the reduction of the RNS content using heat shock treatment a yeast was obtained which
contained 3,05 % RNS in the solids.
After drying of the fermented material, the crude protein (less ammonia) and the ash
contents in the final product accounted for 50 and 5,7 %, respectively. Residual
humidity was 6,2 % (drying tower) and 8 % (roller-drying)
QUALITATSASPEKTE BEI DER BIOPROTEINHERSTELLUNG
The product quality of microbial biomass is influenced by three determinants:
Biosynthesis of the constituent cell components of a microorganism
strain is genetically fixed, in particular the enzymatic
protein pattern. Here, a variation of product properties can only
be achieved through selection, mutation, or controled genetic manipulations.
For a given species this genetic pattern is fixed. Within
the genetically predetermined limits analytical variation can be
achieved by means of differing fermentation parameters. The nutritive
and biological product value, however, results more than from fermentation
from the techniques of harvesting and processing. This aspect
is the most important one and has been neglected so far in basic
research. This calls for an intensified systematic investigation program,
which may enable bioprotein to open up the full potential of
its utilisation in animal feeding, food technology and as for technical
functionalities
SOFTWARE ENGINEERING: EXPERIENCE IN A CONSULTING AGENCY
The paper emphasizes the importance of a methodology in programming, on the opi—
nion that we are in the proper times to develop a "new deal" in EDP. The methodology
is defined as a habit of systematic reasoning which faces problem complexity
by a proper interpretation of structured design and by "egoless" programming. The
development of this habit of thought is based upon the choice of a proper programming
language and upon a new team organization: the adaptive team. The result and
the use of these ideas, in the design and implementation of production planning has
been very successful in reducing the time required to test and debug large software
systems
APPLICATION OF AN ON-LINE SYSTEM OF COUPLED COMPUTERS TO FERMENTATION MODELLING
The application of the method of quasilinearization to fermentation modelling is discussed. The text includes a tutorial explanation of the method. Examples include
parameter identification in a growth model and both parameter and state estimation
in a column reactor. The latter example was performed by an on-line system
of coupled process and time-sharing computers
RICHTLINIEN FUR DIE BEWERTUNG VON EINZELLERPROTEIN: BERICHT AUS DER TÄTIGKEIT DER IUPAC COMMISSION ON BIOTECHNOLOGY
Since many years the "Commission on Biotechnology'' of IUPAC is concerned
with setting up guidelines for the evaluation of single cell
proteins (SCP). In the first general rules for SCP as animal food,
mainly hydrocarbons were considered in addition to carbohydrates as
starting materials for SCP from yeasts and bacteria. The rules have
been published as IUPAC Technical Reports - Number 12 in 1974. The
use of methanol as an additional important raw material for SCP
demanded an adequate extension of these rules. As a result the
"Guidelines for Testing of Single Cell Protein, destined as Protein
Source for Animal Feed - II' were published in 1979. In cooperation
with the "Commission on Food" of IUPAC and the "Protein
Advisory Group" of WHO/FAO, the commission is now preparing once
more adapted recommendations for SCP as protein supplements, destined
for human consumption
Pentacyclic Triterpenoids, Phytosteroids and Fatty Acid Isolated from the Stem-bark of Cola lateritia K. Schum. (Sterculiaceae) of Cameroon origin; Evaluation of Their Antibacterial Activity
The phytochemical investigation on the chemical constituents of dichloromethane-methanol (1:1) stem-bark extract ofCola lateritiaK. Schum. (Sterculiaceae) led to the isolationand characterization of five pentacyclic triterpenoids, one fatty acid and two phytosteroids. Thecompounds were identified as heptadecanoic acid (1), maslinic acid (2), betulinic acid (3), lupenone(4), lupeol (5), friedelin (6),b-stigmasterol (7) andß-sitosterol-3-O-ß-D-glucoside (8). Their struc-tures were determined by NMR analysis (1H,13C, DEPT-135, COSY, HMBC and HSQC), high-resolution mass spectrometry (HR-ESI-MS) and comparisons with published data in the literature.This work, to the best of our knowledge, is the first isolation and identification of these compoundsin pure forms fromCola lateritia. Also, compounds1–3are reported for the first time fromColagenus.In vitroantibacterial activity of the isolated compounds (1–8) and the crude extract wereevaluated againstBacillus subtilis,Staphylococcus epidermidis,Enterococcus faecalis,Mycobacterium smegmatis,Staphylococcus aureus,Enterobacter cloacae,Klebsiella oxytoca,Proteusvulgaris,Klebsiella pneumonia,Escherichia coli, Proteus mirabilisandKlebsiella aerogeneswithstreptomycin, nalidixic acid and ampicillin as standard antibacterial drugs. Compound2was activeagainstE. faecalis(MIC = 18.5mg/mL), and it was 6.9 and 28 times lower and active than that ofstreptomycin (MIC 128mg/mL) and nalidixic acid (MIC>512mg/mL) respectively. INDUZIEREN EINZELLERPROTEINE MUTATIONEN?
Single cell protein (SCP) - a product containing 80 % raw protein obtained
from bacteria grown on a methanol substrate - was tested for
mutagenicity in 5 in vivo tests in different mammalian species. The
animals were fed either a standard laboratory diet, a semi-synthetic
diet, diets containing 12.5 and 25 % SCP or pure SCP. The following
investigations were carried out: dominant-lethal-test; host-mediated
assay; micronucleus test; chromosome studies in bone marrow celis;4in
