    Im allgemeinen sind Fermentationslösungen schwierig zu analysierende Dreiphasensysteme. Sie sind komplexe Lösungen anorganischer und organischer Komponenten, physikalisch gelöster Gase und suspendierter Feststoffe unterschiedlicher Größe wie Mikrooganismen oder Cellulosepartikel. Die kontinuierliche Online-Bestimmung der wichtigsten Prozeßparameter ist eine wesentliche Voraussetzung für eine effektive Kontrolle und Regelung von Fermentationsprozessen. Wegen der hohen Anforderungen, die an interne Sensoren gestellt werden (Dampfsterilisierbarkeit, stabile Kalibrierung, hohe Selektivität), ist die Zahl der kommerziell im Fermenter einsetzbaren Sonden begrenzt. Um externe Detektionsverfahren nutzen zu können, werden Membranprobenahmesysteme verwendet. Hierdurch wird die Kontamination des Kulturmediums vermieden und die Anzahl bestimmbarer Komponenten erheblich vergrößert. Uber Erfahrungen bei der Probenahme zur Online-Messung von Substrat- und Produktkonzentrationen wird ein Überblick vermittelt. Eingegangen wird auf die Standzeit der Membranen, Filtratleistung und Antwortzeit

    Bei hoher Zelldichte kann das Wachstum tierischer Zellkulturen in Suspension durch die Versorgung mit Sauerstoff limitiert werden. Während in kleineren Volumina der Sauerstoffeintrag über die Oberfläche in der Regel ausreicht, ist für Suspensionskulturen im größeren Maßstab die gleichzeitige Erfüllung folgender Forderungen notwendig: - Homogene Suspendierung von Zellen oder Microcarriern; - Geringe Scherkräfte beim Rühren und Begasen unter Gewährleistung ausreichender Sauerstoffversorgung auch für Zellen mit hohem 02-Bedarf; - Hohe Mischgeschwindigkeit; - Möglichkeit der Maßstabsvergrößerung; - Möglichkeit der Nachrüstung an herkömmlichen Rührkessel-Fermentern.     Two kinds of experimental efforts which might lead to the increase in the mass-productivity of an enzyme-modified FET biosensor are reported. Firstly, a new enzyme membrane deposition technique has been developed ; an enzyme membrane is photolithographically deposited on the specific area of an FET using a water soluble photoresist. Secondly, a new flow-through cell has been deviced for exo vivo applications of an enzyme-modified FET. It allows one to use an FET sensor without any polymeric encapsulation and wire bonding

    Starter cultures are contributions of modern research in biotechnology to the industrial production of fermented foods. These products represent an impressive number among our best traditional foods and are fermented by a corresponding multitude of microorganisms. During the period of the past 100 years starter cultures have been developed which have shown their special value for attaining a constant high food quality and which are applied with more or less success in the various fields of food production. They are essential in dairy and brewing technology and a similar trend can be observed for the production of Taw sausages, sour dough and wine. Limitations of a general application of starters exist when fermentation processes are extended over longer periods (weeks and months) and changes in the microbial population take place. The development of the ideal starter cultures requires not only a profound knowledge in production processes and the characteristics of food quality but also in microbial ecology and physiology. There is a gap of knowledge especially in these latter fields which also extends to genetics. With the proceeding improvement of this knowledge, the starter technology will also be raised to a higher status. In this development dairy technology is leading since it applied starters first consistently and has worked consequently on improving starter technology

    The utilization of a thermophilic Glucose isomerase and a psychrophilic B-Galactosidase in the food industry is discussed. Both enzymes show several advantages for their specific field of use. In the case of ßB-Galactosidase the only possible way to come to an industrial application seems to be the cloning of the enzyme in a microorganism, that is already used in the food industry. ZUSAMMENFASSUNG Die Einsatzmöglichkeiten einer thermophilen Glukose-Isomerase sowie einer psychrophilen ß-Galaktosidase in der Lebensmittelindustrie werden dargestellt. Beide diskutierten Enzyme bringen für die jeweiligen Einsatzmöglichkeiten vorteilhafte Eigenschaften mit. Im Falle der B-Galaktosidase scheint aber die einzige Möglichkeit der industriellen Verwertung die Clonierung in einem in der Lebensmittelindustrie bereits eingesetzen Mikroorganismus zu sein
