DETERMINATION OF TRANSLATIONAL DIFFUSION ON THE MODEL OF AMPEROMETRIC ENZYME ELECTRODE USING ESR-ZEUGMATOGRAPHY
In this paper we report the construction of an amperometric enzymic
microelectrode for parallel detection of both the electrochemical and
the electron spin resonance (ESR) signal. The electrode ig arranged
within a continuous-flow System in order to change the substrate
without disturbing the sample position in the ESR cavity. By means of
an ESR zeugmatography device one-dimensional distributions (projections)
of charge carriers (unpaired electrons) were recorded. The
diffusion profile of motion through the enzyme layer perpendicular to
the electrode plane for 4-hydroxy-2,2,6,6,-tetramethylpiperidine-1-
oxyl (TEMPOL) was determined
COVERAGE OF ELECTROCHEMICAL TRANSDUCERS MADE IN Si- TECHNOLOGY WITH ENZYME MEMBRANES
Some methods to cover electrochemical sensors based on silicon
technology with enzyme membranes using different polymer systems
are known. The main requirements of enzyme membranes at
electrochemical sensor elements based on silicon technology, such
as ion sensitive field effect transistors (ISFETs) and thin film
noble metal electrodes (TFMEs), are good adhesion at the sensor
surface and minimal diffusion resistance for substrates and
products. Furthermore the amount of enzyme molecules immobilized
at the active surface of the sensor element influences the
functional Parameters of the biosensor, e.g. lifetime,
sensitivity, linearity, and response time, which should be in
the same range as for the enzyme membranes of first generation
biosensors. In this paper we describe new covering procedures
that have been studied in order to find out technologies
compatible to microelectronic Production technology used for the
fabrication of TFMEs (FIG 1) and multigate ISFETs
HYDROXY FATTY ACIDS AS VALUABLE CHEMOSYSTEMATIC MARKERS IN GLIDING BACTERIA AND FLAVOBACTERIA
The fatty acid patterns of Cytophaga-like bacteria are dominated by 2- and 3-hydroxy
fatty acids with iso-branched carbon-chains. Several flavobacteria with DNA in
the low GC-range have patterns nearly identical to those from cytophagas. With other
flavobacteria, particularly such with DNA in the high GC-range, fatty acids with
branched chains are almost completely lacking, and hydroxy fatty acids are present in
small quantities only. Myxobacteria clearly form two groups with respect to their
fatty acid patterns. While the representatives of the suborder Cystobacterineae (Myxococcus,
Cystobacter, Stigmatella) contain substantial quantities of hydroxy fatty
acids, the representatives of the suborder Sorangineae (Sorangium, Nannocystis) contain
no hydroxy fatty acids at all, at least not in total cell hydrolysates. The main
fatty acids of all myxobacteria have iso-branched carbon chains.
As has been known for some time, the fatty acid patterns of gliding bacteria are
usually dominated by compounds with (iso-) branched carbon chains /3, 9, 10, 11, 12,
13/. This is uncommon for strictly aerobic gram-negative bacteria. Recently another
peculiarity of the fatty acid patterns of gliding bacteria has been discovered: i.e.
that many of these organisms contain substantial amounts of hydroxy fatty acids /3/.
In some cases up to 50 % of the total fatty acids were hydroxylated. While up to this
time hydroxy fatty acids had only been demonstrated in lipopolysaccharide preparations
from gliding bacteria /9/, they have now been found even in total cell hydrolysates,
and in addition to the more common 3-hydroxy compounds, 2-hydroxy fatty acids
could be identified. Branched-chain 2-hydroxy fatty acids are quite unusual and seem
not to have been isolated from living organisms before. It was interesting to see,
that the different groups of gliding bacteria each seem to have their own special fatty
acid patterns, with the characteristic differences usually in the hydroxy acid fraction.
Thus Flexibacter elegans and several other Cytophaga-like bacteria (CLB) have
as the main components (besides Cig: C161 and iso Cis) 2- and 3-0H iso Cis> and 3-0H
iso C17 acids, while the myxobacteria Myxococcus fulvus and Stigmatella aurantiaca
yielded mainly (besides Ce: Ge: and iso Cis) iso C17 and 2-0H iso C7 acids. It
thus appeared to us that the fatty acid pattern might be used in this group of bacteria
as a systematic lead. We therefore analyzed more strains and species of gliding
bacteria for their fatty acid patterns, and also included several flavobacteria in
this study, because there may be taxonomic relations between certain flavobacteria
and some of the CLB
CLINICAL SIGNIFICANCE AND ANTIMICROBIAL SUSCEPTIBILITY OF FLAVOBACTERIA
Flavobacteria are not frequently isolated from patient material. F. meningosepticum
is a well-known agent of neonatal meningitis which has a
poor prognosis. Colonization of compromised hosts in the respiratory
tract may occur under epidemic situations. Clinically significant isolates
from other sources are in the minority. F. odoratum and II-F have
been most frequently isolated from the urogenital tract, and II-J from
dog or cat bites. With the exception of the penicillin-sensitive II-F
and II-J, most flavobacteria are resistant to many antimicrobials. Effective
compounds include rifampin, nalidixic acid, novobiocin, and
sulfamethoxazole-trimethoprim. There may be considerable discrepancies
between the results of disc and tube dilution tests
ANWENDUNG VON MEMBRANVERFAHREN BEI DER SIMULTANEN HERSTELLUNG VON MANNITOL UND GLUCONSÄURE AUS SACCHAROSE DURCH KONJUGIERTE NAD *-ABHÄNGIGE DEHYDROGENASEN
Es wird ein Verfahren zur simultanen und kontinuierlichen Herstellung
von Gluconsäure und Mannitol durch konjugierte NAD(H)-abhängige Dehydrogenasen
innerhalb eines Membranreaktors beschrieben. Die Synthese
löslicher polymergebundener von Glucose- und Mannitol-Dehydrogenase
akzeptierter Coenzyme, sowie die Herstellung von Hohlfasermembranen,
die zu ihrer Rückhaltung innerhalb des Reaktors geeignet sind, werden
dargestellt. Erstmals wird eine Mannitol-Dehydrogenase mit NAD(H) -Spezifität
beschrieben. Die Einsatzmöglichkeiten der Elektrodialyse
zur Aufrechterhaltung des optimalen pH und als Stufe der Produktaufarbeitung
werden diskutiert. Experimentelle Daten zur Reaktorperformance
werden mit Modellrechnungen verglichen.
Die in der Nahrungsmittelindustrie zum Einsatz kommenden enzymtechnologischen
Verfahren beschränken sich bisher fast ausschließlich auf
coenzymunabhängige Reaktionen (Hydrolysen, Isomerisierungen). Erst in
den letzten Jahren wird versucht, insbesondere NAD (P)*-abhängige Enzyme
für die Synthese von wirtschaftlich interessanten Produkten wie
z.B. L-Aminosäuren oder Derivaten von Monosacchariden zu nutzen. Um
mit derartigen Enzym-Coenzym-Systemen im größeren Maßstab ökonomisch
arbeiten zu können, ist es erforderlich, die Coenzyme - wie auch die
Enzyme - in nur katalytischen Konzentrationen einzusetzen und sie
durch kontinuierliche Regeneration für eine möglichst große Zahl von
Reaktionszyklen wiederverwendbar zu machen. Prinzipiell kommen enzymatische
und nicht-enzymatische (chemische, elektrochemische) Verfahren
fiir die Coenzymregeneration in Betracht. In der Praxis haben sich die
enzymatischen Cofaktor-Regenerationssysteme am besten bewährt. Bei der
Entwicklung enzymatischer Syntheseverfahren für L-Aminosäuren mittels
reduktiver Transaminierung entsprechender durch chemische Synthese
leicht zugänglicher Ketocarbonsäuren konnten Wandrey und Mitarbeiter
/1-3/ erstmals das Prinzip der Cofaktorregeneration für einen Redoxprozeß
in einem über die Laboranlage hinausgehenden Maßstab realisieren.
Jedoch muß man bei diesen Prozessen in Kauf nehmen, daß die das Coenzym
regenerierende Hilfsreaktion ein nicht verwendbares Produkt liefert
(Abb. 1, Beispiel 1
ENZYM-MEMBRAN-REAKTOR
Die Immobilisierung von Enzymen und die technische Verwendung immobilisierter Enzyme
wird in der BAYER AG von der Verfahrensentwicklung Biochemie seit mehr als 15 Jahren
intensiv bearbeitet. So wird als Zwischenprodukt zur Synthese halbsynthetischer
Penicilline im Werk Wuppertal 6-Aminopenicillinsäure aus Penicillin unter Verwendung
immobilisierter Penicillinacylase im großtechnischen Maßstab hergestellt. Zur
Entwicklung neuer Verfahren müssen angepaßt an das Substrat und das Produkt der
Träger, die Immobilisierungsmethode und der zur Biotransformation verwendete Reaktor
entwickelt und optimiert werden
POLYMERE FUR SYNTHETISCHE MEMBRANEN UND IHRE CHEMISCHE MODIFIKATION
Ohne das klassische Membranpolymer Cellulose und ihre Derivate wäre die Entwicklung
der Theorien zum Transport durch Membranen und auch die praktische Anwendung
undenkbar. Vorzügliche Dienste leistete in der Anfangsphase die Nitrocellulose,
zu Beginn der sechziger Jahre war jedoch Cellulose-2,5-acetat dominant, dem wiederum
vollsynthetische Polymere überlegen sind, zumal durch chemische Modifikation
das Anwendungsspektrum verbreitert werden kann. So ist z.B. Polysulfon ein geeignetes
Material für die Herstellung von Ultrafiltrationsmembranen; nach Sulfonierung
eignet es sich als Membranpolymer für die Hyperfiltration und, da es nunmehr negative
Festionenladung trägt, auch zur Herstellung homogener Elektrodialysemembranen.
Chemische Modifikation von Membranpolymeren zielt auf Verbesserung der Gebrauchseigenschaften
ab, etwa hinsichtlich Rückhalt oder Volumenfluß aber auch zur Erhöhung
der Hydrophilie der resultierenden Membranen, zur Verbesserung der Kompatibilität,
wenn Interpolymermembranen hergestellt werden sollen oder zur Einführung von
geeigneten Gruppen, die eine chemische Fixierung von Enzymen an der Membran ermöglichen
MEMBRANEN IN DER BIOTECHNOLOGIE
Membranen und Membranprozessen wird heute im Zusammenhang mit biotechnologischen
Produktionsverfahren ein ganz erhebliches Interesse entgegengebracht. Obgleich die heute
verfiigbaren Membranen und Membranprozesse sowohl in ihrer Funktion als auch in ihrer
Anwendung sehr unterschiedlich sind, haben sie eine Reihe von Eigenschaften gemeinsam, die
ihre Nutzung in der Biotechnologie besonders geeignet erscheinen läßt. Eine konsequente
Nutzung von Membranen in der Biotechnologie läßt eine vereinfachte Prozeßführung und damit
preisgünstigere und qualitativ bessere Produkte erwarten.
Membranen können an verschiedenen Stellen in einem mikrobiologischen Produktionsprozeß
eingesetzt werden. So z. B. bei der Sterilisierung der in einen Bioreaktor einzubringenden
Substrate, oder bei der Aufarbeitung von Bioreaktorinhaltsstoffen. Sie können aber auch als
wesentliches Element eines Sensors zur Online-Analyse von Reaktorinhaltsstoffen, oder
aber auch als sogenannter Membranreaktor für eine kontinuierliche Ausschleusung der Reaktionsprodukte
oder unerwünschter Nebenprodukte genutzt werden.
Trotz der offensichtlich günstigen Voraussetzungen ist der Einsatz der Membranen in der
Biotechnologie aus verschiedenen Gründen bisher weit hinter den Erwartungen zurückgeblieben.
Einmal stellt die Stofftrennung mit Hilfe von Membranen noch eine relativ neue Technologie
dar, die auch mit den entsprechenden Anfangsschwierigkeiten behaftet ist, zum anderen sind
die heute kommerziell verfügbaren Membranen für ganz bestimmte Stofftrennprozesse wie z.
B. für die Entsalzung von Meerwasser entwickelt worden und müssen, um erfolgreich in der
Biotechnologie eingesetzt zu werden, den speziellen Problemen der Biotechnologie angepaßt
werden.
Im -vorliegenden Referat wird der heutige Stand der Membranentwicklung zusammenfassend
dargestellt sowie Möglichkeiten und Probleme, die sich beim Einsatz der Membranen in der
Biotechnologie ergeben, diskutiert
ENZYME IMMOBILIZED MEMBRANE PATTERNING METHOD FOR SOS/ISFET BIOSENSORS
A new method for preparing enzyme immobilized membranes of miniature biosensors
has been developed and demonstrated by fabricating a glucose sensor, based on the ion
sensitive field effect transistor (ISFET) . The enzyme solution, containing glutaraldehyde,
was spin coated onto a wafer covered with a patterned photoresist. The
resulting enzyme immobilized membrane was lifted off by ultrasonic vibration in
acetone, except for the membranes on the pH-sensitive regions of the transistors.
The deposited membranes were precisely patterned and hardly peeled off. The method
is compatible with the IC processes and applicable to biosensor mass-production
FIBER OPTICAL pH-SENSOR BASED ON LASER-INDUCED FLUORESCENCE
A fiber optical pH-sensor based on laser-induced fluorescence was developed. The fluorescence
was ww. with the light of a Helium-Neon-Laser (5°mW, Polytec model PL 7504)
emitting at X4 632.8 nm. The dye used was tetra-(p-sulfophenyl)-porphyrine (Ventron),
‘whieh-has in an aqueous solution an extinction coefficient at A = 632.8 nm of e = 4365
I/mol - cm. Its fluorescence behaviour changes at different-pH-values- (figure). It is a
water-soluble dye and the sulfo-groups give the possibility to immobilize this dye on anion
exchange resins. The fluorescence spectrum of this dye has a maximum at pH = 6.8 at
A= 675 nm and A= 700 nm, respectively, which offers the advantage of a ratio-measurement
