Isolation and Characterization of Lytic Bacteriophages from Wastewater and its Application in Pathogen Reduction
Present work wasperformedto isolate, enrich and characterizebacteriophageusing basic laboratory set-up, and to demonstrate their lytic potential in pathogen reduction. Following standard microbiological and molecular biological procedures, we examined cultivable bacteria and bacteriophages in wastewater sample. Subsequently, from cultures of wastewater sample, we isolated and characterized bacterial isolate, which was thenusedtoenrichlytic bacteriophages, using a combination of double layer plaque-assay, isolation and genetic techniques to isolate bacteriophage against Pseudomonas aeruginosa. Subsequently decontamination capability of this phage was assessed in form of spray and matrix adsorbed formulations. Storage stability of phagepreparations at different temperatures was also studied by calculating reduction in phage titer. Infra Red Spectra of Different Species of Cultivated Oyster Mushrooms Possible Tool for Identifying Bioactive Compounds and Establishing Taxonomic Linkage
Mushrooms are macrofungi that serve as a vegetarian source of protein along with various bio-active molecules of primary health importance. The activity of the bio-active molecules range from antioxidant, immunomodulation, hepatoprotection. Cultivated oyster mushrooms are also rich in these components which may be estimated quantitatively by skill intensive ‘destructive’ chemical techniques. Infra Red spectroscopy provides a non-destructive user friendly technique to quickly assess the presence of bio-active compounds in mushroom species to be used as a quality control measure as this non-destructive tool can be used to segregate mushroom harvest according to availability of bioactive compounds. IR spectra based strain classification and taxa delimitation of mushroom samples are also attempted vis a vis DNA sequence based phylogeny analysis of the same, but no correlation is observed between the two types of phylogeny analysis
Development and Standardisation of a Method for Inflicting Frostbite Injury in Rats and Formulation of Essential Oil in Treatment of Frostbite
Frostbite is a cold induced injury which occurs due to exposure of a particular site of body to sub-zero temperature. One of the primary reasons for lack of proper studies about the underlying mechanism of frostbite injury is due to non-availability of any reliable animal model and method for inflicting frostbite. In our current research, a device was designed and standardised to inflict frostbite wound in wistar rat. A formulation comprising different combination of essential oils was also developed and its activity was assessed and found effective in the treatment of frostbite wound
Flow among Scientists A Job Demands Resources Perspective
The study was conducted to investigate the relationship of flow at work of scientists with their job demands, job resources and personal resources. 64 scientists comprised the sample. Personal data sheet, work-related flow inventory, general self-efficacy scale and Copenhagen psychosocial questionnaire (second version) were the instruments used in the study. Descriptive statistics, t-test, ANOVA, correlation analysis and hierarchical regression were the statistical techniques used. The results of the study suggested that scientists of various age-groups and length of service did not differ from each other on job demands, job resources, personal resources and flow at work. It was also found that flow at work was significantly positively related to job resources and personal resources but not significantly related to job demands. Job resources and personal resources were found to be significant positive predictors of flow at work whereas job demands were not found to have a significant impact on flow at work

The northeastern part of India (NE India) is an important biodiversity hotspots and is blessed with a huge diversity of flora and fauna along with other natural resources. Being quite distinct in natural features from rest of the country, NE India has its own unique challenges and opportunities. This part of India is geographically isolated from China by Eastern Himalayas, while tropical forests separate this region from Myanmar and Bangladesh. The armed forces deployed in the NE region have to operate in a wide variety of environments ranging from high altitudes, to tropical rain forests, which pose several operational challenges
Growing Cabbage (Brassica oleracea var capitata L.) in Cold Winter under Passive Solar Greenhouse in Trans Himalayan Ladakh Region
Greenhouse experiments were conducted in three seasons (2017-18, 2018-19, 2019-20) to study the feasibility to grow cabbage, a temperature sensitive crop, during freezing winter months at high altitude (elevation 3340 m) trans-Himalayan Ladakh region. Three varieties viz. Golden Acre, Videshi and Megaton were studied under an improvised passive solar greenhouse. Head was formed in all the varieties despite the temperature extremes (0.0±1.6 to 39.5±0.9 ºC) inside the greenhouse. The mean marketable head weight ranged from 428.6±72.1 g to 831.2±193.0 g, depending on the variety. The mean head weight of Golden Acre was 831 g, which is 15-fold higher as compared to that of the crop grown under traditional greenhouse in Ladakh. However, the marketable head weight was lower as compared to the yield potential of the varieties. It took 133-163 days for Golden Acre to reach maturity for harvest as compared to the expected 60-65 days. Dramatic declines in intercellular CO2 concentration, photosynthesis rate and water-use efficiency were observed at 2:00 PM, which indicated that the plants were severely affected by high temperatures inside the greenhouse. High temperature inside the greenhouse resulted in bolting, head-splitting and loose head forming, and it ranged from 8-36% of the crop depending on the year and variety. The study suggested that cabbage can be successfully grown under improvised passive solar greenhouse during severe winter months in the trans-Himalayan Ladakh
Optimisation of Processing Parameters for the Preparation of Vegetable Curd from Decorticated Sesame Seeds (Sesamum indicum)
Current study aims to develop and optimise the processing parameters for the preparation of imitated curd by using vegetable milk extracted from decorticated sesame seeds (Sesamum indicum). Experiments were planned to optimise curdling process for sesame milk with respect to inoculum (1-5%), lactose (1-5%), added proteins (1-3%). It was established that total solid content in a range of 9-10%, dispersion stability with 30-45% scored well (7.8±0.2) on preferential sensory test i.e. nine-point hedonic scale. Sesame milk supplemented with 5% lactose, 1% electrolyte salt mix, 1% sucrose was subjected to fermentation (10±1.0) hrs at 35±2°C) with 5% inoculum (lactic acid bacteria culture) resulted into a vegetable curd with desired consistency (855.6±47.5 g.sec), acidity (0.57±0.02% lactic acid equivalent; LAE), flavor and overall acceptability (OAA). The Solid Not Fat (SNF) content of optimised vegetable curd was found to be 22.20±0.40%, with protein, fat and ash content as 6.70±0.06%, 6.91±0.06% and 0.92±0.01% respectively. The characteristics of vegetable milk (non-dairy) and toned milk (dairy) were well comparable on various aesthetic parameters (fluidity, color, flavor and taste). Proportional characterisation of vegetable (non-dairy) curd with dairy curd relating to its nutritional profile, color analysis, textural and rheological parameters established its similarity. Such dairy analogues may be considered as substitute to various dairy products where milk-based products are scanty or non-available
In Vitro Antioxidant Activity, Total Phenolic, Flavonoid and Tannin Contents in the Ajuga Bracteosa Wall. Ex Benth, Grown at Middle Hill Climatic Condition of Western Himalayas
The antioxidant activity of aqueous and alcoholic extracts of different plant parts viz, leaves, flower, stem and root of Ajuga bracteosa was investigated against various in-vitro antioxidant assays. The total phenolic, flavonoid and tannin contents also estimated. The results revealed the significant antioxidant potential and variation in the IC50, EC50 and phytochemical contents among all the plant parts. The aqueous extract of leaves exhibited significantly (P<0.05) highest antioxidant activity on 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acixd), 2,2-diphenyl-1-picryl hydrazyl, and potassium ferricyanide reducing power assay, with IC50 values of 0.2707±0.0008, 0.4409±0.0020, and EC50 value 0.3413±0.0030 mg/mL, respectively, followed by the other parts of the plant. The leaves extract also possess the highest total phenolics, flavonoid and tannin contents among all the parts. Similarly, the aqueous extract is better than the alcoholic extract of different parts as far as phytochemical contents and antioxidant activity concerned. The present study revealed that the aqueous extract of leaves had the highest antioxidant potential, which correlated with the high level of total phenolic fl flavonoid, and tannin contents. Therefore, higher the phytochemical contents, higher will be the antioxidant potential
Comparative Allelopathic Effects of Two Weed Extracts on Seed Germination and Seedling Growth of Vigna unguiculata (L.) Walp. and Abelmoschus esculentus L.
Pot culture experiment and Petri dish bioassay were conducted to assess the allelopathic potential of Cyanthillium cinereum and Lantana camara on seed germination and seedling growth of Vigna unguiculata and Abelmoschus esculentus. Aqueous leaf and stem extracts of weed were used for treatment. The differential inhibitory effect was observed for two weed plants on two tested crops. The experimental results revealed that in case of pot culture experiment, lowest germination percentages (40.8±0.49%, 63.6±0.60% and 83±0.78%) were recorded in L. camara leaf extract treated set of A. esculentus and maximum decrease in seedling length (0.31±0.05 cm, 4.18±0.07 cm, 6.42±0.08 cm and 13.51±0.07 cm) was observed in stem extract treated a set of A. esculentus. L. camara stem and leaf extract induced a more negative effect on seedling length in both A. esculentus and V. unguiculata. For petridish bioassay experiment, lowest germination percentages (54.60±0.40%, 59.20±0.49%, 66.20±0.74% and 69.80±0.38%) were observed in L. camara leaf extract treated a set of A. esculentus. In V. unguiculata, lowest germination percentage (45.8±0.49%, 75.4±0. 40%, 84.4±0.25% and 89±0.32%) was observed in stem extract treated set. Maximum suppressive effect on seedling length was recorded for stem and leaf extract of C. cinereum of A. esculentus and leaf extract of L. camara and stem extract of C. cinereum of V. unguiculata
