Treatment of soybean seeds with molybdenum and inoculant: nitrate reductase activity and agronomic performance
The aim of this study was to analyze the effects of molybdenum and inoculant application via seed treatment in soybean, and their role in the nitrate reductase enzyme activity, agronomic traits and physiological quality of the produced seeds. The experiment was conducted at the Federal University of Pelotas (UFPel), Rio Grande do Sul state, Brazil. The treatments were shaped by five doses of molybdenum (zero; 16; 32; 48; 64 g.100kg-1 of seeds) combined with the presence and absence of liquid inoculant (Bradyrhizobium japonicum), at a dose of 200 mL per 100 kg of seeds. The evaluations of nitrate reductase activity, agronomic traits and physiological seed quality were performed in the present work. The measured agronomic traits were: plant height, number of pods per plant, number of seed per plant and thousand-seed weight. Standard germination test was executed to evaluate the produced seed viability. First germination counting, accelerated aging test, cold test, plantlet shoot and root length, plantlet shoot and root dry mass, and field seedling emergence were utilized to measure the seed vigour. Molybdenum, both in presence and absence of inoculant, positively influenced the enzymatic activity, for both vegetative and reproductive stages. The addition of inoculant and molybdenum provided increase in the vigour of the produced seeds. The enzyme activity was positively correlated with the agronomic traits and vigour tests. The addition of inoculant and molybdenum in the seed treatment provides a better expression of nitrate reductase activity and vigour of the produced seeds. The aim of this study was to analyze the effects of molybdenum and inoculant application via seed treatment in soybean, and their role in the nitrate reductase enzyme activity, agronomic traits and physiological quality of the produced seeds. Glycoalkaloids as medicinal agents from callus and regenerated plants of Solanum nigrum var. judaicum
The target of this study is production of glycoalkaloids from cultures of Solanum nigrum var. judaicum Besser. Further, to evaluate their therapeutic effects. S. nigrum var. judaicum leaves were implanted in MS media containing growth regulators for in vitro study. HPLC analyses were applied for qualitative and quantitative determination of glycoalkaloids. Cytotoxic effects against human carcinoma cell lines were evaluated. In addition, antiviral, antioxidant, anti-inflammatory and antiparasitic activities of the formed glycoalkaloids were estimated. HPLC data indicated the success of in vitro solasodine and solanidine glycosides production. Solasonine represented the highest concentration. Biological assays illustrated that obtained glycoalkaloids exhibited cytotoxic activity against human carcinoma cell lines that may be attributed to free radical scavenging activity (69.98%). Strong antiherps performance was observed (94%). In addition, the glycoalkaloids showed in vitro schistomicidal (IC50 76.4 ppm) and fasciolicidal (IC50 76.6 ppm) activities. In vivo anti-inflammatory assay revealed potent activity against carrageenan induced edema. Glycoalkaloids were formed 2-5 folds that of intact plant pointed to the efficiency of the cultures. The present findings referred to the pronounced biological performance of the produced glycoalkaloids including antiviral, cytotoxic anti-inflammatory and antiparasitic activities. Botanical derived medication from S. nigrum var. judaicum could be accomplished guided with the present data.

Growth and nutrient accumulation in three peach rootstocks until the grafting stage
Analysing growth and nutrient accumulation is generally employed for guiding the fertilisation practices in the seedling-production chain. In the production of peach rootstock, such evaluations can provide information to aid can to aid in discrimination available genotypes, especially regarding the efficient use and conversion of fertiliser into dry matter, which can reduce the time required to obtain plants suitable for grafting. The aim of this study, was to investigate the differences in growth and nutrient accumulation in three peach-rootstock cultivars. A completely randomised experimental design was used, in a 3 x 4 factorial scheme, including three peach rootstocks (\u27Flordaguard\u27, \u27Capdeboscq\u27 and \u27Okinawa Roxo\u27) and four periods of growth evaluation (20, 40, 60 and 80 days after transplanting - DAT), giving a total of 12 treatments, with four replications, one for each plant. The Flordaguard and Capdeboscq cultivars showed greater height and stem diameter as well as a greater accumulation of total dry matter at 80 DAT, and both followed the distribution: leaves (67%) > roots (33%), while total dry-matter production in \u27Okinawa Roxo\u27 was less, following the order: leaves (71%) > roots (29%). The rootstock under evaluation showed variations in growth, confirmed by the physiological indices, except for the leaf area and specific leaf area. In general, macronutrient accumulation in the shoots and roots of the cultivars during each growth period was different, suggesting intrinsic differences in each of the genetic materials regarding nutrient demand for maintaining the metabolic processes which are vital to plant growth.Analysing growth and nutrient accumulation is generally employed for guiding the fertilisation practices in the seedling-production chain. Correlations and path analysis in soybean progenies with resistance source to cyst nematode (race 3)
Path analysis is an important study that slices the correlation coefficients between two variables to evaluate whether the relationship between them is of cause and effect. This study aimed to estimate the phenotypic and genotypic correlations between agronomic traits and perform a path analysis in order to identify variables for indirect selection aiming at a higher grain yield. Fourteen soybean F6 lines from the soybean breeding program of FCAV–UNESP, Jaboticabal, São Paulo, Brazil, were analyzed. The experimental design was a randomized block design with three replications. The agronomic traits plant height at maturity (PHM), first pod height (FPH), lodging (Ld), agronomic value (AV), number of pods per plant (NP), number of seeds per plant (NS), and grain yield (GY) were evaluated. Overall, the genotypic correlations were higher than their corresponding phenotypic correlations. The genotypic correlations between grain yield and the traits agronomic value, number of pods per plant, and number of seeds per plant were positive, significant, and of high magnitude. Path analysis showed that the trait number of seeds per plant had the highest direct effect on grain yield, while the trait number of pods per plant had the highest indirect effect through the number of seeds per plant on grain yieldPath analysis is an important study that slices the correlation coefficients between two variables to evaluate whether the relationship between them is of cause and effect. Nitrate and ammonium proportions in sesame cultivation under irrigation with saline waters
This research was conducted with the aim of evaluating the water relations, growth and the production components of the sesame cv. CNPA G3, in function of the irrigation with waters of different salinities and fertilization with different proportions of nitrate (N-NO3-) and ammonium (N-NH4+). The study was performed in drainage lysimeters in conditions of greenhouse, in the municipality of Campina Grande, PB, Brazil. The adopted design was in randomized blocks, in a factorial scheme of 5x5 with three repetitions, whose treatments were constituted of five levels of electrical conductivity of the irrigation water - ECw (0.6; 1.2; 1.8; 2.4 and 3.0 dS m-1) and five proportions of nitrate and ammonium (200:0; 150:50; 100:100; 50:150; 0:200 mg kg-1 of soil). The irrigation with ECw superior to 0.6 dS m-1 negatively affected the growth and production of the sesame; the fertilization with N-NH4+ and N-NO3- in the proportion 200:0 resulted in a higher percentage of cell membrane damage; the utilization of N-NH4+ and N-NO3- in the proportion 50:150 provided increase in the relative water content in the leaf blade and in the mass of a hundred seeds; N-NO3 and N-NH4+ proportions of 200:0 and 150:50 and water with ECw of 0.6 dS m-1 promoted the highest values for the content of chlorophyll a and total number of sesame seeds. This research was conducted with the aim of evaluating the water relations, growth and the production components of the sesame cv. Morphological and molecular characterization of powdery mildew on watermelon plants in São Paulo state
Powdery mildew is one of the most commom foliar diseases of cultivated and wild cucurbits worldwide. Six different fungal species have been reported to be associated with powdery mildew of cucurbits, such as Podosphaera xanthii and Golovinomyces cichoracearum. The aim of the study was to identify the causal agent of the disease in watermelon plants (Citrullus lanatus) cultivated in greenhouses of the State of São Paulo. DNA total was extracted using Chelex method and the ITS region was amplified via standard PCR reaction using the specific primers PMITS1 and PMITS2. A fragment of 700pb was amplified and the sequence showed 99% of identity with P. xanthii (accession number JQ728480). Morphological observation showed that the conidia were ellipsoidal-ovoid, had fibrosin bodies and a bifurcated germinative tube in the lateral position. These characteristics and the molecular analyses confirmed P. xanthi as the etiological agent of powdery mildew in watermelon plants in São Paulo State.

Morphological characterization of vaginal epithelial cells of santa inês ewes subjected to estrus synchronization
Vaginal cytology analysis has been used to evaluate the different stages of estrous cycle of several species; it presents a direct correlation with the animal’s hormonal state and provides essential information about the female reproductive tract conditions. Two staining methods were tested to evaluate the vaginal epithelial cell morphology of nulliparous and multiparous ewes during the estrus period. An intravaginal device impregnated with medroxyprogesterone acetate was kept into 10 nulliparous and 10 multiparous ewes for 14 days for estrus synchronization. Then, the progesterone device was withdrawn, and 300 IU of eCG was administered intramuscularly. Vaginal smears were prepared for posterior staining with Panotico or Giemsa stains when estrus was detected. The cells were classified into nucleated superficial, anucleate superficial, intermediate, parabasal, and basal. The Panotico and Giemsa staining of the different cell types studied were satisfactory. A predominance of intermediate epithelial cells (p<0.05) was found after staining. No difference in percentages of the different types of vaginal epithelial cells between nulliparous and multiparous ewes were found. Therefore, both staining methods were efficient, and a predominance of intermediate cells is found in nulliparous and multiparous ewes during the estrus period.Vaginal cytology analysis has been used to evaluate the different stages of estrous cycle of several species; it presents a direct correlation with the animal’s hormonal state and provides essential information about the female reproductive tract conditions. Mapping of the current land use in part of the Irrigated Perimeter Nilo Coelho, Petrolina-PE, Brazil
The objective of this study was to quantify and spatialize the agricultural composition and current land use in part of the Irrigation perimeter Nilo Coelho from the Landsat 8 satellite digital image classification. At first we performed the unsupervised classification image related to January 2014, identifying the separability within natural vegetation, water bodies, agricultural crops and anthropized areas. Then the supervised classification Maxver model was performed where it was observed that the fruit classes “Grape” (5.41%) “Acerola” (3.75%), “Coconut” (6.14%) “Guava” (6.17%), “Banana” (2.29%) and “Mango” (8.18%) showed dominance in the study area. These classes stood out in terms of yield in the area, pointing out that in that expansion which occurs in an area with agricultural crops in the irrigated Nilo Coelho, crops of mango, coco, guava, acerola, banana and grape also grow. In addition, we observed the emergence of new areas with passion fruit crops, melon, watermelon, corn, beans, onions, pumpkin, sweet potatoes, tomatoes, bell peppers, among other crops. Furthermore, we also found the occurrence of pastures containing buffel and elephant grasses, intercropping or not. The estimated overall accuracy was 80.52%, and the mapping defined as very good, with Kappa index equal to 0.7783.The objective of this study was to quantify and spatialize the agricultural composition and current land use in part of the Irrigation perimeter Nilo Coelho from the Landsat 8 satellite digital image classification. Induction of tolerance to salt stress in soursop seedlings using hydrogen peroxide
The exogenous use of hydrogen peroxide is an alternative in the acclimatization of plants to salt stress due to the greater activity of antioxidant enzymes. In this perspective, this study aimed to evaluate the gas exchange and the growth of soursop seedlings under salt stress using hydrogen peroxide. The study was conducted under greenhouse conditions. The treatments were distributed in randomized blocks, in a 5 x 2 factorial arrangement, related to five levels of electrical conductivity of the irrigation water - ECw (0.6, 1.2, 1.8, 2.4, and 3.0 dS m-1) and two concentrations of hydrogen peroxide - H2O2 (0 and 20 μM), with four replicates and two plants per plot. The effects of the treatments were evaluated by the variables of gas exchange and growth. The increase in water salinity negatively affected the gas exchange and the growth of the soursop seedlings. The gas exchange and growth variables presented deleterious effects caused by the salinity of the irrigation water, mitigated by the exogenous application of hydrogen peroxide at the concentration of 20 μM. The exogenous use of hydrogen peroxide at the concentration of 20 μM can be used to induce salt tolerance in soursop seedlings. The exogenous use of hydrogen peroxide is an alternative in the acclimatization of plants to salt stress due to the greater activity of antioxidant enzymes.
