Indanone-based Mannich bases: Design, synthesis, in-silico molecular docking, ADME predictions and biological evaluation including carbonic anhydrases, acetylcholinesterase inhibition and cytotoxicities
This study examined the rational design, synthesis, carbonic anhydrases (CAs), acetylcholinesterase (AChE) inhibitory effects, and cytotoxicity of 2-(2-hydroxy-3-(aminomethyl))benzylidene)-2,3-dihydro-1H-inden-1-one (1-6) and 2-(2-hydroxy-3,5-bis(aminomethyl))benzylidene)-2,3-dihydro-1H-inden-1-one (7-11). All compounds exhibited significant inhibitory activity against hCA I, hCA II, and AChE enzymes. Among them, compound 9 demonstrated potent inhibition of hCA I (Ki = 46.828 f 11.32 nM) and AChE (Ki = 0.9820 f 0.402 nM), while compound 11 showed strong inhibition against hCA II (Ki = 24.683 f 6.216 nM). For comparison, the reference compound acetazolamide (AZA) inhibited hCA I and hCA II with Ki values of 183.390 f 19.71 nM and 104.60 f 27.60 nM, respectively. Regarding AChE inhibition, the reference drug Tacrine exhibited a Ki value of 58.85 f 12.1 nM. These results indicate that compound 9 is significantly more potent than AZA against hCA I and markedly outperforms tacrine in inhibiting AChE. Similarly, compound 11 shows superior inhibitory activity compared to AZA against hCA II. The compounds' cytotoxicity against four human oral squamous cell carcinoma (OSCC) cell lines was compared to that of three normal oral cells. The compound 3, 2-(2-hydroxy-3-(piperidin-1ylmethyl)benzylidene)-2,3-dihydro-1H-inden-1-one, stood out for its promising selectivity in cytotoxicity, demonstrating the highest SI (9.5 toward HSC-2), TS1 (4.8), and PSE (4.8) compared to other studied compounds. Compound 3 showed 5-fold higher tumor specificity than 5-FU when using epithelial normal (human oral keratinocyte) and four human OSCC cell lines. Molecular docking results demonstrated that molecule 9 exhibited a strong binding affinity to AChE (-12.3 kcal/mol) with a remarkably low inhibition constant (Ki = 0.963905 nM), suggesting its potential as a potent inhibitor despite the absence of conventional hydrogen bonding. Conversely, molecule 11 showed enhanced selectivity toward hCAII, forming two hydrogen bonds and displaying a binding affinity of 10.4 kcal/mol. These findings indicate that both molecules possess promising inhibitory potential and can serve as valuable candidates for further experimental validation. Overall, this study highlights the effectiveness of molecular docking as a predictive tool in drug discovery and supports the further development of these lead compounds for therapeutic applications. SAR-based structural changes of compounds 3, 9, and 11 have the potential to generate novel, highly effective molecules, providing alternative treatments for a wide range of therapeutic areas, including cancer and neurological illnesses

Investigation of Electrocatalytic Applications of Various Advanced Nanostructured Alloys-An Overview
Cyclic voltammetry (CV) is one of the advanced techniques used to determine various bioactive molecules, organic dyes, pesticides, veterinary drugs, heavy metals, toxic chemicals, etc. To determine all the above analytes, one needs an electrocatalyst for their electrochemical redox reaction. Many researchers have reported the use of metal nanomaterials, metal oxide nanomaterials, metal-organic frameworks, surfactants, polymers, etc., as modifiers in carbon paste electrodes to enhance their current response, stability, sensitivity, and repeatability. But some of the emerging, cost-effective, and highly efficient electrocatalysts are advanced nanostructured alloy powders. These advanced alloys are used as a modifier to determine various bioactive analytes. These alloy-modified carbon paste electrodes (MCPEs) show excellent selectivity, sensitivity, and stability due to their extraordinary electrochemical properties, as the compositional elements of most of the alloys belong to d-block elements in the periodic table, and these transition elements are famous for their brilliant electrocatalytic properties. The present review article mainly focuses on the determination of dopamine, AA (AA), uric acid, methylene blue, methyl orange, Rhodamine B, and the L-Tyrosine amino acid by various alloys like stainless steel, high-entropy alloys, and shape-memory alloys and how these alloys could change the perception of metallurgists and electrochemists in the future. These alloys could be potential candidates for the development of various electrochemical sensors because of their high porosity and surface areas
An example of a suitability analysis supported by Geographic Information Systems in the evaluation of agricultural production-oriented urban production landscapes: Bartın
Rapid population growth in cities has raised the necessity of taking measures regarding resource use. Especially during the COVID-19 pandemic, this necessity reminded society that the agricultural sector is one of the most crucial sectors for the future. In this study, suitable agricultural areas within the municipal boundary of Bart & imath;n province were identified using Geographic Information Systems (GIS) and the Analytic Hierarchy Process (AHP). Based on interviews with 15 experts, various factors such as land use type, slope, aspect, soil groups, land capability classes, and erosion risk were evaluated, and suitability maps were created. The findings indicate that 53.04% of the agricultural land within the municipal boundary is classified as highly suitable, 24.43% as suitable, and 22.45% as less suitable or not suitable. The suitability of public spaces (public areas, active and passive green spaces, healthcare facilities, places of worship, and educational areas) and arable lands was also analyzed. Among public spaces, 1.09% were classified as not suitable, 22.30% as less suitable, 66.22% as suitable, and 10.40% as highly suitable. In this context, it is argued that public spaces possess physical characteristics suitable for agricultural activities and can be considered production-oriented areas that enhance urban resilience
The effect of foot reflexology on pain intensity, sleep quality and bowel functions in patients undergoing appendectomy surgery
Anahtar kelimeler: Ağrı, apendektomi, ayak refleksolojisi, bağırsak fonksiyonları, hemşirelik, uyku kalitesiThis study aimed to evaluate the effect of foot reflexology on pain intensity, sleep quality, and bowel function in patients who underwent appendectomy for appendicitis. The study population consisted of 178 patients who underwent appendectomy due to acute appendicitis. The sample of the study was determined as 120 with ?=0.05, 0.60 effect size, 90% power by performing power analysis in G power 3.1.9.7 programme. In the postoperative period, the patients were divided into intervention (n=60) and control (n=60) groups by applying double block randomisation with a computer-aided program (www.randomizer.org). The study was conducted with patients who agreed to participate in the study and who had appendectomy surgery with the inclusion criteria. Data were collected using a patient identification form, the Visual Analogue Scale, the Richard Campbell Sleep Questionnaire (RSCQ), and the Bowel Function Monitoring Form. In the intervention group, the information of the patients was first recorded by means of a patient identification form. Pain intensity was evaluated just before foot reflexology was performed. Reflexology was applied to each foot of the patient for 10 minutes at the 1st hour and 3rd hour after the operation, for a total of 20 minutes. At the 1st and 3rd hours, the pain intensity of the patients was measured 5, 30, and 60 minutes after the application of each foot reflexology. 24 hours after the application, the patients' sleep quality and bowel functions within 24 hours and the time they first needed analgesics were evaluated. The control group was measured at the same period without foot reflexology. Sleep quality, bowel function within 24 hours, and the time of first analgesic requirement were evaluated in both groups after 24 hours. Number, percentage, mean, and standard deviation were used as descriptive statistical methods in the evaluation of the data. Chi-square tests analysed differences between the rates of categorical variables in independent groups. Data that were found to be normally distributed were analysed by independent groups t-test, dependent groups t-test, repeated measures ANOVA test, complementary Bonferroni test, and ANCOVA test. In the study, it was determined that pain levels tended to decrease over time after the application in the intervention group, and this situation became especially evident in the measurements after the third hour, while it increased in the control group (p<0.001). Repeated measures ANOVA results showed that pain levels changed significantly over time and that this change was due to the group effect (Fb=351.014 and 105.707; p=0.000). Bonferroni multiple comparison results also showed that pain levels decreased rapidly immediately after the application and this decrease was stronger in the intervention group (?²=0.856 intervention, ?²=0.642 control). The mean total Richard Campbell Sleep Questionnaire scores of the patients in the intervention group (83.200±9.271) were found to be significantly higher than those of the control group (65.717±13.128) (t=8.426; p=0.000). There was no statistically significant difference between the groups in terms of the time of first flatulence (t=1.076; p=0.284) and time to defecation (t=2.237; p=0.064). This study showed that foot reflexology applied in the early postoperative period to patients undergoing appendectomy surgery had a positive effect on the patients' pain levels and sleep quality, but had no effect on bowel functions. Analyzing Academic Achievement and Overconfidence in Database Management Systems Course: A Case Study of Computer and Instructional Technology Education Students
Decision-making tends to be more accurate and of higher quality when there's a sensible harmony between self-confidence and actual capabilities. Overconfidence makes it difficult to set realistic goals in academic settings and increases the likelihood of facing failure. In this study, the academic achievements and overconfidence of students enrolled in the Database Management Systems course were examined. The research also aimed to determine whether there is a difference between midterm and final exams in terms of these variables. The participants were comprised of students enrolled in the Computer and Instructional Technologies Education department of a state university throughout the 2021-2022 academic year. The results indicated that approximately two-thirds of the students did not achieve satisfactory academic scores. Students struggled to accurately assess their exam performances, and a significant number of them overestimated their positions in both the midterm and final exams. Furthermore, there was no significant change between the midterm and final exams for any of the three factors
The Effect of STEM Applications on Mathematics Achievement: A Meta-Analysis Study
This study aims to determine the effect of STEM applications on mathematics achievement using the meta-analysis method. For this purpose, postgraduate theses in Türkiye were analysed. As a result of the review, 25 postgraduate theses related to the topic were initially identified, and data from 14 theses that met the inclusion criteria were included in the meta-analysis. Data analyses were carried out using the Comprehensive Meta-Analysis statistical program. While applying the meta-analysis method, homogeneity test was performed by calculating the effect sizes of the studies. Since the studies showed a heterogeneous structure as a result of the homogeneity test, the random effects model was used to calculate the overall effect size. Hedges's g coefficient was used to determine the effect sizes. Funnel plot and Rosenthal's safe N statistic were used to examine publication bias. The classification of Thalheimer and Cook (2002) was used to interpret the effect sizes obtained as a result of the analyses. In the study, it was determined that the effect sizes of all studies included in the meta-analysis were positive. Only two of the studies had a small effect, while the other studies showed at least a medium effect. The overall effect size value for mathematics achievement was found to be 0.901. Based on these results, it can be said that STEM activities have a positive and large effect on increasing mathematics achievement
The relationship between recreational sport well-being and academic belonging among university students
The aim of this study is to examine university students' recreational sports well-being and academic belonging in relation to various demographic variables and to test the relationships among these variables. The population of the research group consists of 874 students enrolled in the Faculty of Sport Sciences at Bartın University during the 2023-2024 academic semester. A sample of 509 students was selected using the convenience sampling method. In the study, the relational survey model, one of the general survey models, was employed. In the study, the "Recreational Sports Well-Being Scale (RSWBS)" and the "Academic Belonging Scale (ABS)" were used as data collection instruments. In the analysis of the data, an independent samples t-test, one-way ANOVA, and Pearson correlation analyses were utilized. The findings of the study indicate significant relationships in certain sub-dimensions of the RSWBS and ABS based on the correlation test results for the variables of age and weekly participation in recreational activities. According to the t-test results for the gender variable of the participants, a significant difference was found in the "Class Participation, Library and Resource Access" sub-dimension of the ABS, while no significant differences were detected in the other sub-dimensions of the ABS or in the RSWBS and its sub-dimensions. According to the ANOVA test results for the participants' department, leisure-time competence, and perceived family income variables, no significant differences were found among the sub-dimensions of the RSWBS and ABS. According to the correlation analysis conducted to determine the relationship between the RSWBS and ABS, all sub-dimensions were found to have low to moderate positive correlations with each other. In conclusion, it can be stated that participation in recreational sports activities enhances individuals' psychological, social, and physical well-being, thereby strengthening their sense of academic belonging. Engaging in structured recreational activities during their leisure time may contribute to university students establishing a stronger connection with the academic environment, identifying with institutional identity, and participating more actively in the educational process. Key Words: Academic belonging, recreational sport well-being, university students
The First Chalcone Derivatives of Valine-Based Spiro-Cyclotriphosphazenes: In Vitro Cytotoxic Properties, Molecular Docking and DNA Damage Mechanism Studies
Cancer treatment requires novel compounds with potent cytotoxic and genotoxic properties to effectively target cancer cells. In this study, new hybrid cyclotriphosphazene compounds were synthesized, characterized, and evaluated for their biological activity. Cytotoxicity against A2780 and Caco-2 cancer cell lines was assessed via the MTT assay, while genotoxic effects at 60-70% cell viability were examined using the Comet assay. Apoptotic cells were identified through TUNEL analyses, and reactive oxygen species levels were measured. Results showed that these compounds significantly reduced cell viability through DNA damage mechanisms. At high doses (50-100 mu M), BV, BVK1, BVK2, and BVK4 decreased A2780 cell viability by 30-65%, whereas VPA had a milder effect (15-25%). In Caco-2 cells, viability was reduced by 10-35%. The compounds exhibited varying cytotoxicity across different cancer cell lines, reflecting cancer cell heterogeneity. Significant DNA damage, including changes in tail length, tail density, and tail moment, was observed in A2780 cells, confirming cell death via DNA damage. Molecular docking analyses further supported the potential of cyclotriphosphazene compounds (BV and BVK2) as targeted cancer inhibitors. Molecular docking revealed BVK2's high selectivity for Bcl-2, mutant p53, and VEGFR2. BVK2 and BV demonstrate strong binding affinities with key cancer-related targets, indicating their potential as multi-targeted inhibitors that regulate apoptosis, cell cycle control, and angiogenesis, making them promising candidates for targeted cancer therapy.Scientific and Technical Research Council of Turkiye (TUBITAK) [122Z060]This study was supported by a grant from the Scientific and Technical Research Council of Turkiye (TUBITAK, Grant Number: 122Z060)
