The study of the method of fight against formation of polarizing layer at the process of ultrafiltration concentration of the skim milk
The description of experimental set and the method of processing of the results of the study of the method of fight against formation of polarizing layer in the process of ultrafiltration concentration of skim milk raw material were presented. The results of experimental studies relative to the sparging method application for the prevention of polarizing layer in the process of membrane processing of the skim milk and its influence on the capacity of ultrafiltration membranes were offered. The mathematical models were constructed on the base of regressive equations of factorial experiment using the method of the raw material sparging above the membrane surface for selection of technological parameters of the process of ultrafiltration concentration of the skim milk. The rational working parameters of the process of ultrafiltration concentration of the skim milk using the sparging of the skim milk by the gas bubbles in the direct closeness to the membrane surface were determined. Such working parameters are: pressure– 0,4...0,5 MPa, temperature of skim milk– 40...50 ºС, frequency of sparging of skim milk– 0,10...0,15 min-1, pressure of sparging must be 0,56...0,58 MPa
ARGINASE-BASED AMPEROMETRIC BIOSENSOR FOR MANGANESE IONS ANALYSIS
The development of simple cost-effective sensitive enzymatic methods for analysis of toxic metallic ions is an actual problem. Promising tools for elaboration of such methods are Mn2+-dependent enzymes. A novel manganese(II)-sensitive amperometric bi-enzyme biosensor based on of recombinant human arginase I (arginase) isolated from the gene-engineered strain of methylotrophic yeast Hansenula polymorpha and commercial urease is described. The biosensing layer with urease and apo-enzyme of arginase was placed onto a polyaniline-Nafion composite platinum electrode. The developed sensor revealed a high sensitivity to Mn2+-ions – 9200±20 A/(M∙m2)with the apparent Michaelis-Menten constant derived from Mn2+-ions calibration curve of 11.5±1.0 µM. A linear concentration range was observed from 1 µM to 6,5 µM MnCl2, a limit of detection being of 0.15 µM and a response time – 2.5 min. The proposed biosensor may be useful to monitor manganese compounds in laboratories of medicine, food industry and environmental control service
THE PROTEIN METABOLISM IN SMALL ANIMALS WITH CHRONIC RENAL FAILURE AND ITS CORRECTION
Chronic renal failure (CRF) is often diagnosed in cats and dogs of old age. CRF is a consequence of metabolic deviations because of diseases (for example, generalized atherosclerosis, hypertensive disease, diabetes and others) and incorrect feeding. CRF is also a result of the delayed diagnosis of diseases of the urinary system and/or inadequate treatment.
Aim of research. To study the biochemical parameters of protein metabolism that are important for determination of CRF stages and sub-stages in old small animals during correction of metabolic deviations.
Materials and methods. There were analyzed biochemical indices (creatinine concentration in the blood serum; the ratio of protein/creatinine (UP/C) in the urine) and measured arterial blood pressure in 5 cats and 5 dogs with chronic renal failure selected from patients of the clinic according to medical history, clinical examination and special analysis.
Results. It was analyzed CRF stages and sub-stages classification according to the International Renal Interest Society (IRIS). According to the date of creatinine concentration in the blood serum, CRF of the second stage was observed in 5 cats (140-250 μmol/L) and 3 dogs (125-180 μmol/L), and of the third stage - in 2 dogs (181-440 μmol/L). The CRF sub-stage as «without proteinuria» (UP/C < 0.2) was identified in 1 cat, «marginal proteinuria» (UP/C 0.2-0.4) – in 1 cat and «proteinuria» (UP/C > 0.4) - in 3 cats, and as «proteinuria» (UP/C > 0.5) - in 5 dogs.
Conclusions. An important step of the successful treatment of CRF is the definition of the stages and sub-stages according to IRIS based mostly on the measurements of biochemical indices in the blood and urine of sick animals ‒ indicators of creatininemia and proteinuria, a change of which was caused mainly by metabolic processes of proteins under influence of exogenous factors (the traditional treatment of CRF plus enalapril and diet nutrition)
THE STUDY OF SORPTION OF THE MILK IONIZED CALCIUM BY SODIUM ALGINATE
The possibility of regulation of defatted cow milk salt composition (fat content– 0,05 %) with the use of sodium alginate as a natural ion-exchanger was studied. Realization of the properties of sodium alginate as to the bounding of calcium ions allows receive the systems on the base of cow milk, stable in time and at the thermal processing. The studies established the influence of technological factors on the sorption of ionized calcium by the solution of complex-creator by sodium alginate. It was established, that the important factors that influence the process are the active acidity and conditions of the process, namely, the phased addition of sorbent that favors the equal speed of process and, as the result, the equal speed of sorption during the whole process. At the same time the study of influence of sorption area and temperature demonstrates that these factors are not determining ones in this process. It was established, that rationalization of parameters of ionized calcium sorption results in the raise of thermal stability of defatted cow milk and the systems on its base. Materials, given in the work, are the base for elaboration and introduction of technology of dessert production (ice-cream, creams, puddings, cocktails and so on), in which composition the colloid stability is provided at the joint use of milk and fruit-berry raw material that can be used in food industry. 
THE HOLSTEIN CATTLE BREEDING PARTICULARITIES IN RUSSIAN SMALL AND MEDIUM ENTERPRISES’ CONDITIONS
IT- information technologies play a crucial role in the animals breeding value of different breeding, production, morphological, physiological and other livestock groups determining on operation farms. All of these groups are genetic (30 % – in countries with highly developed dairy cattle breeding, 5–25 % in herds of Russia) and technological reserves of milk production. Particularly its exploitation allows to create financial flows that determine the milk production success and farm’s future prospects. The main objective of selection is economics, so livestock owner supports those milk production areas on his farm that give optimum profits. There are at the milk production a lot of such areas: feeding, keeping, excess or lack of staff, etc., Our research, based on experimental data (n=25000) and the literature review had showed that the main of them is the genotype of the animal, so as the low-productive cattle overfeeding doesn’t contribute more additional milk to obtain. The genotype influence is determined by technological, social, financial and other factors that allows to create a harmonious system of their interrelation (r=0,01–0,9). Our researches had allowed to find a model that combines seemingly incompatible factors, and in the individual breeding programs form for the qualitative and quantitative genetics successful application regularities with the dominance of the latter, on small and medium-sized milk production enterprises it to apply
THE STUDY OF FATTY AND ORGANIC ACIDS COMPOSITION IN QUINCE LEAVES AND FRUITS (CYDONIA OBLONGA MILL)
The composition of fatty and organic acids in quince leaves and fruits (Cydonia oblonga Mill.) of selection of M.M. Grishko National Botanical Garden of National Academy of Sciences of Ukraine sorts was determined by gas chromatography-mass spectrometry method. Six sorts of quince were examined: ‘Academichna’, ‘Kashchenka N18’, ‘Studenrka’, ‘Oranzheva’, ‘Maria’, ‘Darunok onuku’. The presence of 36 fatty acids and 32 organic acids in leaves and fruits of the quince were identified and determined their content. Therefore, main acids are oxalic, citric, palmitic and linolenic acids in quince leaves and malic, palmitic and linoleic acids in quince fruits. The highest content of all acids were identified in the quince leaves of sort ‘Maria’. Moreover, a quite high content of succinic acid in the quince leaves was detected
CYTOGENETIC EVALUATION OF THE DRINKING WATER TOXICITY
There was considered the use of biotesting for the assessment of the quality of drinking water from the different water supply sources (artesian, packaged and water-pipe one). The method consists in determination of the toxicants action on the specially selected organisms in the standard conditions with registration of changes at behavior, physiological, cellular and subcellular level using hematological indices of fish, frogs, rats and the lymphocyte cultures of the peripheral human blood. Physical-chemical methods determine only the presence and number of chemical elements in the tested water samples because of the very large number of possible combinations of chemical compounds in water solutions (more than 75 million combinations), including behavior of anthropogenic compounds and the natural vulnerability of the water ecosystems to the combined effects from its toxic influence.
As the optimal set of determination of the some structural and functional changes of cell genome as the result of the toxic influence of combination of the chemical compounds in the water solutions was offered the micronuclear test and leukocytic formula of the fish, frog and rat blood as biomarker. The reaction of fish, frog, rat test-organisms on the toxic irritation is presented in the change of qualitative content of the cells of peripheral blood. There were demonstrated the prospects of the use of hematological indices of the following test-organisms: Danio rerio fishes, Xenopus clawed frogs, Wistar rats and also the lymphocytes cultures of the human peripheral blood. The special attention was paid to the assessment of the risk for human health of the toxic substances in drinking water, genotoxicity and cytotoxicity that are revealed using hematological indices of animal cells. The universality of cellular organization opens the wide possibilities for toxicological studies using peripheral blood of the different groups of animals (fish, frogs, rats), human lymphocyte cultures and allow assume the following possibility of extrapolation of the received results on human organis
THE STUDY OF MAIN PHYSICAL-CHEMICAL PARAMETERS OF CHAENOMELES AND PRODUCTS OF ITS PROCESSING
In the article were considered the topical problems, connected with health worsening and human existence. The use of vegetable raw material as a source of biologically active components is prospective in production foodstuff. There were offered the rational methods of processing of chaenomeles for getting juice and puree. There were carried out an analysis of expedience of using chaenomeles and products of its processing for enrichment foodstuff. It was established, that maximal amount of phenol substances is contained in fruit peel and L-ascorbic acid - in flesh. It was determined, that apple acid prevails among organic ones, fructose - among sugars and phenol substances are mainly presented by procyanidins. Using chromotographic analysis in the products of chaenomeles processing were identified 48 names of aromatic compounds, alcohols, acids, ethers and unsaturated carbohydrates prevail among them. There were studied the physical-chemical parameters of fruit sauces and flour products with addition of chaenomeles processing products. The received results prove that the ready foodstuff that contain puree and juice of chaemomeles have the high organoleptic, physical-chemical parameters and also heightened biological value
THE STUDY OF PROPERTIES OF MINCES IN BOILED SAUSAGES WITH FUNCTIONAL FOOD COMPOSITION USE
The priority direction of innovative activity of meat-processing enterprises is a production of accessible goods of stable and high quality. The attainment of set aim is provided by introduction of diverse food additives (emulsifiers, stabilizers, stiffeners and so on) that replace the significant part of the main raw material for improvement of its functional-technological properties at creation of food systems.
The influence of elaborated functional food composition on functional-technological and structural-mechanical properties of minces in boiled sausages was studied. It was established, that its introduction into the content of meat minces improves the indices of water-binding ability by 3,5–5 % percents comparing with the control sample. Emulsifying ability in the studied samples increases on the average by 5 %, and emulsion steadiness by 22,5 %. It is conditioned by the ability of protein preparations and hydrocolloids, included in the composition together with meat proteins, to encapsulate the fat drops and to keep them during the whole technological process. Determination of the influence of elaborated functional mixture on structural-functional properties of minces demonstrated, that effective viscosity for the studied samples increased on the average by 48,5 %, and the index of extreme tension of the shift decreased on the average by 40,5 %.
The received results prove the perspective of replacement of the part of meat raw material (up to 30 %) by created composition and improvement of functional-technological and structural-mechanical properties of minces. It provides the stable high quality of ready goods and allows improve the effectiveness of production
STUDY OF RHEOLOGY OF YEAST DOUGH WITH PROTEIN-CARBOHYDRATE ADDITIVE
The aim of the studies was the establishing of rational concentration of the dry protein-carbohydrate half-finished product (DPCHFP) in technology of yeast dough, received in accelerated way. The dough structural-mechanical properties were studied using Brabendar’s farinograph and extensograph, the firm-elastic and rheological dough properties at DPCHFP presence were also studied. It was established, that DPCHFP usage in technological process of yeast dough production gives a possibility to correct the flour and purposefully influence its rheological properties. The rational DPCHFP concentration at this stage of research is 15 % for the flour mass
