    Extracellular vesicles released from Marek’s disease virus-transformed T-cells impact immune cell proliferation

    International audienceMarek’s disease virus (MDV) is an alphaherpesvirus responsible for the development of T-cell lymphoma in chickens. Despite the identification of several pro-oncogenic viral molecules encoded by MDV, the processes leading to tumourigenesis remain poorly understood. Extracellular vesicles (EVs) are important mediators of intercellular communication, carrying bioactive molecules that can elicit profound physiological changes in recipient cells. Tumour cells can release significant amounts of EVs, which influence tumour development and growth, metastatic processes and resistance to cancer therapies. These EVs favour cancer cells to evade the immune response, particularly by establishing an immunosuppressive microenvironment. Here, we investigated whether EVs produced by MDV-transformed T lymphocytes affect the proliferation of avian immune cells, a determining feature in neoplastic processes. EVs were purified from an MDV-transformed cell line cultured in vitro . Using a proteomic approach, we confirmed the presence of specific markers and identified a panel of cellular proteins enriched in these EVs. Notably, no viral proteins were detected in the purified EVs. We also demonstrated that EVs are rapidly internalized by recipient chicken cells. Moreover, these EVs can induce a decrease in primary chicken B-cell proliferation, while promoting primary chicken T-cell proliferation. Our findings suggest that EVs released by MDV-transformed cells may contribute to immunosuppression and potentially facilitate lymphoma progression by enhancing T-cell proliferation

    International audienceMetabolomics is an approach to systems biology which studies the metabolic profiles of biological fluids, cells, tissues or organisms as a function of pathophysiological conditions or in response to different stimuli or treatments. Despite its development in research, its use in laboratory medicine remains limited and the contours of clinical metabolomics are not clearly defined. In June 2024, the French Society of Clinical Biology (SFBC)'s'Clinical Metabolomics in Laboratory Medicine' working group conducted an online survey to establish the current status of clinical metabolomics players in France and to better define the issues and expectations involved. Of the 37 specialists who responded, 75% were affiliated to laboratory medicine, while the others were only affiliated to a research unit. The results revealed a wide range of analytical approaches, with mass spectrometry being the most widespread, with targeted and untargeted approaches depending on the objectives of the laboratories. The most frequently mentioned clinical areas of application were hereditary metabolic diseases (45%), metabolic diseases (45%), nutrition (35%), oncology (32%) and neurology (26%). The definitions of clinical metabolomics varied widely depending on the context in which the participants practiced. In this publication, the SFBC Working Group aims to define the scope and objectives of clinical metabolomics in laboratory medicine, relying in particular on a glossary designed to harmonize dedicated terminology

    International audienceWith the acceleration of climate change, the ecological transition is becominga necessity. In this context, forest plantations are particularly encouraged.However, they raise questions from a landscape and environmental viewpoint,especially when they are planted with fast-growing species such as poplar andmaritime pine. This article focuses on the plantations formed by these species.Research was carried out in the Centre-Val de Loire region. The aim was toinvestigate the existence of recreational activities, such as hiking, in or nearplantations, and to analyze the representations of potential users. Research was based on a spatial analysis and, above all, interviews conducted in situ. It shows that hiking trails run alongside some of these plantations, without these guiding their routes. The hikers interviewed reveal a variety of views about plantations, ranging from neutrality to opposition or defense.À l’heure où le changement climatique s’accélère, où la transition écologique s’impose comme une nécessité, les plantations forestières sont particulièrement encouragées. Pourtant, elles questionnent sur les plans paysagers et environnementaux, surtout lorsqu’elles sont formées d’espèces à croissance rapide, comme le peuplier et le pin maritime, sur lesquels l’article se centre. Les recherches, conduites en région Centre-Val de Loire, visaient à questionner l’existence d’activités récréatives, telles que les randonnées, dans ou près des plantations et à analyser les représentations que les éventuels usagers ont de ces dernières. Elles se fondent sur une analyse spatiale et surtout des entretiens menés in situ. Elles montrent que des sentiers de randonnée longent bien des plantations, sans pour autant que ces dernières guident les tracés. Les randonneurs interrogés dévoilent des regards variés vis-à-vis des plantations, entre neutralité, opposition ou défense

    « Appartenir » : tension de la forme héritée chez Jean-Jacques Goldman

    International audienceCette communication propose d’analyser la manière dont la production de Jean-Jacques Goldman témoigne d’un travail important sur la forme héritée, en particulier l’omniprésente structure couplet/refrain. Si cette dernière constitue son cadre dominant, Goldman recourt également à des organisations strophiques, ou ponctuellement rhapsodiques, ainsi qu’à des schémas plus rares dans la chanson française, tels que les formes AAB ou AAAB, qu’il mobilise fréquemment, notamment à des fins narratives : la chanson se déploie souvent comme un parcours, non comme une simple juxtaposition strophique, et emploie des zones transitoires (ponts, solos) qui viennent enrichir la structure attendue ou préétablie. En étudiant un corpus de 116 chansons correspondant aux chansons qu’il a composées et interprétées en studio (dans sa carrière post-Taï Phong, en solo ou avec Carole Fredericks et Michael Jones, c’est-à-dire entre 1981 et 2001), l’objectif est de dégager les stratégies mises en œuvre par Goldman pour structurer ses chansons, depuis le parcours tonal et les progressions harmoniques jusqu’aux enjeux timbraux et instrumentaux. Il s’agira donc enfin de déterminer le degré d’appartenance de la production de Goldman à une forme pop héritée. La focalisation se fera, dans le temps imparti de la communication, sur certaines chansons "extrêmes" des albums : chansons d'ouverture ou de fermeture
