    In vivo AGO-APP identifies a module of microRNAs cooperatively preserving neural progenitors

    International audienceMicroRNAs are essential regulators of gene expression. Their function is particularly important during neurogenesis, when the production of large numbers of neurons from a limited number of neural stem cells depends on the precise control of determination, proliferation and differentiation. However, microRNAs can target many mRNAs and vice-versa, raising the question of how specificity is achieved to elicit a precise regulatory response. Here we introduce in vivo AGO-APP, a novel approach to purify Argonaute-bound, and therefore active microRNAs from specific cell types. Using AGO-APP in the larval Drosophila central nervous system, we identify a module of microRNAs predicted to redundantly target all iconic genes known to control the transition from neuroblasts to neurons. While microRNA overexpression generally validated predictions, knockdown of individual microRNAs did not induce detectable phenotypes. In contrast, neuroblasts were induced to differentiate precociously when several microRNAs were knocked down simultaneously. Our data supports the concept that at physiological expression levels, the cooperative action of miRNAs allows efficient targeting of entire gene networks

    Decompensated esophoria is a binocular vision anomaly characterized by a latent tendency of the eyes to converge beyond the patient’s fusional compensation abilities. This condition can lead to various symptoms such as intermittent diplopia, visual fatigue, and headaches, thereby affecting patients’ quality of life. This thesis explores the different management modalities for decompensated esophoria through a literature review and a clinical study conducted on 30 patients. The analysis of preoperative results highlights the clinical characteristics of patients and the factors influencing decompensation. Several therapeutic approaches are examined, including the use of prisms integrated into optical correction, orthoptic rehabilitation, and surgery. The effectiveness of the prismatic adaptation test is also studied to determine whether it optimizes preoperative evaluation and thus improves surgical planning.The results show that surgery, involving medial rectus muscle recession, Is an effective option, providing stable and long-lasting correction of the deviation as well as an improvement in functional symptoms. However, the study emphasizes the importance of an individualized approach and thorough assessment to maximize treatment effectiveness and reduce the risk of recurrence. This research contributes to a better understanding of decompensated esophoria and suggests improvements in its management within orthoptics.L'ésophorie décompensée est une anomalie de la vision binoculaire caractérisée par une tendance latente des yeux à converger, dépassant les capacités de compensation fusionnelle du patient. Cette condition peut engendrer divers symptômes tels que la diplopie intermittente, la fatigue visuelle et des céphalées, altérant ainsi la qualité de vie des patients. Ce mémoire explore les différentes modalités de prise en charge de l'ésophorie décompensée à travers une revue de la littérature et une étude clinique menée sur 30 patients. L'analyse des résultats préopératoires met en évidence les caractéristiques cliniques des patients et les facteurs influençant la décompensation. Plusieurs approches thérapeutiques sont examinées, comme le port de prismes intégrés dans la correction optique, la rééducation orthoptique ainsi que la chirurgie. L'efficacité du test d'adaptation prismatique est également étudiée dans le but de voir s'il permet d'optimiser l'évaluation préopératoire et donc la planification chirurgicale. Les résultats montrent que la chirurgie, avec le recul des muscles droits médiaux, est une option efficace, permettant une correction stable et durable de la déviation ainsi qu'une amélioration des signes fonctionnels. Toutefois, l'étude souligne l'importance d'une approche individualisée et d'une évaluation rigoureuse pour maximiser l'efficacité de ce traitement pour réduire les risques de récidive. Cette recherche contribue ainsi à une meilleure compréhension de l'ésophorie décompensée et propose des pistes d'amélioration pour sa prise en charge en orthoptie

    Réémergence de la diphtérie en Guinée : une analyse des perspectives de vaccination et de contrôle de la maladie

    International audienceAbstract Background This study presented the key characteristics of patients who tested positive for diphtheria during the outbreak in the Republic of Guinea in 2023 and assessed the influence of some risk factors on disease development. Methods The clinical diagnosis of diphtheria was confirmed by detecting diphtheria toxin genes in nasopharyngeal samples collected from suspected patients via two reverse transcription-quantitative polymerase chain reaction tests. Bivariate analyses with the chi-square test and the Fisher’s exact test were conducted to explore possible associations between diphtheria positivity and various sociodemographic, clinical, and exposure factors. Results In total, 444 samples obtained from suspected cases were analyzed. In 90 (20.3%) cases, the condition was confirmed using quantitative polymerase chain reaction, with an overall fatality rate of 8.9% (n = 8). On average, deaths occurred 2 days after admission, with 6 (75.0%) of 8 (six girls and two boys) deaths recorded within the first 3 months after the epidemic onset. The clinical characteristics included sore throat (91%), fever (90%), whitish throat membrane (83 %), throat redness (81%), and dyspnea (28%). The risk factors were age <15 years, no prior vaccination, and contact with a patient with diphtheria. A whitish throat membrane and dyspnea were significantly associated with diphtheria positivity. Conclusions This study emphasized that diphtheria remains a major and potentially fatal disease, despite vaccination and early symptom recognition. The identification of characteristic signs—particularly a whitish throat membrane and dyspnea—is important for reducing disease severity and mortality.Résumé: Arrière-planCette étude a présenté les principales caractéristiques des patients testés positifs à la diphtérie lors de l’épidémie en République de Guinée en 2023 et a évalué l’influence de certains facteurs de risque sur le développement de la maladie.Matériel et méthodes: Le diagnostic clinique de diphtérie a été confirmé par la détection des gènes de la toxine diphtérique dans des échantillons nasopharyngés prélevés chez des patients suspects via deux tests quantitatifs de réaction en chaîne par polymérase par transcription inverse. Des analyses bivariées avec le test du chi carré et le test exact de Fisher ont été effectuées pour explorer les associations possibles entre la positivité de la diphtérie et divers facteurs sociodémographiques, cliniques et d’exposition.Résultats:Au total, 444 échantillons prélevés sur des cas suspects ont été analysés. Dans 90 cas (20,3 %), l’affection a été confirmée par réaction en chaîne par polymérase quantitative, avec un taux de létalité global de 8,9 % (n = 8). En moyenne, les décès sont survenus 2 jours après l’admission, avec 6 (75,0 %) des 8 décès (six filles et deux garçons) enregistrés dans les 3 premiers mois suivant le début de l’épidémie. Les caractéristiques cliniques comprenaient des maux de gorge (91 %), de la fièvre (90 %), une membrane de la gorge blanchâtre (83 %), une rougeur de la gorge (81 %) et une dyspnée (28 %). Les facteurs de risque étaient l’âge <15 ans, l’absence de vaccination préalable et le contact avec un patient atteint de diphtérie. Une membrane de la gorge blanchâtre et une dyspnée étaient significativement associées à la positivité de la diphtérie.Conclusions:Cette étude a souligné que la diphtérie reste une maladie majeure et potentiellement mortelle, malgré la vaccination et la reconnaissance précoce des symptômes. L’identification des signes caractéristiques, en particulier une membrane de la gorge blanchâtre et une dyspnée, est importante pour réduire la gravité de la maladie et la mortalité

    Le dessin ostéopathique des atteintes corporelles : une clinique émersive

    International audienceObjective. – Osteopathy is the most popular alternative healthcare practice in France. To update and improve this practice, which is already part of patient care pathways, it is necessary to explore and understand bodily damage via a central idea in osteopathy: somatic dysfunction.Methods. – First-person data were collected using a consciousness drawing approach followed by explanatory interviews. During their first two years of osteopathic studies, students engage in a reflective practice around somatic dysfunction. This study utilized a qualitative method with a thematic analysis of the above interviews and drawings.Results. – The thematic analysis identified the existence of four axes: the reading of the bodily injury, the materialization of the bodily injury, the transition from local bodily injury to global bodily injury, and the impact of the bodily injury on the osteopath in the context of emerging intercorporeality. There were three categories of drawings: (drawings) of consciousness, drawings (of) consciousness, and drawings of (consciousness).Discussion. – Exploring knowledge about somatic dysfunction has highlighted that mixed epistemology is needed to propose a model of osteopathic clinical practice that incorporates elements of a bio- psychosocial approach. Touch seems to allow the identification of intercorporeality in the experiences of the therapist and patient, thanks to an emersive clinical experience of the body.Conclusion. – This study explores the knowledge and theories of osteopathic students in early training. Their conception of somatic dysfunction was expressed through several drawings of consciousness and through a narrative arising from bodily reflexivity. This work seems conducive to the better integration of psychosocialcriteria and a holistic understanding of the body, with a view to enhancing the contribution of osteopathic touch as care within a patient-centered approach.L’ostéopathie est la pratique de soins non conventionnelle la plus sollicitée par les Franc ¸ ais. Afin d’actualiser et faire progresser cette pratique déjà présente dans le parcours de soin des patients, ilsemble nécessaire de mieux explorer et comprendre l’atteinte corporelle à travers une notion centrale de la discipline ostéopathique : la dysfonction somatique.Méthode. – Une collecte de données en première personne est réalisée à partir de dessins de conscience puis d’entretiens d’explicitation. Au cours de leurs deux premières années en cur-sus ostéopathique, des étudiants sont engagés dans une pratique réflexive vis-à-vis de la dysfonction somatique. Cette étude repose sur une méthode qualitative avec une analyse thématique des entretiens et des dessins.Résultats. – L’analyse thématique permet d’identifier quatre axes : la lecture de l’atteinte corporelle, la matérialisation de l’atteinte corporelle, le passage d’une atteinte corporelle locale à une atteinteglobale et l’impact de cette atteinte corporelle sur l’ostéopathe dans une intercorporéité émergente. Les dessins révèlent trois catégo- ries : (dessin) de conscience, dessin (de) conscience et dessin de (conscience).Discussion. – L’exploration des connaissances de la dysfonction somatique permet de mettre en lumière la nécessité de proposer une épistémologie mixte pour proposer une modélisation de la pratique clinique ostéopathique intégrant les éléments d’une approche biopsychosociale. Le toucher semble permettre l’identification d’une intercorporéité entre le vécu du thérapeute et du patient, ceci grâce à une clinique émersive du corps.Conclusion. – Cette étude explore les connaissances et les théories du corps d’étudiants en début de cursus ostéopathique. Leur conception de la dysfonction somatique s’exprime au travers de plusieurs dessins de conscience ainsi que d’une narration dans une démarche de réflexivité corporelle. Ceci semble propice à une meilleure intégration de critères psychosociaux et une connaissance du corps holiste afin de valoriser la place de l’expérience vécue par le soin tactile ostéopathique dans une approche centrée sur le patient

    Nonreciprocal Breathing Solitons

    International audienceBreathing solitons consist of a fast beating wave within a compact envelope of stable shape and velocity. They can propagate and carry information and energy in a variety of contexts such as plasmas, optical fibers, and cold atoms, but propagating breathers have remained elusive when energy conservation is broken. Here, we report on the observation of breathing, unidirectional, arbitrarily long-lived solitons in nonreciprocal, nonconservative active metamaterials. Combining precision desktop experiments, numerical simulations, and perturbation theory on generalizations of the sine-Gordon and nonlinear Schrödinger equations, we demonstrate that unidirectional breathers generically emerge in weakly nonlinear nonreciprocal materials, and that their dynamics are governed by an unstable fixed point. Crucially, breathing solitons can persist for arbitrarily long times provided that (i) this fixed point displays a bifurcation when a delicate balance between energy injection and dissipation is struck and (ii) the initial conditions allow the dynamics to reach this bifurcation point. Importantly, discrete effects tend to stabilize these nonreciprocal breathers over a wider range of initial conditions. Our work establishes nonreciprocity as a promising avenue to generate stable nonlinear unidirectional waves and could be generalized beyond metamaterials to optics, soft matter, and superconducting circuits

    Impact of swift heavy ions irradiation on the microstructural and electrochemical properties of vanadium nitride thin films for micro-supercapacitors

    International audienceThe role of defects in pseudocapacitive electrode materials has been poorly investigated due to the poor control of their introduction in a pure phase material. Chemical defects are difficult to monitor although it seems they have an obvious influence on the electrochemical properties of the pristine material. The controlled introduction of defects should provide a more rational approach to understand their role in charge storage mechanisms. This study investigates the impact of swift heavy ions irradiation on the electrochemical properties of pseudocapacitive vanadium nitride (VN) thin films, a promising material for micro-supercapacitor electrodes. VN films were irradiated with 129 Xe 19+ ions at an energy of 70.95 MeV and a fluences up to 10 14 ions⋅cm -2 . The effects of irradiation were compared to those observed in a previous study using lower-energy As + ions (20-150 keV) [1]. Scanning electron microscopy (SEM), X-ray diffraction (XRD) and X-ray photoelectrons spectroscopy (XPS) analyses revealed significant structural changes in the irradiated films, including densification, amorphization and oxidation of VN film surface. The electrochemical performance of the irradiated films was evaluated using cyclic voltammetry. While capacitance was reduced at low scan rates, a notable improvement was observed at high scan rates, which is attributed to the increase in electrical conductivity resulting from irradiation. This study deepens our understanding of the effects of high-energy ion irradiation on electrode materials and highlights the potential of this technique as a tool for guiding the development of more efficient energy storage devices
