Investigating Broadband Variability Of The Tev Blazar 1es 1959+650
We summarize broadband observations of the TeV-emitting blazar 1ES 1959+650, including optical R-band observations by the robotic telescopes Super-LOTIS and iTelescope, UV observations by Swift Ultraviolet and Optical Telescope, X-ray observations by the Swift X-ray Telescope, high-energy gamma-ray observations with the Fermi Large Area Telescope, and very-high-energy (VHE) gamma-ray observations by VERITAS above 315 GeV, all taken between 2012 April 17 and 2012 June 1 (MJD 56034 and 56079). The contemporaneous variability of the broadband spectral energy distribution is explored in the context of a simple synchrotron self Compton (SSC) model. In the SSC emission scenario, we find that the parameters required to represent the high state are significantly different than those in the low state. Motivated by possible evidence of gas in the vicinity of the blazar, we also investigate a reflected emission model to describe the observed variability pattern. This model assumes that the non-thermal emission from the jet is reflected by a nearby cloud of gas, allowing the reflected emission to re-enter the blob and produce an elevated gamma-ray state with no simultaneous elevated synchrotron flux. The model applied here, although not required to explain the observed variability pattern, represents one possible scenario which can describe the observations. As applied to an elevated VHE state of 66% of the Crab Nebula flux, observed on a single night during the observation period, the reflected emission scenario does not support a purely leptonic non-thermal emission mechanism. The reflected emission model does, however, predict a reflected photon field with sufficient energy to enable elevated gamma-ray emission via pion production with protons of energies between 10 and 100 TeV
Combined metagenomic and phenomic approaches identify a novel salt tolerance gene from the human gut microbiome
In the current study, a number of salt-tolerant clones previously isolated from a human gut metagenomic library were screened using Phenotype MicroArray (PM) technology to assess their functional capacity. PM\u27s can be used to study gene function, pathogenicity, metabolic capacity and identify drug targets using a series of specialized microtitre plate assays, where each well of the microtitre plate contains a different set of conditions and tests a different phenotype. Cellular respiration is monitored colorimetrically by the reduction of a tetrazolium dye. One clone, SMG 9, was found to be positive for utilization/transport of L-carnitine (a well-characterized osmoprotectant) in the presence of 6% w/v sodium chloride (NaCl). Subsequent experiments revealed a significant growth advantage in minimal media containing NaCl and L-carnitine. Fosmid sequencing revealed putative candidate genes responsible for the phenotype. Subsequent cloning of two genes did not replicate the L-carnitine-associated phenotype, although one of the genes, a σ54-dependent transcriptional regulator, did confer salt tolerance to Escherichia coli when expressed in isolation. The original clone, SMG 9, was subsequently found to have lost the original observed phenotype upon further investigation. Nevertheless, this study demonstrates the usefulness of a phenomic approach to assign a functional role to metagenome-derived clones
Learning in Action: Implementing a Facilitated Learning Programme for Tourism Micro-Firms
This paper presents findings from research associated with a tourism micro-firm facilitated learning programme, carried out over a four-year period. Considering traditional educational interventions have had limited impact on micro-firm activities; the researchers propose an alternative approach to meeting the learning needs of micro-firms which encompass local and self-developed knowledge and the successful completion of learning cycles. Using a behavioural lens, the researchers’ consider the experiential learning impact of facilitated learning programmes on observed micro-firm owner-managers. Adopting an action learning (AL) approach, the researchers explore the design, development, implementation and impact of this programme and contemplate the observed micro-firms’ focus on and approach to learning in their business setting. Having analysed the micro-firms’ learning-enhanced development strategies, the researchers propose a cohort-specific AL framework based on the findings and highlight the research implications prior to setting out avenues of further research
An Exploration of the Role of Food Tourism in Sustaining Cultural Authenticity in Ireland
Authenticity is often a motivating factor for tourists. In many situations what occurs, however, is staged authenticity. Food is one of the few entities left that indicates true cultural authenticity. Food can bridge the gap between tourists and their understanding of and involvement in a place. Native cultures that are struggling to preserve their heritage and ways of life from globalising forces can use food as a defensive mechanism. This paper is part of a wider postgraduate study on food tourism and culture so will focus on the research completed to date, mainly the literature reviewed as well as initial primary research findings. The town of Kinsale, Co. Cork was the focus of the pilot case study, within which interviews were conducted with those involved in the food tourism sector including; restaurateurs, farmers’ market stall-holders, food trail guides and food festival organisers. Preliminary results indicate that food has played a key role in forming Kinsale’s identity and continues to do so, allowing traditions and culture to be maintained and rejuvenated
Niche Markets in Irish Tourism: The Case of North American Coach Tourists
In 2010, 115,000 coach tourists came from North America to Ireland, averaging a length of stay of 9 nights. This paper applies factor analysis to a survey of North American coach tourists to Ireland (with 486 responses). It identifies the most significant factors that have common characteristics amongst the tourists. Factor analysis, although popular in tourism research, has never been applied to an inbound tourist market to Ireland, so this research fills an important gap in Irish segmentation research. The factor analysis of the survey identifies a number of important characteristics of this market. For example, the North American coach tourists are predominately under 65 years, are drawn to historical destinations, and active, with a strong preference for cycling and water sports. The findings in this paper suggest tourism policies can now be directed more precisely to cater for a specific type of coach tourist visiting Ireland rather than supplying a generic tourist product. Factor analysis shows that coach tour packages should be targeting a particular kind of tourist, not simply the generic tourist to Ireland
Transcriptome analysis of Listeria monocytogenes exposed to biocide stress reveals a multi-system response involving cell wall synthesis, sugar uptake, and motility
Listeria monocytogenes is a virulent food-borne pathogen most often associated with the consumption of “ready-to-eat” foods. The organism is a common contaminant of food processing plants where it may persist for extended periods of time. A commonly used approach for the control of Listeria monocytogenes in the processing environment is the application of biocides such as quaternary ammonium compounds. In this study, the transcriptomic response of a persistent strain of L. monocytogenes (strain 6179) on exposure to a sub-lethal concentration of the quaternary ammonium compound benzethonium chloride (BZT) was assessed. Using RNA-Seq, gene expression levels were quantified by sequencing the transcriptome of L. monocytogenes 6179 in the presence (4 ppm) and absence of BZT, and mapping each data set to the sequenced genome of strain 6179. Hundreds of differentially expressed genes were identified, and subsequent analysis suggested that many biological processes such as peptidoglycan biosynthesis, bacterial chemotaxis and motility, and carbohydrate uptake, were involved in the response of L. monocyotogenes to the presence of BZT. The information generated in this study further contributes to our understanding of the response of bacteria to environmental stress. In addition, this study demonstrates the importance of using the bacterium\u27s own genome as a reference when analysing RNA-Seq data
The implications of regional and national demographic projections for future GMS costs in Ireland through to 2026
As the health services in Ireland have become more resource-constrained, pressure has increased to reduce public spending on community drug schemes such as General Medical Services (GMS) drug prescribing and to understand current and future trends in prescribing. The GMS scheme covers approximately 37% of the Irish population in 2011 and entitles them, inter alia, to free prescription drugs and appliances. This paper projects the effects of future changes in population, coverage, claims rates and average claims cost on GMS costs in Ireland
Profiling Tourism Entrepreneurs and their Enterprises along the Wild Atlantic Way
This paper discusses the profile of tourism entrepreneurs and their enterprises along the WAW (Wild Atlantic Way). Ireland had witnessed less numbers of some holidaymakers in particular the British market visiting over the past five years, resulting in a significant loss of market share from GB. This research undertook a proactive approach to investigating regional qualitative stakeholders profiles while identifying factors that impact on the management of tourism Small and Medium sized Enterprises (SMEs) to develop the International market. The research provides the industry with a snapshot of a current typology of successful tourism entrepreneurs. Furthermore, the research develops a profile of their tourism enterprises and the factors that impact on the management of these businesses in order to identify what contributes to their success. Many Irish tourism SMEs are developing an innovative approach in order to grow their tourist market. This may provide valuable insight and guidance to the tourism enterprises, local and regional authorities. The communication of these findings may facilitate multi-sector intercollaboration regarding best practice solutions with the evolving tourism industry
Antimycobacterial and Photosynthetic Electron Transport Inhibiting Activity of Ring-Substituted 4-Arylamino-7-Chloroquinolinium Chlorides
In this study, a series of twenty-five ring-substituted 4-arylamino-7-chloroquinolinium chlorides were prepared and characterized. The compounds were tested for their activity related to inhibition of photosynthetic electron transport (PET) in spinach (Spinacia oleracea L.) chloroplasts and also primary in vitro screening of the synthesized compounds was performed against mycobacterial species. 4-[(2-Bromophenyl)amino]-7-chloroquinolinium chloride showed high biological activity against M. marinum, M. kansasii, M. smegmatis and 7-chloro-4-[(2-methylphenyl)amino]quinolinium chloride demonstrated noteworthy biological activity against M. smegmatis and M. avium subsp. paratuberculosis. The most effective compounds demonstrated quite low toxicity (LD50 \u3e 20 μmol/L) against the human monocytic leukemia THP-1 cell line within preliminary in vitro cytotoxicity screening. The tested compounds were found to inhibit PET in photosystem II. The PET-inhibiting activity expressed by IC50 value of the most active compound 7-chloro-4-[(3-trifluoromethylphenyl)amino]quinolinium chloride was 27 μmol/L and PET-inhibiting activity of ortho-substituted compounds was significantly lower than this of meta- and para-substituted ones. The structure-activity relationships are discussed for all compound
