    Aislamiento e identificación del virus de la lengua azul por primera vez en Cuba

    Bluetongue (BT) is a viral disease that affects domestic and wild ruminants. The etiological agent of this disease is bluetongue virus (BTV) which is mostly transmitted by blood-borne vectors of the Culicoides genus and has a global distribution. Cuba is located in a zone of endemic circulation of the virus. Although the presence of bovine reactors has been reported to the World Organization for Animal Health (WOAH) since 2007, there is no evidence of clinical manifestations of the disease and the virus has not been isolated. This work aimed to isolate and identify bluetongue virus from the blood of a naturally infected calf from Mayabeque province in 2018. The virus was isolated in chicken embryonated eggs and in cell culture from the blood of the calf at 21 days of age, which tested positive for the presence of virus by a nested reverse transcription polymerase chain reaction assay (nRT-PCR). The inoculated embryos showed embryonic mortality, decreased size, reddish coloration and/or edema as indicative of viral multiplication. In cell culture, rounded and refractile cell foci were observed. Virus identification was performed by nRT-PCR assays, a commercial real-time RT-PCR kit (rRT-PCR) and rRT-PCR based on SYBR Green-I for specific detection of BTV.La lengua azul (LA) es una enfermedad viral que afecta a los rumiantes domésticos y salvajes. El agente etiológico de esta enfermedad es el virus de la lengua azul (VLA) que se transmite principalmente por vectores hematófagos del género Culicoides y tiene una distribución global. Cuba se encuentra en una zona de circulación endémica del virus y a pesar que desde el año 2007 se reporta a la Organización Mundial de la Sanidad Animal (OMSA) la presencia de bovinos seroreactores, no existen evidencias de manifestaciones clínicas de la enfermedad ni se ha aislado el virus. El objetivo del presente trabajo fue aislar e identificar el virus de la lengua azul a partir de la sangre de un ternero naturalmente infectado de la provincia Mayabeque en 2018. El virus fue aislado en huevos embrionados de pollo y en cultivo celular a partir de la sangre del ternero a los 21 días de edad que resultó positiva a la presencia de virus por un ensayo de reverso transcripción acoplada a la reacción en cadena de la polimerasa anidada (nRT-PCR). Los embriones inoculados mostraron mortalidad embrionaria, disminución de la talla, coloración rojiza y/o edema como indicativo de multiplicación viral. En el cultivo celular se observaron focos de células redondeadas y refráctiles. La identificación del virus se realizó por los ensayos de nRT-PCR, un Kit comercial de RT-PCR en tiempo real (rRT-PCR) y rRT-PCR basado en SYBR Green-I para la detección especifica del VLA

    Factores de riesgo de mastitis asociada a Mollicutes en rebaños lecheros de la provincia Zamora-Chinchipe, Ecuador

    Bovine mastitis is a sanitary and economic challenge for animal production, not only because it affects animal health and welfare, but also because of the threat it represents to public health, as well as the millions of losses caused by treatment costs and the milk discarded, indispensable food to the expanding population globally. Mycoplasma spp. were previously identified in tank milk and individual cows in Zamora-Chinchipe province. To analyze the risk factors associated with this problem, semi-structured questionnaires were applied to the herds previously tested. The variables with p-valor<0,2 in regression analysis were selected and included in the multivariate model, finally handle adjusted by backward procedure elimination variables with no significance (p<0,05). The risk factors for mollicutes in herds were the higher quantity number of zoothecnic categories (RP=1,691, IC95%: 1,293 – 2,210), the presence of Holstein race (RP=15,89; 1,44 – 175,45), no drinkable water use (RP=4,37; 1,306 – 14,620) and animal purchase into the province (RP=4,28; 1,416 – 12,939). Meanwhile, hand washing by the milker between cows or between groups showed a 57% and 67.6% lower prevalence, respectively, compared to herds where this practice is not followed. Additionally, drying the udders with paper before milking (with a 69.6% lower prevalence) is also a protective factor against Mollicutes in individual cows.La mastitis bovina es un desafío sanitario y económico para la producción animal, no solo porque afecta la salud y bienestar de los animales, sino también por la amenaza que representa para la salud pública, sin descontar las pérdidas millonarias que ocasionan los costos de tratamiento y el descarte de leche, un alimento por demás imprescindible para una población en plena expansión a nivel global. En Zamora-Chinchipe se demostró previamente Mollicutes en leche de tanque y vacas individuales. Para analizar los factores de riesgo asociados a esta problemática se aplicaron cuestionarios semi-estructurados a los rebaños previamente investigados. Las variables con p-valor<0,2 en el análisis univariado se seleccionaron e incluyeron en el modelo multivariado respectivo, que finalmente se ajustó manualmente mediante el procedimiento de eliminación hacia atrás (backward) para retirar las variables no significativas (p<0,05). Son factores de riesgo de Mollicutes en los rebaños un número mayor de categorías zootécnicas en la finca (RP=1,691; IC95%: 1,293 - 2,210), la presencia de ganado Holstein (RP=15,89; 1,44 - 175,45), fuentes de abasto de agua diferentes al acueducto (río, pozo, presa) (RP=4,37; 1,306 - 14,620) y las fincas que compran animales dentro de la misma provincia (RP=4,28; 1,416 - 12,939). En tanto, el lavado de las manos del ordeñador entre vacas o entre grupos mostró una prevalencia 57% y 67,6% menor, respectivamente, que en hatos donde esta medida no se cumple. Además, el secado de los cuartos con papel previo al ordeño (prevalencia 69,6% menor) es también un factor de protección de Mollicutes en vacas individuales

    Efecto del biocarbón en invertebrados de suelo: resultados preliminares

    The soil mesofauna comprises a group of arthropods, whose body size ranges from 0.2 to 2.0 mm, and which are important in transforming soil organic matter, recycling nutrients, and improving physical properties of the ground. Within the faunal groups that inhabit the soil, oribatid mites and springtails stand out because of their number, diversity, abundance of species and activity; great aptitude for speciation, short life cycle and little dispersion of species adapted to edaphic life and to different types of soils. Due to these characteristics, these organisms are used as biogeographic-ecological indicators and indicators of the soil quality and health. The use of biochar (Biocarbón in Spanish), as a soil amendment, to improve fertility and mitigate climate change is becoming a common practice. Rice husk biochar was used in three concentrations. Four tomato seedlings (Solanum lycopersicun L.) cultivar L43 were placed in each experimental unit. A composite sample of each treatment was taken weekly and placed in Berlese-Tullgren funnels for seven days. Subsequently, the collected samples were examined under the stereomicroscope and the individuals were separated by morphospecies for their subsequent slide mounting and identification. Arthropods belonging to the groups Insecta (Hymenoptera, Hemiptera, Coleoptera, Lepidoptera), Acari, and Collembola were identified. An increase in the number of Collembola was observed in the treatment with 75% Biochar (rice husk) after 28 days. In the case of mites, the most represented orders were Oribatida (Cryptostigmata), Prostigmata, and Mesostigmata, with an increase in their populations in the containers with 25% Biochar after 28 days. A positive influence of biochar on soil arthropods was demonstrated, expressed by the increase in the number of springtails and soil mites with respect to the control, with an outstanding recovery of soil biological activity in the short term.La mesofauna del suelo comprende un grupo de artrópodos, cuyo tamaño del cuerpo oscila entre 0,2 y 2,0 mm. Estos son importantes en la transformación de la materia orgánica, el reciclaje de nutrientes y el mejoramiento de las propiedades físicas del suelo. Los ácaros oribátidos y los colémbolos destacan dentro de los grupos faunísticos que habitan el suelo por su número, diversidad, abundancia de especies y actividad, así como por su ciclo de vida corto, gran aptitud para la especiación y poca dispersión de las especies adaptadas a la vida edáfica y a diferentes tipos de suelos. Debido a las características antes mencionadas, estos artrópodos se utilizan como indicadores biogeográficos-ecológicos; de la calidad y salud de los suelos. El empleo de biocarbón (en inglés Biochar) como enmienda de suelos para mejorar la fertilidad y mitigar el cambio climático, es una práctica común. Se utilizó biocarbón de cascarilla de arroz en tres concentraciones. En cada unidad experimental se colocaron cuatro plántulas de tomate (Solanum lycopersicun L.) cultivar L43. Semanalmente, se tomó una muestra compuesta de cada tratamiento y se colocaron durante siete días en embudos de Berlese-Tullgren, Posteriormente, las muestras recolectadas se revisaron al estereomicroscopio, separando los individuos por morfoespecies para su posterior montaje e identificación. Se identificaron artrópodos pertenecientes a varios grupos: Insecta (Hymenoptera, Hemiptera, Coleoptera, Lepidoptera), Acari y Collembola. Se observó un incremento del número de Colémbolos a los 28 días en el tratamiento, con 75 % Biochar (cascarilla de arroz). En el caso de los ácaros, los órdenes más representados fueron Oribatida (Cryptostigmata), Prostigmata y Mesostigmata, con un incremento de sus poblaciones a los 28 días en los contenedores, con 25 % Biochar. En el control se demostró una influencia positiva del biocarbón en los artrópodos del suelo, expresado en el incremento de la cantidad de colémbolos y ácaros, destacándose la recuperación de la actividad biológica del suelo, a corto plazo

    The objective of this study was to evaluate the effect of the combined application of biochars and the nematophagous fungus Pochonia chlamydosporia (Goddard) Zare and Gams on growth of tomato plants (Solanum lycopersicum L.) cv. ‘Elbita’ and their protection against Meloidogyne incognita (Kofoid and White) Chitwood. The biochars were produced from: rice husk (Oryza sativa L.), branches of leucaena (Leucaena leucocephala (Lam.) de Wit.) and marabou (Dichrostachys cinerea Wight et Am.), mango seeds (Mangifera indica L.), and coconut fiber (Cocos nucifera L.), using Kon-Tiki technology. The biochars and P. chlamydosporia strain IMI SD 187 (dose of 5x103 chlamydospores.g-1 of substrate) were added to the substrate consisting of red ferralitic soil: cattle manure: biochar (2:1:1 v/v). The experiment was carried out in 60 cell trays , which were randomly distributed in biological isolators. Control treatments without the fungus were included. After 30 days, vegetative growth and colonization of the substrate and roots by P. chlamydosporia were evaluated. Subsequently, five plants from the rice husk biochar + P. chlamydosporia treatment were transplanted to pots containing one kg of sterile substrate (soil: cattle manure 1:1 v/v), inoculated with 500 juveniles of a pure population of M. incognita, and to control pots without nematodes and left to grow for 60 days. The vegetative growth and the nematode gall index were evaluated. The application of biochars and P. chlamydosporia did not affect rhizosphere colonization by the fungus and showed a positive effect on growth of tomato plants. The fungus with rice biochar achieved early protection against M. incognita.El objetivo de este estudio fue evaluar el efecto de la aplicación combinada de biocarbones y el hongo nematófago Pochonia chlamydosporia (Goddard) Zare y Gams sobre el crecimiento de plantas de tomate (Solanum lycopersicum L.) cv. ‘Elbita’, y la protección frente a Meloidogyne incognita (Kofoid y White) Chitwood. Los biocarbones se produjeron a partir de cáscara de arroz (Oryza sativa L.), ramas de leucaena (Leucaena leucocephala (Lam.) de Wit.) y marabú (Dichrostachys cinerea Wight et Am.), semillas de mango (Mangifera indica L.) y fibra de coco (Cocos nucifera L.) mediante la tecnología Kon-Tiki. Los biocarbones y P. chlamydosporia cepa IMI SD 187 (dosis de 5x103 clamidosporas.g-1 de sustrato) se adicionaron en la preparación de los sustratos consistentes en suelo ferralítico rojo: estiércol vacuno: biocarbón (2:1:1 v/v). El experimento se realizó en bandejas multiceldas de 60 alveolos y se distribuyeron al azar en aisladores biológicos. Se prepararon tratamientos controles sin el hongo. A los 30 días, se evaluó el crecimiento vegetativo y la colonización de P. chlamydosporia en el sustrato y las raíces. Posteriormente, se trasplantaron cinco plantas del tratamiento biocarbón de cáscara de arroz + P. chlamydosporia, hacia macetas de un kg de sustrato (suelo: estiércol vacuno 1:1 v/v) estéril, donde se inocularon 500 juveniles de una población pura de M. incognita, y controles sin nematodos, durante 60 días. Se evaluó el crecimiento vegetativo y el índice de agallamiento del nematodo. La aplicación de biocarbones y P. chlamydosporia no afectó la colonización del hongo en la rizosfera y mostró un efecto positivo en el crecimiento de las plantas de tomate.     Biocarbón, humus y micorrizas: efecto sobre Dioscorea alata L. e infestación y reproducción de nematodo agallero en condiciones semicontroladas

    The objective of the research was to determine the effect of biochar enriched with worm humus and arbuscular mycorrhizal fungi (AMF) on growth and galling of yam plants and M. incognita reproduction. The studywas developed under semi-controlled conditions in a biological isolator of the Instituto de Investigaciones de Viandas Tropicales (INIVIT), Santo Domingo, Villa Clara Province, Cuba, in 2022. A completely randomized design with seven replicates was used. The treatments consisted of different combinations of soil, biofertilizers, and biochar, with and without inoculation of a pure population of M. incognita established at the institute. The best development of yam plants was obtained in the treatment where, in the absence of M. incognita, the plants were grown in a substrate containing a mixture of biochar +humus and mycorrhizae as inoculant. It was evident that M. inognita parasitism affected the development of the cultivar \u27INIVIT Ñ-2008\u27 because of the decreases produced in plant stem length and fresh and dry matter of the foliar part. The potentialities of incorporating AMF, humus, and biochar in the production technology of D. alata were evidenced by the positive effects on plant growth and development and reduction of M. inognita galling index and reproduction on this cultivar, which was cataloged as a good host and susceptible to this nematode; results which will be corroborated in subsequent field studies.El objetivo de la investigación fue determinar el efecto del biocarbón enriquecido con humus de lombriz y Hongos Micorrizógenos Arbusculares (HMA) en el crecimiento y agallamiento del ñame; así como en la reproducción de M. incognita. El estudio, en condiciones semicontroladas, se desarrolló en el aislador biológico del Instituto de Investigaciones de Viandas Tropicales (INIVIT) en Santo Domingo, provincia de Villa Clara, Cuba. El experimento tuvo un diseño completamente aleatorizado, con ocho tratamientos, los cuales consistieron en diferentes combinaciones de suelo, biofertilizantes y biocarbón, con y sin de una población pura de Meloidogyne incognita (Kofoi y White) Chitwood. Se realizaron siete replicas de cada uno. Los datos se analizaron por ANOVA y las medias se compararon (Duncan, 95 % de confianza). El mejor desarrollo de las plantas de ñame se obtuvo en el tratamiento donde, en ausencia de M. incognita, las plantas crecieron en un sustrato que contenía una mezcla de biocarbón, humus y micorrizas. Resultó evidente que el parasitismo por M. inognita afectó el desarrollo del cultivar ‘INIVIT Ñ-2008\u27, pues se produjeron disminuciones en los parámetros longitud del tallo y la masa fresca y seca foliar. Se evidenciaron los beneficios de incorporar HMA, humus y biocarbón en la tecnología de producción de D. alata, dado sus efectos positivos en el crecimiento y desarrollo de las plantas, así como en la disminución del Indice de Agallamiento (IA) y la reproducción de M. incognita en este cultivar, catalogado como buen hospedante y susceptible a este nematodo; resultados que se corroborarán en posteriores estudios de campo

    Actividad antifúngica in vitro de nanopartículas de plata a partir de hojas de Leea coccinea L. frente a Fusarium spp. y Penicillium spp.

    The objective of this work was to determine the antifungal activity of three batches of AgNPs obtained from Leea coccinea L. on mycelial growth of Fusarium spp. and Penicillium spp. associated with common bean (Phaseolus vulgaris L.) and potato (Solanum tuberosum L.) seeds and spore germination of of Fusarium spp. Seven concentrations (0.485, 0.242, 0.121, 0.060, 0,03, 0,.015, and 0.007 mg/ml) of the AgNPs suspensions were evaluated using the serial dilution method. The Minimum Fungicide Concentration (MFC) of the AgNPs against the different microorganisms was determined. The results showed that the application of the AgNPs was effective in inhibiting growth of Fusarium spp. and Penicillium spp. The CMF of the AgNPs from the three batches ranged between 0.485 and 0.242 mg/ml against the phytopathogenic agents. The inhibition of mycelial growth of phytopathogens increased with increasing concentrations. The suspension of AgNPs from L. coccinea affected spore germination of the isolates of F. oxysporum and Fusarium spp. No. 1 in the two highest concentrations (0.485 and 0.242 mg/ml). It was confirmed that the higher the AgNPs concentration, the greater the antifungal effect on mycelial growth and spore germination of the phytopathogens studied. The results obtained denote the efficacy of this product as an alternative in the fight against phytopathogenic fungi.El presente trabajo tuvo como objetivo, determinar la actividad antifúngica de tres lotes de nanopartículas de plata (AgNPs) obtenidas de Leea coccinea L., sobre el crecimiento micelial de aislamientos de Fusarium spp. y Penicillium spp. asociados a semillas de frijol común (Phaseolus vulgaris L) y papa (Solanum tuberosum L.), y la germinación de esporas de Fusarium spp. Se evaluaron siete concentraciones (0.485; 0.242; 0.121; 0,060; 0.030; 0.015 y 0.007 mg/ml) de las suspensiones de AgNPs mediante el método de diluciones seriadas. Se determinó la Concentración Mínima Fungicida (CMF) de las AgNPs frente a los diferentes microorganismos. Los resultados mostraron que la aplicación de las AgNPs fue efectiva en la inhibición del crecimiento de los aislados de Fusarium spp. y Penicillium spp. Las CMF de las AgNPs de los tres lotes oscilaron entre 0,485- 0,242 mg/ml frente a los agentes fitopatógenos. La inhibición del crecimiento micelial de los fitopatógenos por las AgNPs se incrementó con el aumento de las concentraciones. La suspensión de AgNPs de L. coccinea afectó la germinación de esporas de los aislamientos de F. oxysporum y Fusarium spp. No. 1 en las dos concentraciones más altas (0,485 y 0,242 mg/ml). Se confirmó que a mayor concentración de las AgNPs de L. coccinea, mayor fue el efecto antifúngico sobre el crecimiento micelial y germinación de las esporas de los fitopatógenos estudiados. Los resultados obtenidos denotan la potencia de este producto, como alternativa, en el enfrentamiento contra hongos fitopatógenos

    Seguridad e inmunogenicidad en lechones con el candidato vacunal E2-CD154, una vacuna de subunidades contra la peste porcina clásica. Resultados del ensayo clínico de fase III

    E2-CD154 is a subunit vaccine candidate that has been proven to be safe and to protect piglets from classical swine fever (CSF). In this study, those previous findings were confirmed and extended to a larger number of animals in a phase III clinical trial conducted on two production farms in Pinar del Río province. All animals in both farms were vaccinated with two doses of E2-CD154 on days 0 and 21. The study extended up to 60 weeks. The vaccine was well tolerated in piglets between 15 and 28 days of life, with neither local nor systemic side effects documented. Immunized pregnant sows were capable of transmitting high levels of maternally-derived neutralizing antibodies (MDNAs) to their offspring (Unit A, geometric mean titer = 1:1295, minimum value 1:100 and Unit B geometric mean titer = 1:474, minimum value 1:150), well above the protection threshold (1:50). These high MDNA titers in the piglets did not interfere with the immunogenicity of the candidate. All vaccinated piglets evaluated at random (more than 10% of 2804 vaccinated) developed protective neutralizing antibody titers higher than 1:400 at the four time points analyzed (nine, 21, 41, and 44 weeks) in both farms. The results of this study confirm the safety, immunogenicity and robustness of this vaccine candidate in this sensitive pig category in the field.El candidato vacunal E2-CD154 es una vacuna de subunidad cuya seguridad y capacidad protectora en crías frente a la peste porcina clásica ha sido demostrada en estudios previos. Esos hallazgos se confirman en el presente estudio de fase III realizado en dos unidades de producción porcina en la provincia de Pinar del Rio con un número mayor de animales. Todos los animales en ambas granjas recibieron dos dosis de E2-CD154 los días 0 y 21. El estudio se extendió por 60 semanas, la vacuna fue bien tolerada en las crías vacunadas entre los 15 y 28 días de edad, no se documentaron efectos adversos locales o sistémicos. Las cerdas gestantes inmunizadas fueron capaces de trasmitir altos títulos de anticuerpos a sus crías (Unidad A, media geométrica = 1:1295, valor mínimo 1:100 y Unidad B, media geométrica = 1:474, valor mínimo 1:150), muy por encima del umbral de protección (1:50). Estos elevados títulos de anticuerpos neutralizantes derivados de la madre no interfirieron con la inmunogenicidad del candidato vacunal en estudio. Todas las crías vacunadas estudiadas al azar (más del 10% de las 2804) desarrollaron títulos de anticuerpos neutralizantes mayores de 1:400 en los cuatro muestreos realizados (nueve, 21, 41, y 44 semanas) en ambas granjas. Los resultados de este estudio confirman la seguridad, inmunogenicidad y robustez del candidato vacunal en esta sensible categoría en condiciones de campo

    Introducción del primer núcleo genético de cabras de la raza Murciano-Granadina en Cuba

    Animal imports increase the risk of exposure of local or susceptible livestock to pathogens if the risk in imported animals is not properly managed. The combination of risk analysis, diagnosis and quarantine facilities is often the main strategy to mitigate such risk. The aim of this work was to ensure, from the sanitary point of view, the introduction of the first genetic nucleus of Murciano-Granadina goats in Cuba. To this end, sanitary requirements were harmonized between the Veterinary Authorities of exporter and recipient countries. Pre-shipment diagnostic tests were carried out and, once transported to Cuba, they were kept in a quarantine unit for 60 days during which time further research was carried out. Diagnostic testing at origin and destination showed no evidence of active infection. Viability of the imported goats was effectively preserved in the quarantine unit until their transfer to post-quarantine. There was an acceptable level of risk reached for the release of Murciano-Granadina goat nucleus for use in the Genetic Development Program of the species, which has been validated after animals have been in post-quarantine conditions for one year, showing good productive performance with no relevant health incidences.La importación de animales puede exponer al patrimonio zootécnico autóctono susceptible a patógenos si no se maneja apropiadamente este riesgo en los animales importados. La combinación del análisis de riesgo, el diagnóstico y facilidades de cuarentena suele ser la principal estrategia para mitigar tal riesgo. El objetivo del presente trabajo fue asegurar, desde el punto de vista sanitario, la introducción del primer núcleo genético de cabras de la raza Murciano-Granadina en Cuba. Para ello se compatibilizaron requisitos sanitarios entre las Autoridades Veterinarias del exportador y el receptor, se efectuaron pruebas diagnósticas preembarque y una vez trasladados hasta Cuba se mantuvieron en una unidad de cuarentena por 60 días durante los cuales se les practicaron otras investigaciones. Las pruebas diagnósticas en origen y destino no evidenciaron presencia de infección activa. La viabilidad de las cabras importadas se preservó de forma efectiva en la unidad de cuarentena hasta su traslado a post-cuarentena. Se logró un nivel de riesgo aceptable para la liberación del núcleo de cabras Murciano-Granadinas para su uso en el programa genético de desarrollo de la especie, lo cual se ha validado después de un año de los animales en post-cuarentena con buen desempeño productivo sin incidencias de salud relevantes
