    Evaluación del cumplimiento de normativas establecidas para la acreditación de ensayos ecotoxicológicos en el Centro de Bioactivos Químicos

    In the new economic scenario, companies must not only survive, but also succeed. That is why accreditation has become a first-order necessary. Considering this requirement, it is intended that the biological laboratory of the Center for Bioactive Chemicals (CBQ), demonstrates that it operates competently and generates valid results, under an integrated management system. This research was aimed at evaluating compliance with the standards established for the accreditation of the bioassay service for toxicity testing with Lactuca sativa L. seeds at CBQ. Diagnosing the degree of compliance with the regulatory requirements applicable to the bioassay service was designed from the analysis of each stage of the service process to the identification and processing of the problems that prevent the accreditation of the testing service. Tools such as blueprinting, checklist and Pareto Chart were used for this purpose. Out of 286 requirements evaluated, 97 (34 %) were not complied with. They were mainly related to the requirements related to: testing service process, elements of the management system, impartiality, and confidentiality. Additionally, an action plan was established to solve the unfulfilled requirements which represented the highest percentage of the problems that prevented the start of the accreditation process.En el nuevo escenario económico, las empresas no pueden existir para sobrevivir, sino para tener éxito. Es por ello que la acreditación se ha convertido en una necesidad de primer orden. Teniendo en cuenta esta premisa, se pretende que el laboratorio biológico del Centro de Bioactivos Químicos (CBQ), bajo el alcance de un sistema de gestión integrado, demuestre que opera de forma competente y genera resultados válidos. Esta investigación tiene como objetivo evaluar el cumplimiento de las normas establecidas para la acreditación del servicio de bioensayo de toxicidad con semillas de Lactuca sativa L. en el CBQ. El diagnóstico del grado de cumplimiento de los requisitos de las regulatorias aplicables al servicio de bioensayo estuvo concebido desde el análisis de cada una de las etapas del proceso de servicio hasta la identificación y el procesamiento de los problemas que impiden la acreditación del servicio de ensayo. Para ello se utilizaron herramientas como fueron el blueprinting, la lista de chequeo y el diagrama de Pareto. De 286 requisitos evaluados, se incumplieron 97 (34 %), los cuales se relacionaron fundamentalmente con requisitos relativos a: proceso del servicio de ensayo, elementos del sistema de gestión, imparcialidad y confidencialidad. Además, se confeccionó un plan de acción para darle solución a los requisitos incumplidos que representan el mayor porcentaje de los problemas que impiden el inicio del proceso de acreditación

    Artrópodos depredadores asociados a especies arbóreas en un agroecosistema tabacalero del municipio Pinar del Río, Cuba

    The research was conducted in the tobacco farm “La Rosa” in the municipality of Pinar del Río, Cuba, with the objective of determining the diversity and abundance of predatory arthropods associated with arboreal species. A floristic inventory of the arboreal diversity was carried out to family and species level, and samples of the predatory arthropods were collected for their transfer and processing at the entomology laboratory of the University of Pinar del Río. Taxonomic keys were used to identify the arthropod species. Twelve tree species belonging to eleven botanical families were inventoried, with the greatest representation of fruit trees (58 %). The fourteen species of predatory arthropods identified presented regulatory activity on insects and/or mites, being pests on fruit trees, tobacco (Nicotiana tabacum L.), and other economically important crops. The most represented orders were Hemiptera, Diptera and Mesostigmata, while the families Coccinellidae and Phytoseiidae presented the highest species richness. It is also noteworthy that five groups of identified predators (phytoseiid mites, spiders, myrids, coccinellids, and syrphid flies) reached relative abundance ≥ 10 %.La investigación se desarrolló en la finca tabacalera “La Rosa” del municipio Pinar del Río, Cuba, con el objetivo de determinar la diversidad y abundancia de artrópodos depredadores asociados a especies arbóreas. Se realizó un inventario florístico de la biodiversidad arbórea, hasta nivel de familia y especie; se recolectaron muestras de los artrópodos depredadores para su traslado y procesamiento al Laboratorio de Entomología de la Universidad de Pinar del Río. Para la identificación de las especies de artrópodos se utilizaron claves taxonómicas. Se inventariaron 12 especies arbóreas pertenecientes a 11 familias botánicas, con mayor representación de frutales (58 %). Las 14 especies de artrópodos depredadores identificadas presentaron actividad reguladora sobre insectos y/o ácaros que constituyen plagas en frutales, tabaco (Nicotiana tabacum L.) y otros cultivos de importancia económica. Los órdenes más representados fueron Hemiptera, Diptera y Mesostigmata; mientras que, las familias Coccinellidae y Phytoseiidae presentaron la mayor riqueza de especies. También se destaca que cinco grupos de depredadores identificados (ácaros fitoseidos, arañas, míridos, coccinélidos y moscas sírfidas) alcanzaron abundancia relativa ≥ 10 %

    Efecto de aceites esenciales y componentes seleccionados sobre Heterorhabditis amazonensis Andaló et al. cepa HC1

    The effect of four essential oils and eleven selected components on the infective juveniles (IJ) of the entomopathogenic nematode Heterorhabditis amazonensis Andaló et al. strain HC1 was studied under laboratory conditions. Susceptibility of this strain to seven organic solvents and surfactants was determined. The lethal and sublethal toxic effects on IJ exposed directly to the essential oils and components were evaluated in 24 well plates for 24 h. Data oflethal toxic effects were subjected to ANOVA and the means compared by Duncan´s multirange test at 0,05 %, whereas data of sublethal toxic effects were analyzed by the multiple comparison of proportions (Wald´s Method, confidence level of 0,05). Toxicity of Triton X-100 at 0,5 % on H. amazonensis strain HC1 was low and could be used in bioassays of chemical product compatibility with this nematode strain. The essential oils of Melaleuca quinquenervia (Cav) S.T. Blake and Citrus sinensis (L.) Osbeck caused no significant lethal toxic effect on IJ of H. amazonensis (strain HC1), while those of Piper aduncum subsp. ossanum (C. DC.) Saralegui and Piper auritum Kunth produced moderate mortality. No sublethal effects were caused by these four oils on the infectivity of IJ of H. amazonensis strain HC1. The essential oil components camphene, p-cymene and piperitone did not affect IJ of H. amazonensis strain HC1; 1,8-cineole, limonene, camphor, and methyl chavicol showed moderate toxicity, and carvacrol, eugenol, linalool, and thymol were highly toxic. According to the parameters evaluated, the essential oils from M. quinquenervia and C. sinensis and the components camphene, p-cymene, and piperitone were compatible with H. amazonensis strain HC1.Se investigó en laboratorio el efecto de cuatro aceites esenciales (Melaleuca quinquenervia (Cav) S.T. Blake; Citrus sinensis (L.) Osbeck; Piper aduncum subsp. ossanum (C. DC.) Saralegui y Piper auritum Kunth) y 11 componentes seleccionados sobre los juveniles infectivos (JI) Heterorhabditis amazonensis Andaló et al. cepa HC1. Se determinó la susceptibilidad de esta cepa a siete disolventes orgánicos y tensoactivos por inmersión. Los efectos tóxicos letales de los aceites y componentes sobre los JI se evaluaron por exposición directa en placas de 24 pocillos (24 h). El bioensayo de infectividad Uno-Uno se utilizó para evaluar el efecto tóxico subletal de los aceites esenciales sobre los JI. Los datos de los ensayos del efecto tóxico letal de disolventes orgánicos /tensoactivos y de componentes de aceites esenciales se sometieron a ANOVA y las medias se compararon (Duncan; 0, 05 %); mientras que, los del ensayo del efecto tóxico subletal, se analizaron mediante comparación múltiple de proporciones (Método de Wald, nivel de confianza de 0,05). El Tritón X-100 al 0,5 % evidenció baja toxicidad sobre los JI y se puede utilizar en ensayos de compatibilidad de productos químicos con esta cepa. Los aceites esenciales de M. quinquenervia y C. sinensis no provocaron un efecto tóxico letal significativo sobre los JI de H. amazonensis cepa HC1; los aceites de P. aduncum subsp. ossanum y P. auritum produjeron moderada mortalidad. Estos cuatro aceites esenciales no causaron efectos subletales sobre la infectividad de los JI de H. amazonensis cepa HC1. Los componentes de aceites esenciales canfeno, p-cimeno y piperitona no afectaron a los JI de H. amazonensis cepa HC1; 1,8-cineol, limoneno, alcanfor y metil chavicol mostraron toxicidad moderada y carvacrol, eugenol, linalol y timol fueron altamente tóxicos a los JI. De acuerdo con los parámetros evaluados, los aceites esenciales de M. quinquenervia y C. sinensis y los componentes canfeno, p-cimeno y piperitona son compatibles con H. amazonensis cepa HC1

    Factores de riesgos asociados a la prevalencia de patologías podales en vacas Siboney de Cuba

    In Cuba, there is little information on the foot pathologies affecting cattle, and these constitute a problem of animal welfare under production conditions. In the present study, 384 dairy Siboney de Cuba cows were examined, which represented 100 % of milking cows from six dairy farms in Artemisa province. The prevalence of lesions was calculated and the logistic regression analysis was performed to determine the risk factors associated with these pathologies. Fifty-six cows were detected with foot lesions (14.58 %), of which 53.57 % presented first degree of lameness. The most affected members were the hind legs (66.26 %) and, of these, the side hooves with the highest number of lesions. The main pathologies diagnosed were: pathological deformations of the hoofs, ulceration of the sole and white line disease. It is concluded that poor drainage and sleeves in poor condition were the main risk factors for the presentation of foot lesions in the cows studied.En Cuba existe poca información sobre las patologías podales que afectan el ganado bovino, y estas constituyen un problema de bienestar animal en las condiciones de producción. En el presente estudio se examinaron 384 vacas lecheras de la raza Siboney de Cuba que representaron el 100 % de las vacas en ordeño de seis vaquerías de la provincia Artemisa. Se calculó la prevalencia de las lesiones y se realizó análisis de regresión logística para determinar factores de riesgo asociados a estas patologías. Se detectaron 56 vacas con lesiones podales (14,58 %), de las cuales el 53,57 % presentaron claudicación de primer grado. Los miembros más afectados fueron los posteriores (66,26 %) y, de estos, las pezuñas laterales con mayor número de lesiones. Las principales patologías diagnosticadas fueron: deformaciones patológicas de las pezuñas, úlcera de la suela y enfermedad de la línea blanca. Se concluye que el mal drenaje y los caminos y mangas en mal estado fueron los principales factores de riesgo para la presentacion de lesiones podales en las vacas estudiadas

    Ocurrencia de estructuras compatibles con hemoplasmas en caninos no domiciliados de Cuba

    Canine hemoplasmosis is a disease caused by hemotrophic mycoplasmas (hemoplasmas) that affects dogs regardless of breed or age. These agents (formerly belonging to the genera Haemobartonella and Eperythrozoon) are obligate eperythrocytic bacteria that can induce anemia in the infected host and are recognized for their zoonotic potential. The objective of the present study was to detect the presence of hemoplasma compatible structures in populations of non-domiciled dogs in Havana province, Cuba. Samples were collected from 124 dogs from 11 municipalities of this province. Diagnosis was carried out by observation of blood smears stained with Giemsa and by polymerase chain reaction (PCR), amplifying a region of the 16S rRNA gene of the class Mollicutes. Hemoplasma compatible structures were detected in 23 % of the dogs tested and Mollicutes was diagnosed in 41 % of the samples processed by PCR. No association was found between the presence of hemoplasma compatible structures and anemia in the animals. The spatiotemporal pattern of hemoplasma compatible structures was identified, indicating that dogs infected with them were detected in all the municipalities included in this study. No association was found between hemoplasma infection and the presence of anemia. This result is the first study of this type to be carried out in Cuba and evidences the need to confirm, by other molecular assays, the identification of hemoplasma and the development of a prevention and control strategy for this entity.La hemoplasmosis canina es una enfermedad causada por micoplasmas hemotróficos (hemoplasmas) que afectan a los canes sin distinción de raza o edad. Estos agentes (antiguamente pertenecientes a los géneros Haemobartonella y Eperythrozoon) son bacterias epieritrocitarias obligadas que pueden inducir anemia en el huésped infectado y que se reconocen por su potencial zoonótico. El objetivo del presente estudio fue detectar la presencia de estructuras compatibles con hemoplasmas (ECHM) en poblaciones de perros no domiciliados de la provincia La Habana, Cuba. Se colectaron muestras de 124 perros procedentes de 11 municipios de esta provincia. El diagnóstico se realizó a través de la observación de frotis sanguíneos teñidos con Giemsa y por reacción en cadena de la polimerasa (PCR), amplificando una región del gen ARNr 16S de la clase Mollicutes. En el 23 % de los perros analizados se detectó la presencia de ECHM y en el 41% de las muestras, procesadas por PCR, se diagnosticó Mollicutes. No se encontró asociación entre la presencia de ECHM y la anemia que tenían los animales. Se identificó el patrón espaciotemporal de ECHM, el cual indica que en todos los municipios incluidos en este estudio se detectaron canes infectados con ECHM. Este resultado constituye el primer estudio de este tipo que se realiza en Cuba y evidencia la necesidad de confirmar, por otros ensayos moleculares, la identificación de hemoplasma y el desarrollo de una estrategia de prevención y control sobre esta entidad

    Dogs can be affected by several species of the genus Babesia causing important diseases in different regions of the world. Among these species are Babesia canis, B. vogeli, B. rossi, B. gibsoni, and B. vulves. The aim of this study was to detect the presence of Babesia spp. in stray dogs from Havana province. Sixty dogs were studied and subjected to haematological examination, blood smear and PCR, which amplified 18S rRNA. Three animals (5 %) showed hematocrit values higher than reference values while 23 (38.3 %) showed lower hematocrit values, indicating the presence of an anemic syndrome in those dogs. According to the leukogram analysis, five dogs (8.3 %) had leukocytosis and 7 (11.6 %) had leukopenia. Thirty-one animals were infested with Rhipicephalus sanguineus sensu lato ticks and 10 were co-infested with Rhipicepahalus microplus ticks. Twenty percent of the samples tested positive by PCR, while microscopic examination revealed Babesia spp. merozoites in only 13.3 % of the samples. A relationship was observed between PCR-positive animals and the decrease in hematocrit value. There was no association between the presence of ticks and sex in animals positive to Babesia spp. The present study demonstrates the presence of Babesia spp. in stray dogs from Havana province.Los perros pueden ser afectados por varias especies del género Babesia que causan enfermedades importantes en diferentes regiones del mundo. Dentro de estas especies se encuentran Babesia canis, B. vogeli, B. rossi, B. gibsoni y B. vulves. El presente trabajo tuvo como objetivo detectar la presencia de Babesia spp. en perros sin dueño de La Habana. Se estudiaron 60 perros, a los que se les realizó examen hematológico, frotis sanguíneo y PCR, que amplifica el ARNr 18S. Tres animales (5 %) presentaron valores de hematocrito superiores a los valores de referencia y 23 (38,3 %) mostraron valores de hematocrito inferiores, lo que indica la presencia de un síndrome anémico en estos perros. Como resultado del leucograma, cinco perros (8,3 %) tenían leucocitosis y 7 (11,6 %) leucopenia; 31 animales estaban infestados con garrapatas Rhipicephalus sanguineus sensu lato y 10 estaban coinfestados con Rhipicepahalus microplus. El 20 % de las muestras fueron positivas por PCR, mientras que en la examinación microscópica solo en el 13,3 % de las muestras se observaron merozoitos de Babesia spp. Se observó relación entre los animales positivos por PCR y la disminución del valor del hematocrito. No existió asociación entre la presencia de garrapatas y el sexo en los animales positivos a Babesia spp. El presente estudio demuestra la presencia de Babesia spp. en perros sin dueño de La Habana

    Primer registro de Podogaster Brullé (Hymenoptera: Ichneumonidae: Anomaloninae) en cultivo de morera en Cuba

    The Ichneumonidae family is considered the most diverse within the order. However, despite of the existence of many endemic species, knowledge of their distribution and biodiversity is limited in Latin America. To identify if species from this group are associated to lepidoptera present in mulberry tree (Morus alba L.), during 2018-2019 period, prospections were carried out in plantations located in agricultural areas of northeastern Havana, Cuba. Larvae of Glyphodes sibillalis Walker (Lepidoptera: Crambidae) were collected and transferred to the Entomology Laboratory at National Center of Health for Plants and Animals (CENSA) to complete their lifespan and verify the parasitoid presence. The identification of the emerged specimens was carried out according to the morphological criteria defining this group. For this, the corresponding dichotomous keys and an NSZ-606 stereoscope with 10x resolution were used. The study allowed identifying the genus Podogaster Brullé, first reported in Cuba, as well as its association with G. sibillalis and mulberry tree.La familia Ichneumonidae se considera la más diversa dentro del orden. Sin embargo, a pesar de la existencia de muchas especies endémicas, el conocimiento de su distribución y biodiversidad es limitado en América Latina. Para determinar si las especies de este grupo se asocian a los lepidópteros presentes en la morera (Morus alba L.), durante el período 2018-2019 se realizaron prospecciones en plantaciones situadas en áreas agrícolas del noreste de La Habana (Cuba). Se recogieron larvas de Glyphodes sibillalis Walker (Lepidoptera: Crambidae) y se transfirieron al Laboratorio de Entomología del Centro Nacional de Sanidad Agropecuaria (CENSA) para completar su vida útil y verificar la presencia del parasitoide. La identificación de los especímenes emergidos se llevó a cabo de acuerdo con los criterios morfológicos que definen a este grupo. Para ello, se utilizaron las correspondientes claves dicotómicas y un estereoscopio NSZ-606 con una resolución de 10x.     Incidencia de Alternaria spp. en semillas de cereales, vegetales, frutales y ornamentales

    The aim of this work was to determine the Alternaria species associated with seeds of cereals, vegetables, fruits and ornamentals. For this purpose, 2503 seed lots from 21 countries were examined. From each lot, 400 seeds were analyzed by blotter test method. Alternaria species were identified morphologically. For each specie, the occurrence frequency and infestation index by seed type were calculated. A total of 18 species of Alternaria, of the sections Alternaria, Brassicicola, Dianthicola, Gypsophilae, Infectoriae, Japonicae, Porri, Radicina, and Sonchi, were detected. The most frequently detected species was A. alternata. This species constituted 59% of all Alternaria spp., followed by A. brassicicola (10%), T. padwickii (7%), A. tenuisima (7%), A. brassicae (2%), and A. radicina (2%). The observed infestationindexes of A. alternata and A. tagetica endanger the phytosanitary quality of the seeds and the crop health.El objetivo del trabajo fue determinar las especies de Alternaria asociadas a semillas de cereales, vegetales y ornamentales. Se examinaron 2503 lotes de semillas procedentes de 21 países. Por cada lote, se analizaron 400 semillas, mediante el método del "blotter test". Las diferentes Alternaria spp. se identificaron morfológicamente. Para cada especie se calculó la Frecuencia de Aparición por tipo de semilla y el Índice de infestación por cada lote analizado. Se detectaron 18 especies del género Alternaria, de las secciones Alternaria, Brassicicola, Dianthicola, Gypsophilae, Infectoriae, Japonicae, Porri, Radicina y Sonchi. También se detectó a Alternariaster helianthi (Hansf.) E.G. Simmons, Prathoda longissima (Deighton & MacGarvie) E.G. Simmons y Trichoconiella padwickii (Ganguly) B.L. Jain, anteriormente incluidas dentro del género. La especie con mayor frecuencia de detección fue A. alternata. Esta especie constituyó el 59 % de las detecciones de Alternaria spp., seguida por A. brassicicola (10 %), T. padwickii (7 %), A. tenuisima (7 %), A. brassicae (2 %) y A. radicina (2 %). Los índices de infestación observados de A. alternata y A. tagetica alcanzaron valores que comprometen la calidad fitosanitaria de la semilla y la sanidad del cultivo

    First report of Pyrrhocalles antiqua orientis (Skinner) (Lepidoptera: Hesperidae) infesting the Bermuda turfgrass in Cuba

     Dear editor:Butterflies have proven to be excellent indicators of the disturbance and biodiversity of habitats worldwide. Skipper (Hesperiidae) remains one of the most diverse and least known butterfly group. Pyrrhocalles Mabille (1904) (Lepidoptera: Hesperiidae) is a small genus endemic to the Greater Antilles, and Pyrrhocalles antiqua Herrich-Schäffer is found across the Caribbean. Three subspecies have been described, and Pyrrhocalles antiqua orientis Skinner is an endemic subspecies of Cuba. However, the knowledge about its distribution, population size, and caterpillar host plants is limited. In 2007, P. antiqua orientis caterpillars have been reported on Roystonea regia (H.B.K.), family Arecaceae, in Camaguey province, Cuba. Recently, samples collected from the Varadero golf field, Matanzas province, showed P. antiqua orientis caterpillars to be present causing defoliations on Bermuda turfgrass (Cynodon dactylon (L.) Pers), family Poaceae. Several pupae and twenty-six adults were also captured on greens. To our knowledge, this is the first evidence of P. antiqua orientis infesting Bermuda turfgrass in Cuba. The occurrence of P. antiqua orientis in coastal ecosystems, including the north littoral of the island, has been described. Caterpillars of the subfamily Hesperiinae have been reported feeding only on gramineas (Poaceae). These results suggest the need for carrying out observations of P. antiqua orientis populations and including this insect in the management strategies of the Varadero golf field.Sincerely

    Actividad antifúngica de compuestos volátiles producidos por especies endófitas de Bacillus sobre Moniliophthora roreri H.C Evans et al.

    The aim of this work was to determine the antifungal activity of Bacillus subtilis 9-2, Bacillus mycoides 29-2, Bacillus sp. 33 and 72, Bacillus cereus 45-1, 51-2, 53-4 and Bacillus amyloliquefaciens 24 on mycelial growth and sporulation of the phytopathogenic fungus Moniliophthora roreri H.C Evans et al. The confrontation experiments were carried out in Petri dishes with two compartments. PDA culture medium was added to one side of the plate and TSA to the other. The bacterial strains were individually inoculated (100 µl; 1×108 CFU ml-1) on the face of the cap with TSA culture medium; while on the other lid with PDA culture medium, a mycelial disk (5 mm in diameter) of the strain of the phytopathogenic fungus was placed. Sterile distilled water was used as a control. All plates were sealed with Parafilm and incubated at 28°C for 15 days. The experiments were repeated twice. All strains of bacteria produced volatile compounds that inhibited mycelial growth and the formation of M. roreri spores. B. subtilis 9-2, B. mycoides 29-2, B. sp. 33, Bacillus cereus 45-1, 51-2 and Bacillus amyloliquefaciens 24 inhibited mycelial growth and sporulation of M. roreri by more than 60%. These findings offer new possibilities to control frosty pod rot caused by M. roreri using Bacillus endophytes strains producing antifungal volatile compounds.El presente trabajo tuvo como objetivo determinar la actividad antifúngica de Bacillus subtilis 9-2, Bacillus mycoides 29-2, Bacillus sp. 33 y 72, Bacillus cereus 45-1, 51-2, 53-4 y Bacillus amyloliquefaciens 24 sobre el crecimiento micelial y esporulación del hongo fitopatógeno Moniliophthora roreri H.C Evans et al. Los experimentos de enfrentamiento se realizaron en placas Petri con dos compartimentos. En una cara de la placa se añadió medio de cultivo PDA y en la otra TSA. Las cepas bacterianas se inocularon individualmente (100 µl; 1×108 UFC ml-1) sobre la cara de la tapa con medio de cultivo TSA; mientras que, sobre la otra tapa con medio de cultivo PDA, se colocó un disco de micelio (5 mm de diámetro) de la cepa del hongo fitopatógeno. Como control se usó agua destilada estéril. Todas las placas se sellaron con Parafilm e incubaron a 28°C durante 15 días. Los experimentos se repitieron dos veces. Todas las cepas de bacterias produjeron compuestos volátiles que inhibieron el crecimiento micelial y la formación de esporas de M. roreri. B. subtilis 9-2, B. mycoides 29-2, B. sp. 33, Bacillus cereus 45-1, 51-2 y Bacillus amyloliquefaciens 24 inhibieron en más de 60 % el crecimiento micelial y la esporulación de M. roreri.
