    A SEM study of the aedeagus and spermatheca of Cassida viridis Linnaeus, 1758 (Coleoptera: Chrysomelidae: Cassidinae) from Turkey

    It is accepted male genitalia are not diagnostic, but spermathecae are partly diagnostic within the genus Cassida Linnaeus, 1758 (Coleoptera: Chrysomelidae: Cassidinae). However, studies on genitalia have been based only on stereomicroscopic examination. The ultrastructure of genitalia has mostly not been studied. The aim of this study was to determine whether this ultrastructures could be diagnostic. Aedeagi and spermathecae of 63 specimens of Cassida viridis Linnaeus, 1758 were collected from Qankiri, Hatay, Kayseri, Osmaniye and Zonguldak Provinces in Turkey in 2003, 2007 and 2015, and examined for the first time. Thus, new diagnostic characters were obtained and it revealed that it was diagnostic to either species or subgenus.     IMPACT OF IZBAN ON NEIGHBORHOOD POPULATION CHANGE IN THE IZMIR CITY REGION

    This study aims to find out the effects of a suburban railway investment on population change. Based on this quest, we hypothesize that reliable and fast rail transit service, which connects main destinations in an urban region has a significant role in population increase nearby its stations. In this vein, we use IZBAN Suburban rail as fast rail transit service in Izmir urban region as case study. Accordingly, population changes between 2013 and 2018 in the neighborhoods, where there is direct access to IZBAN stations, are examined in relation to other neighborhoods by Analysis of Covariance (ANCOVA) statistical models. Two model groups are devised in the study. Basic Models examine the impact of IZBAN Neighborhoods considering Izmir sub-regions, i.e., North, Metropolitan Area and South Sub-Region, while Expanded Models estimate interaction variables to account for spatial variation in population changes. We find significant relative increase in the populations of non-metropolitan neighbourhoods served by IZBAN stations. There is high variation between different sub-regions and IZBAN sections. Population increase is higher in the Northern and Southern sub-regions, which leads us to reach IZBAN-induced decentralization in the metropolitan area. Although, IZBAN exemplify an incipient transit-oriented development in Turkey, it falls short of its counterparts in the world. Strategic planning decisions are needed to support transit-oriented development around IZBAN stations

    Investıgatıon Of Bıocıde And Antıbıotıc Resıstance Profıles In Gram Posıtıve Bacterıa Isolated From Food

    Günümüzde hayvan yetiştiriciliğinde gerek hastalıkların tedavisi ve önlenmesi, gerekse hayvansal gıdaların verimini arttırmak amacıyla antibiyotik ve biyositlerin kullanımının oldukça yaygın olması ekonomi ve halk sağlığı açısından ciddi sorunlar yaratmaktadır. Araştırmamızda, Ankara ilinde çeşitli marketlerde satışa sunulan 132 et ve süt ürünü örneğinden izole edilen 77 Staphylococcus izolatında, antibiyotik ve biyosit direnç dağılımları ve antibiyotik direnç ve efluks pompa genlerinin varlığı araştırılmıştır. Ayrıca efluks pompa inhibitörü rezerpinin antimikrobiyal direncine etkisi araştırılmıştır. Staphylococcus izolatlarında en yüksek oranda direnç tetrasiklin (%24,7)’e karşı belirlenmiştir. İzole edilen 77 Staphylococcus izolatının 29’u bir veya birden fazla antibiyotiğe dirençli olarak tespit edilmiştir. Bu izolatların BBL crystal Gram pozitif tanımlama kiti ve 16S rDNA dizi analizi yöntemleriyle tür bazında tanımlamaları yapılmıştır. Test edilen biyositlerin hepsine yüksek MİK değerleri gösteren izolatlar mevcuttur. Rezerpinin Staphylococcus’larda ampisilin duyarlılığını %25, tetrasiklin duyarlılığını %27,8, triklosan duyarlılığını %20,7, povidon iyodin duyarlılığını %34,5 arttırdığı belirlenmiştir. Çalışılan 29 Staphylococcus izolatında tetK (%88,9), bla (%53,8) antibiyotik direnç genleri yüksek oranda tespit edilmiştir. Ayrıca efluks pompa genlerinden acrB (%31), norE (%79,3), sugE (%44,8), smr (%86,2), efrA (%79,3), efrB (%72,4), mdeA (%82,8), sepA (%51,7) yüksek oranda tespit edilen genlerdir. Araştırmamızda tüketime sunulan hayvansal gıdalardan izole ettiğimiz Staphylococcus’ların antimikrobiyal direnç genleri bulundurmaları, bu direnç genlerinin kromozomal ve kromozom dışı genetik materyallerle patojenlere ve normal flora bakterilerine aktarmaları riski sebebiyle, halk sağlığı açısından önemli bir tehdit oluşturmaktadır. Efluks pompası kaynaklı antimikrobiyal direncinin, efluks pompası inhibitörü rezerpin kullanımı ile belli bir oranda azaltılabileceği fakat bu alanda daha kapsamlı çalışmaların yapılması gerektiği düşünülmektedirThe widespread use of antibiotics and biocides in animal husbandry, both for the treatment and prevention of diseases and to increase the yield of animal foods, creates serious problems in terms of economy and public health. In this study, antibiotic and biocide resistance profile and antibiotic resistance and efflux pump genes were investigated in 77 Staphylococcus isolated from 132 meat and dairy products sold in various markets in Ankara. In this study, the effect of efflux pump inhibitor reserpine on antimicrobial resistance was investigated. The highest resistance was determined against tetracycline (24.7%) in Staphylococcus isolates. Of the 77 Staphylococcus isolated, 29 were found to be resistant to one or more antibiotics. These isolates were identified on the basis of species by BBL crystal Gram positive identification kit and 16S rDNA sequence analysis methods. Isolates showing high MICs to all of the biocides tested were available. Reserpine increased ampicillin sensitivity 25%, tetracycline sensitivity 27.8%, triclosan sensitivity 20.7%, povidone iodine sensitivity 34.5% in Staphylococcus isolates. The antibiotic resistance genes tetK (88,9%), bla (53,8%) were found to be high in 29 Staphylococcus isolates. Furthermore, among the efflux pump genes acrB (31%), norE (79.3%), sugE (44.8%), smr (86.2%), efrA (79.3%), efrB (72.4%), mdeA (82.8%), sepA (51.7%) are highly detected. The presence of antimicrobial resistance genes of Staphylococcus isolated from foods of animal origin presented in our study is an important threat to public health due to the risk that these resistance genes are transferred to pathogens and normal flora bacteria by chromosomal and extrachromosomal genetic materials. It is thought that animicrobial resistance from efflux pump can be reduced to a certain extent with the use of efflux pump inhibitor reserpine, but further studies are needed in this area
