    Digitalt erfaringsbibliotek

    Byggebransjen er preget av prosjektbasert arbeid, høy kompleksitet og stramme tidsfrister, noe som ofte fører til at verdifull erfaring går tapt. Effektiv erfaringsoverføring mellom prosjekter er derfor avgjørende for å sikre kvalitet, redusere feil og forbedre læring. Likevel viser praksis at erfaringsoverføring ofte skjer uformelt og lite systematisk, og at dokumentasjon er fragmentert eller utilgjengelig. Denne masteroppgaven undersøker hvordan et digitalt erfaringsbibliotek kan effektivisere overføringen av erfaringer i byggeprosjekter, gjennom å tilby en strukturert, søkbar og brukervennlig digital plattform for dokumentasjon og deling av prosjekterfaringer. Formålet med oppgaven er å vurdere hvordan et slikt bibliotek kan bidra til å redusere kunnskapstap, forbedre beslutningsgrunnlaget, støtte raskere opplæring og stimulere til kontinuerlig læring i virksomheten. Studien besvarer tre forskningsspørsmål: * Hvordan er dagens praksis i forhold til erfaringsoverføring i dag? * Hvilke fordeler kan et erfaringsbibliotek gi for byggebransjen? * Hvordan bør et erfaringsbibliotek designes for å støtte effektiv erfaringsoverføring og hvilke forutsetninger og implementeringsutfordringer kan den ha? Oppgaven benytter en kvalitativ metode, basert på semistrukturerte intervjuer med ansatte i ulike roller i byggeprosjekter. Analysen viser at dagens praksis preges av informasjonsfragmentering og manglende struktur. Informantene etterspør et lett tilgjengelig og søkbart system som gjør det mulig å finne relevante erfaringer raskt, og som samtidig er enkelt å bruke og integrert i arbeidshverdagen. Basert på funnene presenteres en skisse til hvordan et slikt system kan struktureres og organiseres for å møte disse behovene. Teoretisk bygger oppgaven på erfaringsoverføringskultur, Lean-filosofi, sosioteknisk systemteori (STS), Project Management Office (PMO) sin rolle i erfarinsgsoverføring, single-loop og double-loop learning, og SECI-modellen og Kolbs læringssirkel som er sentrale modeller innen kunnskapsledelsesfagområde. Disse teoriene og modeller belyser hvordan teknologi, organisasjonskultur og læring må samvirke for å sikre en vellykket erfaringsoverføring. Funnene viser at et digitalt erfaringsbibliotek kan redusere behovet for prøving og feiling, støtte opplæring, forbedre samarbeid på tvers av avdelinger og bidra til mer effektive prosjekter. Samtidig forutsetter en slik løsning at det finnes en aktiv delingskultur, tydelige rutiner og en forankring i ledelsen. Studien er avgrenset til intern kunnskapsdeling og omfatter ikke erfaringsoverføring til eller fra eksterne aktører. Metodisk er studien begrenset av et lite antall informanter og ulik rollefordeling, noe som svekker generaliserbarheten. Likevel gir oppgaven praksisnære innsikter i hvordan digitale løsninger kan forbedre erfaringsutveksling i prosjekter. Oppgaven gir et konkret rammeverk som kan danne grunnlag for videre forskning eller fungere som utgangspunkt for virksomheter som ønsker å utvikle eller teste et digitalt erfaringsbibliotek tilpasset egne behov. Funnene viser at et digitalt erfaringsbibliotek kan bidra til raskere onboarding av nyansatte, redusert risiko for gjentakelse av feil, mer effektiv beslutningsstøtte og bedre samarbeid på tvers av prosjekter og avdelinger. Oppgaven kombinerer teoretiske perspektiver med empiriske funn og identifiserte behov, og presenterer på dette grunnlaget et forslag til hvordan et digitalt erfaringsbibliotek kan designes og implementeres i praksis. Til tross for metodiske begrensninger viser funnene et tydelig potensial for å styrke læringskulturen og forbedre prosjektresultater gjennom systematisk og digitalisert erfaringsoverføring.The construction industry is characterized by project-based work, high complexity, and tight deadlines, which often leads to the loss of valuable experience. Effective transfer of experience between projects is therefore essential to ensure quality, reduce errors, and enhance learning. Nevertheless, practice shows that experience transfer often occurs informally and unsystematically, and that documentation is fragmented or inaccessible. This master’s thesis investigates how a digital experience library can improve the transfer of experience in construction projects by offering a structured, searchable, and user-friendly digital platform for documenting and sharing project experiences. The aim of the thesis is to assess how such a library can help reduce knowledge loss, improve the basis for decision-making, support faster onboarding, and stimulate continuous learning within the organization. The study answers three research questions: * What is the current practice in relation to experience transfer? * What benefits can an experience library offer the construction industry? * How should an experience library be designed to support effective experience transfer, and what prerequisites and implementation challenges might it face? The thesis employs a qualitative method, based on semi-structured interviews with employees in various roles in construction projects. The analysis shows that current practice is characterized by fragmented information and a lack of structure. The informants express a need for an easily accessible and searchable system that allows them to quickly find relevant experiences, and which is also easy to use and integrated into the daily workflow. Based on the findings, a draft is presented outlining how such a system can be structured and organized to meet these needs. Theoretically, the thesis draws on knowledge sharing culture, Lean philosophy, sociotechnical systems theory (STS), Project Management Office (PMO), single-loop and double-loop learning, the SECI model, and Kolb’s learning cycle, which are all central models within the field of knowledge management. These theories and models highlight how technology, organizational culture, and learning must interact to ensure successful experience transfer. The findings show that a digital experience library can reduce the need for trial and error, support training, improve collaboration across departments, and contribute to more effective projects. At the same time, such a solution requires an active culture of sharing, clear routines, and anchoring in management. The study is limited to internal knowledge sharing and does not cover experience transfer to or from external actors. Methodologically, the study is constrained by a small number of informants and varied role distribution, which limits generalizability. Nevertheless, the thesis provides practice-oriented insights into how digital solutions can improve experience sharing in projects. The thesis provides a concrete framework that can serve as a basis for further research or act as a starting point for organizations seeking to develop or test a digital experience library tailored to their own needs. The findings show that a digital experience library can contribute to faster onboarding of new employees, reduced risk of repeated mistakes, more effective decision support, and better collaboration across projects and departments. The thesis combines theoretical perspectives with findings from current practice and identified needs, and on this basis proposes how a digital experience library can be designed and implemented in practice. Despite methodological limitations, the findings show a clear potential to strengthen the learning culture and improve project outcomes through systematic and digitalized experience transfer

    Space and the Alteration of Warfare: Satellites and the war in Ukraine

    How has the proliferation of satellites altered warfare and the military power of states on Earth? This thesis studies the development of space asset systems and how they have affected the world. The study finds that satellites have come to play a significant part in national and transnational infrastructure and security. Threats against the integrity of these space-based systems therefore also run the risk of compromising essential functions of our daily lives. At the same time, attempts to legislate and regulate space have been persistently forwarded for decades. Success has however been limited, given that these regulations are usually formulated by international institutions with a highly limited ability to enforce them. Private businesses and commercial actors have also grown increasingly prominent in the space sector. This introduces additional stakeholders with an interest in maintaining peace in space, but it also comes with its own risks and worries. The analysis finds that the proliferation of satellites has increased insecurity across the world, due to a higher degree of vulnerability to threats, a lack of ability to enforce consequences, and a disproportionate degree of collateral damage. The precedent this sets for the future remains uncertain, however

    Effekt av tidlig og sen separasjon fra mor på opphold nær bingeskillet hos NRF-kalver

    Ku–kalv-samvær i melkeproduksjonen utvikles for å kunne tilfredsstille samfunnets ønsker samt imøtekomme nye og kommende krav om bedret dyrevelferd. Den vanligste praksisen per i dag er å skille ku og kalv kort tid etter kalving. Ettersom ku og kalv viser mer intense og mer vedvarende stressrespons ved separasjon etter forlenget samvær enn om de blir separert rett etter kalving, er det behov for separasjonsmetoder som kan dempe denne atferdsresponsen. Gradvis avvenning med nedtrapping av kontakt før full separasjon i tillegg til supplerende melk, viser seg å være med på å dempe separasjonsstress hos kalven, men optimal lengde og timing for gradvis avvenning er per nå uavklart. Det ble i denne studien antatt at kalver med lenger samvær med mor og kortere separasjonsfase, sammenlignet med kortere samvær og lenger separasjonsfase, ville bruke mer tid nær bingeskillet og utføre mer pacing i separasjonsfasene. I tillegg ble det sett på om kalvenes inntak fra melkefôringsautomaten hadde en sammenheng med opphold nær bingeskillet. Det ble registrert atferd fra videoopptak i et ku-drevet ku–kalv-system med 30 kalver (Norsk rødt fe) fordelt på 4 runder. De to behandlingene bestod av fire faser hvor kyrne hadde tilgang til kalvene i de tre første fasene. Kyrne i første fase hadde tilgang til kalvene i 24 t/d før redusering til 12 t/d (12-timers fasen) og 6 t/d (6-timers fasen). Første fase i den ene behandlingen varte i 4 uker (etter kalving), andre og tredje fase i 14 dager hver; tidlig separering/lang tilvenning (TL). Den andre behandlingens første fase varte i 6,5 uke (etter kalving), andre og tredje fase i 5 dager hver; sen separering/kort tilvenning (SK). Siste fase («ingen tilgang»-fasen), hvor kyrne ikke hadde tilgang til kalvene (0 t/d), bestod av 7 dager i begge behandlingene. Det ble registrert atferd i 5 døgn fra første separasjonsdag i 12- og 6-timers fasene, samt 7 døgn i «ingen tilgang»-fasen. Det ble ikke funnet forskjeller mellom behandlingene i 12-timers fasen eller 6-timers fasen. I «ingen tilgang»-fasen hadde SK-kalvene signifikant lenger gjennomsnittlig opphold nær bingeskillet (P = 0,001) sammenlignet med TL-kalvene, henholdsvis 5,4±1,6 minutter per time (gjennomsnitt ± SD) og 3,6 ± 1,1 minutter per time. Det ble kun registrert noen få tilfeller av pacing, og det ble ikke funnet sammenheng mellom melkeinntak og opphold nær bingeskillet. Det ble konkludert med at effekten av lenger gradvis separasjon som starter når kalven er yngre ser ut til å kunne dempe separasjonsstress sammenlignet med kortere gradvis separasjonsfase som starter når kalven er eldre. Pacing ser ikke ut til å være en god indikator for separasjonsstress. Det ble ikke funnet sammenheng mellom opphold nær bingeskillet og inntatt melkemengde fra melkefôringsautomat

    Deciphering ADA2 Deficiency: Creating an ADA2 deficiency model using CRISPR/Cas genome editing

    Mangel på adenosindeaminase 2 (DADA2) er en sjelden autoinflammatorisk lidelse forårsaket av bialleliske mutasjoner i adenosindeaminase 2 (ADA2) genet. Pasienter med DADA2 viser et bredt spekter av kliniske manifestasjoner, og tilstanden kan være livstruende. Sykdomsmekanismene til DADA2 er i stor grad ukjente og for å øke kunnskapen i dette feltet, hadde denne studien som mål å lage DADA2-cellemodeller og studere effekten av nedslått ADA2 genuttrykk på to sentrale immune celle mekanismer: proliferasjon av monocytter og makrofagpolarisering. For å gen redigere ADA2 ble guide RNA (gRNA) og CRISPR-assosiert protein 9 (Cas9) introdusert inn i THP-1 celler ved bruk av elektroporering. De gen redigerte cellene ble videre sortert ved bruk av fluorescensaktivert cellesortering (FACS), hvor til slutt fire av de utvalgte klonene viste en homozygot delesjon i ADA2-genet som er forventet å forårsake en leseramme forskyvning i budbringer-RNA (mRNA). En proliferasjon analyse som ble utført på ADA2-redigerte, ikke-sorterte celler ved bruk av Incucyte S3 viste at de redigerte cellene hadde en signifikant lavere proliferasjon sammenlignet med kontroll celler. Effekten av DADA2 på makrofagpolarisering ble testet på sorterte ADA2-redigerte celler og evaluert med spesifikke markører ved hjelp av kvantitativ PCR (qPCR). Cellene ble først differensiert til M0 og deretter polarisert til M1 og M2a makrofager. Resultatene indikerte at DADA2-cellemodeller viste en favorisert polarisering mot M1 makrofager. Eksperimentelle begrensninger oppsto, og ytterligere testing må gjøres for å trekke pålitelige konklusjoner

    Med kjøpesenter i sentrum: En historisk analyse av utviklingen av bygulvet i Fredrikstad og Ski, i lys av konkurranse fra kjøpesentrene

    I dagens urbane virkelighet bevitner vi en postindustriell kontekst der sentrum har fått en fornyet betydning som et sentralt knutepunkt for både kommersielle og kulturelle aktiviteter. Tidligere preget av industri, består sentrum nå av et mangfold av forretninger, restauranter, kafeer og møteplasser. Denne endringen representerer en ny æra i byutviklingen, der fokuset har flyttet fra produksjon til forbruk, med handel som en ledende aktivitet. Samtidig legges det stor vekt på kvalitetene på byrommene, med ønske om innbydende omgivelser og møteplasser for å fremme interaksjon I dette studiet har vi undersøkt endringer i bygulvet i Fredrikstad og Ski gjennom et historisk perspektiv, i lys av konkurranse fra kjøpesentre lokalisert i sentrum. Begge byene har kjøpesentre integrert i sentrum, etablert på 1980- og 1990-tallet. Inntoget av kjøpesentre til sentrumskjernen har ført til at flere sentrumsområder har tapt validitet. Likevel antyder enkelte studier at kjøpesentre som er integrert i sentrum kan ha en positiv innvirkning på bysentrum. Dette paradokset la grunnlag for vår forskning. For å analysere utviklingen av bygulvet fra før etableringen av kjøpesentrene og frem til i dag har vi studert tre hovedfaktorer. Vi har undersøkt endringer i vare- og tjenestehandel på bygulvet, samt hvordan gentrifisering og opplevelsesøkonomi har påvirket denne utvikling. Videre har vi vurdert den betydningsfulle rollen eierskap og koordinert aktørsamarbeid spiller for utforingen av bygulvet. Disse analysene er forankret i fem teoretiske perspektiver. Analysene avslørte betydelige forskjeller i utviklingen av bygulvet mellom Fredrikstad og Ski. I Fredrikstad så vi en økning i tjenestehandelen og en tydeligere overgang til opplevelsesøkonomi, mens forholdet mellom vare- og tjenestehandel i Ski var relativt stabilt over kartleggingsperioden. Tendenser til gentrifisering, gjennom revitalisering av sentrum, var tydeligere i Fredrikstad enn Ski. Dette kan forklares i manglende utvikling av Ski sentrum. Våre analyser indikerer også at eierskapsstruktur og koordinert eiendomsutvikling spiller en viktig rolle i utviklingen av bygulvet. Vi kan ikke med sikkerhet fastslå om endringene vi har observert skyldes kjøpesenteretableringene, men vi har støttet oss til kvalifiserte antakelser basert på våre funn. Studien indikerer at kjøpesentrene har påvirket utviklingen av bygulvet i de to byene, men det er viktig å erkjenne at de ikke har vært den eneste faktoren som har formet dette landskapet. Andre faktorer, som eiendomsutvikling, eierskap og samfunnsmessige endringer, har også spilt en rolle i denne utviklingen.n today's urban reality, we witness a post-industrial context where city centers have regained significance as central hubs for both commercial and cultural activities. Once dominated by industry, city centers now consist of a variety of shops, restaurants, cafes, and meeting places. This shift represents a new era in urban development, transitioning the focus from production to consumption, with commerce as a leading activity. Additionally, there is a strong emphasis on the quality of urban spaces, aiming to create inviting environments and meeting places that foster interaction. This study examines changes in the ground floors of Fredrikstad and Ski from a historical perspective, in light of competition from centrally located shopping centers. Both cities have shopping centers integrated into their downtown areas, established in the 1980s and 1990s. The influx of shopping centers into the city centers has led to the decline of several downtown areas. However, some studies suggest that shopping centers integrated into city centers may have a positive impact on urban centers. This paradox formed the basis for our research. To analyze the development of the ground floors from before the establishment of shopping centers to the present day, we have studied three main factors. We have examined changes in retail and service activities in the ground floors, as well as how gentrification and the experience economy have influenced this development. Furthermore, we have assessed the significant role that ownership and coordinated collaboration among stakeholders play in shaping the ground floors. These analyses are grounded in five theoretical perspectives. The analyses revealed significant differences in the development of the ground floors between Fredrikstad and Ski. In Fredrikstad, we observed an increase in service activities and a clearer transition to the experience economy, while the balance between retail and service activities in Ski remained relatively stable over the study period. Trends of gentrification, through the revitalization of the city center, were more pronounced in Fredrikstad than in Ski. This may be explained by the limited development of Ski's city center. Our analyses also indicate that ownership structure and coordinated property development play a crucial role in the development of the urban fabric. We cannot definitively attribute the observed changes to the establishment of shopping centers, but we have relied on qualified assumptions based on our findings. The study suggests that shopping centers have influenced the development of the urban fabric in the two cities, but it is important to recognize that they have not been the sole factor shaping this landscape. Other factors, such as property development, ownership, and societal changes, have also played a role in this development

    Samutråtning av ulike substrater i biogassproduksjon - En LCA studie av klimapåvirkning og biorestkvalitet

    Biogass regnes som en karbonnøytral gass og bringer med seg en rekke miljøfordeler ved at den kan erstatte fossile energikilder. Biogass kan benyttes til oppvarming, produksjon av elektrisitet eller drivstoff. Samtidig gir også biogassproduksjon et næringsrikt restprodukt; biorest. Biogassproduksjon har et stort potensial for å bidra til å nå klimamålene i Norge. I dag produseres det kun 0,6 TWh biogass i Norge, Lyng og Berntsen (2023) viser til at potensialet, med dagens teknologi, ligger på 5,5 TWh. Formålet med oppgaven har vært å analysere potensialet for å redusere klimaavtrykket fra biogassproduksjonen ved Fredrikstad vann-, avløp- og renovasjonsforetak (FREVAR KF), og samtidig se på hvilke mulige positive eller negative effekter ulik sammensetning av substrat kan ha å si på næringssammensetningen i bioresten. Biorest er i utgangspunktet et avfallsprodukt fra biogassproduksjonen. Riktig næringsinnhold kan bidra å øke verdien til biorest som produkt. FREVAR produserer biogass som i hovedsak benyttes til drivstoff, og avvannet biorest går til jordforbedring. Dagens produksjon har samråtning med avløpsslam og matavfall. I 2026 skal nytt avløpsrenseanlegg stå klart hos FREVAR. Avløpsrenseanlegget vil produsere biogass sammen med det eksisterende biogassanlegget, men avløpsslammet vil bli behandlet separat fra andre substrater. Dette vil frigjøre kapasitet i det eksisterende anlegget og gi økt mulighet for å utnytte andre typer substrat. Analysene er gjennomført i biogassmodellen Bio Value Chain og viser klimapåvirkningen fra biogassproduksjonen. Det er utført analyser av fem scenarioer, hvor scenario S1 er dagens produksjon, S2 inkluderer produksjon med det nye avløpsrenseanlegget, S2.1 er samråtning av matavfall og husdyrgjødsel fra storfe og svin, S2.2 er samråtning med matavfall og husdyrgjødsel fra storfe, svin og fjørfe, og S2.3 er kun matavfall. I tillegg er det utført beregninger for innhold av nitrogen (N), fosfor (P) og kalium (K) i bioresten fra de fem ulike scenarioene. Resultatene viser at samråtning med matavfall og husdyrgjødsel gir høyest klimagevinst av de analyserte scenarioene. Scenario S2.1 er det scenarioet som gir høyest netto klimagevinst, sammenlignet med dagens produksjon (S1) og utgjør 2 550 tonn CO2-ekv. årlig. De beregnede N-P-K-verdiene viser at scenario S2 gir mest næringsrikt substrat, dernest scenario S1. Verdiene for N-P-K varierer ut ifra mengde og type substrat. I beregningene kommer det tydelig frem at det er høyest andel av nitrogen i alle scenarioene. Biogassproduksjonen ved FREVAR har et betydelig potensial for å redusere klimagassutslipp og produsere verdifull biorest. Samråtning med matavfall og husdyrgjødsel kan gi økt klimagevinst og økt kvalitet på biorest. Det nye avløpsrenseanlegget åpner for muligheten til å optimalisere substratsammensetningen og ytterligere øke bærekraften til biogassproduksjonen ved FREVAR.Biogas is considered a carbon-neutral gas and offers several environmental benefits by replacing fossil fuels. Biogas can be used for heating, electricity generation, or fuel. Biogas production also generates a nutrient-rich digestate. Biogas production has great potential to contribute to achieving Norway's climate goals. Today, only 0,6 TWh of biogas is produced in Norway, while Lyng og Berntsen (2023) show that the potential, with current technology, is 5,5 TWh. The objective of this project was to analyze the potential to reduce the climate footprint of biogas production at Fredrikstad vann-, avløp- og renovasjonsforetak (FREVAR KF), while also examining the possible positive or negative effects of different substrate compositions on the nutrient composition of digestate. Digestate is primarily a waste product from biogas production. Proper nutrient content can help to increase the value of digestate as a product. FREVAR produces biogas that is mainly used for fuel, and dewatered digestate is used for soil improvement. Current production co-digests sewage sludge and food waste. In 2026, a new wastewater treatment plant will be ready at FREVAR. The wastewater treatment plant will produce biogas together with the existing biogas plant, but the sewage sludge will be treated separately from other substrates. This will free capacity in the existing plant and provide increased opportunities to utilize other types of substrates. The analyses were carried out in the biogas model Bio Value Chain and show the climate impact of biogas production. Analyses were performed for five scenarios, where scenario S1 is current production, S2 includes production with the new wastewater treatment plant, S2.1 is co-digestion of food waste and manure from cattle and swine, S2.2 is co-digestion with food waste and manure from cattle, swine, and poultry, and S2.3 is only food waste. In addition, calculations were performed for the content of N-P-K in the digestate from the five different scenarios. The results show that co-digestion with food waste and manure gives the highest climate gain of the analyzed scenarios. Scenario S2.1 is the scenario that gives the highest net climate gain, compared to current production (S1) and amounts to 2 550 tonnes CO2-eq. annually. The calculated N-P-K values show that scenario S2 gives the most nutrient-rich digestate, followed by scenario S1. The N-P-K values vary depending on the amount and type of substrate. The calculations clearly show that there is highest proportion of nitrogen in all scenarios. Biogas production at FREVAR has significant potential to reduce greenhouse gas emissions and produce valuable digestate. Co-digestion with food waste and manure can lead to increased climate gain and improved quality in digestate. The new wastewater treatment plant opens a possibility of optimizing substrate composition and further increasing the sustainability of biogas production at FREVAR

    Phase Segregation Mapping in Perovskite Solar Cells Using Hyperspectral Photoluminescence Imaging

    Hybrid organic-inorganic lead halide perovskites represent a class of novel solar cell materials that have recently gained interest due to their excellent optoelectronic properties. However, their poor stability and fast degradation hinder commercialization. This work investigates one instability mechanism, light-induced phase segregation, by analyzing hyperspectral photoluminescence (HSPL) images of mixed-halide perovskite solar cells. The present study aims to use HSPL imaging and accompanying data analysis techniques to map light-induced phase segregation in mixed-halide perovskite solar cells. To achieve this, planar regular (n-i-p) perovskite solar cells were fabricated in REALTEK’s laboratories. HSPL images of these cells were then analyzed using data analysis pipelines comprising some preprocessing steps in addition to principal component analysis (PCA) and multivariate curve resolution (MCR). Supplementary X-ray fluorescence (XRF) images of the samples were also taken in an attempt to assess whether the HSPL images can be used for mapping phase segregation-caused regions of higher and lower concentrations of the perovskite’s halide species (iodide and bromide). The samples’ photoluminescence (PL) spectra comprise several peaks and shoulders that may originate from phases of different halide stoichiometries. However, it can not be concluded that the observed PL signals are caused by phase segregation since the XRF images from the present XRF setup could not be used for validation. Also, the literature suggests that similar spectral features may be caused by other mechanisms, like re-absorption effects. A thorough analysis of the origins of the various PL signals must be conducted, and a suitable characterization method for validation of the HSPL analysis must be in place before the method can be developed further. Since the literature suggests that the iodide-rich domains are small compared to the resolutions of the present XRF and HSPL setups, increasing the resolution may be necessary to facilitate the localization of such domains

    The urgency of the green transition is undeniable. As the global climate crisis looms, businesses face mounting pressure to address sustainability. Efforts to promote sustainable development have been steadily increasing since the Brundtland report in 1987. In Norway, companies are coping with new regulations and heightened expectations from stakeholders. Among these businesses, small and medium-sized Enterprises (SMEs) play a crucial role in realizing national sustainability goals. However, despite their significance, SMEs still struggle to fully integrate environmental and social considerations into their operations. The objective of this study is to explore the strategies employed by SMEs in Viken to overcome sustainability-related barriers and, more specifically, to focus on identifying those that surpass mere compliance. It aims to address the following research question: How do SMEs in Viken currently respond to the changing regulatory environment and increased demand from stakeholders, and what are the implications of these responses? To achieve this, an inductive qualitative method was employed, based on an exploratory research design. Eight semi-structed, in-depth interviews were conducted with sustainability officers in SMEs and sustainability advisors in Viken county. The findings indicate that the main barriers that SMEs in Viken face are a lack of specialized expertise in sustainability, as well as a general lack of awareness and understanding throughout the organization. In addition, all the SMEs experience challenges with the regulations due to a perceived lack of standardization and framework. Nevertheless, SMEs are adopting strategies beyond mere compliance, focusing on internal measures such as organizational involvement, collaboration, and investing in sustainability expertise. The role of a dedicated Sustainability Officer emerges as crucial, but challenges remain in securing management support and avoiding unrealistic expectations. Proactive approaches, including sustainable innovation, are highlighted as valuable for SMEs to differentiate themselves and achieve long-term sustainability goals. The study underscores the importance of prioritizing internal measures as a foundation for successful sustainability integration.The urgency of the green transition is undeniable.     Samspillet mellom LPMOer og cellulaser under enzymatisk nedbrytning av cellulose : effekter av tid, lignin, lys og substratkonsentrasjon

    The recent discovery of lytic polysaccharide monooxygenases (LPMOs) marked a paradigm shift in our understanding of enzymatic saccharification of lignocellulosic biomass. Under aerobic conditions, interactions between oxygen and redox-active components such as ascorbic acid or lignin produce the LPMO co-substrate H2O2. The presence of H2O2 is pivotal for maintaining LPMO activity during saccharification processes, but in situ production is challenging to regulate. This is problematic since H2O2 accumulation in the system may lead to reactive oxygen species production and inactivation of the enzymes. LPMOs’ flat active site surface facilitates direct oxidation in crystalline cellulose, which makes the substrate accessible to classical hydrolytic cellulases. However, the interplay between cellulases and LPMOs is still not fully understood, which restricts practical applications. The aim of this thesis was to improve our understanding of the LPMO-cellulase interplay by investigating the effects of time, light, lignin, and substrate concentration on LPMO activity and saccharification performance. This study is based on four research articles: In Paper I, a positive effect of LPMO pretreatment on the productive binding capacity of a reducing end cellulase, TlCBHI, was demonstrated. Despite the fact that LPMO inactivation occurred before the first time point (5 h), a pronounced enhancement of TlCBHI's productive binding capacity on LPMO-pretreated cellulose was observed after 24 h. This indicates that the LPMO cleavage of the crystalline cellulose does not directly serve as new access points for cellulases but that this is followed by a non-enzymatic amorphization that makes the substrate accessible for cellulase activity. In Paper II, the importance of maintaining LPMO activity throughout the reaction to ensure efficient cellulase activity, especially at high substrate loadings, was demonstrated. The positive effect of including LPMOs in a cellulase cocktail increased with dry matter concentrations and reaction time. The impact of LPMO regioselectivity varied depending on the substrate, but a clear increase in cellulose conversion was observed when combining C1 and C4 LPMO activity. The study also indicates that chelating of free copper with EDTA might hinder transition metal-induced side reactions following LPMO inactivation. In Papers III and IV, light-exposed lignin was shown to promote LPMO activity on cellulose by improving in situ generation of H2O2 compared to similar reactions in the dark. These studies further demonstrated that the H2O2 production and thus LPMO activity could be controlled by lignin concentration, light intensity, and light-specific wavelengths. The findings of Paper III indicate that when lignin is exposed to light, it enhances LPMO activity by converting O2 to H2O2, most likely via O2•-. In contrast, the results in Paper IV showed that light exposure of lignin-containing reactions negatively affected cellulose saccharification by cellulolytic enzyme cocktails. Interestingly, LPMO activity could help mitigate this negative effect on the glucose yield by their productive H2O2 consumption, delaying the accumulation of reactive oxygen species and subsequent enzyme inactivation.Den nylige oppdagelsen av lytisk polysakkarid monooxygenaser (LPMOer) markerte et paradigmeskifte i vår forståelse av enzymatisk sakkarifisering av lignocellulose. Under aerobe forhold blir LPMOenes ko-substrat, H2O2, produsert gjennom interaksjoner mellom oksygen og redoksaktive komponenter som askorbinsyre eller lignin. Tilstedeværelsen av H2O2 er avgjørende for å opprettholde LPMO-aktiviteten under enzymatisk sakkarifisering, men in situ produksjon av H2O2 er utfordrende å regulere. Dette er problematisk siden akkumulering av H2O2 i systemet kan føre til produksjon av reaktive oksygenarter og inaktivering av enzymene. LPMOenes flate aktive sete tillater direkte oksidasjon i krystallinsk cellulose og gjør dermed substratet mer tilgjengelig for klassiske hydrolytiske cellulaser. Imidlertid er samspillet mellom cellulaser og LPMOer fortsatt ikke helt forstått, noe som begrenser praktiske anvendelser. Målet med denne avhandlingen var å forbedre forståelsen vår om samspillet mellom LPMOer og cellulaser ved å undersøke effekter av tid, lignin, lys-eksponering og substratkonsentrasjon på LPMO-aktivitet og glukose-utbytte. Denne studien er basert på fire forskningsartikler: I Artikkel I ble det vist at LPMO-forbehandling økte den produktive bindingskapasiteten til TlCBHI, en cellulase med affinitet for reduserende celluloseender. En markant økning i den produktive bindingskapasiteten til TlCBHI var synlig etter 24 t med LPMO-forbehandling, og dette til tross for at LPMOene var inaktivert allerede etter 5 t. Dette tyder på at LPMO-kutt i den krystallinske celluloseoverflaten ikke direkte fungerer som nye tilgangspunkter for cellulasene, men at LPMO-oksideringen er etterfulgt av en ikke-enzymatisk dekrystallisering som gjør substratet mer tilgjengelig for cellulase-aktivitet. I Artikkel II ble det vist at stabil LPMO-aktivitet gjennom hele reaksjonsforløpet fremmer cellulase-aktivitet, spesielt ved høye substratkonsentrasjoner. Den positive effekten av å inkludere LPMO i cellulase-cocktailer økte i takt med substratkonsentrasjonen og gjennom reaksjonsforløpet. Effekten av C1- vs C4-aktive LPMOer varierte avhengig av substratet, men en tydelig økning i glukose-utbytte ble observert ved å kombinere C1- og C4-aktive LPMOer. Studien viste også at chelatering av fri kobber med EDTA kan forhindre sidereaksjoner med innskuddsmetaller som en følge av LPMO-inaktivering. I Artikkel III og IV ble det vist at lys-eksponering av lignin fører til økt in situ H2O2-produksjon og dermed økt LPMO-aktivitet sammenlignet med reaksjoner i mørket. Disse studiene viste også at H2O2-produksjonen og LPMO-aktiviteten kunne kontrolleres ved hjelp av ligninkonsentrasjon, lysintensitet og lys-spesifikke bølgelengder. Artikkel III viste at lyseksponering av lignin økte omdannelsen av O2 til H2O2, mest sannsynlig via O2•-, og ga derfor økt LPMO-aktivitet. Samtidig viste resultatene i Artikkel IV at lys-eksponering av reaksjoner med lignin hadde en negativ påvirkning på cellulase-aktivitet. Tilstedeværelsen av LPMO kunne hjelpe til med å motvirke den negative effekten på glukoseutbyttet til cellulasene. Dette er mest sannsynlig et resultat av LPMOens konsumering av H2O2, som vil forsinke produksjonen av reaktive oksygenarter og enzym inaktivering
