Cytotoxic, Antioxidant, and Anti-Genotoxic Properties of Combretastatin A4 in Human Peripheral Blood Mononuclear Cells: A Comprehensive In Vitro Study
Despite significant advances in drug discovery and the promising antitumor potential of combretastatin A4 (CA-4), which selectively targets rapidly dividing cancer cells, CA-4’s effects on non-dividing human cells, such as peripheral blood mononuclear cells (PBMCs), remain unclear. The aim of this study is to evaluate the in vitro bioactivity of CA-4 in human PBMCs, focusing on its antigenotoxic and antioxidant properties, while comparing its cytotoxic potency against PBMCs, cancer cell lines (JAR and HeLa), and the normal trophoblast cell line HTR-8/SVneo. Cell viability and metabolic activity were evaluated using the MTT assay. ROS production in PBMCs was measured using the H2DCFDA assay, and DNA damage was assessed using the Comet assay. CA-4 showed cytotoxicity in PBMCs and HTR-8/SVneo cells at concentrations above 200 µM, while cancer cells, JAR and HeLa, showed cytotoxicity at 100 µM and 1 µM, respectively. CA-4 also reduced ROS levels in PBMCs under oxidative stress and showed antioxidant effects at concentrations from 1 to 200 µM. In addition, CA-4 showed antigenotoxic effects against H2O2-induced DNA damage in PBMCs at concentrations of up to 1 µM. CA-4 exhibited lower cytotoxicity in human PBMCs compared to cancer cells, inhibited ROS production, and showed antioxidant and antigenotoxic properties, providing insight into its potential therapeutic efficacy and safety
Antibacterial electrospun poly(ε-caprolactone) nanofiber mats loaded with cefazolin and meropenem for wound healing
Nanofiber wound dressings represent a promising approach to wound care, offering anadaptable platform for promoting faster and more effective wound healing. The goal of thisresearch was the development of poly(ε-caprolactone) (PCL) nanofiber mats with incorporatedantibiotics cefazolin (CEF) and meropenem (MER) for wound healing treatments.Nanofiber mats based on PCL (8 wt.% PCL prepared in 2,2,2-trifluoroethanol (TFE)) withcefazolin or meropenem (10 % of the polymer's weight) were produced using the blendelectrospinning method. For comparison, PCL nanofibers without antibiotics were prepared bydissolving PCL pellets in TFE. The electrospinning parameters were a flow rate of 2.7 ml/h forPCL/CEF and 3 ml/h for neat PCL and PCL/MER solutions. The distance from the collector was10 cm, and the voltage was 29 kV for all solutions.In the processed samples, cefazolin and meropenem maintained their biological activity anddid not react with the PCL. The drug release profiles showed that the PCL/CEF released about60 % of the drug, while the PCL/MER released about 40 % after 48 hours of incubation inMueller-Hinton broth. Both PCL/CEF and PCL/MER nanofiber mats showed similar growthdelays of Gram-positive clinical strains (S. aureus and S. epidermidis) compared to neat PCL.However, PCL/MER nanofiber mats efficiently inhibited the growth of Gram-negative clinicalstrains (E. coli, Klebsiella, Enterobacter spp., and Acinetobacter spp.). The effective antibacterialaction of PCL/MER nanofiber mats could minimize infection-related inflammation, reducingthe risk of complications like delayed healing, sepsis, and chronic wound formation.The overall results of this study have shown that novel PCL nanofiber mats could be used as arelevant drug scaffold, keeping the wound infection-free and promoting faster and moreeffective tissue repair

The complex responsibility of national public health associations – a case from Serbia
The rapidly evolving landscape of public health challenges requires national public health associa-
tions (NPHA) to take leadership and play a multifaceted role (1). NPHAs usually serve as primary
non-governmental advocates for population health and manage complex intersections of policy,
education, and community engagement to address both traditional and emerging public health
threats. One of their key roles is to shape strategies that promote equitable access to healthcare,
thereby significantly impacting public health outcomes. NPHAs exert their influence across local,
regional, and global levels, advancing global health agendas.
The establishment of the Serbian Public Health Association (SPHA) in 2003 was a significant mile-
stone in the country’s public health history. SPHA added new value to traditional partnerships for
public health, led by the Network of Institutes of Public Health, along with the Serbian Medical
Chamber, academic institutions, United Nations agencies, Serbian Medical Society, civil society
organizations, and interested groups and individuals. SPHA has been active in various public health
areas, including strengthening community resilience by supporting families and visiting nurses,
promoting the response to the COVID-19 pandemic, providing continuous medical education, and
facilitating public dialogue on health reforms.
Today, SPHA is a national, not-for-profit association of current and future public health professionals
and other individuals devoted to promoting scientific evidence and multidisciplinary collaboration
in health promotion and protection (2). SPHA is a permanent member of the European Public
Health Association and the World Federation of Public Health Associations (3). It brings together 96
members of public health practitioners from various backgrounds and affiliations, working together
to protect and improve the health of all individuals and reduce health inequalities
Research proposal: investigating effectiveness of training videos on medical professional communication – relying on sfl analysis and politeness theory
The proposal aims to document whether training videos on medical professional communication may be effective in identifying and improving the communication skills of doctors and other healthcare practitioners with patients. The strategy may be of particular importance for LSP students, medicine students, or pharmacy students who are non-native English speakers. After following a typical educational video, and based on their knowledge of communication in healthcare, they will need to identify the key semiotic resources that develop assertiveness and empathy in communication. The investigation should rely on SFL, with an emphasis on speech act theory and politeness theory
Plant-Based Sunscreen Emulgel: UV Boosting Effect of Bilberry and Green Tea NaDES Extracts
Natural deep eutectic solvents (NaDES) were employed for the extraction of bilberry and green tea leaves. This study explored the incorporation of these NaDES extracts into various carrier systems: hydrogels, emulsions, and emulgels stabilized with hydroxyethyl cellulose or xanthan gum. The results demonstrated that, when combined with synthetic UV filters, the NaDES extracts significantly enhanced the SPF and improved the antioxidant properties of the formulation. Although NaDES extracts cannot fully replace synthetic UV filters (homosalate, ethylhexyl methoxycinnamate, and benzophenone-4), they can serve as effective UV boosters, significantly enhancing the SPFs of formulations containing UV filters. Hence, the SPF of the formulation could be improved without increasing the concentrations of synthetic filters. Moreover, NaDES extracts, unlike UV filters, significantly increased the antioxidant potential of the formulations. Among the carriers, hydrogels with xanthan gum and emulgels with hydroxyethyl cellulose achieved the highest SPFs when containing both NaDES extracts and synthetic filters. A texture analysis further revealed that the NaDES extracts positively impacted the mechanical properties of the formulations by increasing their cohesiveness, thus enhancing their physical stability under mechanical pressure. These findings pave the way for further research into NaDES-based formulations, including in vivo testing, to optimize and confirm their efficacy on human skin and validate NaDES extracts as eco-friendly ingredients in cosmetics, with antioxidant and UV boosting potential
The effect of nonionic Brij 35 micelles on the ionization of some drugs used in the treatment of alergic reactions
The pKa values of four compounds used in the treatment of allergic reactions, bilastine, cetirizine,
montelukast and rupatadine have been potentiometrically determined in micellar solutions of
nonionic surfactant Brij 35. Nonionic micelles affected the ionization of investigated compounds
from -1.97 to +2.51 pKa units comparing to “pure” water. The most significant changes in
distribution of the equilibrium forms are observed at pH values of biopharmaceutical importance
which may indicate on potential interactions with noncharged but polar molecules under the

Quinoline-based thiazolyl-hydrazones target cancer cells through autophagy inhibition
Heterocyclic pharmacophores such as thiazole and quinoline rings have a significant role in medicinal chemistry. They are considered privileged structures since they constitute several Food and Drug Administration (FDA)-approved drugs for cancer treatment. Herein, we report the synthesis, in silico evaluation of the ADMET profiles, and in vitro investigation of the anticancer activity of a series of novel thiazolyl-hydrazones based on the 8-quinoline (1a–c), 2-quinoline (2a–c), and 8-hydroxy-2-quinolyl moiety (3a–c). The panel of several human cancer cell lines and the nontumorigenic human embryonic kidney cell line HEK-293 were used to evaluate the compound-mediated in vitro anticancer activities, leading to [2-(2-(quinolyl-8-ol-2-ylmethylene)hydrazinyl)]-4-(4-methoxyphenyl)-1,3-thiazole (3c) as the most promising compound. The study revealed that 3c blocks the cell-cycle progression of a human colon cancer cell line (HCT-116) in the S phase and induces DNA double-strand breaks. Also, our findings demonstrate that 3c accumulates in lysosomes, ultimately leading to the cell death of the hepatocellular carcinoma cell line (Hep-G2) and HCT-116 cells, by the mechanism of autophagy inhibition
Unlocking the Unique Potential of Thymus pannonicus: Exploring the Efficacy of Supercritical CO2 Extraction, with and Without Pre-Treatments
Since ancient times, many plant species within the Thymus genus have been used due to their numerous health benefits, such as antimicrobial, anti-inflammatory, antiseptic, or diuretic activity. While many of the species within this genus were well known and described, Thymus pannonicus All. or Pannonian thyme remains relatively unexplored despite its unique chemical composition and activity. T. pannonicus is a small shrub, spread over central and eastern Europe, with a diverse, location-dependent chemical composition. Within this study, the citral chemotype of T. pannonicus was used, which up to our knowledge was found and described only in Northern Serbia. Therefore, in order to explore the composition of T. pannonicus low-polar extracts and to present a preliminary insight into their bioactivity, supercritical carbon dioxide (ScCO2) extraction was applied (with and without pre-treatments), followed by GC-MS analysis. Different ScCO2 extraction pressures were tested (100, 150, 200, 250, and 300 bar) in order to determine the most suitable conditions for the extraction of T. pannonicus’s low-polar fraction. Additionally, in order to maximize the yield of the extraction, two types of ScCO2 extraction pre-treatments were investigated (enzymatic and microwave). The highest extraction yield obtained from untreated plant material was 3.01% (w/w), and it was obtained at conditions of 150 bar and 40 °C, while the 4% (v/w) enzymatic pre-treatment, with the same conditions, provided a yield of 3.89%. For all of the obtained extracts, the GC-MS analysis showed that oxygenated monoterpenes and sesquiterpenes were the two most dominant groups with principal bioactive compounds such as (E)-citral (18.95–38.17%), (Z)-citral (6.68–14.66%), β-bisabolene (8.2–14.4%), and nerol (6.08–9.67%). The extracts that exhibited the highest concentration of principal bioactive compounds ((Z)-citral, and (E)-citral) were further analyzed for anticancer potential, using short- and long-term cell viability observations on liver cancer cells
