Replisome loading reduces chromatin motion independent of DNA synthesis
Chromatin has been shown to undergo diffusional motion, and during active gene transcription the RNA
polymerase activity negatively affects chromatin motion. However, the relationship between chromatin motion
and other genomic processes remains unclear. Hence, we set out to label the DNA directly in a sequence
unbiased manner and recorded labeled chromatin dynamics in interphase human cells expressing GFP-tagged
PCNA, a cell cycle marker and core component of the replication machinery. We detected decreased
chromatin mobility during S-phase compared to G1 and G2 phases using automated particle tracking. To gain
insight into the organization and dynamics of the genome during DNA replication, we analyzed labeled
chromatin domain size and motion in replicating cells. By correlating chromatin mobility with proximity to sites
of active DNA synthesis, we show that chromatin motion is locally constrained at the sites of DNA replication.
Furthermore, inhibiting DNA synthesis activity leads to increase loading of DNA polymerases and further
restricts local chromatin motion. The polymerases under stress no longer reel DNA through, as they normally
do during DNA synthesis, which may explain the further restriction of chromatin motion and the fact that
loading the helicase/polymerase already restricts it during active DNA synthesis. We, therefore, propose that
increased polymerase loading but not their catalytic activity reduces genome dynamics and its accessibility
and interaction with other genomic regions.Prepublication with Access for Reviewe

Spray_Average - Direct Injecion for Cat-Heating Operating Points
The spray was visualized using high-speed images of the Mie scattering of the fuel droplets. The liquid fuel was illuminated with a volumetrically expanded laser beam through the piston mirror. The same setup of the PIV was used and only the laser cutting optics were adapted according to the expansion of the laser. The images were also taken with the Phantom V2640 high-speed camera, but in single-frame mode with a Sigma lens (focal length 180 mm, f# 5.8). The images were then imported into Matlab and calibrated using a calibration target (LaVision). The motor components were masked and the Mie scattering images of the spray were binarized with a threshold value. Statistical analyses were then carried out, such as the penetration depth of the spray.1.

HYPA-r: Hydrodynamic patterns dataset (raw data)
The HYPA-r dataset contains raw image data of a variety of gravure printed
patterns. The raw image data is created by high-resolution scanning of printed
samples from industrial gravure printing web-presses. Pattern types, which are
represented in the dataset, are dot patterns, finger patterns and mixed
patterns. The HYPA-r dataset mainly aims at data-driven analysis of
hydrodynamic pattern formation in gravure printing.
HYPA-r stands for **hy** drodynamic **pa** tterns. The suffix -r stands for
**r** aw data.
To get a quick impression of the dataset, two example-images are provided
separately for download, named 'G1-01_WCPvbam_V005_ESA0_L01.tif' and
'G1-01_WCPvbam_V005_ESA0_S01.tif'.
Metadata for the HYPA-r dataset can be found in the dissertation of Pauline
Rothmann-Brumm (2023) and in the provided README-file.
