Gonadal absence of ceh-22: Degradation of CEH-22 in the gonads of Caenorhabditis elegans
The reproductive system of hermaphrodite Caenorhabditis elegans (C. elegans) comprises two gonadal arms, each with a terminating distal tip cell (DTC), connected by a common uterus. The DTC has two functions: leader and niche. In C. elegans, ceh-22/nkx2,5 has been found to work together with Wnt signaling to determine the fates of the DTC (Lam et al., 2006). This is an indication that ceh-22 aids in the specification of DTCs. Our research aims to determine the specific function of ceh-22 in the gonad of both male and hermaphrodite C. elegans. This will be accomplished by observing the effect of the absence of CEH-22 protein in the tissue. However, due to the gene’s essentiality in various aspects of the development of C. elegans, the deletion of the gene entirely will result in the death of the animals. To avoid this, we utilize a Degron toolkit to target GFP-tagged proteins for ubiquitination and degradation
Rural, Gifted Student Experience in Appalachian Kentucky
Current literature shows that gifted students\u27 experiences often differ from nongifted peers. Similarly, research shows that rural students’ experiences are unique compared to nonrural peers. This study hopes to address the gap in literature on experiences of students at the intersection of giftedness and rurality with the question: How do rural, gifted students in Appalachian Kentucky experience and perceive K-12 schooling? Rural, gifted students exist in the overlap between two unique groups. Using a multi-case study, participants came from three separate school districts in a region of Appalachian Kentucky, with each district being a separate case. Data collection included observations and interviews of participants across three separate school-based settings. Data were analyzed within cases to start, then compared across cases. The data analysis yielded three central themes: student experience is shaped by gifted stereotypes and expectations, industry shapes academic programming and student goals, and students find a network of support from teachers, friends, and family.
These findings confirm current literature that says students are more goal-oriented when they have strong relationships with teachers. This provides a prospective method to expand upon current literature, using industry-driven pathways as a method to engage the greater community as a resource for students. Future studies might focus on repeating the study for nongifted, rural counterparts in the same schools or nonrural, gifted counterparts, to determine if the experiences of the rural, gifted students were unique. Additional inquiries should be made on how to support and develop teachers’ flexibility and capacity as student champions

Please Don’t Teach to the Writing Test: Suggestions to Improve Writing Instruction
This essay urges teachers and school administrators to move away from the restrictive five-paragraph essay format and the emphasis on standardized writing tests like the Kentucky Standards Assessment (KSA). Based on personal teaching experience and insights from composition and rhetoric scholars, the essay highlights how these practices stifle authentic writing development and fail to prepare students for college and career writing demands. The author advocates for a shift in writing instruction towards focusing on audience and purpose, in line with Common Core Standards and the National Council of Teachers of English\u27s recommendations. By drawing parallels to the successful shift in math education towards conceptual understanding, the essay calls on educators to adopt a more meaningful, audience-driven approach to writing instruction, fostering students\u27 real-world writing skills and preparing them for future success
Simplifying 3D Printing Using Natural Language Processing
3D printing is a crucial technology with many applications in different fields. To be able to use this technology to its full extent, expertise in computer aided design (CAD) technology and 3D modeling is required. Many people interested in 3D printing do not have this expertise and thus cannot build custom models, and are consequently forced to buy them instead. Natural language processing (NLP) is one tool that can vastly simplify 3D modeling for those lacking CAD experience. Using NLP, someone can simply dictate what they want to be able to print, and a computer can then build a 3D model for them. Furthermore, NLP could be used to create simple machine instructions, known as gcode, that direct a 3D printer to produce a particular shape or figure. To test the efficacy of NLP for gcode generation, an application was built to parse English and then generate gcode for the specified model. While this app can only make simple shapes, it shows great potential in using NLP to simplify the 3D printing process
Cyber and Tech-Based Partner Stalking in Jefferson County
Since stalking laws were enacted in the 1990s, they have been historically difficult to enforce. As less and less of our lives become private due to our ever-present devices, stalkers frankly have more access to their targets than ever before. The tenuous nature of stalking laws and the growing resources for stalkers place targets of stalking in a very precarious situation: the legal system has never been adequately equipped to handle stalking cases, and local justice systems do not have the resources to handle tech-based crimes. Through interviews with actors in the criminal justice system, this presentation considers how these systemic issues manifest and seeks actionable solutions in Jefferson County, Kentucky
The Bluey Effect: Using the Children’s Cartoon to Connect Social-Emotional Learning and Core Content in the Classroom
In recent years, there has been a push for incorporating SEL, or social-emotional learning, into the classroom. As important as this movement is, many teachers have expressed concerns about their ability to intentionally address SEL concepts during the school day. This stems from a lack of time, accessible SEL programs, and training for teachers and staff. During this time, studies have also shown that as students go through school into higher education, rates of engagement tend to drop significantly. This has only increased after the COVID-19 pandemic. One way to address both of these challenges is to create a way to engage students, teach SEL skills, and not take away time from important content-related teaching. This project outlines just how that can be achieved by using the children’s cartoon Bluey as the missing puzzle piece. In this project, a unit plan has been created to propose possible lessons framed around Bluey episodes that touch on important SEL topics while also integrating content-related knowledge
Preoperative Predictors for Outcomes Following Total Knee Arthroplasty for Individuals with Degenerative Arthritis
Arthritis is a major concern across the globe and in the United States. This joint disease is a large contributor to debility and loss of function as well as increasing medical costs in this country. While conservative treatments are available for the treatment of arthritis, joint replacement is an option for those for whom these treatments are not successful. Many factors contribute to outcomes following a total knee arthroplasty. Some of the factors previously examined are patient comorbidities, psychological factors, rehabilitation following the procedure, and preoperative functional status, but to date, no research examines how the time of year that surgery is performed affects patient outcomes following a total knee arthroplasty. Clinically, patients often comment they feel stiff or have more trouble with functional tasks on cold, damp days. The purpose of this retrospective study was to use a multivariate model to analyze preoperative function, pain, quadriceps strength, range of motion, and time of year that surgery is performed, to predict patient functional outcomes following total knee arthroplasty. The primary question examined is if these preoperative variables predict functional outcomes following rehabilitation after knee replacement. A retrospective chart review using a random sample was conducted, and charts were stratified by season in which surgery was performed to ensure an equal number of cases per season. All data collected were then analyzed to evaluate outcomes following total knee replacement. Results from the study show that preoperative pain, strength, and range of motion do not influence outcomes following total knee arthroplasty. The season surgery is performed and preoperative function predicted functional outcome after total knee replacement. Comprehensive management of total knee replacement, considering both the timing of surgery and the patient’s function prior to surgery, is indicated. Findings suggest that outcomes following total knee replacement are more favorable when surgeries are performed in
the spring and fall compared to winter and summer. Environmental conditions, seasonal activity levels, and possibly psychological factors related to the time of year may influence recovery. Findings also reveal the importance of teaching future clinicians to recognize how factors beyond biomechanics can impact recovery following total knee replacement. Integrating this evidence into educational curricula encourages a more holistic approach to patient care during the total joint replacement process
APRN Transition to Primary Care Practice Program: A Quality Improvement Project
Transitioning to independent practice presents challenges for advanced practice registered nurses (APRNs) new to primary care. This quality improvement project addressed gaps in training and support through a structured Transition to Primary Care Practice (TTPCP) program across multiple primary care clinics in the southeastern United States. Over 12 months, the program provided weekly didactic sessions, monthly specialty rotations, and bi-weekly feedback. Guided by the Plan-Do-Study-Act framework, the initiative tracked structure, process, and outcome measures. Among 20 participants, attendance and schedule ramp-up reached 100%. Knowledge scores improved by 44%, and self-assessments increased by 23%. All assessed participants were rated Proficient or higher at 12 months. These findings support the integration of structured transition programs to enhance APRN readiness, retention, and care quality

The growth of women’s professional sports in the United States has long been tied to the level and quality of media coverage these leagues receive. This paper examines how media exposure impacts the success of women\u27s sports, with a focused case study on Racing Louisville FC. By analyzing Racing Louisville’s media presence alongside its attendance numbers, this study highlights how limited coverage can stunt fan engagement and broader support. In addition, the paper emphasizes the importance of fair comparisons, suggesting that women\u27s leagues should be evaluated at similar stages of development as men’s leagues, particularly when assessing success at the ten-year mark. A historical overview of women’s basketball and soccer in the U.S. provides essential context, demonstrating both progress and persistent challenges over time. A review of existing literature supports the argument that media visibility is a crucial factor in the sustainability and growth of women’s professional sports. Ultimately, the findings suggest that continued investment in media coverage is not just beneficial but necessary for the future of women’s leagues
