Expression and Epitope Prediction of the Sirohydrochlorin Cobaltochelatase Isolated from a Local Strain of Mycobacterium Tuberculosis
The efficacy of the BCG vaccine, a widely recognized tuberculosis vaccine, has shown varying degrees of effectiveness, ranging from 0% to 80%. Sirohydrochlorin cobaltochelatase (CbiX), found in Mycobacterium tuberculosis, plays a crucial role in the bacteria metabolism, making it a promising target for future vaccine and drug development. Although several studies had been published regarding its role in M. tuberculosis vitamin B-12 metabolism, the potentials of CbiX protein as a vaccine candidate had not been widely discussed nor explored. This study focuses on the cloning and expression of the Rv0259c gene obtained from a clinical isolate of M. tuberculosis, as well as the exploration of CbiX protein epitopes. The Rv0259c gene was isolated by PCR, cloned into the pGEM®-TEasy vector, and subsequently sub-cloned into the pTrcHisA expression vector. Sanger sequencing, followed by BLASTN and BLASTX analyses, confirmed the presence of the CbiX protein-encoding gene. The amino acid sequence was predicted using BioEdit v.7.0.11, and a three-dimensional (3D) model was generated using SwissModel. Exploration for both B and T-cell epitopes was conducted using IEDB Ellipro, MHCI, and MHCII tools, revealing highly immunogenic epitopes, indicating the potential of CbiX as a vaccine candidate. Alignment using MAFFT between the putative amino acid sequence and CbiX proteins available in the NCBI database identified an amino acid variation (A182), situated outside the B-cell epitopes but within the T-cell epitopes. In silicoanalysis of HLA allele frequency predicted vaccine coverage of 86.14%±10.77%, with two significant epitope cores identified: AASAHPHVT and RRVAVASFL (both highly antigenic and showing high-frequency HLA allele binding), suggesting the protein might be a potential antigen for a future vaccine candidate. A New Approach to the Use of Non-Primitive Variables in the Mechanics of Continuous Media
The problem of an approximate solution to hydrodynamic problems is the consideration of pressure. To exclude it from the equations, the transition to "non-primitive variables” (vortex and velocity vector divergence) is made. In this case, there are difficulties in the algorithmization of new equations for solving the inverse problem of hydrodynamics and a lot of internal iterative calculations. The object of this study includes equations in "non-primitive” variables. The research methods are based on the transformation without simplifications and assumptions of hydrodynamic equations into a form containing "non-primitive” variables and the demonstration of the possibilities of solving the equations. The GAMS programming language was used for approximate solutions for the first time. The aim of this paper is to demonstrate the possibility of solving the full equations in "non-primitive” variables for various conditions. The results showed the possibility of considering the compressibility of the medium when solving the inverse problem of hydrodynamics; the identity of solutions of the proposed system of equations and equations using the potential; and the possibility of using optimizing programming languages for hydrodynamics problems. Relationship between the Characteristics of e-Commerce Live Streaming and Purchase Intentions: Moderating Effect of Hedonistic Shopping Value
E-commerce live streaming has experienced a significant and rapid increase in recent years, establishing itself as a vital catalyst for the growth of the e-commerce business. This research delves into the influence of e-commerce live streaming attributes, namely, authenticity, interactivity, and entertainment, on consumers' purchase intentions. It is based on the Stimulus"’Organism"’Response (SOR) theory and focuses on customer flow experience and hedonic shopping value. To evaluate the study model, the authors gathered survey data from 360 live streaming e-commerce customers and used structural equation modeling. The results indicate that client purchase intentions in e-commerce live streaming are positively influenced by authenticity, interactivity, and entertainment. Authenticity, entertainment, and interaction all contribute to enhancing consumers' flow experience in e-commerce live streaming, with interactivity being the most influential factor. The phenomenon of flow experience has a beneficial impact on the purchasing intentions of clients engaged in e-commerce live streaming. The flow experience serves as an intermediary between the characteristics of e-commerce live streaming and consumers' intentions to make purchases. Hedonic shopping value affects the relationship between the flow experience and the customer's intention to make a purchase. Put simply, when customers have a strong perception of the pleasure they derive from shopping, their intention to make purchases becomes more profound when they are influenced by the state of flow. This study not only contributes to the body of knowledge regarding the correlation between the attributes of e-commerce live streaming and consumer receptiveness, but it also provides businesses and platforms that utilize e-commerce live streaming with actionable recommendations. How Do Transformational Leadership and Affective Trust Enhance Creativity?
The pressure of competitive higher education has affected most universities, and whether the utilization of human capital as a resource to foster creativity, increase motivation, and boost innovation is an important topic to investigate. This study aims to explore the relationship between the elements of transformational leadership, affective trust, and intrinsic motivation and their impact on creativity to increase the performance of lecturers at Vietnamese universities, where most of them are also creators. Quantitative research is applied with data collected from full-time lecturers at Vietnamese universities from December 2023 to January 2024. The hypothesized correlations were investigated using structural equation modeling on a sample of 311 participants in Vietnam. The findings reveal substantial causal correlations between transformational leadership, affective trust, intrinsic motivation's determinants, and the creativity to improve performance and positive behavior from the lecturers' perspective. These results imply that leaders should focus on the prefix role of transformational leadership, which is the origin of affective trust, intrinsic motivation, and creativity, to raise the standard of higher education. In particular, affective trust needs more attention because it has the most vital mediating role in enhancing the impact of transformational leadership on lecturers' creativity. Effects of Social Media Marketing Activities on Perceived Values, Online Brand Engagement, and Brand Loyalty
This study aims to validate the model on the effects of social media marketing activities on the perceived values of social media marketing activities and the effects of these perceived values on online brand engagement and, consequently, on brand loyalty. The data used in this study were collected through an online self-administered survey of 501 young social media users in Vietnam. Partial Least Squares Algorithm, Bootstrapping, PLSpredict/CVPAT, and Multi-Group Analysis methods embedded in Smart-PLS software were used to validate the measurement model and test the research hypotheses. The findings confirm that the positive effects of social media marketing activities on brand loyalty are transmitted through the perceived values of these activities and online brand engagement. These effects are more substantial for luxury brands compared to non-luxury brands. Importantly, our study offers a new approach to explaining the impact of social media marketing activities on brand loyalty by focusing on the perceived values of these activities and their effects on online brand engagement. To enhance brand loyalty, businesses should prioritize creating hedonic and utilitarian values through their social media marketing activities and use these values and online brand engagement as key performance indicators for planning and controlling their strategies. University Students' Knowledge about Welfare Technologies within Active Aging Concept: An Online Survey Interview
Welfare technology has made a significant impact on society, supporting people´s communication, independence, and day-to-day living. This study aims to investigate university students' knowledge of welfare technologies related to smart active aging. The research was guided by the following questions: 1: What do Slovak university students know about welfare technologies? 2: How are welfare technologies used in the households of university students and their relatives? An online interview survey with reflexive thematic analysis was conducted with 120 university students at two universities in Slovakia. Findings show four themes that were identified: (1) Different interpretations of welfare technologies. (2) Broadening students' knowledge about welfare smart technologies. (3) Attitudes towards smart and welfare technologies. (4) Gap in practical usage of welfare technologies. Welfare technology is a new topic connected to active aging. Primarily, the younger generation, as proved, has low knowledge on this theme. The novelty of the study lies in connecting the welfare technology concept with the education of the younger generation. Welfare technologies have the potential to promote positive intergenerational relationships, autonomy, and mental and physical well-being among older adults. As for improvements, smart active aging and welfare technology concepts should be incorporated more intensively into higher education. This would help curb the knowledge gap and ensure the development of skills for implementing welfare technologies in the daily lives of their elderly relatives. Framing Assessment Questions in the Age of Artificial Intelligence: Evidence from ChatGPT 3.5
With the rise of artificial intelligence (AI), higher education faces a significant challenge in learning assessment. The emergence of tools like ChatGPT raises concerns regarding the potential for cheating and the reliability of assessment outcomes. This paper aims to address these concerns by proposing a methodology for framing questions that effectively measures learning outcomes while reducing the risk of AI-enabled cheating. To achieve this objective, we employ a methodological approach that involves getting responses from ChatGPT 3.5 to various question prompts across different domains. These responses are then evaluated by faculty members specializing in management education. Through this process, we aim to identify question-framing strategies that effectively assess learning outcomes while minimizing susceptibility to AI Cheating. Our analysis reveals several key findings. Certain question Types (Decision Making, Recent Events, and Experiential Learning) demonstrate greater resilience against AI-generated responses, indicating their potential effectiveness in assessing student learning. The Impact of Artificial Intelligence on Digital Marketing: Leveraging Potential in a Competitive Business Landscape
This study aims to thoroughly investigate how Artificial Intelligence (AI) is strategically integrated into digital marketing practices and its consequential effects on Indonesia's fiercely competitive business environment. Employing a quantitative research approach, this study meticulously examines Indonesian enterprises' prevailing strategies for AI utilization. The research method employed in this study is quantitative, with the unit of analysis being Indonesian companies. The sample size comprises 100 companies selected through the stratified random sampling technique. Analysis of the data is conducted using the SPSS statistical package. Through detailed analysis of survey data and advanced statistical techniques, the research reveals a significant positive correlation between the integration of AI in digital marketing and improved marketing effectiveness. The study highlights a noticeable increase in customer engagement metrics and noteworthy enhancements in conversion rates among businesses proficient in leveraging AI technologies, further reinforcing this correlation. Additionally, the findings suggest that companies embracing AI demonstrate significantly heightened adaptability to the constantly evolving market dynamics, strengthening their competitive positioning. Preparedness for Mandatory CSR Reporting of Multinational Companies: Case of the Czech Republic
The way how multinational corporations integrate CSR activities into their business practices and communicate them to stakeholders can significantly impact their market position and business reputation. This paper evaluates the perceived level of preparedness for non-financial reporting associated with social, environmental, and economic CSR activities in selected multinational corporations operating in the Czech Republic. A questionnaire survey for this research was conducted between September 2021 and January 2022, targeting a specific group of multinational corporations established in EU countries and operating in the Czech Republic. As non-financial reporting is expected to be extended to a considerably large number of companies in the near future, this research has high practical relevance for stakeholders involved in decision-making processes. Peer-Assessment Academic Integrity Scale (PAAIS-24)
This study aims to develop an academic integrity scale to help lecturers evaluate the academic integrity of university students with peer-assessment learning experiences using the ADDIE research and development method. Twenty-four items were designed using the concepts of academic integrity; honesty, trust, fairness, respect, responsibility, and courage validated by six raters and tested on 520 students from various universities in Indonesia. Testing content validity using the Aiken-V formula, construct validity testing using discriminant indexes, factor analysis (Exploratory and Confirmatory Factor Analysis), and concurrent validity testing by correlating PAAIS-24 with the developed Academic Integrity Scale. While the reliability test used Cronbach's alpha formula. The study results proved that each item of content validity meets with an Aiken value >0.80; on construct validity testing, the discriminant index value is >0.50, and the EFA and CFA loading factor values are >0.50. In addition, the model is appropriate because the theoretical model of PAAIS-24 is in accordance with empirical data. Whereas in reliability testing, Cronbach's alpha value is 0.95. Lastly, PAAIS-24 is one of the credible instruments. It contributes to developing knowledge that assists lecturers in measuring, assessing, and evaluating student academic integrity based on peer-assessment to promote increased academic performance.
