    Robot-Assisted Manufacturing Technology for 3D Non-Metallic Reinforcement Structures in the Construction Applications

    Of all industrial sectors, the construction industry accounts for about 37% of carbon dioxide (CO₂) emissions. This encompasses the complete life cycle of buildings, from the construction phase to service life to component disposal. The main source of emissions of climate-damaging greenhouse gases such as CO₂, with a share of 9% of global emissions, is the production of ordinary cement as the main binder of concrete. The use of innovative approaches such as impregnated carbon yarns as non-corrosive reinforcement embedded in concrete has the potential to dramatically reduce the amount of concrete required in construction, since no excessive concrete cover is needed to protect against corrosion, as is the case with steel reinforcement. At the same time, architectural design options are expanded via this approach. This is achieved above all using novel robotic manufacturing technologies to enable no-cut direct fiber placement. This innovative technological approach to fabricating 2D and 3D biologically inspired textiles, including non-metallic structures for textile-reinforced concrete (TRC) components, will promote an automatable construction method that reduces greenhouse gas emissions. Furthermore, the impregnated yarn which is fabricated enables the production of load-adapted and gradual non-metallic reinforcement components. Novel and improved design strategies with innovative reinforcement patterns allow the full mechanical potential of TRC to be realized. The development of a robotic fabrication technology has gone beyond the state of the art to implement spatially branched, biologically inspired 3D non-metallic reinforcement structures. A combined robotic fabrication technology, based on the developed flexible 3D yarn-guiding and impregnation module and a 3D yarn fixation module, is required to implement this sophisticated approach to fabricate freely formed 3D non-metallic reinforcement structures.     A hybrid process for integration of organic electrochemical transistors for high uniformity & reliability

    Photolithography is believed to be a complementary technique to large-area printing, allowing for nanometer-scale integration and offering cost-efficiency. For organic electronics though, adapting photolithography is very challenging due to chemical incompatibilities. However, with the help of Alexander Zakhidov, orthogonal resins opened up the prospect of adapting the well-established process of photolithography for organic electronics. Here, we present a hybrid fabrication method for organic electrochemical transistors by combining orthogonal photolithography and inkjet printing, enabling high uniformity and reliability. We demonstrate how the resolution of each process affects the uniformity, and we explore the advantages of this process for device scaling and circuit integration

    Grundlagen einer nachhaltigen Stadtentwicklung unter Berücksichtigung der Auswirkungen urbaner Güterverkehre

    Der urbane Güterverkehr wird trotz seiner Auswirkungen und seiner Bedeutung nicht ausreichend in die integrierte Stadtentwicklung einbezogen. Dies führt letztendlich zu unterschiedlichen Planungszielen der privatwirtschaftlichen und öffentlichen Akteure. Das Ergebnis dieser Planungsdualität sind nicht aufeinander abgestimmte Güter- und Stadtentwicklungskonzepte in Städten. Diese Arbeit stellt das methodische Konzept ISE+IFUD vor, das die urbane Güterverkehrsplanung mit der integrierten Stadtentwicklung methodisch und modellbasiert verbindet. Die Methodik ISE+IFUD basiert auf einem zweistufigen Integrationsprozess. Im ersten Schritt erfolgt eine Integration des urbanen Güterverkehrs in die Stadtentwicklung. Im zweiten Schritt werden die Handlungsfelder der integrierten Stadtentwicklung und der urbanen Güterverkehre miteinander verknüpft. Für diesen Schritt wird eine informationsbasierte Software Logistics And City Architecture Multilevel Model (LOCAMM) entwickelt. Grundlage für LOCAMM bildet ein agentenbasiertes Güterverkehrsmodell und ein Entscheidungsunterstützungsframework mit nachhaltigen Güterverkehrsindikatoren. Die Grundlage für das Entscheidungsunterstützungsframework stellt ein Multi-Criteria-Decision-Making Problem dar. Dieses Framework ermöglicht eine Bewertung und einen objektiven Vergleich unterschiedlicher Szenarien. Es dient als verlässliche Unterstützung für reale Entscheidungen und Forschungszwecke. Die Funktionalität LOCAMMs wird an der Stadt Hamburg mit reduzierten Gütermengen (1 % der Gütermenge) für zwei Szenarien veranschaulicht. Szenario 0 bildet die derzeitigen Logistikketten des Kurier-, Express-, Paketdienst (KEP)-Verkehrs in Städten mit dem Ausgang peripherer Lieferzentren ab. In Szenario 1 werden die Lieferketten der KEP-Dienstleister durch einBahn-City-Portal (BCP) im Zentrum Hamburgs verändert. Die Ergebnisse des Szenarios mit BCP zeigen vor allem positive Effekte auf die ökonomischen und ökologischen Nachhaltigkeitsindikatoren. Gleichzeitig können entstehende Reboundeffekte, wie eine erhöhte Verkehrsdichte im Umfeld des BCPs beobachtet werden. Diese Erkenntnisse unterstreichen die Notwendigkeit des ISE+IFUD-Ansatzes und die vereinte Betrachtung der integrierten Stadtentwicklung mit den urbanen Güterverkehren. Durch den ISE+IFUD-Ansatz und der Umsetzung in LOCAMM werden sowohl Chancen wie auch Risiken an der Schnittstelle zwischen integrierter Stadtentwicklung und urbaner Güterverkehre effektiv identifiziert und Entscheidungen unterstützt.Despite its impact and importance, urban freight transport is not sufficiently included in integrated urban development. This ultimately leads to different planning objectives on the part of private and public sector players. The result of this planning duality are uncoordinated freight and urban development concepts in cities. This paper presents the methodological concept ISE+IFUD, which combines urban freight transport planning with integrated urban development in a methodical and model-based way. The ISE+IFUD methodology is based on a two-stage integration process. In the first step, urban freight transport is integrated into urban development. In the second step, the fields of integrated urban development and urban freight transport are linked together. For this step, an information-based software Logistics And City Architecture Multilevel Model (LOCAMM) is being developed. LOCAMM is based on an agent-based freight transport model and a decision support framework with sustainable freight transport indicators. The basis for the decision support framework is a multi-criteria decision making problem. This framework enables an evaluation and objective comparison of different scenarios. It serves as a reliable support for real decisions and research purposes. The functionality of LOCAMMs is illustrated for the city of Hamburg with reduced goods volumes (1 % of the goods volume) for two scenarios. Scenario 0 depicts the current logistics chains of courier, express and parcel service (CEP) traffic in cities with the output of peripheral delivery centers. In scenario 1, the supply chains of CEP service providers are changed by a rail city portal (BCP) in the center of Hamburg. The results of the scenario with BCP primarily show positive effects on the economic and ecological sustainability indicators. At the same time, rebound effects, such as increased traffic density in the vicinity of the BCP, can be observed. These findings underline the necessity of the ISE+IFUD approach and the combined consideration of integrated urban development and urban freight transport. The ISE+IFUD approach and its implementation in LOCAMM effectively identifies both opportunities and risks at the interface between integrated urban development and urban freight transport and supports decision-making

    Steps towards higher pressures in low-moment magnetometry: A case study of the 2D antiferromagnet La₂CuO₄

    Magnetization can already be measured in a wide range of temperatures and external fields even with commercial instruments. Contrary to temperature and magnetic field, controlling pressure is more challenging. Commercial solutions are often either limited in their range or are only suitable to measure ferromagnets or superconductors, due to their high magnetic susceptibility. Researchers frequently need to manufacture their own specialized pressure cells, for the study of low moment systems at high pressures. This work evaluates several modifications to one of those specialized cells. The aim is increasing the maximum pressure achievable, while still keeping the background magnetization of the cell small. This would advance research on systems with very low moments, by to reach higher pressures. As a test case, the 2D-Heisenberg antiferromagnet La2CuO4 will be examined in this study. In the chosen measurement configuration, its low moment and high sensitivity to non-hydrostatic conditions make this compound ideal to show the limits of the setup. While a working solution to increase the pressure, was not fully achieved, this work provides valuable insights and is able to point toward the next steps in reaching this goal.Die Magnetisierung kann seit langem in einem weiten Bereich von Temperaturen und externen Feldern gemessen werden, selbst mit handelsüblichen Instrumenten. Kommerzielle Lösungen zur Einstellung von Druck hingegen verfügen oft nicht über einen hohen Druckbereich oder sind nur für die Messung von Ferromagneten oder Supraleitern geeignet, da diese hohe Suszeptibilität aufweisen. Für die Untersuchung von Systemen mit niedrigem Moment bei hohen Drücken müssen Forschende oft ihre eigenen speziellen Druckzellen herstellen. In dieser Arbeit werden mehrere Modifikationen an einer solchen Spezialzelle untersucht. Dabei ist das Ziel, den maximal erreichbaren Druck zu erhöhen und gleichzeitig die Hintergrundmagnetisierung der Zelle gering zu halten. Dies würde die Forschung an Systemen mit sehr niedrigen Momenten bei gesteigertem Druck erlauben. Als Fallstudie wird der 2D-Heisenberg-Antiferromagnet La2CuO4 untersucht. Durch sein geringes Moment und die hohe Anfälligkeit gegenüber nicht-hydrostatischen Bedingungen, ist dieses System besonders geeignet, um Schwächen im Aufbau hervorzuheben. Obwohl diese Arbeit den erreichbaren Druck nur bedingt erhöhen konnte, liefert sie wertvolle Erkenntnisse und kann die nächsten Schritte zur Erreichung dieses Ziels aufzeigen

    Short-Track-Speed Skating (STSS) ist eine olympische Disziplin, in der die anaerobe Leistungsfähigkeit eine zentrale Rolle spielt. Ein etabliertes Testverfahren zur Erfassung der anaeroben Leistungsfähigkeit bei STSS-Athlet:innen ist der Wingate Anaerobic Test (WAnT). Ziel dieser Studie war es, zu untersuchen, ob der WAnT als Prädiktor für die sportartspezifische Leistung bei 500 m-STSS-Rennen genutzt werden kann. Da STSS ein hochintensiver Eissport mit schnellen Rennen und potenziellem Körperkontakt zwischen Athlet:innen ist, besteht ein erhöhtes Risiko für Stürze und Verletzungen. Daher wurde im Rahmen der Nebenfragestellung eine Analyse der orthopädisch-traumatologischen Verletzungen bei deutschen STSS-Athlet:innen durchgeführt. Methoden: Die Daten von 72 Athlet:innen (33 weiblich, 39 männlich), die zwischen 2009 und 2020 Teil der deutschen STSS-Nationalmannschaft waren, wurden retrospektiv analysiert. Die statistischen Analysen zur Hauptfragestellung umfassten deskriptive Verfahren sowie eine linear mixed-model Analyse (LMM). Die Auswertung der Verletzungshistorien erfolgte auf Basis des 2020 vom Internationalen Olympischen Komitee (IOC) veröffentlichten Consensus Statement zur Erfassung von Krankheit und Verletzung im Sport. Ergebnisse: Die Ergebnisse zeigten signifikante Korrelationen zwischen den WAnT-Ergebnissen und der 500 m-Zeiten der STSS-Athlet:innen. Bei männlichen Athleten betrugen die Koeffizienten −0.40 für die relative Mean Power (rMP) und −0.32 für die relative Peak Power (rPP) (p < 0.001). Bei weiblichen Athlet:innen zeigte die rMP keine signifikante Korrelation, während die rPP mit einem Koeffizienten von −0.41 signifikant korrelierte (p < 0.001). Die marginalen R²-Werte lagen zwischen 0 % und 12 %, während die konditionalen R²-Werte je nach Geschlecht und untersuchtem WAnT-Parameter bis zu 76 % erreichten. Die durchschnittliche Karrieredauer der Athlet:innen betrug 10.9 ± 4.4 Jahre, wobei jede Athlet:in durchschnittlich insgesamt 10,116 ± 4,326 Stunden Risikoexposition im Training akkumulierte. Insgesamt wurden 371 orthopädisch-traumatologische Verletzungen, die eine ärztliche Konsultation erforderten, registriert. Dies ergab eine Inzidenz von 0.53 pro 1,000 Stunden Risikoexposition. 69.8 % der Verletzungen betrafen die unteren Extremitäten. Hauptsächlich betroffen waren Knie und Sprunggelenke. Die meisten Verletzungen ereigneten sich während des Eistrainings (45.0 %), gefolgt vom Off-Ice-Training (30.0 %) und Wettkämpfen (25.0 %). Schlussfolgerung: Der WAnT kann als potenzieller Prädiktor für die Zeit in 500 m-STSS-Rennen angesehen werden. Er liefert für sich genommen jedoch nur einen geringen Erklärungswert der sportartspezifischen Leistung. Die Studie stellt die erste Implementierung der Empfehlungen des IOCs im STSS dar. An established assessment tool for anaerobic performance in STSS athletes is the Wingate Anaerobic Test (WAnT). Therefore, the aim of this study was to investigate whether WAnT could be used as a predictor of sport-specific performance in the STSS 500-m races. Additionally, medical records of German STSS athletes were analyzed. Since STSS is a high-intensity ice sport characterized by fast-paced races and proximity among athletes, STSS inherently poses a high risk for falls and injuries. This paper presents a comprehensive analysis of orthopedic-traumatological injuries in German national STSS athletes with comparisons to previous studies. Methods: Data from 72 German national STSS athletes (33 female, 39 male), who were part of the German ST national team from 2009 to 2020, were analyzed retrospectively. The statistical analyses for the primary study objective included descriptive procedures as well as linear mixed-model analyses. For the secondary question medical records were analyzed based on a new standardized injury surveillance system published by the International Olympic Committee in 2020. Results: The results showed significant correlations between WAnT outcomes and 500-m performance of STSS athletes. In male athletes, the coefficients were −0.40 for relative Mean Power (rMP) and −0.32 for relative Peak Power (rPP) (p < 0.001). For female athletes, the rMP showed no significant correlation whereas rPP was significantly correlated with a coefficient of −0.41 (p < 0.001). Marginal R2 values ranged between 0% and 12%. Conditional R2 reached up to 76% depending on sex and WAnT parameter investigated. The average career duration of athletes on the German STSS national team was 10.9 ± 4.4 years, with each athlete accumulating 10,116 ± 4326 h of exposure to risk during training. A total of 371 orthopedic- traumatological injuries requiring medical consultation were recorded, with an estimated incidence of 0.53 per 1000 h of exposure. 69.8 % of the observed injuries affected the lower extremities, primarily involving the knee and ankle. Joint injuries were the leading cause of IRMCs, accounting for 16.3 % of cases. The majority of injuries occurred during on-ice training (45.0 %), followed by off-ice training (30.0 %), and competitions (25.0 %). Conclusion: In conclusion while WAnT can be regarded as a potential predictor of time in 500-m STSS races, it should be used in conjunction with other performance metrics and individualized assessments to better predict performance. This was the first implementation based on a standardized injury surveillance system in STSS. Important information about career-related injuries and injury patterns resulted from the analyses.     Development of impedance-based sensing systems

    This work demonstrates how impedance-based sensors can be transformed into adaptable biomedical tools, emphasizing the need to rethink how emerging technologies are integrated into healthcare systems. The future of diagnostics is moving away from centralized laboratory settings toward decentralized, accessible solutions that are versatile, customizable, and easily integrated into both clinical workflows and individual health management. This thesis presents the development, characterization, and validation of three impedance-based sensing platforms: a nanocytometer for label-free discrimination of subpopulations of peripheral blood mononuclear cells (PBMCs) and myeloblasts, a nanobiosensor for the detection of antigens and antibodies of SARS-CoV-2, and a flexible pH sensor for real-time monitoring of microphysiological systems (MPSs). First, the nanocytometer, based on interdigitated gold nanowires (Au NWs) placed in a microfluidic channel, discriminated between PBMCs subpopulations and myeloblasts according to their inherent dielectric properties, offering a prospective tool for hematological malignancy diagnosis. Next, the nanobiosensor, also using the Au NWs and a functionalization approach base on a selfassembled monolayer and EDC/NHS chemistry, enabled the detection of SARS-CoV-2 antigens and antibodies, with the addition of an on-chip calibration technique for semi-quantitative analysis. Finally, the flexible pH sensor, fabricated with a chemiresistive PEDOT:PSS film, provided continuous pH monitoring in MPS environments, demonstrating seamless integration into various in vitro cell culture platforms. These sensing platforms address some of the main limitations in biosensing, including miniaturization, sensitivity, and real-time analysis, and demonstrate the versatility of impedance-based technologies in diagnostics and in vitro monitoring. By bridging the fields of materials science, micro- and nanofabrication, and biology, this work contributes to the advancement of impedance-based sensing, promoting the development of more accessible, rapid, and reliable biomedical sensing solutions. The unique features of these technologies unlock new possibilities, not as standalone solutions but as integral components of a larger network of innovations. Die Zukunft der Diagnostik geht weg von zentralisierten Laboreinrichtungen und somit hin zu dezentralen, zugänglichen Lösungen, die vielseitig, anpassbar und nahtlos in klinische Arbeitsabläufe sowie in das individuelle Gesundheitsmanagement integriert werden können. Diese Dissertation präsentiert die Entwicklung, Charakterisierung und Validierung von drei impedanzbasierten Sensortechnologien: einem Nanocytometer zur markierungsfreien Unterscheidung von Subpopulationen peripherer mononukleärer Blutzellen (PBMCs) und Myeloblasten, einem Nanobiosensor zur Detektion von SARS-CoV-2-Antigenen und -Antikörpern sowie einem flexiblen pH-Sensor zur Echtzeitüberwachung mikrophysiologischer Systeme (MPSs). Das Nanozytometer, basierend auf in einem mikrofluidischen Kanal eingebetteter interdigitalen Gold-Nanodrähten (Au NWs), ermöglichte die Unterscheidung zwischen PBMC-Subpopulationen und Myeloblasten aufgrund ihrer intrinsischen dielektrischen Eigenschaften und bietet damit ein potenzielles Instrument für die Diagnose hämatologischer Malignome. Der Nanobiosensor, der ebenfalls auf Au NWs basiert und eine funktionalisierte Oberfläche auf Grundlage einer selbstorganisierten Monoschicht (SAM) und EDC/NHS-Chemie nutzt, ermöglichte die Detektion von SARS-CoV-2-Antigenen und -Antikörpern, ergänzt durch eine neuartige On-Chip-Kalibrierungstechnik zur semiquantitativen Analyse. Der flexible pH-Sensor, hergestellt mit einer chemiresistiven PEDOT:PSS-Schicht, ermöglichte eine kontinuierliche pH-Überwachung in MPS-Umgebungen und zeigte eine nahtlose Integration in verschiedene In-vitro-Zellkultursysteme. Diese Sensortechnologien adressieren einige der zentralen Herausforderungen in der Biosensorik, darunter Miniaturisierung, Sensitivität und Echtzeitanalyse, und unterstreichen die Vielseitigkeit impedanzbasierter Methoden in der Diagnostik und in vitro-Überwachung. Durch die Verknüpfung der Fachgebiete Materialwissenschaften, Mikro- und Nanofabrikation sowie Biologie trägt diese Arbeit zur Weiterentwicklung der impedanzbasierten Biosensorik bei und fördert die Entwicklung zugänglicher, schneller und zuverlässiger biomedizinischer Sensorlösungen.     Supraleitende Nanostrukturen für skalierbare Quantentechnologien

    Das Quantencomputing gilt als potentiell disruptive Quantentechnologie, die Umsetzung steht jedoch vor grundlegenden Skalierungsherausforderungen. Die Erforschung von alternativen Materialien und innovativen Bauelementen in einer skalierbaren Fertigungsumgebung kann einen entscheidenden Beitrag auf dem Weg zu universellen Quantencomputern leisten, analog zur wegweisenden Integration des Feldeffekttransistors in der Halbleiterindustrie. Diese Arbeit befasst sich mit der Entwicklung, Fabrikation und Charakterisierung supraleitender Dünnschichten und Nanostrukturen auf 300-mm-Wafern im Kontext der industriellen Halbleiterfertigung. Im Fokus stehen die Übergangsmetallnitride Hafniumnitrid (HfN) und Zirkoniumnitrid (ZrN), welche aufgrund ihrer Materialeigenschaften und potentiellen CMOS-Kompatibilität vielversprechend erscheinen. Im Rahmen der Arbeit erfolgt die Entwicklung von reaktiven Magnetronsputterprozessen zur Abscheidung von HfN- und ZrN-Dünnschichten und eines monolithischen, subtraktiven Strukturierungsverfahrens für ZrN-Nanostrukturen. Außerdem erfolgt die Konzeptionierung und Inbetriebnahme eines kryogenen Messsystems für die Charakterisierung von Dünnschichten und Testchips. Die resultierenden HfN- und ZrN-Dünnschichten werden umfassend hinsichtlich ihrer strukturellen, chemischen und supraleitenden Eigenschaften analysiert. Die Ergebnisse zeigen, dass beide Materialien kompatibel mit der skalierbaren Prozesstechnologie und als Dünnschichten supraleitend sind – mit kritischen Temperaturen von bis zu 5,8 K (HfN) bzw. 7,3 K (ZrN). ZrN-basierte Dayembrücken und Quanteninterferometer weisen konventionelle, auf Josephson-Effekten basierende Quanteneffekte auf. In planaren, ZrN-basierten 'gate controlled superconductivity' (GCS)-Transistoren wird die Modulation des kritischen Stroms durch Gate-Spannungen und Leckströme beobachtet. Die Unterdrückung der Supraleitung wird mit der Feldemission hochenergetischer Elektronen sowie Ausbreitung und Relaxationen von supraleitenden Quasiteilchen und Phononen in Verbindung gebracht. Insbesondere ZrN hat sich als vielversprechendes Material für die skalierbare Herstellung supraleitender Quantentechnologien im Kontext der Halbleiterfertigung erwiesen und eröffnet damit die potentielle Anwendung in der Quanteninformationsverarbeitung

    Alterations of cohesin complex genes in acute myeloid leukemia: differential co-mutations, clinical presentation and impact on outcome

    Functional perturbations of the cohesin complex with subsequent changes in chromatin structure and replication are reported in a multitude of cancers including acute myeloid leukemia (AML). Mutations of its STAG2 subunit may predict unfavorable risk as recognized by the 2022 European Leukemia Net recommendations, but the underlying evidence is limited by small sample sizes and conflicting observations regarding clinical outcomes, as well as scarce information on other cohesion complex subunits. We retrospectively analyzed data from a multi-center cohort of 1615 intensively treated AML patients and identified distinct co-mutational patters for mutations of STAG2, which were associated with normal karyotypes (NK) and concomitant mutations in IDH2, RUNX1, BCOR, ASXL1, and SRSF2. Mutated RAD21 was associated with NK, mutated EZH2, KRAS, CBL, and NPM1. Patients harboring mutated STAG2 were older and presented with decreased white blood cell, bone marrow and peripheral blood blast counts. Overall, neither mutated STAG2, RAD21, SMC1A nor SMC3 displayed any significant, independent effect on clinical outcomes defined as complete remission, event-free, relapse-free or overall survival. However, we found almost complete mutual exclusivity of genetic alterations of individual cohesin subunits. This mutual exclusivity may be the basis for therapeutic strategies via synthetic lethality in cohesin mutated AML
