    Fluorescent Analysis of Protein Localization in Saccharomyces cerevisiae

    A principal objective of cellular biology is to identify the function of proteins as they interact with their environment. Cellular localization provides context for the function and interactions of proteins. In this study, we seek to identify the localization of three proteins within Saccharomyces cerevisiae through C-terminal green fluorescent protein epitope tagging and subsequent fluorescent microscopy. As 75% of the yeast proteome has been classified, this project seeks to fill the remaining gap in unidentified cellular localization by investigating the proteins fsh3, gid10, and ade13. The analysis of high-resolution localization and comparative phylogenetic data reveals potential cellular environments and protein functions

    Taulkinham’s Guardian: The Character of Enoch Emery in Flannery O’Connor’s Wise Blood

    Flannery O’Connor’s first novel Wise Blood follows the story of Hazel Motes as his journey in the hostile city of Taulkinham transforms him from a defiant defector of the divine to a stoic and resigned blind prophet. For many readers, Hazel’s evolution labels him as the sole owner of wise blood in the novel, a conclusion that fails to acknowledge Enoch Emery’s obedience to his own innate wisdom. Enoch’s bumbling behavior and animalistic drive cause many O’Connor writers and scholars, such as Patrick O’Donnell and Jason C. Lee, to interpret him as little more than a comic simpleton. However, careful analysis reveals that his character is more complex and far-reaching. The wisdom that drives Enoch’s actions is not intellectual in nature but is instead based on action and result, situated within what is worldly as opposed to what is spiritual. Enoch therefore becomes Taulkinham’s mirror and guardian of hope, the only character capable of conceiving, confronting, and presenting the brokenness of the world around him. This leads Enoch to truths that others cannot see or refuse to acknowledge, and the transformation caused by obedience to his wise blood assigns him the duty of leading others to see these distasteful truths

    Many scholars focus on J.R.R. Tolkien\u27s personal history and attempt to locate his own trauma in the texts of his works. However, this focus often overlooks the role that trauma plays in the reshaping of individual and cultural identity within the works of Tolkien. Tolkien uses a number of methods to communicate trauma throughout his works, but these methods often have roots in Anglo-Saxon Literature. This study analyzes the various ways that Tolkien adapts Anglo-Saxon works to communicate trauma while simultaneously using the traumatic events to help communicate healing through the interaction of the traumatized with their community

    Adults\u27 Recollections and Reflections on the Impacts of Grade Retention

    This qualitative study chronicles the experiences of retention as told by adults who experienced retention. Through criterion sampling and snowball sampling, ten participants were identified and interviewed to learn more about their experiences with retention. Through one-on-one interviews, the researcher sought to examine what characterizes the experiences and responses of adults who were retained during their K-12 academic career. Their in-depth responses revealed participants’ perceptions of retention and the effects that it produced in their lives. This study humanized the data by allowing participants to tell an account of retention that coincides with past research. The findings of this study reflected previous studies that found retention produces temporary gains followed by lifelong detriments to participants’ social, emotional, and behavioral well-being. This study brought new insight to the retention debate by giving voice to participants

    Community and Population Dynamics of Diatoms in Recovering Agricultural Stream and Ubiquitous Taxon in Low-Impact Sites Across Southeastern United States

    Streams play vital roles to surrounding communities and provide essential ecosystem services. The protection of streams is important, legally protected under the Clean Water Act (CWA) and can be monitored through the continuous analyses of biological data, like algal or other aquatic communities. Diatoms are a group of algae characterized by rigid glass cell walls and ubiquitous distribution. They are typically identified using the morphological species concept and an ever-varying range of sizes may pose challenges in the identification process. However, when identified and applied correctly, diatoms are robust bioindicators. This research identifies and analyzes low-impact sites across the Southeastern United States in attempts to identify reference sites. Three study sites within two different projects were analyzed: Tobler Creek, a recovering agricultural stream on the site of Andalusia Farm; Upper Three Runs Creek (UTRC), a reference site protected by the Department of Energy (DOE); and Murder Creek, a low-impact stream located that has been continuously monitored within the Oconee National Forest. Given a 31- year recovery period (2011), Tobler Creek did not meet nutrient reference criteria but did with an additional 11-year recovery (2022). From 2011 to 2022, Tobler Creek was found to display positive ecological values, such as species richness and diversity, in response to recovery. However, degradation was still signaled from diatom metrics, detecting high percentage of sediment tolerant taxa dominating communities. Gomphonema parvulum, a taxon traditionally thought to be tolerant of high nutrients, was in high abundance in 2011 and low in 2022. Analyses of Southeastern low-impact streams found G. parvulum still dominating UTRC in 2018, a reference site. This opens new questions about adaptations, ecological preferences, and life history of this taxon. Morphological responses were assessed across the Southeastern United States, and found that in UTRC, G. parvulum had a significant reduction in frustule sizes from 1956 to 2018, at the population level. However, in Tobler Creek, frustule lengths significantly varied monthly whereas there was no significant difference from 2011 to 2022. When cultured, frustules had significantly varied morphological responses from populations sampled from natural environments. Monocultures provide evidence that environmental conditions influence diatom morphology to some extent, as well as rates of sexual and asexual reproduction, and evolution of population morphologies of G. parvulum in UTRC is detected. Tobler Creek can serve as a model for recovery of abiotic and biotic properties in degraded streams whereas G. parvulum populations in UTRC is a case-study for population responses of this ubiquitous taxon to environmental conditions

    Measuring the Relationship between the Big Five Personality Traits and Time Spent on Social Media

    This study examined the effects of personality on social media usage. Ninety-seven participants completed a Social Media Survey that asked them to use the iPhone “Screen Time” settings to record the time spent on social media applications in a given week. They then completed the 44-item Big Five Inventory to measure their personality based on openness, contentiousness, extraversion, agreeableness, and neuroticism. It was hypothesized that people with higher levels of all big five traits will have higher social media usage. Similarly, it was hypothesized that people who score higher on openness, agreeableness, and extraversion will use Snapchat the most. It was also hypothesized that those who score higher on conscientiousness and neuroticism will use Instagram the most. Finally, it was hypothesized that TikTok will be the most used application overall. There were no significant correlations between social media usage and any of the five personality traits. Also, the only significant finding between any of the social media applications and personality traits was between YikYak and neuroticism. As hypothesized, TikTok was the most used social media application

    Blockchain technology, cryptocurrencies and transforming accounting fees

    This study investigated the accounting treatment of Bitcoin, considering whether it should be classified as an asset or a currency. The study also explored the role of blockchain technology in promoting transparency and the potential separation of blockchain from Bitcoin. The study found that cryptocurrencies are significantly impacted by political, economic, and regulatory factors. The study also found that it is challenging to control cryptocurrencies and cross-border transactions in the absence of accounting standards. The study recommends that regulators and accounting standards setters should establish new regulations and accounting standards for cryptocurrencies. Finally, this study identifies the vast majority of the existing literature as lacking adequate, well-rounded knowledge about cryptocurrencies or access to adequate resources, despite their clearly understanding the fundamental concepts of cryptocurrency. Further, the theoretical part of this paper is there to establish some type of accounting approach for Bitcoin

    COVID-19 and the “Golden Era”: Turning the page on rural, weekly newspaper production

    The rural weekly newspaper industry rooted in traditional geographically bounded U.S. communities was long thought to have been shielded from industry dilemmas—revenue and circulation—at larger mainstream daily newspapers. However, recent data show a shift in these rural community journalism trends, particularly since the COVID-19 pandemic. Using normative theory and diffusion of innovations, this study examines how rural weekly newspaper publishers perceived, nearly 18 months after the onset of the pandemic, how COVID-19 impacted their newspapers’ advertising, news, technology, and community involvement. Survey results reveal short-term and long-term content and financial challenges plague U.S. rural grassroots information sources with circulation less than 3,000—indicating the end of the “golden era” of weekly newspapers. Despite the constraints, publishers remain optimistic and dedicated to their audiences, albeit reluctant to swift change, as they perceive themselves as the only reliable information source for their rural communities

    Lie -isoclinism in Leibniz n-algebras

    We study the notion of Lie-isoclinism on Leibniz n-algebras. This is isoclinism on Leibniz n-algebras relative to Filippov algebras. We provide equivalent conditions under which Leibniz n-algebras are Lie-isoclinic. Also, we provide a characterization of Lie-stem Leibniz n-algebras, and show that each Lie-isoclinic class of Leibniz n-algebras contains a Lie-stem Leibniz n-algebra. Moreover, this Lie-stem Leibniz n-algebra has minimum dimension among finite dimensional Leibniz n-algebras in that class. Finally, we define the notion of factor sets on Leibniz n-algebras and analyze their connection with Lie-stem Leibniz n-algebras
