Turbulent exchange of energy, water and carbon between crop canopies and the atmosphere : an evaluation of multi-year, multi-site eddy covariance data
The increase of anthropogenic CO2 emissions and other greenhouse gases has raised concern about climate change. Climate change has manifold impacts on yield and yield quality, crop rotations, carbon and nitrogen cycling, water regime and agricultural production systems. To understand its consequences on environmental systems, measuring the matter and energy exchange at the land surface provides data to help validate and inform a wide range of process models. Such flux measurements at the land-surface provide an opportunity to test simulations of processes in the soil-plant-atmosphere continuum. Currently, such measurements are mainly based on the eddy covariance (EC) method, for the quality of which the energy balance closure (EBC) is a problem. The EBC significantly influences the calibration and validity of land-surface models, especially in regard to the energy and water balance at the Earths surface. The EBC quantifies the deviation between turbulent fluxes and available energy. It is crucial to obtain high-quality EC measurements to determine the reasons for the EBC.
The research aims of this dissertation were: 1) to clarify the role of minor storage and flux terms in the energy balance, 2) to determine the possible reasons for the energy imbalance using a long-term dataset (2010-2017) from agricultural croplands, and 3) to investigate the effects of region, site, year and crop type on carbon fluxes and budgets.
In the first study (Chapter 2) the contribution of minor storage terms to the EBC were investigated. I also determined the contribution of ground heat fluxes calculated by different methods. A harmonic analysis method was used to calculate ground heat fluxes from measurements of heat flux plates and soil temperature sensors. Soil heat storage and enthalpy change in the plant canopy were determined at different locations within the EC footprint. Considering minor storage terms improved the energy balance closure on average by 5.0 % in 2015 and by 6.8 % in 2016. The greatest energy balance closure improvement occurred in May of both study years. The dominant fraction of minor energy storage was energy uptake and release through photosynthesis and respiration. Additionally, the energy fluxes related to soil temperature change were also observed. The ground heat flux calculated by harmonic analysis from soil heat flux plates narrowed the EBC by 3 % compared to the calorimetric method. The results indicated that the typical correction approach to achieve energy balance closure, i.e. the Bowen-ratio method, overestimated the turbulent fluxes.
The second study (Chapter 3) investigated the effects of crop type, site characteristics, wind directions, atmospheric conditions and footprint on the EBC. The long-term evaluation of EC measurements showed that, with the EC method, 25 % of the available energy could not be detected. Decreasing the flux footprint area increases the chance of a more homogeneous area. Homogeneity plays an important role in achieving a better energy balance closure. The synthesis of long-term EC data indicated that the sonic anemometer is very sensitive to orientation, not allowing accurate measurements from all wind directions. Discarding the measurements from wind directions 0° and 90° at EC4 improved the EBC from 80 to 84 %.
In the third study, presented in Chapter 4, a long-term and multi-site experiment was evaluated to clarify the effects of site, year and region on the CO2 fluxes and budgets in agroecosystems. The net ecosystem exchange of CO2 fluxes measured on six sites during eight years was comprehensively examined. Winter rapeseed had the lowest CO2 uptake, cropping of silage maize resulted in the highest C losses. The management of harvest residues was the most effective means of controlling the C budgets. Comparing the CO2 fluxes processed with the recently developed ogive optimization method versus the conventional calculation showed that eliminating low-frequency contributions had a considerable effect. On average, the ogive optimization method delivered 6.9 % higher net ecosystem exchange rates than the conventional method.
This dissertation provides new insights into how to obtain better measurements of matter and energy fluxes from EC measurements by a) considering storage terms otherwise neglected, b) using harmonic analysis for calculating ground heat fluxes, c) discarding fluxes from behind the anemometer and d) applying the ogive optimization method.Der Anstieg der anthropogenen, klimawandelverursachenden Emissionen von Kohlenstoffdioxid und anderer Treibhausgase löst Besorgnis aus. Der Klimawandel hat vielfältige Auswirkungen auf Ertrag und Ertragsqualität, Fruchtfolgen, Kohlenstoff- und Stickstoffkreislauf, Wasserregime und insgesamt das landwirtschaftliche Produktionssystem. Um die Auswirkungen auf Umweltsysteme zu verstehen, werden zur Optimierung und Validierung von Prozessmodellen Messungen des Stoff- und Energieaustauschs an der Landoberfläche durchgeführt. Dies ermöglicht es, Prozesse im Boden-Pflanze-Atmosphäre-Kontinuum detaillierter zu simulieren. Gegenwärtig basieren solche Messungen hauptsächlich auf der Eddy-Kovarianz-Methode (EC), im Rahmen derer die nicht vollumfängliche Energiebilanzschließung (EBC) ein methodisches Problem darstellt. Die EBC beeinflusst signifikant die Kalibrierung und Validierung von Landoberflächenmodellen, insbesondere hinsichtlich der Wasser- und Energiebilanz. Die EBC quantifiziert die Abweichung zwischen turbulenten Flüssen und verfügbarer Energie. Qualitativ hochwertige EC-Messungen sind wichtig, um die Gründe für die nicht vollständige EBC zu bestimmen.
Die Forschungsziele waren: 1) Klärung der Rolle von geringfügigen Speicher- und Flusstermen zur Verbesserung der Energiebilanzschließung, 2) Ermittlung der möglichen Gründe für die nicht vollständig gegebene EBC anhand eines Langzeitdatensatzes (2010-2017) von landwirtschaftlichen Nutzflächen, und 3) Untersuchung der Auswirkungen von Region, Standort, Jahr und Fruchtart auf Kohlenstoffflüsse und -bilanzen.
In der ersten Studie (Kapitel 2) wurde der Beitrag von geringfügigen Speichertermen zur EBC untersucht. Dabei wurde der Beitrag des Bodenwärmeflusses ermittelt, welcher mit verschiedenen Methoden berechnet wurde. Die Harmonische Analyse wurde verwendet, um den Bodenwärmefluss aus Messungen von Wärmeflussplatten und Bodentemperatursensoren zu berechnen. Die Wärmespeicherung des Bodens und die Enthalpieänderung im Pflanzenbestand wurden an verschiedenen Stellen innerhalb des EC-Fußabdrucks ermittelt. Unter Berücksichtigung der geringfügigen Speicherterme verbesserte sich die Schließung der Energiebilanz im Jahr 2015 um durchschnittlich 5,0 % und 2016 um 6,8 %. Die größte Verbesserung der Energiebilanzschließung trat im Mai beider Untersuchungsjahre auf. Der dominierende Anteil der geringfügigen Speicherterme war die Energieaufnahme und -freisetzung durch Photosynthese und Atmung. Zusätzlich wurden die Energieflüsse, die mit der Veränderung der Bodentemperatur zusammenhängen, untersucht. Der durch die harmonische Analyse berechnete Bodenwärmefluss erhöht die EBC um 3 % im Vergleich zur kalorimetrischen Methode. Die Ergebnisse zeigen, dass der typische Korrekturansatz für die Schließung der Energiebilanz, das Bowen-Verhältnis-Verfahren, die turbulenten Flüsse überschätzte.
In der zweiten Studie (Kapitel 3) wurden die Auswirkungen von Fruchtart, Standorteigenschaften, Windrichtung, sonstigen atmosphärischen Bedingungen und des EC-Messbereichs auf die EBC untersucht. Die Langzeitauswertung zeigte, dass mit der EC-Methode 25% der verfügbaren Energie nicht erfasst werden konnten. Durch die Verringerung des EC-Messbereichs wird die Wahrscheinlichkeit einer homogeneren Fläche erhöht. Diese Homogenität spielt eine wichtige Rolle für die Erreichung einer besseren Energiebilanzschließung. Die Synthese von Langzeit-EC-Daten zeigte, dass auch die Orientierung des Ultraschallanemometers das Messergebnis stark beeinflusst. Durch Ausklammern der Messungen aus den Windrichtungen zwischen 0° und 90° bei EC4 verbesserte sich beispielsweise die EBC von 80 auf 84 %.
In der dritten Studie, die in Kapitel 4 vorgestellt wird, wurde eine Langzeit- und Multi-Site-Untersuchung durchgeführt, um die Auswirkungen von Standort, Jahr und Region auf die CO2-Flüsse und Budgets in Agrarökosystemen zu analysieren. Der Netto-Ökosystemaustausch von CO2-Flüssen, gemessen an sechs Standorten während acht Jahren, wurde umfassend untersucht. Es zeigte sich, dass Winterraps die geringste CO2-Aufnahme aufweist und Silomais die höchsten C-Verluste verursacht. Das Management der Ernterückstände war das wirksamste Instrument zur Steuerung der Kohlenstoffbilanz. Ein Vergleich der CO2-Flüsse, die mit der kürzlich entwickelten Ogive-Optimierungsmethode verarbeitet wurden, zeigte im Vergleich zur herkömmlichen Berechnung, dass die Eliminierung von Niedrigfrequenzflüssen eine beträchtliche Wirkung hatte. Im Durchschnitt lieferte die Ogive-Optimierung 6.9 % höhere Netto-Ökosystem-Austauschraten als die konventionelle Methode.
Diese Dissertation liefert neue Erkenntnisse darüber, wie man bessere Stoff- und Energieflussmessungen an der Landoberfläche erzielen kann: a) durch Berücksichtigung von vernachlässigten Speichertermen, b) durch Verwendung der Harmonischen Analyse zur Berechnung des Bodenwärmestroms, c) durch Ausschluss von Flüssen, die aus dem rückwärtigen Bereich des Anemometers stammen, und d) durch Anwendung der Ogive-Optimierungsmethode
Cooperative approaches to face the challenges of services of general interest in rural areas
Vor dem Hintergrund des gesellschaftlichen, (infra-)strukturellen, landwirtschaftlichen und demografischen Wandels entstehen für die ländlichen Räume neue Herausforderungen. Besonders kleinere Gemeinden, ohne leistungsfähige Infrastruktur, sind vom demografischen Wandel betroffen (IREUS 2011, S. 64 f.). Eine zentrale Herausforderung, die sämtliche (Lebens-)Bereiche betrifft stellt dabei die flächendeckende Daseinsvorsorge dar.
Ausgehend von den Herausforderungen der flächendeckenden Daseinsvorsorge wurde ein bereits stellenweise praktizierter Lösungsansatz analysiert und weiterentwickelt. Aus dem vergangenen Jahrzehnt liegen Beobachtungen, Hinweise und statistische Aufzeichnungen vor, die davon zeugen, dass die vielfältigen Aufgaben der Daseinsvorsorge partiell von Genossenschaften übernommen werden (vgl. BLOME-DREES et al. 2015; DOLUSCHITZ et al. 2013a; GOESCHEL 2012, S. 51; KLEMISCH und BODDENBERG 2012, S. 570; KLUTH 2017; MARTENS 2012, S. 145; MÜNKNER 2012, S. 332; STAPPEL 2016, S. 66), mit dem Ziel, spezifische lokale Probleme zu lösen (HAUNSTEIN und THÜRLING 2017, S. 2).
Aufgrund mangelnder Grundlage spezifisch notwendiger Sekundärdaten wurde ein umfassender Datensatz deutschlandweit erhoben, mithilfe dessen die Spezifika sowie die Hintergründe der Entstehung der neu gegründeten Genossenschaften der Daseinsvorsorge analysiert werden konnten.
Die gewonnenen Ergebnisse bekräftigen die in der Literatur genannten Hintergründe der Gründungen wie der Wunsch nach einer angemessenen Infrastruktur, regionale Defizite (BLOME-DREES et al. 2015, S. 134), Bedrohung der Infrastruktur, gesellschaftliche Herausforderungen (vgl. WIEG 2016, S. 78) und spezifische lokale bzw. regionale Probleme (HAUNSTEIN und THÜRLING, 2017, S. 2). Die Analyse der Intentionen, welche hinter den Gründungen von Genossenschaften stehen und zu diesen geführt haben, liefert darüber hinaus jedoch ergänzende und neue Befunde. Weitere Intentionen sind zum einen karitative Gründe wie zum Beispiel die Schaffung von sozialen Anknüpfungspunkten und zum anderen die Gründung aufgrund des Wunsches/der Initiative von Gleichgesinnten zur Organisation von z. B. Fortbildungen. Rund zwei Drittel der antwortenden Genossenschaften der Daseinsvorsorge, speziell in den ländlichen Räumen, schaffen ein neues Angebot für ihre Mitglieder sowie für Nicht-Mitglieder. Rund ein Drittel der analysierten Genossenschaften soll bestehende Einrichtungen und Leistungen erhalten oder wieder aktivieren, die aufgrund unterschiedlicher Herausforderungen wegzubrechen drohen oder bereits weggebrochen sind.
Ausgehend von der unzureichenden Versorgungssituation in den ländlichen Räumen sowie den Herausforderungen bestehender Genossenschaften der Daseinsvorsorge wurde ein Modell entwickelt, welches die Kooperation mehrerer Primärorganisationen unterschiedlicher Sparten beinhaltet. Das Modell ist als eine genossenschaftliche ,Second-Level-Kooperation, ähnlich einer Holdingstruktur zu verstehen. Da im Kern mehrere Primärorganisationen unter einem gemeinsamen Dach zusammenarbeiten, wird das Modell ,Geno-Haus genannt. Das gemeinsame Dach des Geno-Hauses kann sinnbildlich aber auch ausdrücklich z. B. in Form eines gemeinschaftlich genutzten Gebäudes oder Räumlichkeiten verstanden werden.
Ein zweiter mit der Daseinsvorsorge verbundener Aspekt dieser Dissertation liegt auf der Digitalisierung des Agrar- und Ernährungssektors. Insgesamt zeichnet sich hinsichtlich der Digitalisierung entlang der Wertschöpfungskette ein heterogenes Bild ab. Im Ergebnis gliedern sich die analysierten Unternehmen in Pioniere, Einsteiger und Folger. Im Zuge einer Befragung von praktizierenden Landwirten hinsichtlich der Nutzung von E-Commerce Strukturen beim Bezug von landwirtschaftlichen Betriebsmitteln wurden Nutzerstrukturen analysiert. Diese erwiesen sich als alters-, betriebsgrößen-, ortsunabhängig und losgelöst von der Betriebsausrichtung. Als Hauptmotiv für die Beschaffung von Betriebsmitteln über E-Commerce-Kanäle wurden auf Basis der Befragungen Zeitvorteile ermittelt, was im Widerspruch zu den in der Literatur genannten Kostenvorteilen steht (vgl. CLASEN 2005, S. 54 f.).In the context of social, (infra-)structural, agricultural and demographic change, new challenges are emerging for rural areas. Demographic change especially impacts on smaller communities without an efficient infrastructure (IREUS 2011, p. 64 f.). One central challenge that affects all areas of life is the nationwide provision of public services.
Based on the challenges of providing public services on a nationwide basis, an approach already practised in some locations was analysed and further developed. Observations, infomation and statistical records from the past decade indicate that the wide-ranging tasks involved in providing public services are partially performed by cooperatives (cf. BLOME-DREES et al. 2015; DOLUSCHITZ et al. 2013a; GOESCHEL 2012, p. 51; KLEMISCH and BODDEN-BERG 2012, p. 570; Kluth 2017; MARTENS 2012, p. 145; MÜNKNER 2012, p. 332; STAPPEL 2016, p. 66) with the aim of addressing specific local problems (HAUNSTEIN and THÜRLING 2017, p. 2).
Due to the lack of a basis of specifically required secondary data, a comprehensive data set was collected throughout Germany that enables analysis of the specific details and background to the emergence of newly-founded public service cooperatives.
The results obtained confirm the background to these foundations as described in the literature, such as the desire for an appropriate infrastructure, regional deficits (BLOME-DREES et al. 2015, p. 134), threats to the infrastructure, social challenges (cf. WIEG 2016, p. 78) and specific local and regional problems (HAUNSTEIN and THÜRLING, 2017, p. 2). However, an analysis of the underlying the establishment of cooperatives also provides fresh, supplementary findings. On the one hand, further goals reflect a charitable purpose such as the creation of social points of contact, while on the other hand there are efforts to organise further training programmes, for example, based on the wishes/initiative of like-minded individuals. About two thirds of the responding public service cooperatives, especially in rural areas, are creating a new scheme for both members and non-members. Approximately one third of the public service cooperatives analysed aim to maintain or reactivate existing facilities and services that risk being discontinued or where discontinuation has already occurred as a result of various challenges.
A second aspect of this dissertation relating to public services is the digitisation of the agricultural and food sectors. All in all, a heterogeneous pattern emerges with regard to digitisation along the value chain. As a result, the companies analysed are divided into pioneers, newcomers and followers. User structures were analysed by means of a survey among practising farmers that focused on the use of e-commerce structures when purchasing agricultural resources. These structures proved to be independent of age, farm size, location and farm orientation. On the basis of the surveys, time benefits were identified as the main source of motivation behind the procurement of resources through e-commerce channels, which contradicts the cost benefits described in the literature (cf. CLASEN 2005, p. 54 f.)
Mapping Stratification : the industry-occupationspace reveals the network structure ofinequality
Social stratification is determined not only by income, education, race, and gender, but also by an individuals job characteristics and their position in the industrial structure. Utilizing a dataset of 76.6 million Brazilian workers and methods from network science, we map the Brazilian Industry-Occupation Space (BIOS). The BIOS measures the extent to which 600 occupations co-appear in 585 industries, resulting in a complex network that shows how industrial-occupational communities provide important information on the network segmentation of society. Gender, race, education, and income are concentrated unevenly across the core-periphery structure of the BIOS. Moreover, we identify 28 industrial occupational communities from the BIOS network structure and report their contribution to total income inequality in Brazil. Finally, we quantify the relative poverty within these communities. In sum, the BIOS reveals how the coupling of industries and occupations contributes to mapping social stratification
Spatial and temporal variations of microorganisms in grassland soils : influences of land-use intensity, plants and soil properties
Grassland ecosystems provide a wide range of services to human societies (Allan et al., 2015) and plants and soil microorganisms have been identified as key drivers of ecosystem functioning (Soliveres et al., 2016). Therefore, understanding soil microbial distributions and processes in agricultural grassland soils is crucial for characterizing these ecosystems and for predicting how they may shift in a changing environment. Yet we are only beginning to understand these complex ecosystems, which account for about 26% of the worlds terrestrial surface (FAOSTATS, 2018), making it especially urgent to gain better insights into the effects of land-use intensity on soil microbial properties and plant-microbe interactions. This thesis was conducted to evaluate the impact land-use intensity has on soil microbial biogeography of grasslands with respect to both spatial patterns and temporal changes in soil microbial abundance, function (in terms of enzyme activities), and community composition. It also investigated the relationships between plants and the spatial and temporal distributions of soil microorganisms. Thereby both, land-use intensity effects and plant-microbe interactions, were assessed in light of ecological niche and neutral theory. This thesis is based on three observational studies conducted on from one to 150 continuously farmed, un-manipulated grassland sites in three regions of Germany within the Biodiversity Exploratories project (DFG priority program 1374).
The first study assessed the effects of land-use intensity and physico-chemical soil properties on the spatial biogeography of soil microbial abundance and function in 18 grasslands sites from two of the three regions, sampled at one time point. The second study analyzed spatial and temporal distributions of alpha- and beta-diversity of arbuscular mycorrhizal fungi in a low land-use intensity grassland with six sampling time points across one season. The third study investigated both legacy and short-term change effects of land-use intensity, soil physico-chemical properties, plant functional traits, and plant biomass properties on temporal changes in soil microbial abundance, function, and community composition in 150 grassland sites across three regions, with particular regard to direct and indirect land-use intensity effects.
Although the three studies used different approaches and assessed different soil microbial properties, general patterns were detectable. Abiotic soil properties, namely pH, nitrogen content, texture, and bulk density played fundamental roles for spatial and temporal microbial biogeography. Since these factors were specific and unique for each investigated site, they formed the background based on which other processes occurred. In addition to abiotic soil properties, impacts of land-use intensity and plants were detected, though to various degrees in the three studies. Land-use intensity played a much smaller role than anticipated in the first and third study. No influence on the spatial distribution of soil microbial abundance and function could be detected in the first study. In the third study, short-term changes in and legacy effects of land-use intensity played a minor role with respect to short-term changes in soil microbial abundance, function, and community composition. Where detected, changes in land-use intensity had a direct and negative effect on soil microbial properties in structural equation modelling; i.e., increases in land-use intensity reduced, e.g., soil microbial enzyme activities, while legacy effects of land-use intensity were shown to act both directly and indirectly on soil microbial properties. Thereby indirect legacy effects were mediated via plant functional traits. Only one of the three studies detected minor plant diversity effects on soil microbial properties. Instead, functional properties of the plant communities, i.e., plant functional traits, biomass, and nutritional quality, were significantly related to spatial and temporal distributions of soil microorganisms. Finally, the findings of the three studies suggest that processes related to niche and neutral theory both drive spatial and temporal patterns of soil microbial properties at the investigated plot scale (up to 50 m × 50 m).
This thesis concluded that in order to gain deeper insights into the complex functions and processes occurring in grassland ecosystems, a multidisciplinary approach investigating fundamental physico-chemical site characteristics, microbial soil properties, and plants is necessary. The results of the thesis suggest that focus be turned to functional properties of plant and microbial communities, as they are closely intermingled, provide more detailed insights into plant-microbe interactions, and are able to reflect effects of human impacts on grassland soils better than diversity measures.Die vorliegende Dissertation diente dem Zweck, die Einflüsse der Landnutzungsintensität von Grünländern auf die Biogeographie von Bodenmikroorganismen in Bezug auf die räumliche Verteilung und zeitliche Veränderung von mikrobieller Biomasse, Funktion und Gemeinschaftsstrukturen aufzudecken und die Beziehungen zwischen Pflanzen und den räumlichen und zeitlichen Verteilungsmustern von Bodenmikroorganismen zu untersuchen. Dabei wurde auch betrachtet, inwieweit die Verteilung der Mikroorganismen den Konzepten der ökologischen Nischen- und Neutraltheorie entspricht. Die Dissertation basiert auf drei beobachtenden Studien, die auf einer bis 150 dauerhaft bewirtschafteten und nicht manipulierten Grünlandflächen durchgeführt wurden. Die Flächen sind verteilt auf drei Regionen in Deutschland und Teil des Schwerpunktforschungsprogramms Biodiversitäts-Exploratorien der Deutschen Forschungsgemeinschaft (DFG 1374).
Die erste Studie untersuchte die Effekte von Landnutzungsintensität und physikalisch-chemischen Bodeneigenschaften auf die räumliche Biogeographie von mikrobieller Biomasse und Funktion zu einem einzelnen Zeitpunkt in 18 Grünlandflächen, die auf zwei Regionen verteilt sind. Die zweite Studie analysierte die räumliche und zeitliche Verteilung der Alpha- und Betadiversität arbuskulärer Mykorrhizapilze (AMF) in einem extensiv genutzten Grünland mittels sechs Probennahmezeitpunkten verteilt über die Vegetationsperiode eines Jahres. Die dritte Studie untersuchte sogenannte Legacy und kurzfristige Effekte von veränderter Landnutzungsintensität, physikalisch-chemischen Bodeneigenschaften, funktionellen Pflanzeneigenschaften und Charakteristika der pflanzlichen Biomasse auf zeitliche Veränderungen von mikrobieller Biomasse, Funktion und Gemeinschaftsstruktur in 150 Grünlandböden verteilt auf drei Regionen, sowie direkte und indirekte Effekte der Landnutzungsintensität.
Obwohl alle drei Studien unterschiedliche Ansätze verfolgten und verschiedene bodenmikrobielle Eigenschaften untersuchten, sind generelle Muster erkennbar: abiotische Bodeneigenschaften, namentlich pH-Wert, Stickstoffgehalt, Textur und Lagerungsdichte, waren für die räumliche und zeitliche Biogeographie der Bodenmikroorganismen von fundamentaler Bedeutung. Sie waren spezifisch für jede der untersuchten Flächen und bildeten den Hintergrund, vor dem sich andere Prozesse abspielten. Zusätzlich zu den abiotischen Bodeneigenschaften wurden, wenn auch in unterschiedlichem Maße, Einflüsse von Landnutzungsintensität und Pflanzen in den drei Studien detektiert. Die Landnutzungsintensität spielte dabei eine wesentlich geringere Rolle als ursprünglich angenommen. Sie hatte keinen Einfluss auf die räumliche Verteilung der bodenmikrobiellen Biomasse oder Funktion in der ersten Studie. In der dritten Studie waren kurzfristige Veränderungen und Legacy-Effekte nur in geringem Maße mit kurzfristigen Veränderungen von bodenmikrobieller Biomasse, Funktion und Gemeinschaftsstruktur assoziiert. Die Strukturgleichungsmodelle zeigen, dass sich die Effekte von kurzfristigen Veränderung der Landnutzungsintensität, dort wo sie auftraten, direkt und negative auf die kurzfristigen Veränderungen bodenmikrobieller Eigenschaften auswirkten. Dahingegen wirkten sich Legacy-Effekte der Landnutzungsintensität sowohl direkt als auch indirekt auf bodenmikrobielle Eigenschaften aus. Die indirekten Legacy-Effekte wurden dabei über funktionelle Pflanzeneigenschaften auf die Mikroorganismen übertragen. Nur eine der drei Studien fand einen marginalen Einfluss der Pflanzendiversität auf die bodenmikrobiellen Eigenschaften. Stattdessen waren funktionelle Eigenschaften der Pflanzengemeinschaften sowie deren Biomasse und Futterqualität signifikant mit der räumlichen und zeitlichen Verteilung von Bodenmikroorganismen verbunden. In Bezug auf die Nischen- und Neutraltheorie sprechen die Ergebnisse der drei Studien dafür, dass sowohl Prozesse, die mit der Nischentheorie zusammenhängen, als auch solche, die mit der Neutraltheorie in Verbindung stehen, die räumliche und zeitliche Verteilung von bodenmikrobiellen Eigenschaften auf der untersuchten Plotskala (bis 50 m × 50 m) steuern.
Diese Dissertation zieht die Schlussfolgerung, dass multidisziplinäre Forschung notwendig ist, um die komplexen Funktionen und Prozesse von Grünlandökosystemen zu erforschen. Diese müssen sowohl die fundamentalen physikalischen und chemischen Eigenschaften der Böden, als auch die Eigenschaften der Bodenmikroorganismen und Pflanzengemeinschaften umfassen. Dabei sprechen die Ergebnisse dieser Arbeit dafür, dass ein besonderes Augenmerk auf die funktionellen Eigenschaften von Pflanzen- und Mikrobengemeinschaften gelegt werden sollte, da diese eng miteinander verflochten sind und bessere Einblicke in die Interaktionen zwischen Pflanzen und Mikroorganismen gewähren sowie besser in der Lage sind die Effekte von menschlichen Einflüssen auf Grünlandböden wieder zu spiegeln, als die bisher oft üblichen Diversitätsmessungen

Evaluation of the new cropping practices in sugar beet (Beta vulgaris L.) cultivation in the central black soil region of Russian Federation
In recent years Russian sugar production has exceeded the country needs with a surplus of 500.000 tons. Sugar producers and traders are forced to start trading on the global markets. However, ineffective production, caused by low yields of sugar puts the price of Russian sugar 35 to 50 % higher than the leading sugar exporters.
Weeds belong to one of the main factors reducing sugar beet yield. Weeds that survive control operations may cause significant crop yield reductions. Rapid emergence and homogeneous crop stand are very important for competition with weeds. At the same time, the crop may be suppressed by selective herbicide application, if herbicides are applied during suboptimal weather conditions. This study evaluated glyphosate-tolerant sugar beet technology and seed priming for the possibility to increase the productivity of sugar beet cultivation.
A series of studies were carried out in different environments in Germany and in Russian Federation. The scope of the studies was dealing with:
i) comparison of the conventional weed control technology with technology, based on glyphosate applications;
ii) a study of the crop weed interaction in German environment and in the environment of the Central black soil region of the Russian Federation;
iii) tests of sugar beet seed priming for the speed of germination by means of growth chamber test, and for the speed of emergence in the soil seedbed, by means of greenhouse and field experiments.
The results of the conducted experiments are concentrated in three scientific articles that have been published in the international peer-reviewed journals:
The purpose of the first article was to analyse the weed control efficacy of weed control schemes with one, two and three applications of glyphosate and compare it with the conventional weed control technology. The results show that the application of glyphosate supplied significantly higher efficacy of weed control than conventional herbicides. In five out of 7 experiments single application of glyphosate gave the same weed control efficacy as two or three applications. No significant differences in weed control efficacy were observed between two different dosages of glyphosate 900 and 1350 g a.e. ha-1. The variants treated with conventional herbicides and with two and three glyphosate applications showed no significant differences in white sugar yield. In one location, the variant with one application of glyphosate resulted in lower white sugar yield due to delayed application of herbicide, and longer time of crop and weed interference.
The aim of the second article was to study the yield loss caused by the competition with weeds in different environments. In the Russian locations Chenopodium album L. and Amaranthus retroflexus L. caused serious yield reduction already at low plant densities. White sugar yields harvested at Russian locations were approximately 45 % lower than in German locations. At Russian locations 50 % of the maximum weed population has caused more than 80 % yield reduction. Relative weed cover was the best predictor of the sugar beet yield loss. Weed biomass and weed density gave less accurate predictions.
The third article was focused on testing of priming technology on the seed performance and crop establishment. In the controlled environment, primed seeds needed 10 days to reach full germination percentage of the seeds, for not primed seeds it took between 12 and 14 days. Primed seeds produced significantly larger area of plant foliage area and amount of dry biomass than non-rimed seeds. In uncontrolled environments, findings of previous experiments could not be confirmed. Priming did not influence the weed suppressive ability of the crop and the white sugar yield.
The general conclusion of this dissertation is that GT technology may help to improve the profitability of sugar beet cultivation for Russian growers by reducing the number of herbicide applications and increase weed control efficacy. Seed priming can give benefits for crop establishment, however only under specific environmental conditions.In den letzten Jahren übertraf die russische Zuckerproduktion den Bedarf des Landes mit einem Überschuss von 500.000 Tonnen. Zuckerproduzenten und -händler sind gezwungen, auf den Weltmärkten zu handeln. Ineffektive Produktion, verursacht durch niedrige Zuckererträge, führt jedoch dazu, dass der russische Zuckerpreis um 35 bis 50% höher liegt als die führenden Zuckerexporteure.
Unkräuter, welche Bekämpfungsmaßnahmen überleben, gehören zu den wichtigsten Faktoren, die den Zuckerrüben erheblich reduzieren. Ein schneller Feldaufgang und ein homogener Rübenpflanzenbestand sind für die Konkurrenz mit Unkräutern sehr wichtig. Allerdings kann die Kulturpflanze durch Herbizide negativ beeinflusst werden, wenn das Herbizid nicht unter optimalen Wetterbedingungen angewendet wird. In der vorliegenden Studie wurden die Glyphosat-tolerante Zuckerrübentechnologie und die Saatgutaktivierung auf mögliche Produktivitätssteigerungen im Zuckerrübenanbau untersucht.
Eine Reihe von Untersuchungen wurde in verschiedenen Umgebungen in Deutschland und in der Russischen Föderation durchgeführt. Der Umfang der Studien befasste sich mit:
- i) Vergleich der konventionellen Unkrautbekämpfungstechnologie mit der auf Glyphosat-Anwendungen basierenden Technologie;
- ii) eine Untersuchung der Interaktion zwischen Kulturpflanzen und Unkräuter in der deutschen Umwelt und in der Umgebung der zentralen Schwarzbodenregion der Russischen Föderation;
- iii) Tests der Aktivierung von Zuckerrübensamen auf die Keimungsgeschwindigkeit mittels Klimakammerversuch und auf die Geschwindigkeit des Aufgangs im Bodensaatbett durch Gewächshaus- und Feldversuche.
Die Ergebnisse der durchgeführten Experimente sind in drei wissenschaftlichen Veröffentlichungen zusammengefasst, die in internationalen, Peer-Reviewed Zeitschriften veröffentlicht wurden:
Ziel der ersten Veröffentlichung ist es, den Erfolg der Unkrautkontrolle mit einer, zwei und drei Anwendungen von Glyphosat mit der konventionellen Technologie der Unkrautkontrolle zu vergleichen. In fünf von sieben Versuchen ergab die einmalige Anwendung von Glyphosat die gleiche Effizienz der Unkrautkontrolle wie bei zwei oder drei Glyphosatanwendungen. Es wurden keine signifikanten Unterschiede in der Effizienz der Unkrautkontrolle zwischen zwei verschiedenen Dosierungen von Glyphosat (900 und 1350 g a.e. ha-1) beobachtet. Die mit konventionellen Herbiziden und mit zwei und drei Glyphosat-Anwendungen behandelten Varianten zeigten keine signifikanten Unterschiede im Zuckerertrag. An einem Standort führte die Variante mit einer einmaligen Glyphosatanwendung zu einem niedrigeren Zuckerertrag aufgrund einer verzögerten Herbizidbehandlung und einer längeren Konkurrenzdauer zwischen Kulturpflanzen und Unkräutern.
Ziel der zweiten Veröffentlichung war es, den Ertragsverlust zu untersuchen, der durch die Konkurrenz mit Unkraut in verschiedenen Umgebungen verursacht wurde. An den Standorten in Russland führten Chenopodium album L. und Amaranthus retroflexus L. bereits bei niedrigen Pflanzendichten zu einer erheblichen Ertragsreduzierung. Die an russischen Standorten geernteten Zuckererträge waren etwa 45 % niedriger als an deutschen Standorten. An russischen Standorten haben 50 % der maximalen Unkrautpopulation eine Ertragsminderung von mehr als 80 % verursacht. Die relative Unkrautbedeckung war der beste Prädiktor für den Ertragsverlust der Zuckerrüben. Mit Hilfe der Unkrautbiomasse und der Unkrautdichte waren die Verlustprognosen weniger Präzis.
Die dritte Veröffentlichung konzentrierte sich auf die Bewertung des Effekts der Priming-Technologie auf die Leistung des Saatguts, sowie Etablierung und der Produktivität von Zuckerrüben. In der kontrollierten Umgebung benötigte aktiviertes Saatgut 10 Tage, um den vollen Keimungsprozentsatz zu erreichen. Für nicht aktiviertes Saatgut dauerte es zwischen 12 und 14 Tagen. Aktiviertes Saatgut erzeugte eine wesentlich größere Blattfläche und Menge an trockener Biomasse als Saatgut ohne Aktivierung. In Feldversuchen, in unkontrollierten Umgebungen konnten frühere Entdeckungen nicht bestätigt werden. Die Unkrautunterdrückungsfähigkeit der Kulturpflanzen und der Zuckerertrag wurden durch die Aktivierung nicht beeinflusst.
Die allgemeine Schlussfolgerung dieser Dissertation lautet, dass die GT-Technologie russischen Zuckerrübenanbauern dabei helfen kann, die Rentabilität des Zuckerrübenanbaus zu verbessern, indem die Anzahl der Herbizidbehandlungen reduziert und die Wirksamkeit der Unkrautkontrolle erhöht wird. Saatgutaktivierung kann Vorteile für die Etablierung von Kulturpflanzen bieten, jedoch nur unter bestimmten Umweltbedingungen
Functional analysis of the genes Brachyury, Goosecoid and Myosin1d in early patterning and establishment of the body axes during the embryogenesis of the clawed-frog Xenopus laevis
Während einer fundamentalen Phase der Wirbeltier Embryonalentwicklung, der Gastrulation, wird die Anlage des zukünftigen Körperbauplans determiniert. Dieser Prozess kontrolliert die Musterbildung der Antero-Posterioren (AP) und Dorso-Ventralen (DV)- Achse, sowie der sich im Inneren offenbarenden Links-Rechts (LR)-Achse. Die Etablierung der Körperachsen wird initial über den Spemann-Organisator (SO) gesteuert, wobei die Determinierung der LR-Achse als letztes stattfindet. Die Ausbildung der Lateralität während anschließender Neurulation findet dabei durch einen, bei Fischen, Amphibien und Säugern, hoch konservierten Mechanismus des Symmetriebruchs statt. Eine extrazelluläre, Monocilien-getriebene Flüssigkeitsströmung (Flow) definiert dabei die spätere Lateralität des Embryos, durch links-asymmetrische Expression des Morphogens Nodal. Verantwortlich für den Flow ist ein ciliertes Epithel im postero-dorsalen Bereich des Embryos, das während der Gastrulation seine korrekte Positionierung erfährt und im Frosch als GRP (gastrocoel roof plate) bezeichnet wird. Letztlich beschreibt dieses konservierte Gewebe den Links-Rechts-Organisator (LRO) und garantiert den ordnungsgemäßen Organsitus. Den Ursprung des LRO kann man auf einen epithelialen Zellverband an dorsaler Oberfläche der Gastrula zurückführen, der durch die Expression des kanonischen Wnt-Zielgens foxj1 charakterisiert wird. Der Spezifizierung dieses Superfiziellen Mesoderms (SM) folgend, wandern die zukünftigen Zellen des LRO während der Gastrulation gerichtet in den Embryo ein. Die Bewegungsmuster verlangen dabei einer Umstrukturierung des Aktin-Cytoskeletts. Diese Wanderungen werden durch den PCP- (planar cell polarity) Signalweg gewährleistet. Dieser nicht-kanonische Wnt-Signalweg garantiert die polarisierte Ausrichtung und Migration von Zellen entlang der Körperachsen, die konvergente Extension (CE). Als ein Organisator-Gen charakterisiert die Expression von Goosecoid (Gsc) den SO und seine Eigenschaften. Entgegen der Annahme, dass ein Funktionsverlust von Gsc somit die Gastrulation inhibiert, wurde in der Knock-Out Maus bzw. dem Knock-Down in Xenopus jedoch keinerlei Beeinträchtigung festgestellt. Es waren Funktionsgewinne im Frosch, die die Rolle von Gsc während der Gastrulation analysierten und aufzeigten, dass das Homeobox-Gen eine Funktion bei der Regulation von Zellbewegungen ausüben kann. Überexpressionen führten zu einer gestörten CE dorsaler Gewebe, aufgrund fehlerhafter Lokalisierung von Kernproteinen des PCP-Signalwegs. Gsc-induzierte Missbildungen konnten durch Co-Injektionen assoziierter Komponenten kompensiert werden. Schlussfolgernd wurde somit eine neue Funktion als Inhibitor PCP-abhängiger CE während der Gastrulation nahegelegt. Es sind die morphogenetischen Bewegungen der CE, die für die AP-Elongation und LRO-Positionierung verantwortlich sind. Dabei agieren sowohl mechanische Zugkräfte als auch der Cilien-basierte Flow. Im Invertebraten Drosophila, in dem weder Nodal, noch Cilien exprimiert sind, wird die Organasymmetrie durch interne Chiralität über Motorproteine wie Myosin1d PCP-abhängig gewährleistet. Vergleichbar werden in Vertebraten wie Xenopus durch myo1d asymmetrische Interaktionen des Aktin-Cytoskeletts vermittelt, die die Lateralität des Organsystems garantieren. Die funktionelle Konservierung des nicht-kanonischen Wnt-Signalwegs konnte dabei durch Co-Injektionen mit PCP-Kernproteinen aufgezeigt werden, die eine gestörte Morphologie des LRO wiederherstellten. Die gewonnenen Daten zeigten deutlich die evolutionär manifestierte, speziesübergreifende Regulation der Achsenasymmetrie durch myo1d. Da die strikte räumlich-zeitliche Regulierung von Zellbewegungen zur Musterbildung fundamental ist, wird die CE durch einen weiteren Transkriptionsfaktor propagiert: Brachyury (Tbxt in Xenopus). Dessen Expression induziert und steuert die Differenzierung mesodermaler Zellpopulationen, wie die notochordalen Zellen der dorsalen Mittellinie, somit des LRO. Die Notwendigkeit für die Spezifizierung der Vorläuferzellen im SM konnte dabei ebenfalls aufgezeigt werden, wobei diese dort nicht-zellautonom vermittelt wird, um die Induktion von foxj1 zu garantieren. Auch erfolgt die Determinierung beider Gewebe durch die funktionelle Interaktion von Tbxt mit dem PCP-, sowie dem ß-catenin-abhängigen Wnt-Signalweg. Der Brachyury Verlust manipuliert dabei die Lateralität in Mutanten und Morphanten. Der speziesübergreifende Brachyury-FGF feedback-loop für die Induktion von Nodal und Foxj1 könnte dabei bereits im SM agieren und zeigte, dass sowohl Funktion als auch der Prozess der LR-Entwicklung, wie zwischen Fgf8 und Brachyury nahegelegt, funktionell konserviert scheint. In Xenopus wurde die komplexe Signaltransduktion für die SM und foxj1 Induktion um die Funktion des Wnt-Rezeptors frizzled-8, des Liganden und Tbxt-Zielgens wnt11b, sowie dem ventral agierenden wnt8a in Wohle-Mount Embryonen und Explantat-Co-Kulturen erweitert.During a fundamental phase of vertebrate embryonic development, gastrulation, the reorganization of the future body plan, is determined. This process controls the embryonic patterning of the antero-posterior (AP) and dorso-ventral (DV) axes, as well as the internal left-right (LR) axis. The establishment of the body axes is initially controlled by the dorsally localized Spemann organizer (SO), whereby the determination of the LR axis takes place the latest. The formation of laterality during subsequent neurulation is executed by a highly conserved mechanism of symmetry breakage within fishes, amphibians and mammals. An extracellular, monocilia-driven fluid-flow (Flow") eventually defines the laterality of the embryo, by left-asymmetric gene expression of the morphogen Nodal. Responsible for the Flow is a ciliated epithelium in the postero-dorsal region of the embryo, which undergoes morphological development and correct positioning during gastrulation and is termed GRP (gastrocoel roof plate") in the frog. Ultimately, this conserved tissue describes the left-right organizer (LRO) and guarantees proper organ situs. The origin and function of the LRO can be traced back to an epithelial organized cell structure on the dorsal surface of the gastrula, characterized by the expression of the canonical Wnt target gene and cilia marker foxj1. Following the specification of this Superficial Mesoderm (SM), the future cells of the LRO invaginate in a directed fashion during gastrulation. The patterns of cellular movement require a restructuring of the actin-cytoskeleton. These migrations, executing the future body plan, are ensured by the PCP- (planar cell polarity) signaling pathway. This non-canonical Wnt pathway guarantees polarized alignment and migration of cells along the body axes, namely convergent extension (CE). As an organizer gene, the expression of Goosecoid (Gsc) characterizes SO and its properties. Contrary to the assumption that a Gsc loss of function thus inhibits gastrulation, no visible impairment was detected in the Knock-Out mouse and Knock-Down in Xenopus. Gain-of-function in the frog, which analyzed the role of Gsc during gastrulation finally demonstrated the homeobox-gene exerting a function in regulating cell movements. Overexpression resulted in impaired CE of dorsal tissue due to defective localization of nuclear proteins of the PCP signaling pathway. Gsc-induced malformations could be compensated by co-injections of associated components. In conclusion, a new function as an inhibitor of PCP-dependent CE during gastrulation was suggested. The morphogenetic movements of CE are responsible for the AP-elongation and LRO-positioning. Both, mechanical forces and cilia-based Flow functionally interact for that matter. In the invertebrate Drosophila, where neither Nodal nor cilia are expressed, organ asymmetry is ensured by internal chirality conducted via motor proteins, such as myosin1d, in a PCP-dependent manner. Similarly, in vertebrates such as Xenopus, myo1d mediates interactions of the actin cytoskeleton asymmetrically, that guarantee laterality of the organ system. The functional preservation of the non-canonical Wnt pathway could be demonstrated by co-injections of PCP core proteins, being able to restore disturbed LRO morphology. The obtained data clearly demonstrated the evolutionary, interspecific, regulation of axis asymmetry by myo1d. Since the strict spatio-temporal regulation of cell movements is fundamental for patterning, CE is propagated by another transcription factor: Brachyury (Tbxt in Xenopus). Its expression induces and controls differentiation of mesodermal cell populations, such as the notochordal cells of the dorsal midline, hence LRO. The necessity specifying progenitor cells in the SM could also be highlighted, showing to be mediated non-cell autonomously to guarantee the induction of foxj1. Additionally, the determination of both tissues is exerted by functional interactions of Tbxt with either PCP-, as well as ß-catenin-dependent Wnt signaling pathway. Loss of Brachyury affects laterality in mutants and morphants. The species-spanning Brachyury-FGF feedback-loop" for the induction of Nodal and Foxj1 could already act in the SM and showed that function as well as the process of LR development, as suggested between Fgf8 and Brachyury, to be functionally conserved. In Xenopus, signal transduction for SM and foxj1 induction was extended by the function of the Wnt receptor frizzled-8, the ligand and Tbxt target gene wnt11b, and the ventrally acting wnt8a in Whole-Mount embryos and explant co-cultures
How can miscanthus be integrated most efficiently into agricultural production systems?
The demand for biomass is increasing steadily, as fossil resources are gradually being replaced by biomass within the context of a developing bioeconomy. Plant-based feedstocks currently used for this replacement virtually all come from annual crops. However, perennial crops such as miscanthus are expected to be more environmentally benign due to their generally low-input requirements and high yield potential.
Despite these advantages, the current cultivation area of miscanthus in Europe is quite low. One reason for this is that the cultivation and utilization of miscanthus faces several challenges. For example, the most common propagation method via rhizomes is very labour-intensive and thus expensive, leading to high establishment costs. Seed propagation is a promising option to reduce costs, but is not suitable for sterile genotypes. Another challenge to be overcome is the problem of re-integrating former miscanthus fields into crop rotations. The crop following miscanthus needs to be highly competitive in order not to be impaired by resprouting miscanthus shoots and thus able to achieve high yields. Additionally, there is only little information available on the effect of miscanthus cultivation and its subsequent removal on soil N content. This information is however crucial, for example to avoid environmental problems being caused by a potential nitrogen leaching after a miscanthus removal. If miscanthus is to be utilized as a biogas substrate, there are further challenges to be overcome. Firstly, the optimal harvest date needs to be defined with regard to the methane hectare yield and resilience of the crop to green cutting. Secondly, as a continuous supply of biomass throughout the year is necessary, ensiling will become a relevant topic. However, information is still required on the optimal harvest date to achieve a sufficient silage quality and the effects of ensiling on methane hectare yield. Finally, the suitability of miscanthus for biogas production is also influenced by biomass quality such as the proportions of leaf and stem. This has already been established for miscanthus utilization in combustion but has not yet been sufficiently investigated for anaerobic digestion.
In summary, there are a number of uncertainties involved in miscanthus establishment, removal and utilization, which currently hamper its integration into agricultural production systems.
From a bioeconomic point of view, this integration needs to be conducted as efficiently as possible in terms of nutrient-use, environmental and land-use efficiency. The aim of this study was to contribute to the filling of these knowledge gaps.
To answer these knowledge gaps, several miscanthus field trials and laboratory experiments were conducted: a novel propagation method was tested; the re-integration of miscanthus fields into a crop rotation was analysed; and the effect of genotype, harvest date and ensiling on the digestibility and methane hectare yield was investigated.
The results illustrate some possibilities of improving the nutrient-use, environmental and land-use efficiency of miscanthus biomass production along its supply chain: It was shown that miscanthus propagation via collars is feasible and a promising alternative to rhizome propagation, as the multiplication rate of collars is comparable to that of rhizome propagation. As the harvesting of collars is likely to be less labour-intensive and is less destructive for the mother field than rhizome propagation, this method is more favourable for both economic and ecological reasons.
The re-integration of miscanthus into crop rotations revealed maize to be a suitable crop after miscanthus, as it coped with the prevailing soil conditions and suppressed resprouting miscanthus efficiently, resulting in satisfactory yields. The soil mineral nitrogen (Nmin) content was found to increase during the vegetation period following a miscanthus removal, but was generally on a low level (average: 17.3 kg Nmin ha-1).
Additionally, it was found that, in Germany, miscanthus should be harvested in mid-October to maximize methane yields and nutrient recycling but minimize yield reduction. In addition, silage quality was best when miscanthus was harvested on this date. As leaf proportion correlated positively with substrate-specific methane yield (SMY) and thus genotypes with a higher leaf proportion were found to have a higher SMY, methane hectare yields could be increased even further by using genotypes with a high leaf proportion.
In summary, the approaches developed in this study allow to considerably improve the ecological and economic performance of miscanthus production by increasing nutrient-use,environmental impact and land-use, and thus simplifying implementation into practice.Im Rahmen einer wachsenden Bioökonomie steigt der Biomassebedarf stetig an, da fossile Ressourcen nach und nach durch biobasierte ersetzt werden. Die dafür benötigte Biomasse wird derzeit überwiegend mittels einjähriger Kulturpflanzen gedeckt. Mehrjährige Kulturpflanzen, wie zum Beispiel Miscanthus, bilden aufgrund ihres geringen Ressourcenbedarfs bei einem gleichzeitig hohen Ertragspotential eine interessante, umweltverträglichere Alternative. Trotz dieser Vorteile ist die Anbaufläche von Miscanthus in Europa derzeit gering. Ein Grund hierfür ist, dass der Anbau und die Nutzung von Miscanthus verschiedene Herausforderungen mit sich bringen. So ist zum Beispiel die übliche Vermehrungsmethode über Rhizome sehr arbeitsintensiv, was zu hohen Etablierungskosten führt. Eine Vermehrung über Samen ist zwar im Hinblick auf eine Kostenreduktion vielversprechend, aber diese Methode ist für sterile Genotypen nicht anwendbar. Eine weitere Herausforderung, bei der Integration von Miscanthus in die Praxis, ist die Wiedereingliederung früherer Miscanthusfelder in eine Fruchtfolge. Die Folgefrucht sollte den wiederaustreibenden Miscanthus effektiv unterdrücken und gleichzeitig einen möglichst hohen Ertrag generieren. Darüber hinaus gibt es derzeit nur wenig Information darüber, wie sich der Stickstoffgehalt im Boden nach einem Miscanthusanbau und anschließendem -umbruch verhält. Diese Information ist aber entscheidend, um negative Umweltwirkungen, beispielsweise durch Nitratauswaschung, zu vermeiden.
Weitere Herausforderungen ergeben sich bei der Nutzung der Miscanthusbiomasse, beispielsweise als Biogassubstrat. In einem ersten Schritt muss der beste Erntezeitpunkt im Hinblick auf den Methanhektarertrag und der Widerstandsfähigkeit der Kultur gegenüber einem Grünschnitt bestimmt werden. Des Weiteren ist die Silierung ein wichtiges Thema, da eine kontinuierliche Substratverfügbarkeit über das Jahr gewährleistet sein muss. Jedoch fehlen noch Informationen darüber, wann der optimale Erntezeitpunkt ist, um eine ausreichende Silagequalität zu erzielen, und wie sich die Silierung auf den Methanhektarertrag auswirkt.
Darüber hinaus wird die Eignung von Miscanthus für die Biogasproduktion auch durch die Biomassequalität, wie beispielsweise das Blatt-/Stängelverhältnis, beeinflusst. Ähnliches wurde bereits für Nutzung von Miscanthus für die Verbrennung gezeigt, für die Biogasproduktion wurde diese jedoch noch nicht hinreichend untersucht.
Zusammengefasst gibt es im Bereich der Etablierung, des Umbruches sowie der Nutzung von Miscanthus mehrere Barrieren, die derzeit eine Integration in landwirtschaftliche Produktionssysteme erschweren. Das Ziel dieser Studie war es, die identifizierten Hindernisse zu überwinden und dabei die Integration von Miscanthus in Bezug auf die Nährstoffnutzung, Umweltwirkung und die Landnutzung so effizient wie möglich zu gestalten.
Hierfür wurden mehrere Feld- und Laborversuche durchgeführt: So wurde eine neue Vermehrungsmethode getestet; die Wiedereingliederung von Miscanthusfeldern in die Fruchtfolge wurde untersucht; die Wirkung verschiedener Genotypen,Erntetermine und Silierung auf die Verdaulichkeit und den Methanhektarertrag wurde analysiert.
Die Ergebnisse konnten zeigen, dass die Vermehrung von Miscanthus über Collars möglich und eine vielversprechende Alternative zur Rhizomvermehrung ist. Die Vermehrungsrate ist zwar vergleichbar, die Ernte von Collars ist jedoch weniger arbeitsintensiv und destruktiv für das Mutterfeld. Der Versuch, der sich mit der Wiedereingliederung von Miscanthusflächen in Fruchtfolgen beschäftigte, zeigte, dass Mais eine geeignete Folgefrucht ist. Dieser kam mit den vorherrschenden Bodenbedingungen zurecht, unterdrückte den wiederaustreibenden Miscanthus effizient und lieferte gleichzeitig hohe Biomasseerträge. Es stellte sich heraus, dass in der Vegetationsperiode nach dem Miscanthusumbruch der Bodenstickstoffgehalt (Nmin) zwar anstieg, aber generell auf einem niedrigen Niveau lag (im Mittel 17,3 kg Nmin ha-1). Darüber hinaus wurde gezeigt, dass in Deutschland ein Erntetermin Mitte Oktober optimal ist, um die Methanerträge und die Nährstoffrückverlagerung zu maximieren und Ertragsverluste zu minimieren. Außerdem wurde aufgezeigt, dass die Silagequalität von Miscanthus an diesem Erntetermin am besten war. Des Weiteren konnte in den Versuchen eine positive Korrelation von Blattanteil und dem substratspezifischen Methanertrag (SME) demonstriert werden. Hierbei hatten Genotypen mit einem höheren Blattanteil einen höheren SME. Basierend darauf, könnten Methanhektarerträge durch die Nutzung von Genotypen mit höheren Blattanteilen noch weiter verbesset werden.
Zusammengefasst erlauben die in dieser Studie entwickelten Ansätze das ökologische und ökonomische Leistungsverhalten der Miscanthusproduktion, durch eine Verbesserung der Nährstoffnutzung, Umweltwirkung und Landnutzung, deutlich zu steigern und dadurch die Implementierung in die Praxis zu vereinfachen
Estimating heritability in plant breeding programs
Heritability is an important notion in, e.g., human genetics, animal breeding and plant breeding, since the focus of these fields lies on the relationship between phenotypes and genotypes. A phenotype is the composite of an organisms observable traits, which is determined by its underlying genotype, by environmental factors and by genotype-environment interactions. For a set of genotypes, the notion of heritability expresses the proportion of the phenotypic variance that is attributable to the genotypic variance. Furthermore, as it is an intraclass correlation, heritability can also be interpreted as, e.g., the squared correlation between phenotypic and genotypic values.
It is important to note that heritability was originally proposed in the context of animal breeding where it is the individual animal that represents the basic unit of observation. This stands in contrast to plant breeding, where multiple observations for the same genotype are obtained in replicated trials. Furthermore, trials are usually conducted as multi-environment trials (MET), where an environment denotes a year × location combination and represents a random sample from a target population of environments. Hence, the observations for each genotype first need to be aggregated in order to obtain a single phenotypic value, which is usually done by obtaining some sort of mean value across trials and replicates. As a consequence, heritability in the context of plant breeding is referred to as heritability on an entry-mean basis and its standard estimation method is a linear combination of variances and trial dimensions.
Ultimately, I find that there are two main uses for heritability in plant breeding: The first is to predict the response to selection and the second is as a descriptive measure for the usefulness and precision of cultivar trials. Heritability on an entry-mean basis is suited for both purposes as long as three main assumptions hold: (i) the trial design is completely balanced/orthogonal, (ii) genotypic effects are independent and (iii) variances and covariances are constant.
In the last decades, however, many advancements in the methodology of experimental design for and statistical analysis of plant breeding trials took place. As a consequence it is seldom the case that all three of above mentioned assumptions are met. Instead, the application of linear mixed models enables the breeder to straightforwardly analyze unbalanced data with complex variance structures. Chapter 2 exemplarily demonstrates some of the flexibility and benefit of the mixed model framework for typically unbalanced MET by using a bivariate mixed model analyses to jointly analyze two MET for cultivar evaluation, which differ in multiple crucial aspects such as plot size, trial design and general purpose. Such an approach can lead to higher accuracy and precision of the analysis and thus more efficient and successful breeding programs.
It is not clear, however, how to define and estimate a generalized heritability on an entry-mean basis for such settings. Therefore, multiple alternative methods for the estimation of heritability on an entry-mean basis have been proposed. In Chapter 3, six alternative methods are applied to four typically unbalanced MET for cultivar evaluation and compared to the standard method. The outcome suggests that the standard method over-estimates heritability, while all of the alternative methods show similar, lower estimates and thus seem able to handle this kind of unbalanced data.
Finally, it is argued in Chapter 4 that heritability in plant breeding is not actually based on or aiming at entry-means, but on the differences between them. Moreover, an estimation method for this new proposal of heritability on an entry-difference basis (H_Delta^2/h_Delta^2) is derived and discussed, as well as exemplified and compared to other methods via analyzing four different datasets for cultivar evaluation which differ in their complexity. I argue that regarding the use of heritability as a descriptive measure, H_Delta^2/h_Delta^2, can on the one hand give a more detailed and meaningful insight than all other heritability methods and on the other hand reduces to other methods under certain circumstances. When it comes to the use of heritability as a means to predict the response to selection, the outcome of this work discourages this as a whole. Instead, response to selection should be simulated directly and thus without using any ad hoc heritability measure.In der Humangenetik, Tier- und Pflanzenzüchtung sowie anderen Forschungsdisziplinen, bei denen die Beziehung zwischen Genotypen und Phänotypen im Fokus steht, ist die Heritabilität eine wichtige Maßzahl. Der Phänotyp setzt sich aus einem oder mehreren beobachteten Merkmalen eines Organismus zusammen und wird durch den zugrunde liegenden Genotypen, durch Umwelteinflüsse, sowie durch Genotyp-Umwelt-Wechselwirkungseffekte bestimmt. Die Heritabilität gibt an, welcher Anteil der phänotypischen Varianz genetisch bedingt ist. Sie kann als quadrierte Korrelation zwischen phänotypischen und genotypischen Werte interpretiert werden.
Ursprünglich wurde die Heritabilität in der Tierzüchtung vorgeschlagen, in welcher das einzelne Tier die kleinste Beobachtungseinheit darstellt. Dies steht im Gegensatz zur Pflanzenzüchtung, in der meist wiederholte Versuche durchgeführt werden, so dass derselbe Genotyp reproduziert und mehrfach beobachtet werden kann. Hinzu kommt, dass die Versuche meist in Versuchsserien an mehreren Standorten und über mehrere Jahre hinweg durchgeführt werden. Um also einen phänotypischen Wert je Genotyp zu erhalten, müssen dessen Beobachtungen aggregiert werden, was meist durch eine Form von Mittelwertbildung geschieht. Aus diesem Grund wird Heritabilität in der Pflanzenzüchtung standardmäßig als Heritabilität auf Sortenmittelwertbasis.
Ich sehe zwei Hauptnutzen von Heritabilität in der Pflanzenzüchtung: Zum einen kann mit ihr der Selektionserfolg vorhergesagt werden und zum anderen dient sie als beschreibende Maßzahl für die Präzision und Brauchbarkeit eines Versuchs. Die Heritabilität auf Sortenmittelwertbasis ist für beide Zwecke geeignet solange folgende Bedingungen erfüllt sind: (i) Das Versuchsdesign ist vollkommen balanciert/orthogonal, (ii) die Genotyp-Effekte sind unabhängig und (iii) alle Varianzen, sowie Kovarianzen sind konstant.
In den letzten Jahrzehnten gab es mehrere Weiterentwicklungen in der Methodik des Versuchsdesigns sowie der statistischen Analyse von Pflanzenzüchtungsversuchen. Gemischte Modelle ermöglichen komplexe Varianzstrukturen und unbalancierte Daten auszuwerten In Kapitel 2 wird beispielhaft gezeigt, welche Möglichkeiten und Vorteile in der Anwendung von gemischten Modellen liegen, indem typisch unbalancierte Datensätze von zwei verschiedenen Sortenversuchsserien mithilfe eines bivariaten gemischten Modells gemeinsam ausgewertet werden. Ansätze wie dieser können eine höhere Analyseexaktheit und präzision erzielen und demnach die Effizienz und den Erfolg von Pflanzenzüchtungsprogrammen steigern. Gleichzeitig führt dies dazu, dass die oben genannten Bedingungen nur selten erfüllt sind.
In solchen Fällen ist dann nicht klar, wie eine Heritabilität auf Sortenmittelwertbasis definiert und geschätzt werden kann. Mehrere alternative Methoden wurden vorgeschlagen. In Kapitel 3 werden sechs dieser alternativen Methoden für vier typische Datensätze aus Sortenversuchsserien berechnet und miteinander, sowie mit der Standardmethode verglichen. Die Ergebnisse deuten darauf hin, dass letztere die Heritabilität überschätzt, während alle alternativen Methoden ähnliche, niedrigere Schätzungen zeigen. Dies lässt vermuten, dass diese Methoden besser für die vorliegenden, unbalancierten Daten geeignet sind.
Abschließend wird in Kapitel 4 gezeigt, dass Heritabilität in der Pflanzenzüchtung im Grunde nicht auf Genotypmittelwerten sondern auf deren Differenzen basiert. Hieraus wird eine Methode zur Berechnung einer generalisierten Heritabilität auf Sortendifferenzbasis (H_Delta^2/h_Delta^2) hergeleitet und diskutiert. Vier unterschiedlich komplexe Datensätze von Sortenversuchen werden verwendet und mit alternativen Heritabilitätsschätzern verglichen. Bezüglich der Verwendung der Heritabilität als beschreibende Maßzahl bietet H_Delta^2/h_Delta^2 einen ausführlicheren und bedeutsameren Einblick als die alternativen Heritabilitätsschätzer oder die Standardmethode. Hinzu kommt, dass H_Delta^2/h_Delta^2 die bisher bekannten Methoden nicht nur verallgemeinert, sondern in Spezialfällen exakt abbildet. Basierend auf den Resultaten der gesamten Arbeit rate ich von der Verwendung von Heritabilität als Mittel zur Vorhersage des Selektionserfolges ab. Der Selektionserfolg sollte stattdessen direkt simuliert werden, sodass die Nutzung einer ad hoc Schätzungsmethode der Heritabilität unnötig ist
Yacon (Smallanthus sonchifolius Poepp. & Endl) - the potential of a neglected crop as an alternative sweetener and source of phytochemicals for functional foods
Yacon (Smallanthus sonchifolius Poepp. & Endl.) as an underutilized crop, native to the Andean region, has attracted growing attention. The tuberous roots of yacon have been advertised as an alternative low caloric plant source for replacing sucrose. In fact, yacon has gained recognition based on the fact that its sweet tasting tuberous roots and its leaves have a favourable phytochemical content to be included in a range of functional food products. The leaves on the one hand are a significant source of health promoting phenolic compounds and their extract exerts certain biological activities such as antioxidant activity and hyperglycemic effects. The tubers on the other hand consist of carbohydrates including simple sugars, namely, fructose, glucose, sucrose and fructooligosaccharides (FOS). The FOS - representing the dominant polysaccharide in the tubers - are sweet tasting, prebiotic, and non-digestible oligosaccharides. Therefore, their consumption imposes several health benefits such as lowering the energy intake while enhancing the beneficial microflora of the colon. It is noted that 60-70 % of the dry matter content of yacon tubers is composed of FOS. Besides, yacon tubers are a remarkable source of biological components such as phenolic compounds. Thus, yacon is considered as multifunctional plant food.
The main objectives of this thesis were to 1) differentiate between the quality of young and old yacon leaves of two cultivars (red and white) in terms of their total phenolic content (TPC), total flavonoid content (TFC), antioxidant activity when using ohmic-assisted decoction (OH-DE) and decoction (DE) as well as energy consumption of extraction process, 2) differentiate between various parts of yacon tubers (flesh, peel and whole tuber) of seven cultivars in terms of their simple sugar (fructose, glucose and sucrose) content, TPC, TFC and antioxidant activity, 3) examine the TPC and antioxidant activity of yacon tubers of two cultivars (red and white) one week and three weeks after the harvest and under the influence of different pre-treatments before drying, and 4) determine the effect of drying on quality of yacon chips produced from two cultivars (red and white) at two time intervals after harvest.
Overall, this thesis provided a broad dataset und information with regard to phytochemical contents of yacon leaves and tubers of different cultivars grown under the environmental conditions of southwestern Germany. However, further studies with regard to the determination of individual functional constitutes of leaves and tubers of yacon, their mechanism of action and effectiveness in promoting the health benefits, and their safety is essential. Moreover, with regard to novel product development from yacon leaves and tubers, further studies are strongly suggested to ensure the sustainability of final food products by optimizing energy consumption and environmental impacts of the whole food supply chain for such products as well as their quality.Yacon (Smallanthus sonchifolius Poepp. & Endl.), eine Knollenfrucht, die in der Andenregion heimisch ist, hat sich wachsende Aufmerksamkeit aufgezogen, da die Knollen als alternative, pflanzliche Zuckerquelle beworben wurden. Neben den Knollen stellen die Blätter eine bedeutende Quelle für gesundheitsfördernde phenolische Verbindungen dar und ihr Extrakt weist bestimmte biologische Aktivitäten, wie ein hohes antioxidatives Potenzial und hyperglykämische Wirkung, auf. Die Yaconknollen bestehen aus Kohlenhydraten einschließlich der Einfachzucker Fructose, Glucose,Saccharose und Fructooligosacchariden (FOS). Die FOS gelten als süß schmeckende, präbiotische Oligosaccharide. Ihr Verzehr führt zu einigen gesundheitlichen Vorteilen, wie der Verringerung der Energiezufuhr, während gleichzeitig die vorteilhafte Mikroflora des Dickdarms verbessert wird. Es wurde festgestellt, dass 60-70 % des Trockensubstanzgehalts der Yaconknolle aus FOS besteht. Außerdem sind Yaconknollen eine bemerkenswerte Quelle für biologische Komponenten wie etwa den phenolischen Verbindungen. Yacon gilt somit als multifunktionales pflanzliches Nahrungsmittel.
Die Hauptzielen dieser Dissertation darin bestand: 1) die Qualität junger und alter Yaconblätter zweier Sorten (rot und weiß) hinsichtlich ihres Gesamtphenolgehaltes (TPC), des Gesamtflavonoidgehalts (TFC) und des antioxidativen Potenzials mit Hilfe der Verwendung verschiedener Extraktionsverfahren sowie dem Energieverbrauch des Extraktionsprozesses zu unterscheiden, 2) verschiedene Teile der Yaconknollen (Fruchtfleisch, Schale und ganze Knolle) hinsichtlich ihres Gehalts an Einfachzucker (Fructose, Glucose und Saccharose), TPC, TFC und der antioxidativen Aktivität zu unterscheiden, 3) die TPC- und Antioxidationsaktivität von zweier Sorten (rot und weiß) eine Woche und drei Wochen nach der Ernte und unter dem Einfluss verschiedener Vorbehandlungen vor der Trocknung zu untersuchen und, 4) die Auswirkung der Trocknung auf die Qualität von Yaconchips von zwei Sorten (rote und weiß) in zwei Zeitabständen nach der Ernte zu bestimmen.
Insgesamtdie vorliegende Dissertation bietet eine umfassende Datengrundlage zu den phyotchemischen Gehalten verschiedener Inhaltsstoffe in Yaconblättern und knollen, die unter den gegebenen Umweltbedingungen in Südwestdeutschland erzeugt wurden. Um jedoch, sind weitere Studien zur Bestimmung der Zusammensetzung der Blätter und Knollen von Yacon, ihres Wirkmechanismus und ihrer Wirksamkeit im Hinblick auf ihre ernährungsphysiologischen Vorteile sowie die finale Produktsicherheit von wesentlicher Bedeutung. Im Hinblick auf die Entwicklung neuer Lebensmittelprodukte aus Yaconblättern und Knollen werden weitere Studien empfohlen, um die Nachhaltigkeit der Endprodukte zu gewährleisten und den Energieverbrauch und die Umweltauswirkungen der gesamten Wertschöpfungskette zu optimiere
