    Agricultural education in Honduras : return on investment, quality, and efficiency

    The Agri-food sector, like any other economic sector, requires competent workers and entrepreneurs able to maximize productivity and cope with the various challenges the sector is facing. Agricultural education prepares these actors to acquire knowledge, identify options that optimize their productivity, and adapt to changing environments. However, the importance of agricultural education is declining in education policies, and nowadays, it is no longer an attractive field of study for the youth. There is a need to re-invent agricultural careers to the new demands of the agri-food and labor market sectors. As there is a limited number of studies available that analyze agricultural education in Latin-American and other developing countries, this research provides new insights by examining the determinants for return to investment, quality, and efficiency of agricultural education, taking Honduras as a case study. The present dissertation encompasses three interrelated studies. The first study addresses the research question on how the educational attainment on agricultural education, years of experience, age, sex, geographical region of residence, and work sector, affect the graduates earnings. This study computes the return to agricultural education by educational level, using several econometric techniques, such as the instrumental variables, Heckmans two-step procedure, and the multinomial logit to tackle endogeneity of education, sample selection bias, and selection bias for choosing this field of study, respectively. The results show that regardless of the economic activity in which the individuals work, graduates from agricultural educational programs receive positive returns on their investment in education. High school graduates from these programs, have higher earnings within the economic activity of agriculture than non-graduates. At the university level, the graduates competencies are better paid in economic activities outside agriculture than inside, for example, in fields of manufacturing, public administration, and education. The second study addresses the question of what competencies employers require from graduates of agricultural education programs. Interviews and workshops were conducted, addressing employers from agricultural value chains, purposely selected, and using maximum variation sampling. The results show that graduates perform at an intermediate level in technical competencies. Of the 20 competencies identified, graduates perform poorly in at least six areas of competence, which requires immediate action. Although the analysis of academic curricula shows that the competencies included are relevant for employers to cope with the challenges faced by the agri-food sector, there is room for improvement. Finally, the third study addressed the question about the level of efficiency of the public institutions delivering agricultural educational programs, and the factors influencing it. This study computes the efficiency using the non-parametric approach data envelopment analysis (DEA). The model used is output-oriented, variable return to scale, and two-stage where controllable and non-controllable inputs are included in the first and second stages, respectively. This study follows a double bootstrapping procedure to avoid the caveats of the conventional DEA analysis. The evidence showed that none of the agriculture education centers in the sample is considered a full efficient entity. However, there is a potential to improve the usage of the current resources. It is possible by expanding the outcomes in a range of 1% to 50% without changing the existing resources. In summary, the findings of this dissertation provide compelling evidence that each additional level of agricultural education in Honduras is a worthwhile investment, and that graduates from both education levels, secondary and tertiary levels, are necessary to support Honduras economy. Nevertheless, improvements should be made in terms of the quality and the resources used in the provision of agricultural education. Based on the research findings, it is recommended to revise the academic curricula, with the aim of better matching education and labor market requirements. Secondly, a closer analysis of the implementation of the educational processes is necessary in order to enhance the competencies requested and identify better management of the resources available. Finally, setting formal collaboration mechanisms between the education centers, authorities, and the private sector could strongly improve agricultural education systems contribution to the performance of the sector. To further refine this research, it would be beneficial to increase the sample sizes to expand further the techniques used.

    Comparative transcriptome analysis and functional studies of enterohemorrhagic Escherichia coli after cultivation in plant medium

    Bei enterohämorrhagischen Escherichia coli (EHEC) handelt es sich um humanpathogene Bakterien, welche beim Menschen schwerwiegende gastrointestinale Erkrankungen auslösen können. Der Gastrointestinaltrakt von Rindern wird als Hauptreservoir von EHEC angesehen und die primäre Infektionsquelle des Menschen stellt kontaminiertes, rohes Fleisch dar. Jedoch kam es in den letzten Jahrzehnten vermehrt zu EHEC-Infektionen, welche mit dem Verzehr von rohem Gemüse und Salatpflanzen assoziiert wurden. Heute ist es anerkannt, dass diese Infektionen dadurch begünstigt werden, dass EHEC-Bakterien in der Lage sind, Pflanzen als Sekundärwirte zu nutzen und damit eine Übertragung auf den Menschen erleichtern. Um diese Interaktion zwischen Pathogen und Pflanze besser zu verstehen, sind grundlegende Kenntnisse in der molekularen Anpassung an pflanzliche Bestandteile nötig. Im Rahmen dieser Arbeit wurde die Anpassung verschiedener EHEC Stämme an pflanzliche Bestandteile untersucht, dazu wurden O157:H7 Stamm Sakai, O104:H4 Stamm C227-11phicu und O157:H- Stamm 3072/96 als Prototypen ausgewählt. Zunächst konnte in Wachstumsexperimenten in einem artifiziellen Salatmedium gezeigt werden, dass Inhaltsstoffe des Salatmediums für ein erfolgreiches Wachstum aller drei Stämme ausreichend waren. In der RNA-Sequenzierung wurde die differentielle Genexpression der drei Stämme nach Wachstum in Salatmedium gegenüber dem Wachstum in M9-Minimalmedium bestimmt. Um Gene zwischen den Stämmen anhand einer einheitlichen Genbezeichnung miteinander zu vergleichen, wurde die differentielle Genexpression mit der Grundlage eines geteilten Genoms der drei pathogenen Stämme inklusive Referenzstamm Escherichia coli Stamm K-12 Substamm MG1655 durchgeführt. Analog zum erfolgreichen Wachstum in Anwesenheit pflanzlicher Bestandteile zeichnete sich eine erhöhte Transkription von zahlreichen Genen des Kohlenhydrat- und Peptidmetabolismus in allen drei Stämmen ab. Besonders die Gene des Lactose- (lacZ), Ribose- (rbsAC) und Xylosemetabolismus (xylF) waren in allen Stämmen deutlich hochreguliert. Die deutlichsten transkriptionellen Unterschiede zwischen den Stämmen zeigten sich in der Regulation von Motilitäts- und Chemotaxis-Genen. O104:H4 Stamm C227-11phicu zeigte in Anwesenheit pflanzlicher Bestandteile eine starke Expression aller Gene der drei Flagellenklassen (Klasse I, II und III). Dazu gehören die Gene, welche für den Aufbau der Flagellengrundstruktur (fli, flg), der Ausbildung des Filaments (fliC) sowie des Chemotaxis-Systems (che, tar, tap) verantwortlich sind. Dem entgegen konnte bei O157:H7 Stamm Sakai ausschließlich eine erhöhte Expression der Klasse I und II Flagellengene beobachtet werden. In Übereinstimmung mit den Transkriptomdaten zeigten diese beiden Stämme ein gesteigertes Schwimm- und Schwärmverhalten auf Motilitätsplatten in Anwesenheit von Salatextrakt. Ausschließlich bei dem, ausgelöst durch eine Deletion im Gen flhC, unbeweglichen O157:H- Stamm 3072/96 konnte eine gesteigerte Expression von Virulenzfaktoren der LEE-Pathogenitätsinsel beobachtet werden. Dazu gehörten Gene für den Aufbau des T3SS (esc) sowie T3SS-vermittelten Effektoren (esp). Interessanterweise äußerte sich dieser Stamm als leistungsfähiger Biofilmbildner in M9-Minimalmedium. Des Weiteren konnte gezeigt werden, dass die Komplementierung des intakten flhC-Gens die Beweglichkeit von O157:H- Stamm 3072/96 wieder herstellen konnte. In diesem Zusammenhang konnte ebenfalls festgestellt werden, dass die Deletion im Gen flhC nicht der alleinige Grund für die erhöhte Biofilmbildung dieses Stammes war. Zusätzlich zur Bestimmung der Biofilmbildung wurden die Stämme auf ihre Adhärenz an HT-29 Zellen untersucht. Hierbei konnte für O157:H- Stamm 3072/96 gegenüber den motilen Stämmen eine signifikant erhöhte Adhärenz beobachtet werden, das geringste Adhärenzpotential konnte bei O157:H7 Stamm Sakai festgestellt werden. Die Ergebnisse dieser Studie liefern klare Hinweise, dass die verschiedenen EHEC-Stämme fähig sind, sich an die Nährstoffverfügbarkeiten in einem pflanzlichen Wirt anzupassen. Es kann davon ausgegangen werden, dass Flagellen und das Chemotaxis-System eine entscheidende Rolle in der Auffindung von Pflanzen und deren Erschließung spielen. Außerdem könnten Curli-Strukturen eine elementare Funktion bei der initialen Anheftung und der anschließenden Biofilmbildung auf pflanzlichem Gewebe spielen. Vermutlich sind, neben dem typischen Pflanzen-assoziierten Ausbruchstamm O157:H7 Stamm Sakai, auch weitere EHEC-Stämme in der Lage ihr genetisches Repertoire auszunutzen, um eine Antwort auf die atypische Bedingungen dieser Nische zu gewährleisten. Die in dieser Arbeit gewonnenen Ergebnisse legen nahe, dass die Stämme aufgrund ihrer unterschiedlichen genetischen Ausstattung, neben zahlreichen übereinstimmenden Mechanismen, zusätzlich auch stammspezifische Strategien bei einer Interaktion mit Pflanzen als Sekundärwirte zeigen.Enterohemorrhagic Escherichia coli (EHEC) are human pathogens which are able to cause severe gastrointestinal diseases in humans. The gastrointestinal tract of cattle is considered as the main reservoir for EHEC and contaminated raw meat represents the primary source of infection. Yet there have been increasing reports over the last few decades of EHEC infections that were linked to the consumption of raw vegetables. Today it is generally accepted that EHEC bacteria are able to use plants as their secondary hosts, thus favouring the transmission to humans. To improve the understanding of this pathogen-plant interaction fundamental knowledge about the pathogens molecular adaptions towards plant material is urgently required. In the cope of this study the adaption of different EHEC strains towards components of the plant was examined. Therefore O157:H7 strain Sakai, O104:H4 strain C227-11phicu and O157:H strain 3072/96 were chosen as surrogates. In growth experiments performed with an artificial lettuce medium it could be shown that components of the lettuce were sufficient for the proliferation of the three strains. RNA-sequencing was performed to study the differential gene expression of the three strains after the growth in lettuce medium compared to the growth in M9 minimal medium. In order to compare genes according to standardized gene denotations, the differential gene expression analysis was performed on the basis of a shared genome including the genomes of the three pathogenic strains as well as the genome of Escherichia coli strain K-12 substrain MG1655. Analogous to the successful growth in presence of components of the plant an upregulation of genes involved in carbohydrate and peptide metabolism throughout all three strains was observed. Especially genes involved in the catabolism of lactose (lacZ), ribose (rbsAC) and xylose (xylF) were found to be uniformly upregulated. The greatest differences among the strains accounted for the regulation of motility and chemotaxis genes. O104:H4 strain C227-11phicu showed a strong upregulation of all three classes of the flagellar hierarchy (class I, II and III) in presence of plant derived compounds. These included genes involved in the establishment of the basal body hook structure (fli, flg), the synthesis of the flagellar filament (fliC), and the chemotaxis-system (che, tap, tar). In contrast, O157:H7 strain Sakai only featured upregulation of class I and class II genes. According to the transcriptional data both of these strains also showed increased swimming and swarming behaviour on motility plates in presence of lettuce extract. Solely O157:H- strain 3072/96, which is non-motile due to a deletion in the flhC gene, showed an upregulation of virulence factors encoded on the LEE pathogenicity island, including genes involved in the establishment of the T3SS (esc) and T3SS secreted effectors (esp). Interestingly, it was shown for O157:H- strain 3072/96 to have a powerful capacity to form biofilms in M9 minimal medium. Furthermore it was proven that the complementation of an intact flhC gene restored motility in O157:H- strain 3072/96. In this regard it could be shown that the deletion in flhC was not the mere reason for the augmented biofilm formation capacity. In addition to the biofilm formation, the strains potential to adhere to HT-29 cells was examined. Here a significantly increased adherence potential for O157:H- strain 3072/96 with respect to the motile strains could be observed, the lowest adherence potential was determined for O157:H7 strain Sakai. The results presented in this study clearly indicate that the different EHEC strains are capable to adapt towards the nutrient availability provided by their plantal host. It can be assumed that flagella and the chemotaxis system play a fundamental role in the finding and exploitation of the plant. Furthermore curli structures might play a crucial role in the initial adherence and the subsequent establishment of a biofilm on plant tissues. Presumably, besides the typical plant associated outbreak strain O157:H7 strain Sakai, there are further strains capable of utilizing their genetic repertoire in order to adapt towards the atypical environmental conditions within this niche. The findings of this study suggest that the strains, besides sharing multiple coinciding mechanisms, are able to adapt in a strain specific manner and use different strategies in coping with plants as their secondary hosts

    The effect of aging in the murine gut microbiome

    Aging is characterized by several physiological changes. During the lifespan, the biological systems from the body of humans and other animals remain dynamic. Throughout the early stages of life, the microbiome develops into a complex ecosystem with thousands of species. Variations related to diet, environmental changes, medications affect the diversity and composition of the microbiota through the lifespan. Some old individuals with higher incidence of chronic diseases have a loss of the stability of the microbiome and an imbalance occurs between the different colonizers of the gut, also named dysbiosis. One of the most distinctive changes occurring with age is the prevalent low grade inflammation, which is named inflamm-aging. This not only changes the microbial composition of the GIT but also affects the permeability. Murine models are well established and help us to understand the complex dynamics between the host and the microbial communities inhabiting the gastrointestinal tract. These models allow us to analyze microbial communities from tissue and mucosa, from all sections of the gut, which is limited in humans. Methods standardization is an important topic in microbiome research. In chapter 2 it was compared the efficiency of two sample methods, cotton swab and tissue biopsy, in characterizing the mouth microbiota. In recent years, the mouth microbiome is being seen as a diagnostic tool for not only oral diseases but also systemic diseases. As physiological changes occur with aging, the microbiome from the mouth is affected and there is an increase of pathogens present in the oral surfaces. In murine models, cotton swab is a common tool used for sampling the microbiome of the oral cavity. In our study, we observed similar microbial community structure using both methodologies. However, the species Streptococcus danieliae, Moraxella osloensis, and some unclassified members of Streptococcus were affected by the different sampling procedures. In this trial, we included mice at two different ages, 2 months old being considered young and 15 months old considered middle aged mice. We observed changes in the genera Actinobacillus, Neisseria, Staphylococcus, and Streptococcus related to the age of the animal and the sampling type. These results showed the importance of sampling standardization in microbiome research and that age has a strong effect on the microbial ecology of the oral cavity. In chapter 3, it was studied the bacterial communities from duodenum and colon of mice at 2, 15, 24 and 30 months of age in combination with the results of the expression levels of antimicrobial peptides in small intestine and markers of intestinal barrier function. Besides, in this chapter were also assessed the indices of liver damage, inflammation and expression levels of lipopolysaccharide binding protein (Lbp) as well as of toll-like receptors (Tlr) 1-9 in liver tissues. At 24 and 30 months of age there was an increase in inflammation, they developed fibrosis and the levels of endotoxin in plasma were higher. Regarding changes in the microbiome, the duodenum had more changes than the colon related to age. Allobacullum, Bifidobacterium, Olsenella, Corynebacterium were the genera that differed statistically in the duodenum through the murine lifespan. Fewer changes were observed in the colon, as Allobaculum was the only genus that showed differences between young and old mice. Additionaly, it was analyzed the impact of aging in the active microbial communities of mouth, duodenum and colon at 2, 9, 15, 24 and 30 months of age (chapter 4). Changes were observed at every age and different taxonomical levels, with a greater shift at 15 months of age. This is related to the age of the mice, as at middle age systemic changes related to the aging process start to occur. At old ages, there was an increment of the pathobiontic species Helicobacter hepaticus and Helicobacter ganmani in the duodenum and colon. The oral, duodenal and colonic microbial communities are important pieces of information that can be related to the health status of the host. Research that focuses on assessing the changes in the different niches and not only in the feces, gives a broader overview of the microbial community of the host.Das Altern ist durch verschiedene physiologische Veränderungen gekennzeichnet. Während der Lebensdauer von Menschen und anderen Tieren bleiben die biologischen Systeme des Körpers dynamisch. In den frühen Lebensphasen entwickelt sich das Mikrobiom zu einem komplexen Ökosystem mit Tausenden von Arten. Variationen in Bezug auf Ernährung, Umweltveränderungen und Medikamenten beeinflussen die Vielfalt und Zusammensetzung der Mikrobiota während des gesamten Lebens. Bei einigen alten Personen mit einer höheren Inzidenz chronischer Krankheiten ist ein Verlust der Stabilität des Mikrobioms zu beobachten und es tritt ein Ungleichgewicht zwischen den verschiedenen Darmbesiedlern auf, was auch als Dysbiose bezeichnet wird. Eine der auffälligsten Veränderungen ist eine im Alter auftretende niedriggradige Entzündung, die als inflamm-aging bezeichnet wird. Diese verändert nicht nur die mikrobielle Zusammensetzung des GIT, sondern beeinflusst auch dessen Permeabilität. Mausmodelle sind gut etabliert und helfen uns, die komplexe Dynamik zwischen dem Wirt und den im Magen-Darm-Trakt lebenden mikrobiellen Gemeinschaften zu verstehen. Diese Modelle erlauben es uns, die mikrobiellen Gemeinschaften in Gewebe und Mucosa in allen Abschnitten des Darms zu analysieren, was im Menschen limitiert ist. Ein wichtiges Thema in der Mikrobiomforschung ist die Standardisierung von Methoden. In Kapitel 2 wurde die Effizienz von den zwei Probenmethoden, Wattestäbchen und Gewebebiopsie, bei der Charakterisierung der Mundmikrobiota verglichen. In den letzten Jahren wurde das Mundmikrobiom als diagnostisches Instrument nicht nur für orale Erkrankungen, sondern auch für systemische Erkrankungen eingesetzt. Mit zunehmendem Alter treten physiologische Veränderungen auf, welche das Mikrobiom im Mund beeinflusst und eine Zunahme von Krankheitserregern auf den Mundoberflächen beobachten lässt. In Mausmodellen sind Wattestäbchen ein übliches Werkzeug zur Probeentnahme des Mikrobioms der Mundhöhle. In unserer Studie beobachteten wir mit beiden Methoden eine ähnliche Struktur der mikrobiellen Gemeinschaft. Jedoch wurden die Anteile der Arten Streptococcus danieliae, Moraxella osloensis und einige nicht klassifizierte Mitglieder von Streptococcus von den verschiedenen Probeentnahmeverfahren beeinflusst. In dieser Studie wurden Mäuse zwei verschiedener Altersstufen untersucht, wobei die 2 Monate alten Mäuse als jung und die 15 Monate alten als im mittleren Alter eingestuft wurden. Wir beobachteten Veränderungen in den Gattungen Actinobacillus, Neisseria, Staphylococcus und Streptococcus in Bezug auf das Alter des Tieres und dem Probentyp. Diese Ergebnisse zeigten, dass das Alter einen starken Einfluss auf die mikrobielle Ökologie der Mundhöhle hat und wie wichtig eine standardisierte Probenentnahme in der Mikrobiomforschung ist. In Kapitel 3 wurden die bakteriellen Gemeinschaften aus Zwölffingerdarm und Dickdarm von Mäusen im Alter von 2, 15, 24 und 30 Monaten in Kombination mit den Ergebnissen der Expressionsniveaus von antimikrobiellen Peptiden im Dünndarm und Markern der Darmbarrierefunktion untersucht. Außerdem wurden in diesem Kapitel die Indizes für Leberschäden, Entzündungen und Expressionsniveaus von Lipopolysaccharid-Bindungsprotein (Lbp) sowie von Toll-like-Rezeptoren (Tlr) 1-9 in Lebergeweben bewertet. Im Alter von 24 und 30 Monaten kam es bei den Mäusen zu einem Anstieg der Entzündungswerte, sie entwickelten eine Fibrose und die Endotoxinspiegel im Plasma waren höher. In Bezug auf Veränderungen im Mikrobiom kam es im Zwölffingerdarm zu mehr altersbedingten Veränderungen als im Dickdarm. Allobacullum, Bifidobacterium, Olsenella, Corynebacterium waren die Gattungen, die sich im Zwölffingerdarm über die Lebensdauer der Maus statistisch unterschieden. Im Dickdarm wurden weniger Veränderungen beobachtet, Allobaculum war die einzige Gattung, die Unterschiede zwischen jungen und alten Mäusen zeigte. Zusätzlich wurde der Einfluss des Alterns in den aktiven mikrobiellen Gemeinschaften von Mund, Zwölffingerdarm und Dickdarm im Alter von 2, 9, 15, 24 und 30 Monaten analysiert (Kapitel 4). Veränderungen wurden in jedem Alter und auf verschiedenen taxonomischen Ebenen beobachtet, im Alter von 15 Monaten kam es zu der größten Veränderung. Dies hängt mit dem Alter der Mäuse zusammen, da im mittleren Alter systemische Veränderungen im Zusammenhang mit dem Alterungsprozess auftreten. Im hohen Alter gab es eine Zunahme der pathobiontischen Arten Helicobacter hepaticus und Helicobacter ganmani im Zwölffingerdarm und Dickdarm. Die oralen, duodenalen und kolonalen mikrobiellen Gemeinschaften sind wichtige Informationen, die auf den Gesundheitszustand des Wirts schließen lassen. Forschungen, die sich auf die Bewertung der Veränderungen in den verschiedenen Nischen und nicht nur im Kot konzentrieren, geben einen breiteren Überblick über die mikrobielle Gemeinschaft des Wirts

    Going beyond GDP with a parsimonious indicator : inequality-adjusted healthy lifetime income

    Per capita GDP has limited use as a well-being indicator because it does not capture many dimensions that imply a good life, such as health and equality of opportunity. However, per capita GDP has the virtues of easy interpretation and can be calculated with manageable data requirements. Against this backdrop, a need exists for a measure of well-being that preserves the advantages of per capita GDP, but also includes health and equality. We propose a new parsimonious indicator to fill this gap and calculate it for 149 countries

    The retirement migration puzzle in China

    We examine whether and how retirement affects migration decisions in China. Using a regression discontinuity (RD) design approach combined with a nationally representative sample of 228,855 adults aged between 40 and 75, we find that retirement increases the probability of migration by 12.9 percentage points. Approximately 38% of the total migration effects can be attributed to inter temporal substitution (delayed migration). Retirement-induced migrants are lower-educated and have restricted access to social security. Household-level migration decisions can reconcile different migration responses across gender. Retirees migrate for risk sharing and family protection mechnisms, reducing market production of their families in the receiving households

    Behavioural economic aspects in the strategic development of membership relations in rural cooperatives

    Aufgrund von anhaltenden Wandlungsprozessen verändern sich die Rahmenbedingungen für ländliche Genossenschaften (e.G.) und ihre Mitglieder ständig. Wandlungsprozesse sind von Interesse, weil sie u.a. Bedürfnisse von Menschen und dementsprechend Geschäftsfelder beeinflussen. Im Zuge der Wandlungsprozesse nimmt die Komplexität der landwirtschaftlichen Produktion zu und daraus folgt, dass Landwirte ein gesteigertes Bedürfnis nach Wissen und Informationen haben, um ihren Betrieb weiter zu entwickeln. Eine Organisationsform, welche ihre Mitglieder z.B. in Form von Beratung fördert, sind e.G. Zusätzlich nimmt der Wettbewerbsdruck auf ländliche e.G. zu. Um sich den Veränderungen in der Agrarwirtschaft anzupassen, nehmen ländliche e.G. strukturelle Veränderungen vor. Im Zuge des Konsolidierungsprozesses im Genossenschaftssektor steigt zum einen bspw. die Heterogenität von Mitarbeitern und Mitgliedern an und zum anderen kann es zwischen Organbetrieb und Genossenschaftsmitglied zu einem Verlust von Vertrauen kommen. Folglich kann Selbstverpflichtung und Loyalität des Mitgliedes in Bezug auf seine e.G. abnehmen. Vor dem Hintergrund beschriebener Transformationen Anpassungs- und Innovationsdruck von deutschen Landwirten und des Konsolidierungsprozesses im Genossenschaftssektor und deren strategischer Bedeutung für die Agrar- und Ernährungsbrache sowie der Ansicht, dass die Genossenschaftsforschung eine anwendungsorientierte Wissenschaft ist, verfolgt die vorliegende Dissertation das Ziel, ländlichen e.G. praxisrelevante Fragestellungen im Rahmen ihres Mitgliedermanagements zu beantworten. Dadurch kann ein Erkenntnistransfer von der Forschung in die Praxis ermöglicht sowie eine nachhaltige Entwicklung von ländlichen e.G. begünstigt werden. Demzufolge wurden auf Grundlage der Neuen Institutionenökonomie, verhaltensökonomischer Ansätze und mit Hilfe analytischer Tests, wie des U-Tests nach Mann und Whitney, sowie Regressionsanalysen, folgende Forschungsfragen (FF) behandelt: FFi: Was verstehen Mitglieder ländlicher Genossenschaften unter der von ihrer Genossenschaft ausgeführten Servicequalität? FFii: Wodurch lassen sich Vertrauensbeziehungen zwischen Mitgliedern und Mitarbeitern in ländlichen Genossenschaften erfolgversprechend gestalten? FFiii: Welche Förderleistungen können Genossenschaften zur Entwicklung ländlicher Regionen übernehmen? Die in dieser kumulativen Dissertation aufgeführten Studien beruhen vermehrt auf Datensätzen, welche im Bundesland Baden-Württemberg mittels quantitativer Erhebungsinstrumente wie schriftlichen Fragebögen, im Zeitraum 2016 bis 2017, erhoben wurden. U.a. findet das MemberValue Modell Anwendung. Hierbei wird die Wirksamkeit des spezifischen Förderauftrages der jeweiligen e.G. anwendend analysiert. Die unterschiedlichen Stichproben der dieser Dissertation beiliegenden Studien setzen sich aus Befragungen von Mitarbeitern, Mitgliedern und leitenden Mitgliedern von gemeinwesenorientierten und einer ländlichen e.G. zusammen. Mitarbeiter stellen einen notwendigen Faktor zur Erfüllung der Mitgliederzufriedenheit und loyalität dar. Nach derzeitigem Wissen des Autors wurde daher erstmalig die Mitglieder MitarbeiterBeziehung in letztgenannter ländlicher e.G. näher beleuchtet. Diese kumulative Dissertation liefert neue Erkenntnisse für das innerorganisationale Verständnis von e.G. im deutschsprachigen Raum insbesondere in Bezug auf weiche Faktoren wie Partizipation, Vertrauen und Zufriedenheit.Due to ongoing processes of change, the frame conditions for rural cooperatives (coop) and their members are constantly changing. Change processes are of interest because they influence, among other things, the needs of people and, accordingly, business areas. The complexity of agricultural production is increasing in the course of these processes of change and, as a result, farmers have an increased need for knowledge and information in order to further develop their business. One form of organisation that supports its members, e.g. in the form of advice, is the registered association. Additionally, the competitive pressure on rural registered associations is increasing. In order to adapt to the changes in the agricultural economy, rural cooperatives make structural changes. In the course of the consolidation process in the cooperative sector, the heterogeneity of employees and members increases on the one hand and on the other hand, there can be a loss of trust between the organ company and the cooperative member. As a consequence, the commitment and loyalty of the member with regard to his or her coop may decrease. Against the background of the described transformations (pressure of adaptation and innovation from German farmers and the consolidation process in the cooperative sector) and their strategic importance for the agricultural and food sector as well as the view, that cooperative research is an application-oriented science, the present dissertation aims to answer rural coop practice-relevant questions within the framework of their membership management. Thus, a transfer of knowledge from research to practice can be enabled and a sustainable development of rural cooperatives can be promoted. Consequently, the following research questions (rq) were addressed on the basis of New Institutional Economics, behavioural economic approaches and with the help of analytical tests, such as the Mann and Whitney U-test, and regression analyses: rqi: What do members of rural cooperatives understand by the quality of service provided by their cooperative? rqii: How can relationships of trust between members and employees in rural cooperatives be shaped successfully? rqiii: What member value can cooperatives provide for the development of rural regions? The studies listed in this cumulative dissertation are increasingly based on data sets that were collected in the federal state of Baden-Württemberg using quantitative survey instruments such as written questionnaires in the period 2016 to 2017. Among others, the MemberValue model is applied. Here, the effectiveness of the specific funding mandate of the respective coop is analysed by applying it. The different samples of the studies accompanying this dissertation are composed of employees, members and leading members of community oriented and a rural coop. Employees represent a necessary factor for the fulfilment of member satisfaction and loyalty. According to the current knowledge of the author, the member-employee relationship in the latter rural coop was therefore examined in more detail for the first time.     Biotechnological conversion of lignocellulose hydrolyzates : model microorganisms for a bio-based economy

    Lignocellulose has substantial potential as a carbon source in a bio-based economy. It is the most abundant renewable raw material on earth and is available in large quantities as waste from the agriculture, food and wood industry. It is composed mainly of the polymers lignin, cellulose and hemicellulose. In contrast to glucose derived from cellulose, hemicellulose sugars often remain unused although 60 billion tons of hemicelluloses are produced annually. Hemicelluloses are a group of heterogeneous polysaccharides consisting of different monomers such as D xylose, D arabinose, D mannose and D galactose. Lignocellulose is mostly depolymerized in order to obtain fermentable sugars. During the depolymerization process, inhibitors such as organic acids or furan aldehydes can be formed or released, which could be problematical for biotechnological processes. The aim of this thesis was to develop and evaluate bacterial-based biotechnological processes capable of using hemicellulose sugars as a source of carbon. First, Pseudomonas putida KT2440 was chosen. Pseudomonades are claimed as a promising chassis in biotechnology due to their versatile and robust metabolism. Unlike other Pseudomonades, the strain KT2440 is classified as biosafety level 1 in the American Type Culture Collection (ATCC). However, these bacteria can metabolize glucose as the only lignocellulose monosaccharide. Cellvibrio japonicus was the second selected bacterium. This strain is not yet established as a microbial host in biotechnology, but can degrade a huge portfolio of plant cell wall polysaccharides and is also classified as biosafety level 1 in ATCC. The topic of the first publication was to engineer P. putida KT2440 strains for metabolizing the hemicellulose monosaccharides xylose and arabinose and characterize their growth behavior. Initially, an arabinose metabolizing strain with the araBAD operon and a xylose metabolizing strain with xylAB operon was constructed. Later on, these strains were cultivated in minimal salt medium with glucose, xylose and arabinose as carbon sources in Erlenmeyer flasks. The recombinant P. putida KT2440 strains metabolized xylose and arabinose with high growth rates comparable to glucose. It turned out that both engineered strains were able to grow on both pentoses as well as on mixtures of glucose xylose and arabinose. The intent of the second publication was to evaluate P. putida KT2440 as a platform model organism for bioconversion of lignocellulose hydrolyzates. Strains were cultivated in minimal salt medium with several hydrolyzates as carbon source in Erlenmeyer flask and bioreactor. In addition, the growth-inhibiting effect of major toxic substances contained in lignocellulose hydrolyzates on P. putida KT2440 was analyzed via cultivation experiments. Several suitable hydrolyzates were figured out for this strain. Formic acid and acetic acid proved to be relatively unproblematic under pH neutral conditions, whereas furfural and hydroxymethylfurfural (HMF) had a negative effect on the bacterial growth. A diauxic-like growth behavior was revealed via fed batch bioreactor cultivations, since pentoses were almost not consumed with sufficient glucose supply. Consequently, feed-medium was added step-by-step in the next experiment. The applied feed profile did lead to an almost complete metabolization of xylose. The purpose of the third publication was to evaluate C. japonicus as a potential host strain for the one‐step bioconversion of xylans into rhamnolipids. Cultivation experiments were performed in Erlenmeyer flasks filled with minimal salt medium and containing different carbon sources. Furthermore, the strain was transformed with the plasmid pSynPro8oT carrying rhlA (encodes acetyltransferase) and rhlB (encodes rhamnosyltransferase I) to complete the rhamnolipid metabolism. The strain grew on all main lignocellulose monosaccharides as well as, on different xylans. Mono rhamnolipids were produced with the engineered strain using xylans as carbon source. This is particularly interesting as most industrially relevant bacteria are not able to depolymerize wood polymers. As the product yields were quite low, there are still many challenges in order to achieve an economically efficient process. Nevertheless, to the best of our knowledge, it is the first published one step bioconversion of hemicellulose polymers into rhamnolipids. In total, P. putida KT2440 turned out as a flexible and powerful model organism and two xylose and arabinose metabolizing strains were constructed. Moreover, bioreactor cultivations with lignocellulose hydrolyzates were performed and a feeding strategy to overcome diauxic-like growth behavior was presented. A proof of concept for a one-step bioconversion of xylans into rhamnolipids with a recombinant C. japonicus strain was successfully demonstrated.Im Rahmen einer biobasierten Wirtschaft wird Lignocellulose als Kohlenstoffquelle ein großes Potential zugesprochen. Sie ist der am häufigsten vorkommende nachwachsende Rohstoff auf unserer Erde und steht in großen Mengen als Abfallstoff aus der Agrar-, Lebensmittel- und Holzindustrie zur Verfügung. Lignocellulose besteht hauptsächlich aus den Polymeren Lignin, Cellulose und Hemicellulose. Im Gegensatz zu der aus der Cellulose stammenden Glucose werden die Hemicellulose-Zucker selten als Kohlenstoffquelle in biotechnologischen Prozessen verwendet, obwohl jährlich ca. 60 Milliarden Tonnen Hemicellulosen anfallen. Hemicellulosen sind eine Gruppe von heterogenen Polysacchariden, welche hauptsächlich aus den Monosacchariden D Xylose, L Arabinose, D Mannose D Glucose und D Galactose zusammengesetzt sind. Um fermentierbare Zucker aus der Lignocellulose zu erhalten, muss diese meist depolymerisiert werden. Bei diesem Verzuckerungsprozess können für biotechnologische Prozesse problematische Stoffe wie z.B. organische Säuren oder Furanaldehyde gebildet oder freisetzet werden. Ziel dieser Arbeit war es, bakteriell basierte biotechnologische Prozesse zu entwickeln und zu bewerten, die Hemicellulose-Zucker als Kohlenstoffquelle nutzen können. Zum einen wurde der Bakterienstamm Pseudomonas putida KT2440 ausgewählt. Pseudomonaden gelten aufgrund ihres vielseitigen und robusten Stoffwechsels als vielversprechende Biokatalysatoren. Im Gegensatz zu vielen anderen Pseudomonaden ist der Stamm KT2440 in der ATCC (American Type Culture Collection) unter der Risikogruppe 1 eingeordnet. Allerdings können Pseudomonaden außer Glucose keine weiteren Lignocellulose-Zucker als Kohlenstoffquelle nutzen. Als zweiter Bakterienstamm wurde Cellvibrio japonicus ausgewählt. Dieser Stamm ist als Wirt für biotechnologische Prozesse nicht etabliert, ist aber in der Lage, ein großes Portfolio an Pflanzenzellwand Polysacchariden abzubauen und ist der Risikogruppe 1 zugeordnet. Das Thema der ersten Publikation war die Konstruktion von Xylose- und Arabinose-verstoffwechselnden P. putida KT2440-Stämmen und die anschließende Charakterisierung ihres Wachstumsverhaltens. Zunächst wurde jeweils ein Stamm zur Metabolisierung von Xylose und Arabinose konstruiert. Hierzu wurde das xylAB-Operon und das araBAD-Operon von Escherichia coli verwendet. Zur Analyse des Wachstumsverhaltens wurden die Stämme daraufhin in Minimalmedium und mit Glucose, Xylose und Arabinose als Kohlenstoffquelle in Erlenmeyerkolben kultiviert. Die rekombinanten P. putida KT2440 Stämme wuchsen mit Xylose und Arabinose mit ähnlich hohen Wachstumsraten wie mit Glucose. Zudem konnten beide Stämme die jeweils andere Pentose verstoffwechseln und es konnten Kultivierungen mit Mischungen aus Glucose, Xylose und Arabinose erfolgreich durchgeführt werden. Ziel der zweiten Publikation war es, das Potenzial von P. putida KT2440 als Biokatalysator für die Biokonversation von Lignocellulose-Hydrolysaten zu bewerten. Die Stämme wurden in Minimalmedium mit verschiedenen Hydrolysaten als Kohlenstoffquelle in Erlenmeyerkolben und im Bioreaktor kultiviert. Darüber hinaus wurde mittels Kultivierungsversuchen die wachstumshemmende Wirkung von in Lignocellulose-Hydrolysaten enthaltenen toxischen Substanzen auf P. putida KT2440 untersucht. Es konnten mehrere geeignete Hydrolysate für diesen Stamm ausgemacht werden. Ameisensäure und Essigsäure erwiesen sich als relativ unproblematisch unter pH-neutralen Bedingungen, wohingegen Furfural und Hydroxymethylfurfural (HMF) schon bei niedrigen Konzentrationen einen negativen Effekt auf das Wachstum von P. putida KT2440 hatten. Mittels eines Fed Batch Prozesses im Bioreaktor mit Lignocellulose-Hydrolysaten wurde ein diauxi ähnliches Wachstumsverhalten nachgewiesen. Bei ausreichend verfügbarer Glucose wurde die Xylose kaum verstoffwechselt. Als mögliche Strategie zur Lösung dieses Problems wurde der Feed stufenweise dazugegeben. Dies führte zu einer nahezu vollständigen Verstoffwechselung von Xylose. Im Rahmen der dritten Veröffentlichung wurde ein Prozess zur einstufigen Biokonversion von Hemicellulose-Polymeren zu Rhamnolipiden mit dem Bakterium C. japonicus als Wirtstamm getestet. C. japonicus wurde in Minimalmedium zusammen mit verschiedenen Kohlenstoffquellen in Erlenmeyerkolben kultiviert. Um den Rhamnolipidstoffwechsel zu vervollständigen, wurde der Stamm mit dem Plasmid pSynPro8oT mit rhlA (kodiert Acetyltransferase) und rhlB (kodiert Rhamnosyltransferase I) transformiert. C. japonicus wuchs auf allen wichtigen Lignocellulose-Monosacchariden sowie auf verschiedenen Xylanen. Mit dem rekombinanten Stamm und Xylan als Kohlenstoffquelle konnte Mono-Rhamnolipid hergestellt werden. Dies ist insbesondere interessant, da die meisten industriell relevanten Bakterien nicht in der Lage sind, Holzpolymere zu depolymerisieren und zu verstoffwechseln. Jedoch erwies sich die Transformation als schwierig und es konnten nur niedrige Produktausbeuten erzielt werden. Aufgrund dessen sind noch viele Herausforderungen zu bewältigen, um einen wirtschaftlich rentablen Prozess zu erhalten. Nichtsdestotrotz handelt es sich nach unserem Wissen um die erste einstufige Biokonversation von Hemicellulose-Polymeren zu Rhamnolipiden. Zusammenfassend, erwies sich P. putida KT2440 als flexibler und leistungsfähiger Wirtsorganismus. Xylose und Arabinose metaboliserende Stämme wurden konstruiert und Prozessstrategien für Bioreaktor Kultivierungen wurden entwickelt. Des Weiteren konnte ein Proof of Concept für eine einstufige Biokonversion von Xylanen zu Rhamnolipiden mit einem transformierten C. japonicus Stamm erfolgreich erbracht werd

    Identification of target molecules for innovative concepts in mesothelioma therapy : investigation of tyrosine kinases and members of the NF2/mTOR and p14/p16 signaling pathways

    Pleurale Mesotheliome (MPM) sind diffus wachsende Tumore der Pleura, die in 90% der Fälle auf eine Asbestexposition zurückgeführt werden können. Die Diagnosestellung erfolgt meist spät, wenn eine chirurgische Resektion mit kurativer Intention nicht mehr in Frage kommt. Weil Pleuramesotheliome zudem überwiegend resistent gegenüber Strahlentherapie und Chemotherapeutika sind, zählen sie zu den besonders schwer behandelbaren Tumoren, bei denen weniger als 5% der Patienten länger als fünf Jahre überleben. Die zielgerichtete Therapie, die auf der Inhibierung aberrant aktivierter Moleküle durch spezifische Inhibitoren beruht, stellt eine vielversprechende Behandlungsstrategie dar. In präklinischen Studien konnten spezifische Inhibitoren von mTOR oder einzelnen Kinasen das Wachstum von Mesotheliomzellen hemmen, allerdings war keiner der Inhibitoren in klinischen Studien als Monotherapie effizient genug. Das Ziel dieser Arbeit bestand darin, neue Zielmoleküle für eine gerichtete Therapie des pleuralen Mesothelioms zu identifizieren. Dabei sollten insbesondere Tyrosinkinasen und Komponenten des p14/p16- und NF2/mTOR-Signalwegs und die Effekte einer simultanen Inhibierung untersucht werden. Im Fokus standen Moleküle, die als Teil dieser Signalwege auch in anderen Tumorentitäten aktiv sind und für die deshalb spezifische Inhibitoren verfügbar sind. Das erste Teilprojekt sollte die Frage beantworten, ob die Hemmung der Tyrosinkinase ALK mittels Crizotinib allein oder in Kombination mit dem mTOR-Inhibitor Rapamycin das Wachstum von MPM-Tumoren effizient hemmen kann. Dazu wurde eine in vivo-Therapiestudie mit einem Mesotheliom-PDX-Modell, welches ALK und mTOR koexprimierte, durchgeführt. Crizotinib allein war nicht wirksam, verstärkte jedoch die therapeutische Wirkung von Rapamycin und induzierte - erstmalig nachgewiesen - eine Verringerung des Tumorvolumens in einem MPM-Xenograft-Modell. Die simultane Inhibierung von mTOR und ALK supprimierte nicht nur die mTORC1- und ALK-Signaltransduktion, sondern auch die durch Rapamycin induzierte AKT-Aktivierung. Die Kombinationstherapie inhibierte die Tumorzellproliferation und induzierte Autophagie. Apoptose wurde nicht induziert, jedoch ein Caspase-unabhängiger, nekrotischer Zelltod. Auch wenn nur 5% der MPM-Patiententumore ALK und mTOR gleichzeitig exprimieren, konnte diese Studie zeigen, dass es prinzipiell möglich ist, Mesotheliome durch eine geeignete Kombinationstherapie effizient zu therapieren. Im zweiten Projekt wurde erstmals die Eignung des p14/p16-regulierten Transkriptionsfaktors BMI1 als Zielmolekül für die Therapie von MPM untersucht. Eine erhöhte BMI1-Transkriptexpression wurde in 81,3% der MPM-Patiententumore nachgewiesen, eine Koexpression mit mTOR Voraussetzung für eine Kombinationstherapie in 25% der untersuchten Fälle. In vitro-Therapieversuche mit dem BMI1-Inhibitor PTC-209 und den mTOR-Inhibitoren INK-128 und BEZ235 an MPM-Zelllinien ergaben, dass die Einzeltherapie mit den mTOR-Inhibitoren sowie PTC-209 in allen Zelllinien zu einer dosisabhängigen Reduzierung der viablen Zellen führte. Die kombinierte Therapie mit mTOR-Inhibitoren und BMI1-Inhibitor verstärkte den therapeutischen Effekt um das 2fache (BEZ235) bzw. 10fache (INK-128), verglichen mit der PTC-209 Monotherapie. Die Hemmung von BMI1 insbesondere in Kombination mit einem Inhibitor des PI3K/mTOR-Signalweges stellt somit einen attraktiven Ansatz für eine zielgerichtete Therapie des MPM dar. In einem dritten Teilprojekt sollten neue Zielmoleküle für eine gerichtete Therapie durch eine genomweite, arraybasierte CNV-Analyse an MPM-Patiententumoren identifiziert werden. Dadurch wurden signifikante, bisher nicht für MPM beschriebene genomische Zugewinne, unter anderem auf 9p22.2, 13q34 und Chromosom 19 (19p13.3, 19p13.11 und 19q13.2), identifiziert. Die Verifizierung der kritischen Regionen erfolgte mittels qPCR und FISH. Durch die Reanalyse zweier bereits publizierter Datensätze war es möglich, genomische Daten von insgesamt 182 MPM-Tumoren zu analysieren. Mit dieser Herangehensweise wurden mehrere kritische genomische Regionen mit potentiellen Zielgenen identifiziert, darunter die onkogenen Kinasen AKT1, AKT2, MAP3K10 und PTK2. Für AKT2 und MAP3K10 wurde im Rahmen dieser Studie eine erhöhte Genexpression in MPM-Tumoren nachgewiesen. Insbesondere die AKT-Isoformen AKT1 und AKT2 stellen Targets mit großem therapeutischen Potential dar, weil mit dem pan-AKT-Inhibitor Ipatasertib ein Therapeutikum zur Verfügung steht, das aktuell in klinischen Studien getestet wird. Diese Arbeit liefert somit mehrere Ansatzpunkte für die Entwicklung molekular ausgerichteter Therapieoptionen für pleurale Mesotheliome. Die Ergebnisse der vorliegenden Arbeit zeigen zudem, dass es für die Entwicklung einer wirksamen Therapie sinnvoll ist, eine simultane Inhibierung mehrerer Signalwege anzustreben, da im pleuralen Mesotheliom verschiedene Signalwege häufig gleichzeitig aktiv sind.Malignant pleural mesothelioma is a diffuse growing, rare form of cancer affecting the outer lining of the inner chest wall (pleura). About 90% of the cases are related to a prolonged exposure to asbestos. MPM is difficult to distinguish from other respiratory diseases and is therefore often diagnosed at late stages, when curative surgery is no longer a treatment option. Moreover, radiotherapy and chemotherapy only show limited efficacy, including a less than 5% five year survival rate. Targeting single molecules involved in progression, growth and metastasis with specific inhibitors might be a promising new approach for this tumour entity. However, till date there are no recommendations for targeted therapies in MPM. Although specific inhibitors of mTOR or single kinases suppressed MPM tumor growth in pre-clinical studies, they were not effective in clinical trials. Therefore, there is an urgent need to identify new treatment approaches and targets. The aim of this PhD thesis was to identify new therapeutical targets for the treatment of malignant pleural mesothelioma with special emphasis on the simultaneous inhibition of tyrosine kinases, p14/p16 and NF2/mTOR pathway molecules. Moreover it was focused on molecules within these pathways for which specific inhibitors are already available. One key aspect of this project was to examine mTOR and ALK as potential therapeutic targets, and to determine the efficacy of ALK and mTOR inhibition on tumor cell growth with ALK inhibitor Crizotinib and mTOR inhibitor Rapamycin, in individual and combinatorial treatment. Therefore, an ALK and mTOR co-expressing patient-derived MPM xenograft model was used. Crizotinib alone did not show anti-tumor efficacy whereas combined treatment with Rapamycin enhanced anti-tumor effect of Rapamycin treatment, and lead to a reduction in tumor volume in the MPM xenograft model. Simultaneous inhibition not only suppressed mTOR and ALK pathway transduction but also Rapamycininduced AKT activation. Combined therapy significantly suppressed cell proliferation and induced autophagy as well as caspase-independent, necrotic cell death compared to the single treatment. Although only 5% of MPM tumors coexpress ALK and mTOR, the results of this study confirm that combined treatment with Rapamycin and Crizotinib can be a therapeutic treatment option in pleural mesothelioma. Further analyses included the investigation of p14/p16 regulator BMI1 as a therapeutic target for the treatment of MPM. Increased BMI1 expression was detected in 81.3% of the MPM patient samples analyzed. Coexpression of BMI1 and mTOR, a prerequisite for combined therapy, was present in 25% of the analyzed cases, as detected by qRT-PCR and IHC. Monotherapy with mTORC1/2 inhibitor INK-128, mTOR/PI3K inhibitor BEZ235 and BMI1 inhibitor PTC-209 resulted in dose-dependent reduction of cell viability in three MPM cell lines tested. Co-treatment with mTOR inhibitors enhanced therapeutic effect of BMI1 inhibition 2-fold (BEZ235) and 10-fold (INK-128) respectively, thus representing an appealing approach for targeted therapy in MPM. The last part of the project aimed to identify new therapeutic targets by genome-wide, array-based CNV- analysis of 42 MPM. Array-based ONCOSCAN technology, especially developed for formalin-fixed and paraffin-embedded tissue, and GISTIC algorithm identified new gains on chromosome 9p22.2, 13q34 and chromosome 19 (19p13.3, 19p13.11 and 19q13.2), which were verified via qPCR and FISH. GISTIC-reanalysis of previously published data sets provided genomic data of 182 MPM tumors in total and was used to identify critical regions for further research on new target genes like oncogenic kinases AKT1, AKT2, MAP3K10 and PTK2 among others. AKT2 and MAP3K10 gene and protein expression was elevated in the tested MPM cohort. Especially AKT isoforms AKT1 and AKT2 represent attractive targets with important therapeutic potential, since pan-AKT inhibitor Ipatasertib is currently being tested in clinical trials for other tumors. In conclusion, the results of this PhD thesis demonstrate the potential for small molecule inhibitors as therapeutic strategies for the treatment of malignant pleural mesothelioma. Moreover, the results of this study suggest that the concurrent inhibition of mTOR, ALK and AKT pathways is a viable therapeutic strategy, as these pathways are frequently simultaneously activated in pleural mesothelioma
