    Strategies for sustainable pearl millet hybrid breeding in West Africa

    Pearl millet [Pennisetum glaucum (L.) R. Br.] is grown by >90 million subsistence farmers, mostly in the drylands of Sub-Saharan Africa and India for human consumption and provides additionally fodder and building material. It is commonly grown in regions with 300 500 mm of precipitation, low soil phosphorus levels, and temperatures of >42°C), like its center of origin in West Africa (WA). Pearl millet is a highly heterozygous, diploid (2n = 2x = 14) C4 plant species with outcrossing rates of >70%. Yield levels increased largely in India and the US, while they almost stagnated in WA. Challenging, highly variable environments and a weak seed sector are largely contributing to these differences. To suggest a way forward this thesis was meant to guide heterotic group development for sustainable WA pearl millet breeding. The specific objectives were to (I) facilitate efficient use of pearl millet gene bank accessions, (II) identify diversity patterns, (III) validate the yield superiority and stability of pearl millet population hybrids over OPVs, (IV) derive a more comprehensive picture about combining ability patterns, and (V) develop a unified strategy for heterotic grouping and sustainable hybrid breeding. A total of 81 accessions acquired from the pearl millet reference collection was evaluated for resistance to Striga hermonthica (Del.) Benth. in one artificially infested field in Niger. A subset of 74 accessions was characterized in 2009 in multi-environment trials (MET) under low-input and fertilized conditions. The general superiority of local check varieties compared to the genebank accessions highlighted the importance of local adaptation, possibly lost during the ex situ conservation and regeneration. Nevertheless, the development and preservation of germplasm collections are important to maintain the rich genetic diversity. The MET identified several accessions as sources for specific traits of interest and revealed an immense diversity but also strong admixture. This admixture underlines the need to develop heterotic groups. Therefore, 17 WA open-pollinated varieties (OPVs) were crossed in a diallel mating design and tested together with their offspring in nine environments over two years in Niger and Senegal. Results from these MET verified large panmictic better parent heterosis (PBPH) effects with an average of 18% (147%) for panicle yield. A large G × E interaction variance was confirmed and it was not possible to define repeatable mega-environments. Importantly, yield stability was more pronounced in the population hybrids compared to their parental OPVs. Furthermore, a superior combining ability among selected OPVs from Niger vs. Senegal was revealed and the evaluated OPVs were clearly grouped by origin based on genetic information. Nevertheless, there was no significant relationship between genetic distance among OPVs and PBPH. These and earlier studies showed a large diversity, sufficiently large heterosis effects and high yield stability in experimental pearl millet population and topcross hybrids, offering a great opportunity for a regionally coordinated hybrid breeding approach. Therefore, we suggested a unified strategy with a continuous output of different hybrid types, specifically tailored to WA. First, existing diversity and combining ability pattern information on western WA and eastern WA cultivars forming loose groups that combine well with each other should be used. Selected genotypes with high general combining ability (GCA) and per se performance from eastern and western WA, respectively, are promising founder populations. Initiating a reciprocal recurrent selection (RRS) program, possibly supported by modern breeding tools, will diverge the two groups further, while improving the inter-pool per se performance. RRS in combination with continuous diversification of both pools allows distinct female and male pool development, line development and introgression of a cytoplasmic male sterility system. Creating OPVs and population hybrids early and aiming for long-term development of topcross hybrids from improved OPVs and newly derived lines is possible alongside the heterotic pool development. Additionally, the RRS lays the foundation for possible future single-cross hybrid breeding programs. The suggested framework is highly ambitious and requires long-term commitment, vision and financial resources. Considering the flexibility regarding single steps and the possibility to develop different types of varieties at every stage of the pool diversification, it has the potential to enhance gains from selection and, with the continuous output of new high-yielding and stable cultivars, to improve the livelihood of WA subsistence farmers substantially.Perlhirse [Pennisetum glaucum (L.) Perlhirse ist eine sehr heterozygote, diploide (2n = 2x = 14) C4-Pflanze mit Auskreuzungsraten von >70%. Die Erträge stiegen in Indien und den USA, während sie in WA fast stagnierten. Herausfordernde, sehr variable Umwelten und ein schwacher Saatgutsektor tragen weitgehend zu diesen Unterschieden bei. Daher soll diese Thesis die Entwicklung heterotischer Gruppen für eine nachhaltige WA Perlhirsezüchtung anleiten. Die spezifischen Ziele waren (I) effiziente Nutzung von Perlhirsesorten aus Genbanken zu erleichtern, (II) Diversitätsmuster zu identifizieren, (III) Ertragsüberlegenheit und Stabilität von Perlhirse-Populationshybriden gegenüber OPVs zu validieren, (IV) ein umfassenderes Bild über Kombinierfähigkeitsmuster zu erstellen und (V) eine einheitliche Strategie für die heterotische Gruppierung und nachhaltige Hybridzüchtung zu entwickeln. Perlhirsesorten aus der Referenzsammlung wurden auf Striga hermonthica (Del.) Benth. Resistenz in einem künstlich infizierten Feld im Niger untersucht. Davon wurden 74 Genotypen in mehrortigen Versuchen (MET) in 2009 unter gering und regulär gedüngten Bedingungen charakterisiert. Die generelle Überlegenheit der lokalen Standards gegenüber den Genbank-Genotypen zeigt die Bedeutung der lokalen Anpassung, die möglicherweise durch die ex situ Erhaltung und Regeneration verloren ging. Dennoch sind die Entwicklung und Erhaltung von Keimplasma-Sammlungen wichtig, um die reiche genetische Vielfalt zu erhalten. Die MET identifizierten mehrere Genotypen als Quellen für bestimmte, relevante Merkmale und zeigten eine immense Vielfalt, aber auch starke Vermischung. Diese Vermischung zeigt das Erfordernis, heterotische Gruppen zu entwickeln. Daher wurden 17 WA offen bestäubte Sorten (OPVs) in einem Diallel-Paarungsschema gekreuzt und mit ihren Nachkommen in neun Umwelten über zwei Jahre im Niger und Senegal getestet. Diese Ergebnisse verifizierten große panmiktische Heterosiseffekte relativ zum besseren Elter (PBPH) von durchschnittlichen 18% (1 47 %) beim Rispenertrag. Eine große G × E Interaktionsvarianz wurde bestätigt und es war nicht möglich wiederholbare Mega-Umwelten zu definieren. Wichtig ist, dass die Erträge der Populationshybriden im Vergleich zu ihren elterlichen OPVs stabiler waren. Darüber hinaus zeigte sich eine überlegene Kombinationsfähigkeit bestimmter OPVs aus Niger vs. Senegal und die evaluierten OPVs wurden auf Grundlage genetischer Informationen klar nach Herkunft gruppiert. Dennoch gab es keinen signifikanten Zusammenhang der genetischen Distanz zwischen den OPVs und PBPH. Diese und frühere Studien zeigten eine große Diversität, ausreichend große Heterosis Effekte und eine hohe Ertragsstabilität in experimentellen Perlhirse Populations- und Topcross-Hybriden, was eine große Chance für einen regionalen Hybridzüchtungsansatz bietet. Daher schlugen wir eine einheitliche, speziell auf WA zugeschnittene Strategie mit kontinuierlicher Produktion verschiedener Hybridtypen vor. Zunächst sollten Informationen über die bestehende Vielfalt und die Kombinationsfähigkeitsmuster bei westlichen WA- und östlichen WA-Sorten, die lose Gruppen bilden und sich gut miteinander kombinieren lassen, genutzt werden. Ausgewählte Genotypen mit hoher allgemeiner Kombinationsfähigkeit (GCA) und Eigenleistungen aus Ost- und West-WA sind vielversprechende Gründerpopulationen. Die Initiierung eines Programms zur reziproken rekursiven Selektion (RRS), eventuell unterstützt durch moderne Züchtungsmethoden, wird die beiden Gruppen weiter divergieren und gleichzeitig die Eigenleistung je Genpools verbessern. RRS in Kombination mit einer kontinuierlichen Diversifizierung beider Pools ermöglicht die Entwicklung eines jeweils eindeutig weiblichen und männlichen Pools, Linienentwicklung und Introgression eines zytoplasmatischen männlichen Sterilitätssystems. Außerdem ist es möglich, OPVs und Populationshybride frühzeitig zu erzeugen und auf eine langfristige Entwicklung von Topcross-Hybriden aus verbesserten OPVs und neu entwickelten Linien hinzuarbeiten. Darüber hinaus legt das RRS den Grundstein für mögliche zukünftige Zweiweghybridzüchtungsprogramme. Der vorgeschlagene Rahmen ist sehr ehrgeizig und erfordert langfristiges Engagement, Voraussicht und finanzielle Ressourcen. In Anbetracht der Flexibilität der einzelnen Schritte und der Möglichkeit, in jeder Phase der Pooldiversifizierung verschiedene Sortentypen zu entwickeln, hat es das Potenzial, die Selektionserfolge zu steigern und mit der kontinuierlichen Produktion neuer ertragreicher und stabiler Sorten die Lebensgrundlage der Subsistenzlandwirte in WA erheblich zu verbessern

    Development of data-driven evaluation methodology for analysis and estimation of the reactor output of biogas plants

    Die Biogasherstellung ist ein sehr komplexer Prozess, der durch das Zusammenspiel unterschiedlicher Mikroorganismen und einem mehrstufigen Prozessablauf gekennzeichnet ist. Variationen in der Mikroflora können durch das verwendete Substrat, die Beschickung sowie die Temperatur gegeben sein, wobei ein Großteil der beteiligten Mikroorganismen sogar unbekannt sein kann. Die Überwachung und Analyse solcher Prozesse sind zumeist sehr zeit- und kostenintensiv. Die VDI-Richtlinie 4630 beschreibt analytische Methoden zur Untersuchung der Vergärung organischer Stoffe, Substratcharak¬terisierung, Probenahme, Stoffdatenerhebung und Gärversuche, die zur Optimierung von Biogasanalgen dienen. Die in der Richtlinie beschriebenen Methoden erfordern jedoch eine entsprechende Laborausstattung, dafür ausgebildetes Personal und sind sehr aufwändig. Deshalb ist eine moderne Alternative zur Prozessanalyse und -optimierung dringend erforderlich, um gezielt wesentliche Prozessgrößen schnell, effizient und kostengünstig zu identifizieren. Das Ziel der vorliegenden Dissertation ist es eine Methode zu entwickeln, welche eine schnelle und effiziente Analyse der Biogasproduktionsprozesse ermöglicht. In dieser Arbeit soll mithilfe von computerbasierten Verfahren untersucht werden, ob wesentliche Prozessgrößen identifiziert werden können, deren Messung für die Führung der Biogasanlagen ausschlaggebende Informationen zur Verfügung stellt. Neben mathematischen Modellen werden auch Optimierungsalgorithmen für die Identifikation der wesentlichen Einflussgrößen verwendet. Die für die Modellierung verwendeten Datensätze wurden experimentell im Rahmen des Projektes ,Biogas Biocoenosis (FNR 22010711, Dr. Michael Klocke, Leibniz-Institut für Agrartechnik und Bioökonomie e. V., Potsdam) und Biogas-Enzyme (FKZ 22027707, Dr. Monika Heiermann, Leibniz-Institut für Agrartechnik und Bioökonomie e. V., Potsdam) erhoben. Zusätzlich wurden Prozessdaten mithilfe des Anaerobic Digestion Modell No.1(ADM1) simuliert. Die chemischen Prozessgrößen stellen die unabhängigen Prozessvariablen dar. Die Biogasausbeute wird als die abhängige Zielprozessgröße verwendet. Die Vorhersage der Biogasausbeute wird mithilfe linearer und nicht linearer mathematischer Modelle durchgeführt. Dafür werden die Partial-Least-Squares-Regression (PLSR), die Locally-Weighted-Regression (LWR) und künstliche neuronale Netzte (ANN, Artificial Neural Networks) implementiert. Um die wesentlichen unabhängigen Prozessvariablen zu identifizieren, werden folgende Optimierungsalgorithmen verwendet: ein Ameisen-Kolonie-Optimierungsalgorithmus (ACO) und der genetische Algorithmus (GA). Die Qualität der Vorhersage wird anhand des Vorhersagefehlers (Root Mean Square Error, RMSE) und des Bestimmtheitsmaßes (R2) bewertet. Anhand der entwickelten Methode konnte die gewünschte Zielgröße, Biogasausbeute, vorhergesagt werden. Die durchgeführte Variablenselektion mittels metaheuristischer Optimierungsverfahren verbesserte die Vorhersagekraft der Modelle und reduzierte gleichzeitig die Anzahl der unabhängigen Prozessvariablen. Als wesentliche Prozessvariablen wurden folgende identifiziert: hydraulische Verweilzeit, Trockensubstanz, Gehalt an Neutral-Detergentien-Faser, Säure-Detergentien-Lignin-Gehalt und n-Buttersäure. Die besten Vorhersageergebnisse wurden mit Hilfe der künstlichen neuronalen Netze erzielt. Die Modelle hatten den kleinsten Fehler der Vorhersage und das höchste Bestimmtheitsmaß. Die erfolgreiche Umsetzung der mathematischen Verfahren zeigt, dass moderne mathematische Methoden eine gute Alternative für die Analyse und Optimierung komplexer biologischer Prozesse darstellen. Für eine abschließende Bewertung sind jedoch auch weitere experimentelle Untersuchungen notwendig, in denen die Prozessführung basierend auf den identifizierten Prozessvariablen durchgeführt wird.The production of biogas is very complex process, which runs in some stages involving different microorganisms. Microbiological diversity of the process depends mainly on the composition of substrate and ambient conditions, such as process temperature. The fact is, the development and composition of the microbiological communities of the process are difficult to predict. Thus, the control and evaluation of such complex biological processes are very time consuming and expensive. In Germany the evaluation of the biogas plants can be performed according to the VDI-Norm 4630, which describes the methods for the evaluation of fermentation of organic materials including characterization of the substrate, sampling, collection of material data and fermentation tests. For that specially equipment and skilled personnel are required. Moreover, the evaluation procedure is very time consuming. That is why a new state-of-the-art alternative for the evaluation purposes is necessary to simplify and to speed up the assessment of the biogas production processes. The aim of this doctoral thesis is the development of a fast and reliable method for the evaluation of the biogas production processes. Therefore the mathematical modelling should identify significant process variables able to evaluate the whole process. For the optimization of mathematical models metaheuristic tools were used. In this doctoral thesis two different data sets were used experimental data and simulated data. The experimental data were collected in projects Biogas-Biocoenosis (FKZ 22010711, Dr. Michael Klocke, Leibnitz-Institute für Agrartechnik und Bioökonomie e.V., Potsdam) and Biogas-Enzyme (FKZ 22027707, Dr. Monika Heiermann, Leibnitz-Institute für Agrartechnik und Bioökonomie e.V., Potsdam). The simulated data set was generated using the Anaerobic Digestion Model No.1 (ADM1). The chemical process variables were used as the independent process variable set, while the biogas production output represented the dependent process variable. Prediction of the biogas production was done using linear and nonlinear mathematic models. Here, Partial-Least-Square-Regression (PLSR), Locally-Weighted-Regression (LWR) and Artificial Neural Networks (ANN) were implemented. In order to identify the most significant undependable process variables optimization algorithms were used, Ant Colony Optimization (ACO) and Genetic Algorithm (GA). Prediction capacity was evaluated using two model evaluation variables, Root Mean Square Error (RMSE) and Coefficient of Determination (R2). Figure 1 in Supplementary represents the flow chart of the developed methodology applied for ADM1 generated data set. In Figure 2 (Supplementary) there is a flow chart of the developed methodology applied for the experimentally collected data. The developed approaches could be successfully used for the prediction of the desired process variable, biogas production rate. The variable selection done with the help of metaheuristic optimization algorithms improved the prediction results and reduced number of the independent process variables. Hydraulic retention time, dry matter, neutral detergent fibre, acid detergent fibre and n-butyric acid were identified as the most significant ones. The best prediction was obtained using ANN models. Here, the error of prediction was low and the coefficient of determination high. The successful implementation of the developed methodology proved mathematical models to be an effective alternative method capable to evaluate and to optimize complicated biological processes. Furthermore, it would be mandatory further experimental evaluation of the developed strategy, using the model-based process information

    Trade integration, global capital flows and the link to institutional quality from a North-South perspective

    This doctoral thesis is a cumulative dissertation containing three essays. In the first essay, I create a panel data set of North-South preferential trade agreements (PTAs) building on the comprehensive database on the design of trade agreements (DESTA). I analyze the effects of the depth and number of PTAs signed on the quality of institutions in developing countries, the global South, measured as the political risk component investment profile of the ICRG database. I show that the system GMM is the appropriate estimator to apply for my empirical analysis to account for various sources of endogeneity. I show that signing deep North-South PTAs positively affects institutions in the South. The results differ with respect to the type of agreement and region. The second essay deals with the determinants of PTAs focusing on institutional distance as a driving factor and regarding PTAs as an instrument to compensate for missing institutions. I argue that the effect of institutional distance is specifically important (1) in a North-South trade relationship where institutional distance is particularly large and (2) if countries trade a large share of contract-intensive goods. For this analysis I create a panel data set including a large number of developing countries and a variable to measure the difference of the share of bilateral contract-intensive exports and show that a linear probability model for discrete choice panel data is a suitable estimator to be used. I address endogeneity using an instrument variable (IV) approach. I show that institutional distance promotes the formation of PTAs. Comparing this effect for North-North, North-South, and South-South country pairs reveals that the positive effect of institutional distance on the probability of PTA formation is specifically high for the formation of North-South PTAs. Furthermore, I find that the effect is nonlinear and that trading contract-intensive goods reinforces the positive effect of institutional distance for the formation of North-South PTAs and may offset negative effects. Robustness checks with regard to the underlying sample reveal that the effect of institutional distance is driven by North-South relationships involving the EU. Essay 3 is dedicated to global investment flows and aims at deriving a global model to determine the factors of foreign direct investment (FDI) by considering investment flows between and within North and South. We empirically estimate and assess global FDI models, namely the gravity and knowledge capital (KK) model, based on the new CDIS data set by the IMF, which includes a large number of developing and transition countries. This allows us to detect potential vertical motives for FDI and to address the global trend of increasing FDI from and to the global South. We find the gravity model to achieve the best theory-consistent out-of-sample prediction, particularly when parameter heterogeneity of South and North FDI is allowed for. Controlling for surrounding market potential is important to recover the horizontal effect of the gravity model. Including institutional, cultural, or financial factors does not improve the model performance distinctly although results for those variables are mostly in line with theory.Diese Dissertation ist eine kumulative Dissertation bestehend aus drei Essays. Im ersten Aufsatz erstelle ich einen Panel-Datensatz aus Nord-Süd Präferenzhandelsabkommen (PTAs), der auf den umfassenden Datensatz Design of Trade Agreements Database (DESTA) aufbaut. Auf Basis des Panels analysiere ich die Auswirkungen der Tiefe und der Anzahl der unterzeichneten PTAs auf die Qualität der Institutionen von Entwicklungsländern, dem globalen "Süden", gemessen durch das "Investitionsprofil" des Political Risk Index des International Country Risk Guide (ICRG). Die Verwendung des System GMM zeigt sich als geeigneter Schätzer für die empirische Analyse, um verschiedene Quellen der Endogeneität zu berücksichtigen. Die Ergebnisse unterstützen die Hypothese, dass sich die Unterzeichnung tiefer Nord-Süd-PTAs positiv auf die Institutionen im "Süden" auswirkt. Die Ergebnisse unterscheiden sich in Bezug auf die Art der Abkommen und die Region der Entwicklungsländer. Der zweite Aufsatz befasst sich mit den Determinanten von Handelsabkommen. Der Fokus der Analyse liegt auf der institutionellen Distanz als Faktor und betrachtet Handelsabkommen als rechtliches Instrument zur Kompensation fehlender Institutionen. In meiner empirischen Analyse untersuche ich die Hypothese, dass die Wirkung der institutionellen Distanz insbesondere wichtig ist, wenn (1) Länderpaare in einer Nord-Süd-Handelsbeziehung stehen, in der die institutionelle Entfernung besonders groß ist und (2) Länder einen hohen Anteil vertrags-intensiver Güter handeln. Im Rahmen meiner Analyse erstelle ich einen Panel-Datensatz, der eine große Anzahl von Entwicklungsländern umfasst und eine Variable zur Messung der Distanz des Anteils bilateraler vertrags-intensiver Exporte. Ich zeige, dass ein lineares Wahrscheinlichkeitsmodell für Paneldaten mit diskreter Outcome-Variable ein geeigneter Schätzer ist. Potentielle Endogeneität berücksichtige ich im Rahmen einer geeigneten Instrumentierung. Die Ergebnisse zeigen, dass institutionelle Distanz die Bildung von Handelsabkommen fördert. Ein Vergleich dieses Effekts für Nord-Nord-, Nord-Süd- und Süd-Süd-Länderpaare zeigt, dass der positive Effekt der institutionellen Distanz auf die Wahrscheinlichkeit der Bildung von Handelsabkommen, speziell für die Bildung von Nord-Süd-Handelsabkommen, besonders hoch ist. Darüber hinaus stützen die Ergebnisse meine Annahme, dass der Effekt nichtlinear ist und dass der Handel vertrags-intensiver Gütern den positiven Effekt der institutionellen Entfernung für die Bildung von Nord-Süd-PTA verstärkt und negative Auswirkungen ausgleichen kann. Die Robustheitsprüfungen in Bezug auf die zugrunde liegende Stichprobe zeigen, dass die Auswirkungen der institutionellen Entfernung auf die Nord-Süd-Beziehungen insbesondere auf die Beteiligung der EU zurückzuführen sind. Essay 3 widmet sich den globalen Investitionsströmen und zielt darauf ab, ein globales Modell abzuleiten, um die Faktoren ausländischer Direktinvestitionen (FDI) zu bestimmen, indem die Investitionsströme zwischen und innerhalb des globalen "Nordens" und "Südens" berücksichtigt werden. Wir schätzen und bewerten empirisch globale FDI-Modelle, das Gravity- und Knowledge-Capital-Modell (KK), basierend auf dem neuen CDIS-Datensatz des IWF, der eine große Anzahl von Entwicklungs- und Transformationsländern umfasst. Dies ermöglicht es uns, potenzielle vertikale Motive für FDI zu erkennen und dem globalen Trend steigender FDI-Ströme aus und in den Süden zu erklären. Die Analyse zeigt, dass die Verwendung des Gravity-Modells zu den besten theorie-konsistenten Out-of-Sample-Vorhersagen führt, insbesondere wenn heterogene Faktoren in Süd- und Nord-FDI berücksichtigt werden. Die Berücksichtigung des umgebenden Marktpotenzials trägt insbesondere dazu bei, horizontale Effekte des Gravity-Modells aufzuzeigen. Die Einbeziehung institutioneller, kultureller oder finanzieller Faktoren verbessert die Modellleistung nicht wesentlich, obwohl die Ergebnisse für diese Variablen meist der Theorie entsprechen

    Studies of human genetic diseases and developmental processes with the frog Xenopus laevis

    Next generation sequencing is a driving force behind the identification of genes and alleles that are suspected to cause human genetic diseases. In silico tools are routinely used in the clinical everyday life to characterize unknown genotypes. However, these tools have a limited predictive accuracy and can only provide a first-line assessment. Especially un- or less studied genes require in every case predictive in vivo model systems that allow conclusions about disease associations. Classically, mice and zebrafish are utilized for such research, which concomitantly deepens the understanding of the involved developmental processes. In this collection of studies, the African clawed frog Xenopus laevis was used to explore and promote its suitability for the analysis of potential human disease genes, variants and their associated developmental processes. The first chapters covers potential candidate genes for primary ciliary dyskinesia (PCD). The second chapter addresses if an actin based motor protein and a novel metzincin peptidase, encoded by myosin ID (MYO1D) and leishmanolysin like peptidase (LMLN2)/tout-de-travers (TDT), respectively, are potentially causative for PCD independent laterality defects. The third chapter deals with two candidates for neurodevelopmental disorders, namely hyaluronan mediated motility receptor (HMMR) and progesterone immunomodulatory binding factor 1 (PIBF1).Next generation sequencing ist eine treibende Kraft hinter der Identifizierung von Genen und Allelen, die mutmaßlich humane Erbkrankheiten verursachen. Im klinischen Alltag werden routinemäßig in silico Methoden eingesetzt, um unbekannte Genotypen zu charakterisieren. Diese Methoden haben jedoch eine limitierte Vorhersagegenauigkeit und ermöglichen nur eine erste Beurteilung. Besonders spärlich oder nicht analysierte Gene erfordern in jedem Fall aussagekräftige in vivo Modellsysteme, die Schlüsse über etwaige Krankheitsassoziationen zulassen. Klassischer Weise werden für solche Forschungsbemühungen, die beiläufig auch das Verständnis der daran beteiligten Entwicklungsprozesse vertieft, Mäuse oder Zebrabärblinge verwendet. In dieser hier vorliegenden Studienkollektion wurde der Afrikanische Krallenfrosch Xenopus laevis verwendet, um dessen Eignung für die Analyse potentieller humaner Krankheitsgene, -genvarianten und deren assoziierten Entwicklungsprozesse zu ergründen und dies herauszustellen. Im ersten Kapitel werden Kandidatengene der primären ciliären Dyskinesie (PCD) besprochen. Das zweite Kapitel behandelt die Frage ob ein Aktin basierendes Motorprotein und eine neue Metzinkin Peptidase, jeweils von myosin ID (MYO1D) und leishmanolysin like peptidase 2 (LMLN2)/tout-de-travers (TDT) codiert, potentiell für PCD unabhängige Lateralitätsdefekte verantwortlich sein könnten.     News endorser influence in social media

    Social networking sites have become an online realm where users are exposed to news about current affairs. People mainly encounter news incidentally because they are re-distributed by users whom they befriended or follow on social media platforms. In my dissertation project, I draw on shared reality theory in order to examine the question of how the relationship to the news endorser, the person who shares news content, determines social influence on opinion formation about shared news. The shared reality theory posits that people strive to achieve socially shared beliefs about any object and topic because of the fundamental epistemic need to establish what is real. Social verification of beliefs in interpersonal communication renders uncertain and ambiguous individual perceptions as valid and objectively true. However, reliable social verification may be provided only by others who are regarded as epistemic authority, in other words as someone whose judgment one can trust. People assign epistemic authority particularly to socially close others, such as friends and family, or to members of their in-group. I inferred from this that people should be influenced by the view of a socially close news endorser when forming an opinion about shared news content but not by the view of a socially distant news endorser. In Study 1, a laboratory experiment (N = 226), I manipulated a female news endorsers social closeness by presenting her as an in-group or out-group member. Participants opinion and memory of a news article were not affected by the news endorsers opinion in either of the conditions. I concluded that the news article did not elicit motivation to strive for shared reality because participants were confident about their own judgment. Therefore, they did not rely on the news endorsers view when forming an opinion about the news topic. Moreover, the results revealed that participants had stronger trust in the news endorser when she expressed a positive (vs. negative) opinion about the news topic, while social closeness to the news endorser did not predict trust. On the one hand, this is in line with the social norm of sharing positive thoughts and experiences on social networking sites: adherence to the positivity norm results in more favorable social ratings. On the other hand, my findings indicate that participants generally had a positive opinion about the topic of the stimulus article and thus had more trust in news endorsers who expressed a similar opinion. In Study 2, an online experiment (N = 1, 116), I exposed participants to a news post by a relational close vs. relational distant news endorser by having them name a close or distant actual Facebook friend. There was a small influence of the news endorsers opinion on participants thought and opinion valence irrespective of whether the news endorser was a close or distant friend. The finding was surprising, particularly because participants reported stronger trust in the view of the close friend than in the view of a distant friend. I concluded that in light of an ambiguity eliciting news article, people may even rely on the views of less trustworthy news endorsers in order to establish a socially shared and, therefore, valid opinion about a news topic. Drawing on shared reality theory, I hypothesized that social influence on opinion formation is mediated by news endorser congruent responses to a news post. The results indicated a tendency for the proposed indirect relation however, the effect size was small and the sample in Study 2 was not large enough to provide the necessary statistical power to detect the mediation. In conclusion, the results of my empirical studies provide first insights regarding the conditions under which a single news endorser influences opinion formation about news shared on social networking sites. I found limited support for shared reality creation as underlying mechanism of such social influence. Thus, my work contributes to the understanding of social influence on news perception happening in social networking sites and proposes theoretical refinements to shared reality theory. I suggest that future research should focus on the role of social and affiliative motivation for social influences on opinion formation about news shared on social networking sites.Soziale Netzwerkseiten haben sich zu Orten entwickelt, an denen Nutzer:innen Nachrichten über aktuelle Ereignisse begegnen. Menschen treffen vor allem deswegen auf Nachrichten, weil diese von Personen geteilt werden, mit denen sie befreundet sind. In meiner Dissertation untersuche ich aufbauend auf der Shared Reality Theorie ob die Beziehung zu Nachrichten-Endorser:innen, also denjenigen, die Nachrichten teilen, bestimmt ob deren Ansichten einen Einfluss auf die Meinungsbildung haben. Die Shared Reality Theorie geht davon aus, dass Menschen ein fundamentales epistemisches Bedürfnis danach haben richtige und wahrhaftige Ansichten zu entwickeln. Deshalb streben sie nach Ansichten, die von anderen geteilt werden. Durch interpersonale Kommunikation verifizieren sie ihre Ansichten, wobei aus einer unsicheren und ambigen Wahrnehmung ein valides und objektives Urteil werden kann. Allerdings wird eine Wahrnehmung nur dann soziale verifiziert, wenn Menschen ihr Gegenüber als epistemische Autorität ansehen, also als jemanden, dessen Urteil sie vertrauen. Epistemische Autorität wird gewöhnlich Personen zugeschrieben, die einem nahestehen, z.B. Freunden oder Mitgliedern der eigenen In-Group. Daraus leite ich ab, dass die Meinungsbildung über Nachrichten, die auf sozialen Netzwerkseiten von einer sozial nahestehenden Person geteilt werden, von deren Ansicht beeinflusst sein sollte. Die Ansicht sozial entfernter Nutzer:innen sollte hingegen keinen Einfluss auf die Meinungsbildung haben. In Studie 1, einem Laborexperiment (N=226), manipulierte ich die soziale Nähe einer weiblichen Nachrichten-Endorserin, indem ich sie als In-Group oder Out-Group Mitglied vorstellte. In keiner der Bedingungen hatte die Meinung der Nachrichten-Endorserin einen Einfluss auf die Meinungen der Teilnehmenden über einen Nachrichtenartikel und auf ihre Erinnerung an dessen Inhalt. Daraus schließe ich, dass der Artikel keine Motivation für das Streben nach geteilter Realität ausgelöst hat, sondern die Teilnehmenden sich ihres eigenen Urteils sicher waren. Daher zogen sie die Ansicht der Nachrichten-Endorserin bei der Meinungsbildung nicht in Betracht. Darüber hinaus zeigen meine Ergebnisse, dass die Teilnehmenden dem Urteil der Nachrichten-Endorserin mehr vertrauten, wenn sie eine positive (vs. negative) Meinung über das Nachrichtenthema äußerte. Die soziale Nähe hingegen hatte keinen Effekt auf das Vertrauen in das Urteil der Nachrichten-Endorserin. Dies entspricht einerseits der auf sozialen Netzwerkseiten geltenden Norm, positive Gedanken und Erfahrungen zu teilen. Das Befolgen der Norm führt folglich zu einer positiveren sozialen Bewertung. Andererseits legen meine Ergebnisse nahe, dass die Teilnehmenden generell eine eher positive Meinung über das Thema des Artikels und folglich größeres Vertrauen in die Nachrichten-Endorserin hatten, wenn diese eine ähnliche Meinung äußerte. In Studie 2, einem Onlineexperiment (N=1.116), forderte ich die Teilnehmenden auf, einen nahestehenden oder entfernten Facebook-Freund zu nennen. Anschließend präsentierte ich ihnen einen fiktiven Nachrichten-Post mit einer Meinungsäußerung (positiv vs. negativ) des genannten Freundes/der genannten Freundin. Unabhängig von der sozialen Nähe der Nachrichten-Endorser:innen zeigte sich ein kleiner Einfluss ihrer Meinung auf die Valenz der Gedanken und Meinungen der Teilnehmenden über das Nachrichtenthema. Dieses Ergebnis war überraschend, insbesondere da die Teilnehmenden angaben, größeres Vertrauen in die Ansicht eines nahestehenden Freundes/einer nahestehende Freundin zu haben. Daraus schließe ich, dass sich Menschen angesichts eines ambigen Artikels sogar auf die Ansicht weniger vertrauenswürdiger Nachrichten-Endorser:innen verlassen, um sich eine sozial geteilte und damit valide Meinung über das Thema zu bilden. Zusammenfassend liefern die Ergebnisse meiner Studien erste Erkenntnisse über die Bedingungen, unter denen einzelne Nachrichten-Endorser:innen auf sozialen Netzwerkseiten die Meinungsbildung über Nachrichten beeinflussen. Die Ergebnisse sprechen nur teilweise dafür, dass das Streben nach geteilter Realität der zugrundeliegende Mechanismus dieses sozialen Einflusses ist. Meine Arbeit trägt somit einerseits zum Verständnis sozialer Einflüsse auf die Wahrnehmung von Nachrichten auf sozialen Netzwerkseiten bei. Anderseits zeigt sie Weiterentwicklungsbedarf der Shared Reality Theorie auf. Ich schlage vor, dass zukünftige Forschung insbesondere untersuchen sollte, welche Rolle soziale und affiliative Motive für soziale Einflüsse auf die Meinungsbildung über Nachrichten spielen, die auf sozialen Netzwerkseiten geteilt werden

    Impact of land use change on soil respiration and methane sink in tropical uplands, Southwestern China

    Land use conversion could modulate soil CO2 emissions and the balance between CH4 oxidation and production via changing soil physical, chemical and biological properties. Large areas of natural forests have been converted to rubber plantations in Southeast Asia, but its impact on soil CO2 and CH4 fluxes has not been sufficiently understood. This study was conducted in Xishuangbanna, Southwestern China, aiming to quantify the impact of this land use change on soil CO2 and CH4 fluxes and to clarify mechanisms responsible for the differences between natural forests and rubber plantations. Dynamics of soil respiration rates in two land uses were compared, and a mixed effect model was used in studying the interference of soil moisture on estimating temperature sensitivity (Q10) of soil respiration (Chapter 2). The land use change impact on the ability of soils to function as CH4 sink was firstly assessed with surface CH4 fluxes measured by static chambers, and then assessed with gas concentration profiles determined from soil probes. Confounded controlling factors and land use effects were disentangled, and the pathway of interactions between CH4 processes and mineral nitrogen was identified (Chapter 3). The concentration gradient method and one-dimensional diffusion-oxidation model were applied to quantify the vertical distribution of CH4 uptake in soil profiles, and to separate the relative control by gas diffusivity and by methanotrophic oxidation on CH4 uptake (Chapter 4). Distinct different temporal patterns of soil respiration were observed on sites during most of the rainy season: forest maintained a high soil respiration rate, while soil respiration in rubber plantations became suppressed (by up to 69%). Forest soils thus emitted the highest amount of CO2 with an annual cumulative flux of 8.48 ± 0.71 Mg C ha-1 yr-1, compared to 6.75 ± 0.79, 5.98 ± 0.42 and 5.09 ± 0.47 Mg C ha-1 yr-1 for 22-year-old rubber, rubber-tea intercropping, and 9-year-old rubber, respectively. Adding a quadratic soil moisture term into the regression model accounted for interference of moisture effect on the effect by soil temperature, therefore, improved temperature sensitivity assessments when high soil moisture suppressed soil respiration under rubber plantations. The static chamber method showed that soils under natural forest were stronger CH4 sinks than soils under rubber plantations, with annual CH4 fluxes of -2.41 ± 0.28 kg C ha-1 yr-1 and -1.01 ± 0.23 kg C ha-1 yr-1, respectively. Water-filled pore space was the main factor explaining the differences between natural forests and rubber plantations. Although soils under rubber plantations were more clayey than soils under natural forest, this was proved not to be the decisive factor driving higher soil moisture and lower CH4 uptake in the former soils. Concentration gradients method showed that CH4 consumption in 0-5 cm soil was significantly higher in natural forests than in rubber plantations, with a mean CH4 flux of -23.8 ± 1.0 and -14.4 ± 1.0 ug C m-2 h-1 for forest and rubber plantations, respectively. The atmospheric CH4 oxidized by top 10 cm soil accounted for 93% and 99% of total consumption for forest and rubber plantations, respectively. CH4 diffusivity at four sampled depths were significantly lower in rubber plantation than in forest. This reduced CH4 diffusivity, caused by altered soil water regime, predominately explained the weakened CH4 sink in converted rubber plantations. Estimated isotopic fractionation factor for carbon due to CH4 oxidation was 1.0292 ± 0.0015 (n=12). Modeling 13CH4 distribution in soil profiles using a diffusion-oxidation model explained the observations in the dry season, but suggested CH4 production in subsoil in the rainy season. In summary, converting natural forests into rubber plantations tended to reduce soil CO2 emissions, but this conversion substantially weakened CH4 uptake by tropical upland soils. The altered soil water regime and conditions of soil aeration under converted rubber plantations appear to have a pronounced impact on processes of gaseous carbon fluxes from soils. The clarified mechanisms in this study could improve the regional budget of greenhouse gases emissions in response to land use change and climate change.Landnutzungsänderungen können CO2 Emissionen, sowie das Gleichgewicht zwischen der Oxidation und Produktion von CH4 beeinflussen, indem sie physikalische, chemische und biologische Bodeneigenschaften verändern. Trotz der Umwandlung großer Flächen der natürlichen Wälder Südost Asiens zu Kautschukplantagen, wurde der Einfluss auf die CO2 und CH4 Flüsse des Bodens bisher unzureichend erforscht. Dazu wurde die vorliegende Studie in Xishuangbanna im Südwesten Chinas durchgeführt. Die Ziele der Arbeit waren, den Einfluss der Landnutzungsänderungen auf die CO2 und CH4 Flüsse des Bodens zu quantifizieren, sowie Unterschiede der relevanten Mechanismen zwischen naturnahen Wäldern und Kautschukplantagen zu erklären. Der Verlauf der Bodenatmungsraten der beiden Landnutzungen wurde verglichen. Durch ein Modell mit gemischten linearen Effekten wurde die Interferenz der Bodenfeuchtigkeit bei der Schätzung der Temperaturempfindlichkeit (Q10) der Bodenatmung analysiert (Kapitel 2). Der Einfluss der Landnutzungsänderungen auf die Fähigkeit des Bodens, CH4 aufzunehmen, wurde anhand von CH4 Flüssen der Bodenoberfläche, gemessen durch die statische Kammermethode, bewertet. Zudem wurden Gaskonzentrationsprofile durch Bodensensoren gemessen. Die Landnutzungseffekte wurden um die Störfaktoren bereinigt um die Wechselwirkungen zwischen CH4-Prozessen und mineralischem Stickstoff zu identifizieren (Kapitel 3). Danach wurde die Methode des Konzentrationsgradienten sowie ein eindimensionales Diffusions-Oxidationsmodel genutzt, um die vertikale Verteilung der CH4 Aufnahme in Bodenprofilen zu quantifizieren, und die relativen Einflüsse des Gasdiffusionsvermögens und der methanotrophen Oxidation bei der CH4-Aufnahme abzuschätzen (Kapitel 4). Über weite Teile der Regenzeit hinweg zeigten die zwei Standorte deutlich verschiedene zeitliche Bodenatmungsverläufe. Die Respirationsrate des Waldstandortes war konstant hoch, in den Kautschukplantagen hingegen war sie in der Regenzeit (um bis zu 69 %) reduziert. Waldböden emittierten somit das meiste CO2, mit einem jährlichen kumulativen Fluss von 8.48 ± 0.71 Mg C ha-1 a-1, verglichen mit 6.75 ± 0.79, 5.98 ± 0.42 and 5.09 ± 0.47 Mg C ha-1 a-1 beim ausgewachsenen Kautschuk, der Mischkultur aus Kautschuk und Tee sowie den jungen Kautschukplantagen, respektive. Ein zusätzlicher quadratischer Bodenfeuchtigkeitsterm im verwendeten Modell erklärte den Einfluss der Bodenfeuchte auf den Temperatureffekt und verbesserte dadurch die Analyse der Temperatursensitivität, da hohe Bodenfeuchte die Bodenrespiration in Kautschukplantagen limitierte. Laut den Ergebnissen der statischen Kammermethode, waren die Böden unter naturnahem Wald, mit CH4 Flüssen von -2.41 ± 0.28 kg C ha-1 a-1, stärkere CH4 Senken als Böden unter Kautschukplantagen, welche Flüsse von -1.01 ± 0.23 kg C ha-1 a-1 besaßen. Das wassergesättigte Porenvolumen der Böden war der Hauptfaktor welcher die Unterschiede zwischen naturnahem Wald und Kautschukplantagen erklären konnte. Obwohl die Böden unter den Kautschukplantagen höhere Tongehalte als die Waldböden vorwiesen, war dies nicht der entscheidende Faktor für höhere Bodenwassergehalte und niedrigere CH4 Aufnahmen. Die Konzentrationsgradientenmethode zeigte, dass die CH4 Aufnahme in 05 cm Bodentiefe im naturnahem Wäldern signifikant höher war als in den Kautschukplantagen. Die mittleren CH4 Flüsse lagen bei -23.8 ± 1.0 im Wald und bei 14.4 ± 1.0 ug C m-2 h-1 in den Kautschukplantagen. Das atmosphärische CH4, welches in den obersten 10 cm der Böden oxidiert wurde, machte 93% bzw. 99% der Gesamtaufnahme des Waldes und der Kautschukplantagen aus. Die Diffusität des CH4 in den Kautschukplantagen war in allen vier beprobten Bodentiefen signifikant niedriger als im Wald. Diese verminderte, durch Veränderungen des Wasserhaushalt verursachte, Diffusionsfähigkeit, war der wichtigste Erklärungsfaktor für die geringeren CH4 Aufnahmen der konvertierten Kautschukplantagen. Der geschätzte Isotopenfraktionierungsfaktor von Kohlenstoff bei der CH4 Oxidation war 1.0292 ± 0.0015 (n=12). Die Modellierung der Verteilung von 13CH4 im Bodenprofil unter Verwendung eines Diffusions-Oxidationsmodells konnte zwar die Beobachtungen in der Trockenzeit erklären, sagte für die Regenzeit jedoch eine Produktion von CH4 im Unterboden vorher. In der Tendenz verringerte eine Konvertierung von naturnahem Wald zu Kautschukplantagen die CO2 Emissionen, verringerte gleichzeitig jedoch substanziell die CH4 Aufnahme der tropischen Böden. Die resultierenden Wasser- und Lufthaushaltveränderungen der Böden von konvertierten Kautschukplantagen schienen die Prozesse der gasförmigen Kohlenstoffflüsse von Böden deutlich zu beeinflussen. Das bessere Verständnis der Mechanismen durch diese Studie könnte daher zur Verbesserung der regionalen Budgetierung von Treibhausgasemissionen infolge von Landnutzungsänderungen und des Klimawandels beitragen

    Function and dynamic of a common insertion complex of the Sec translocase and YidC insertase in the bacterial membrane

    YidC/Oxa1/Alb3-Insertasen und das Sec-Translokon sind über alle drei Reiche des Lebens konserviert und stellen den wichtigsten Weg für integrale Proteine in Zellmembranen und Membranen von eukaryontischen Organellen dar. Die Insertion von Membranproteinen in die innere Membran von Gram-negativen Bakterien erfolgt hauptsächlich über die SecYEG-Translokase und die YidC-Insertase, die unabhängig voneinander oder in Kooperation miteinander agieren können. Für die kooperative Insertion wird ein enger Kontakt zwischen SecY und YidC vorausgesetzt. Aufgrund von früheren Interaktions-Studien und einer kürzlich gelösten Struktur des sogenannten Holotranslokons mit geringer Auflösung (14 Å) wird von einem Kontakt zwischen dem lateral gate von SecY und der hydrophoben Substratrutsche von YidC ausgegangen. Welche konkreten Domänen von YidC und SecY dabei direkt miteinander interagieren, war bisher nicht bekannt. Ziel dieser Arbeit war es, den Kontakt zwischen SecY und YidC detaillierter zu beschreiben. In vitro wurde über FRET-Messungen mit Fluoreszenzfarbstoff-markiertem SecY und YidC eine hohe Affinität für die Interaktion der beiden Proteine sowohl in Detergens als auch in DOPC-Proteoliposomen ermittelt. Für die Stöchiometrie der SecY/YidC-Interaktion wurde der Faktor eins bestimmt. Um die genauen Kontakte zwischen SecY und YidC zu identifizieren, wurden in vivo Disulfid-Cross-Linking-Experimente durchgeführt. Es konnten direkte Kontakte zwischen der TM3 und TM8 des SecY lateral gates und der TM3 beziehungsweise TM5 der hydrophoben Substratrutsche von YidC gezeigt werden. Zudem wurde eine YidC-Mutante mit fünf Serin-Substitutionen, die den Wachstumsdefekt eines YidC-Depletionsstamms nicht wiederherstellen konnte, charakterisiert. Obwohl die Serin-Positionen alle in der Mitte und der periplasmatischen Hälfte der hydrophoben Rutsche von YidC liegen und vier der Positionen mit Substrat-Kontaktstellen übereinstimmen, konnte keine Inhibierung der Insertion der YidC-abhängigen Substrate M13 procoat und Pf3 coat durch die 5S Mutante im Vergleich zum Wildtyp beobachtet werden. Für die YidC-only Insertion scheint ein Minimum an Hydrophobizität nötig zu sein, das bei dieser Mutante nicht unterschritten wird. In vitro FRET-Experimente zeigten eine gestörte Interaktion zwischen SecY und dieser YidC 5S Mutante und bestätigten damit erneut eine Beteiligung der hydrophoben Rutsche am SecY/YidC-Kontakt. Anhand der ermittelten Cross-Link-Kontakte und der Ergebnisse der FRET-Messungen konnte ein mögliches Kontaktmodell erstellt werden, in dem das SecY lateral gate der hydrophoben Substrat-Rutsche und der sich zwischen TM3 und TM5 öffnenden hydrophilen Tasche von YidC gegenüber steht, wodurch sich eine gemeinsame SecY/YidC-Pore bildet. Die vorliegende Arbeit liefert damit weitere Beweise, dass das lateral gate der Sec-Translokase direkt mit der hydrophoben Substratrutsche von YidC interagiert. In einem weiteren Projekt wurde ein SecY-YidC-Fusionsprotein zur Sicherstellung eines engen Kontakts, der korrekten Orientierung der Proteine zueinander und der richtigen Stöchiometrie in rekonstituierten Proteoliposomen kloniert. Für eine gemeinsame Studie mit der ETH Zürich wurden Proteoliposomen, die das Fusionsprotein, SecYEG, YidC oder SecYEG zusammen mit YidC enthielten, von mir in Hohenheim hergestellt. Die schrittweise Insertion des Sec/YidC-abhängigen Substrats LacY in diese Proteoliposomen wurde von einer Arbeitsgruppe der ETH Zürich mithilfe der AFM-basierten Einzelmolekül-Kraftspektroskopie beobachtet. Die LacY-Insertion wird im Falle des Fusionsproteins und SecYEG in Kombination mit YidC von der Sec-Translokase dominiert, während YidC vermutlich nur eine unterstützende Rolle bei der Proteinfaltung einnimmt.YidC/Oxa1/Alb3-insertases and the Sec-translocase are conserved across all three kingdoms of life and constitute the most important pathway for integral proteins into cell membranes and membranes of eukaryotic organelles. The insertion of membrane proteins into the inner membrane of Gram-negative bacteria occurs mainly via the SecYEG-translocase and the YidC-insertase acting independently or in cooperation. For the cooperative insertion a close contact between SecY and YidC is assumed. Previous interaction-studies and a recently solved low-resolution structure of the so-called holo-translocon (14 Å) indicate a contact between the lateral gate of SecY and the hydrophobic substrate slide of YidC. Which specific domains of YidC and SecY thereby interact directly with each other was unknown so far. The aim of this study was to describe the contact between SecY and YidC in more detail. A high affinity for the interaction of the two proteins in detergent and in DOPC-proteoliposomes was determined via FRET measurements with fluorescently labeled SecY and YidC. For the stoichiometric ratio of the SecY/YidC-interaction a factor of one was calculated. To identify the specific contacts between SecY and YidC in vivo disulphide cross-linking experiments were performed. Direct interactions between the transmembrane domain (TM) 3 and TM8 of the SecY lateral gate and TM3 and TM5 of the hydrophobic slide of YidC were found, respectively. Furthermore, a YidC mutant with five serine substitutions, which was unable to rescue a YidC depletion strain, was investigated. Even though the serine positions are located in the middle and the periplasmic half of the hydrophobic slide of YidC and four of the positions are identical with substrate contact sites, no inhibition of insertion for the YidC-dependent substrates M13 procoat and Pf3 coat by the 5S mutant compared to the wildtype YidC was observed. For the YidC-only pathway a minimum of hydrophobicity seems to be required sufficient to allow the insertion of these substrates. In vitro FRET measurements showed an impaired interaction between SecY and the YidC 5S mutant and confirmed once again an involvement of the hydrophobic slide in the SecY/YidC-contact. Based on the cross-linking contacts and the results of the FRET measurements a possible model of the SecY/YidC-contact was established, which shows the SecY lateral gate vis-à-vis of the hydrophobic slide and the hydrophilic groove between TM3 and TM5 of YidC generating a combined SecY/YidC-cavity. Taken together, the present study provides further evidence that the lateral gate of the Sec-translocase directly interacts with the hydrophobic slide of YidC. In a further project, a SecY-YidC fusion protein was cloned to ensure the two proteins are in close proximity, the correct orientation and proper stoichiometry after reconstitution into proteoliposomes. For a collaboration with the ETH Zürich, proteoliposomes hosting the fusion protein, SecYEG, YidC or SecYEG and YidC together were prepared by myself in Hohenheim. The stepwise insertion of the Sec/YidC-dependent substrate LacY into these proteoliposomes was observed by a collaborating group of the ETH Zürich using AFMbased single-molecule force spectroscopy. The insertion of LacY was observed for the different cases but for the fusion protein and SecYEG combined with YidC the insertion process is dominated by the Sec-translocase, whereas YidC probably only has a supporting function in the folding of the protein

    Towards a better understanding of land surface exchange processes over agricultural crop stands

    Weather and climate models are useful tools for projecting the influence of global climate change on the regional scale. These models are critically dependent on an accurate representation of soil-plant-atmosphere interactions, which are simulated by Land Surface Models (LSMs). The present PhD thesis was designed to improve the representation of land surface exchange processes of croplands in the Noah-MP land surface model. This thesis aims: a) to elucidate the nature of the energy imbalance over a winter wheat stand and to identify the appropriate post-closure method for the study region Kraichgau, southwest Germany; b) to improve the representation of the green vegetation fraction (GVF) dynamics of croplands in the Noah-MP for a more accurate computation of surface energy and water fluxes; and c) to determine the effect of aggregating different crop types with various shares into a single generic cropland class on the simulation of water and energy exchange between land surface and atmosphere.Wetter- und Klimamodelle sind nützliche Werkzeuge, um den Einfluss des globalen Klimawandels auf die regionale Ebene zu projizieren. Diese Modelle sind entscheidend von einer genauen Darstellung der Wechselwirkungen zwischen Boden, Pflanzen und Atmosphäre abhängig, welche durch Land Surface Models (LSMs) simuliert werden. Die Dissertation wurde erstellt, um die Darstellung von Landoberflächenaustauschprozessen von Ackerland im Noah-MP-Landoberflächenmodell zu verbessern.     Economic evaluation of regional competitive potentials of various agricultural biomasses in the context of bioeconomy with the special focus on Baden-Wuerttemberg

    Die Endlichkeit der fossilen Ressourcen sowie der Klimawandel stellen die globale Gesellschaft vor große Herausforderungen und erfordern eine umfassende Transformation des aktuellen Wirtschaftssystems. In dieser Transformation, die auch als Bioökonomie bezeichnet wird, nimmt der Übergang von einer fossil-basierten zu einer bio-basierten Rohstoffversorgung eine wichtige Rolle ein. Die landwirtschaftliche Produktion ist in diesem Zusammenhang ein wichtiger Rohstofflieferant, die bereits durch eine starke Nutzungskonkurrenz um die knappe Fläche gekennzeichnet ist, die weitere Produktionssteigerungen für die Bioökonomie erschweren. Ökonomische Modelle können in diesem Kontext als wertvolle Methode zu Analyse der Bioökonomie agieren, mit denen einerseits die Wechselwirkungen von verschiedenen Nutzungspfaden abgebildet werden und die andererseits mit einer entsprechenden Szenarienentwicklung auch die Unsicherheit adressieren können. Das Ziel der vorliegenden Arbeit ist die Potentialabschätzung verschiedener landwirtschaftlicher Biomassen für die Bioökonomie und die Analyse der damit verbundenen Auswirkungen auf die landwirtschaftlichen Produktionsstrukturen in Baden-Württemberg. In Kapitel 2 wird das Potential von Grünland als Biogassubstrat analysiert, das in Zukunft eine bedeutende Rolle für die Bioökonomie spielen kann. Aufgrund des aufwändigeren Ernteverfahrens und teilweise ungünstiger Produktionsbedingungen hat Grünland höhere Produktionskosten gegenüber ackerbürtiger Biogassubstrate. Die Berücksichtigung der iLUC Faktoren mit hohen Preisen für THG-Emissionen kann die relative Vorzüglichkeit des Grünlandes soweit verbessern, dass es mit der Produktion von Ackerland konkurrenzfähig ist, auch wenn Silomais aus vielfacher Hinsicht oft das vorzüglichere Biogassubstrat ist, wie das Kapitel 3 anhand einer Standortmodellierung für Biogasanlagen in Baden-Württemberg zeigt. In den Kapiteln 4 und 5 wird das Potential von Stroh zur energetischen und stofflichen Verwertung analysiert . Diese Untersuchungen basieren auf der Verknüpfung von EFEM mit dem techno-ökonomischen Standortoptimierungsmodell BIOLOCATE. Die Ergebnisse zeigen anschaulich die Wechselwirkung zwischen den Skaleneffekten und den steigenden Rohstoffbereitstellungskosten. So sinken mit zunehmender Anlagengröße einerseits die durchschnittlichen Investitionskosten, andererseits steigen die Rohstoffkosten, weil die Transportentfernungen zunehmen und eine steigende Biomassenachfrage höhere Marktpreise zur Folge hat. Die Ergebnisse zeigen auch, dass Stroh grundsätzlich mit der Bereitstellung von regionaler Bioenergie und als Rohstoff für stoffliche Wertschöpfungsketten einen Beitrag zur Bioökonomie leisten kann. Allerdings hat selbst die Nutzung von Nebenprodukten Auswirkungen auf die Anbaustrukturen. In Kapitel 6 werden die Auswirkungen von gesamtwirtschaftlichen Expansionspfaden der Bioökonomie auf die landwirtschaftlichen Produktionsstrukturen in Baden-Württemberg untersucht. Hierfür werden die Ergebnisse einer iterativen Modellkopplung zwischen dem Agrarsektormodell ESIM und dem Energiesektormodell TIMES-PanEU von vier Bioökonomieszenarien mithilfe von EFEM von nationaler Ebene auf regionale und einzelbetriebliche Ebene herunterskaliert. Die Ergebnisse zeigen, dass Betriebe mit vornehmlich extensiven Produktionsverfahren wie der Mutterkuhhaltung durch ungünstige Produktionsbedingungen keinen Nutzen aus der Expansion der Bioökonomie ziehen können werden, während insbesondere große Ackerbaubetriebe in fruchtbaren Regionen überdurchschnittlich profitieren würden. Grundsätzlich legen die Ergebnisse Grenzen bei der Mobilisierung von zusätzlichem Biomassepotential offen. Dies begründet sich in der hohen Anbauintensität der landwirtschaftlichen Produktion in Deutschland, aufgrund dessen die Ausweitung einer Produktion aufgrund der Nutzungskonkurrenz andere Produktionen einschränkt. Für Grünland zeigen die Ergebnisse, dass der Rückgang der grünlandbasierten Rinderhaltung und nachteilige ökonomische Rahmenbedingungen zu einer signifikanten Menge an ungenutztem Grünland führen können. Grünland stellt sich so als vielversprechende Ressource zur Biomasseproduktion für die Bioökonomie dar, da es neben der Rohstoffbereitstellung auch wichtige Ökosystemdienstleitungen (z. B. Artenvielfalt) liefern kann. Hierfür müssten allerdings politische Rahmenbedingungen etabliert werden, die die ökologischen Leistungen entsprechend fördern. Abschließend wird in Kapitel 7 weiterer Forschungsbedarf aufgezeigt, der die Weiterentwicklung des methodischen Ansatzes beinhaltet. Diese umfassen einerseits eine Erweiterung um gesamtwirtschaftliche Modelle, um Wechselwirkungen mit der stofflichen Nutzung detaillierter abzubilden. Andererseits ist Integration von ökologischen Aspekten für die ganzheitliche Analyse im Rahmen der Bioökonomie notwendig.The finite nature of fossil resources and climate change pose major challenges to the global society and require a comprehensive transformation of the current economic system. One important aspect of this transformation, also known as bioeconomy, is the transition from a fossil-based to a bio-based supply of raw materials. In this context, agricultural production represents an important supplier of raw materials, which in Germany is already characterized by a strong competition for the scarce land. The scarce land is a major challenge of the expansion of the use of agricultural biomass for the bioeconomy. Accordingly, the derivation of the potential of agricultural biomass for bioeconomy requires consideration of the tradeoffs between various utilization paths. In this context, economic models can be valuable methods, which on one hand are able to depict the trade-offs of different value chains and can, on the other hand, incorporate the uncertainty by developing suitable scenarios. The aim of this thesis is the evaluation of the potential of different agricultural biomasses for the bioeconomy and to analyze the associated effects on agricultural production structures in Baden-Wuerttemberg. In chapter 2 the potential of grassland as a biogas substrate is evaluated, which might be important for the bioeconomy in the future. Due to the more complex harvesting process and partly unfavorable production conditions, grassland has higher production costs compared to arable biogas substrates. The consideration of iLUC Factors with high prices for GHG emissions could improve the competitiveness of grassland to such an extent that it is competitive with the production of biogas substrates on arable land. However, silage maize is often the more favorable biogas substrate in many respects, as chapter 3 shows by means of a site modeling for biogas plants in Baden-Wuerttemberg. In chapter 4 and 5 the potential of straw for energetic and material use is analyzed. These investigations are based on the combination of EFEM with the techno-economic location optimization model BIOLOCATE. The results clearly show the interaction between the economies of scale and the rising raw material supply costs. On the one hand, the average investment costs decrease with increasing plant size, but on the other hand the raw material costs increase, because the transport distances increase and an increasing demand for biomass results also in higher market prices. Additionally, the results show that straw can make a fundamental contribution to the bioeconomy by providing regional bioenergy and as feedstock for material value chains. However, even the use of by-products can have effects on cultivation structures and thus, reduce the production of agricultural biogas substrates, among other things. In Chapter 6 the effects of macroeconomic expansion paths of the bioeconomy on agricultural production structures in Baden-Wuerttemberg are investigated. For this purpose, the results of an iterative model coupling between the agricultural sector model ESIM and the energy sector model TIMES-PanEU of four bioeconomic scenarios are scaled down from national level to regional and farm level using EFEM. The results show different impacts on farm types and thus illustrate the advantages of a differentiated analysis of the expansion of the bioeconomy. Therefore, farms with mainly extensive production methods such as suckler cow husbandry do not profit from the expansion of the bioeconomy due to unfavorable production conditions, while especially large arable farms in fertile regions would benefit disproportional more than the average. Basically, the results reveal limits to the mobilization of additional biomass potential. The reason for this is the already high cultivation intensity of agricultural production in Germany, in which the expansion of one production restricts production of another due to competition for the limited agricultural land. For grassland, the results show that the decline in grassland-based cattle farming and unfavorable economic conditions can lead to a significant increase of unused grassland. Grassland thus presents itself as a promising resource for biomass production for the bioeconomy, as it can provide important ecosystem services (e.g. biodiversity) in addition to the provision of raw materials. However, a political framework has to be established that promotes ecological services accordingly. Finally, in chapter 7 additional research needs are identified, which include further development of the methodological approach. These comprise an extension of the analysis by macroeconomic models to integrate interactions with the material use in a more detailed way. Furthermore, an integration of ecological parameters is necessary for a holistic analysis in the context of bioeconomy

    The importance of aquaporin interacting proteins for cell death regulation in plants and animals

    Bei der Zelltodinduktion spielt Wasserstoffperoxid sowohl in Pflanzen, als auch in Tieren eine wichtige Rolle. Das Signalmolekül Wasserstoffperoxid muss dabei über verschiedene Membranen an seinen Zielort transportiert werden. Dieser Transport geschieht in Pflanzen und Tieren durch Aquaporine. Diese integralen Membranproteine ermöglichen den Transport von Wasserstoffperoxid über die Membranen zwischen Zellkompartimenten. Die pflanzlichen Aquaporine werden durch die Proteine Aquaporin Interactor 1 und 2 (AQI1 und AQI2) reguliert. AQI2 ist dabei ein pflanzliches Homolog von AQI1. Beide Proteine sind in der Lage an das Aquaporin zu binden und den Kanal zu verschließen. Der Einstrom von Wasser und Wasserstoffperoxid kann dadurch verhindert werden. Aquaporin Interactor 1 bindet dabei bevorzugt an das im Tonoplasten lokalisierte Aquaporin TIP1.1, während Aquaporin Interactor 2 stärker an das in der Plasmamembran lokalisierte Aquaporin PIP2.2 bindet. Aquaporin Interactor 1 ist innerhalb der Vakuole oder an der Tonoplastenmembran lokalisiert. Im Gegensatz dazu wird für Aquaporin Interactor 2 eine Lokalisation im Apoplasten angenommen. So kann AQI1 die Aquaporine der Tonoplastenmembran und AQI2 die Aquaporine der Plasmamembran regulieren. Bekannt ist die humane Aminoacylase 1 für ihre Fähigkeit zur Hydrolyse von N-acetylierten Aminosäuren. Dabei hat das Zink-bindende Metalloenzym ein weites Substratspektrum. Das bevorzugte Substrat ist N-acetyl-Methionin. Auch das pflanzliche Homolog AQI1 ist zur Hydrolyse dieses Substrats fähig und für die Funktion ebenfalls auf Metallionen angewiesen. Aquaporin Interactor 2 besitzt keine solche Aminoacylase Aktivität. aqi1 knock-out Mutanten in Arabidopsis thaliana und Nicotiana tabacum zeigen eindeutig, dass die Hydrolyse von N-acetyl-Methionin ausschließlich von AQI1 bewerkstelligt werden kann. Diese Aminoacylase-Aktivität wird für die Bindung an die Aquaporine aber nicht benötigt. Die Aminoacylase-Aktivität und die Fähigkeit zur Bindung an die Aquaporine sind zwei unabhängige Funktionen von Aquaporin Interactor 1. Nach bisherigem Kenntnisstand muss davon ausgegangen werden, dass AQI2 ausschließlich als Aquaporin-regulierendes Protein fungiert. Nach Pathogenbefall ist im betroffenen Pflanzengewebe eine verstärkte Aminoacylase-Aktivität zu detektieren. Sowohl nach Agrobakterieninfiltration, als auch nach Infektion mit Tabakmosaikvirus, ist diese AQI1 Induktion zu beobachten. Dies demonstriert eine Rolle des Proteins bei der Pathogenabwehr. Ein weiteres Aquaporin-interagierendes Protein ist BHRF1, ein anti-apoptotisches Protein aus dem Epstein-Barr-Virus. Bisher konnte diese Interaktion nur für pflanzliche Aquaporine gezeigt werden. BHRF1-transgene N. tabacum Pflanzen zeigen spontan auftretende Zelltodereignisse. Diese werden vermutlich durch die Interaktion mit den Aquaporinen und mehreren Proteinen des G-Protein Signalwegs hervorgerufen. Durch die Bindung von BHRF1 an die Aquaporine kann es die endogenen Interaktionspartner AQI1 und AQI2 verdrängen. Eine korrekte Regulation der Aquaporine durch AQI1 und AQI2 ist somit nicht mehr gewährleistet. Zudem bindet BHRF1 an das pflanzliche AtRGS1 (regulator of G-protein signalling) Protein, einem Glukosesensor. AtRGS1 ist eine Kombination aus G-Protein gekoppeltem Rezeptor und RGS Protein. Die RGS Domäne bewirkt die Hydrolyse des an die Gα-Untereinheit gebunden GTPs. BHRF1 bindet auch an humane RGS Proteine. BHRF1 hat somit auch im Menschen einen möglichen Einfluss auf den G-Protein Signalweg. Eine genaue Regulation der Aquaporine für die Zelltodregulation ist von großer Wichtigkeit. Eine Fehlregulation, wie sie durch das virale BHRF1 zustande kommt, führt zu Zelltodereignissen. BHRF1 tritt dabei vermutlich in Konkurrenz mit den endogenen Interaktionspartnern der Aquaporine und den Proteinen des G-Protein Signalwegs und verursacht somit eine Fehlregulation verschiedener Signalwege.Hydrogen peroxide plays a crucial role as a signalling molecule in the induction of cell death in plants and animals. To mediate signalling and induce apoptosis in a cell, hydrogen peroxide molecules need to be transported across different membranes to their target site. In plants and animals, integral membrane proteins called aquaporins, facilitate the transport of hydrogen peroxide between cell compartments by channelling the signalling molecule across membranes. Plant aquaporins are regulated by proteins called Aquaporin Interactor 1 and 2 (AQI1 and AQI2). AQI2 is a plant homolog of AQI1. Both proteins function as inhibitors of aquaporins by binding to the channels resulting in prevention of water and hydrogen peroxide influx. Aquaporin Interactor 1 binds preferentially to the aquaporin tonoplast intrinsic protein TIP1.1, while Aquaporin Interactor 2 exhibits a binding preference to the aquaporin plasma membrane intrinsic protein PIP2.2. Aquaporin Interactor 1 is located in the vacuole or associated to the tonoplast membrane. In contrast, results obtained for Aquaporin Interactor 2 suggest that it is located in the apoplast. This is compatible with the hypothesis that tonoplast aquaporins can be regulated by AQI1, whereas plasma membrane aquaporins on the other hand are regulated by AQI2. The enzyme Aminoacylase 1 is known to hydrolyse N-acetylated amino acids. It is a zinc-binding metalloenzyme with a wide range of substrates. However, its preferred substrate is N-acetyl-Methionine. N-acetyl-Methionine can also be hydrolysed by the plant homolog AQI1. The plant enzyme also needs metal ions as co-factors. Of note, no aminoacylase activity was found for AQI2. Experiments using aqi1 knock-out mutants of Arabidopsis thaliana and Nicotiana tabacum clearly show, that hydrolysis of N-acetyl-Methionine can only be accomplished by AQI1. However, the aminoacylase activity of AQI1 is not needed for the ability to bind to aquaporins. The data show that the aminoacylase activity and the ability to bind aquaporins are two separate functions of the protein Aquaporin Interactor 1. Based on current knowledge, it must be assumed, that AQI2 acts only as an aquaporin-regulating protein. After pathogen attack an increased aminoacylase activity could be detected in the affected plant tissue. This AQI1 induction can be observed both after agrobacteria infiltration and after infection with the tobacco mosaic virus. This suggests a role for AQI1 in pathogen defence. Another aquaporin interacting protein is BHRF1, an anti-apoptotic protein originating from the Epstein-Barr virus. To date, an interaction between BHRF1 and aquaporins could only be detected with plant aquaporins. Transgenic BHRF1 N. tabacum plants show spontaneously occurring cell death events apparent by necrotic plant tissue. These necrotic areas are caused by BHRF1 interacting with plant aquaporins and several proteins of the G-protein signalling pathway inducing cell death. By binding to the aquaporins, BHRF1 is able to replace the endogenous aquaporin interaction partners AQI1 and AQI2. Thus, a precise aquaporin regulation by endogenous AQI1 and AQI2 is no longer guaranteed. Moreover, results show that BHRF1 can bind the Arabidopsis glucose sensor AtRGS1 (regulator of G-protein signalling). AtRGS1 is a combination of a G-protein coupled receptor and a RGS protein. The RGS domain causes the hydrolysis of GTP bound to the Gα subunit. Further experiments showed, an interaction of BHRF1 with human RGS proteins. Therefore, BHRF1 could also have a possible effect on G-protein signalling in humans. The results of this study demonstrate the importance of a precise regulation of aquaporins in cell death regulation. Deregulation caused by viral BHRF1, leads to cell death events. BHRF1 presumably competes with the endogenous interaction partners of aquaporins and of the G-protein-signalling pathway, ultimately resulting in the deregulation of various signalling pathways
