    Photo-control of Small G Protein RAS GTPase Using Photochromic molecule

    創価大学博士(工学)The small G protein Ras is a central regulator of cellular signal transduction processes, functioning as a molecular switch. Switch mechanisms utilizing conformational changes in nucleotide-binding motifs have been well studied at the molecular level. Previously, we have demonstrated that HRas GTPase activity is photoreversible regulated by modification Ras functional domain with photochromic compound. However, the efficiency of photoregulation was not sufficient to utilize as a photo-switching. In addition, Direct chemical modification to H-Ras carries risk of inactivating Ras function and chemical modification of H-Ras is hard to apply in vivo experiment. In this study, we tried to photoregulate Ras GTPase activity using Ras-PZ, which Ras fused with photochromic protein photozipper (PZ). We designed H-Ras fusion Photozipper (Ras-PZ) and the recombinant proteins were prepared by E.coli expression system. Ras-PZ showed photo-isomerization accompanied by blue light irradiation and in the dark, and Ras-PZ exhibited dimerization by light irradiation. Moreover, GTPase activity in the presence of regulators GEF and GAP was changed accompanied by isomerization. GTPase activity of Ras-PZ in the dark was approximately 50% of that under blue light irradiation. Additionally, GTPase activity of Ras-PZ was higher than that of HRas. It was clarified that the enhancement of this GTPase activity is caused by the allosteric site of Sos.     Fostering Inclusiveness in Japanese University Classrooms: The Transformative Power of the Hip Hop Cypher

    This research article delves into the exploration of the Hip Hop cypher as a novel pedagogical approach to foster inclusiveness in Japanese university classrooms. Amidst the backdrop of a conservative educational system, the study investigates the implementation and impact of the cypher, anchored in two research questions. An action research approach, including reflective journals, a Personal Inclusiveness Questionnaire (PIQ), and in-depth interviews, was employed to glean insights into the students’ experiences and the transformation of classroom dynamics. The findings reveal the cypher’s potential in facilitating open dialogue, authentic expression, and mutual respect, creating dynamic spaces for engaged learning and interaction. The teacher’s role is highlighted as central in nurturing the cypher’s ethos and adaptability. The triangulation of data reinforces the reliability of the results, highlighting the possibilities of the cypher in global education. This is especially relevant in environments aiming to incorporate inclusivity, student engagement, and comprehensive learning within conventional educational frameworks.departmental bulletin pape
