    Photo-control of Small G Protein RAS GTPase Using Photochromic molecule

    創価大学博士(工学)The small G protein Ras is a central regulator of cellular signal transduction processes, functioning as a molecular switch. Switch mechanisms utilizing conformational changes in nucleotide-binding motifs have been well studied at the molecular level. Previously, we have demonstrated that HRas GTPase activity is photoreversible regulated by modification Ras functional domain with photochromic compound. However, the efficiency of photoregulation was not sufficient to utilize as a photo-switching. In addition, Direct chemical modification to H-Ras carries risk of inactivating Ras function and chemical modification of H-Ras is hard to apply in vivo experiment. In this study, we tried to photoregulate Ras GTPase activity using Ras-PZ, which Ras fused with photochromic protein photozipper (PZ). We designed H-Ras fusion Photozipper (Ras-PZ) and the recombinant proteins were prepared by E.coli expression system. Ras-PZ showed photo-isomerization accompanied by blue light irradiation and in the dark, and Ras-PZ exhibited dimerization by light irradiation. Moreover, GTPase activity in the presence of regulators GEF and GAP was changed accompanied by isomerization. GTPase activity of Ras-PZ in the dark was approximately 50% of that under blue light irradiation. Additionally, GTPase activity of Ras-PZ was higher than that of HRas. It was clarified that the enhancement of this GTPase activity is caused by the allosteric site of Sos.     Brazil’s Climate Change Policy Evolution Between COP 21 (2015) and COP 27 (2022)

    The research assesses Brazil’s climate action from a policy perspective. The Paris Agreement Nationally Determined Contributions (NDC) are used to evaluate the evolution of the Brazilian ambition to mitigate the effects of climate change and reach the commitments made to the United Nations Framework Convention on Climate Change. This article uses data from Brazil’s greenhouse gas emissions and deforestation rates, to determine if the current policies are adequate to implement measures aligned with the targets, projects, instruments, and funding for the achievement of the Paris Agreement goals. It is fair to say that Brazil needs more ambitious commitments and more importantly requires persistent and tenacious policies and political will to transform commitments made in reports and conferences into actual benefits for the planet's future.departmental bulletin pape

    創価大学におけるワクチン接種率上昇のための要因分析

    ワクチン接種率上昇の要因を調べるために、期待利得の分析、アナウンス効果の分析、アンケート調査による選択型コンジョイント分析という3 つの分析を行った。金銭的インセンティブという面から期待利得の分析を行った結果、学生は合理的に行動しているわけではないことがわかった。また、クーポン配布についてアナウンス効果があったことがわかった。選択型コンジョイント分析から、創価大学におけるワクチン接種率上昇のためには、クーポン配布と寄付を選べる形にするほうがよいことがわかった。また、学期中よりも長期休暇の接種にインセンティブをもたせるほうがよいことがわかった。例えば、新学期の職域接種に加えて、地元で接種した人も、接種証明があれば対象とするなどが考えられる。departmental bulletin pape

    Foreign Language Anxiety and Psychological Safety

    While foreign language anxiety (FLA) has been a hot topic in second language acquisition (SLA), relatively little is known about the process of how language learners overcome the negative effects of FLA and obtain psychological safety. To fill this gap, this current study aims to examine to what extent language learners experience a change in FLA quantitatively, and what factors cause FLA and contribute to creating psychological safety in learners’ minds. To meet these objectives, 42 Japanese English as a foreign language (EFL) learners took part in this research. This study comprises three survey instruments: a consent form, Foreign Language Classroom Anxiety Scale (FLCAS) translated into Japanese, and a semi-structured interview. A one-way repeated measures ANOVA indicated that there was a statistically significant change in FLA levels during the academic year 2022. Furthermore, a qualitative data analysis suggests that interacting with peers, instructors’ teaching approaches, and promoting resilience might play an important role in easing EFL learners’ FLA. In line with these findings, implications for EFL instructors are discussed.departmental bulletin pape
