Study on the quality of ruminal fermentations and ration digestibility: a field trial
The ruminal environment is characterized by a highly complex ecosystem where fermentations
can be controlled with the aim of: improving the health and welfare of the animal, the efficiency
of synthesis and degradation processes, and the quality of products. A parameter that allows us
to evaluate rumen efficiency is the microbial biomass produced as a result of the proliferation of
ruminal microorganisms. Its nutritional value is very high, as it provides between 50 and 80% of
the essential amino acids to cattle, necessary to improve nitrogen utilization efficiency and at the
same time control feed costs. The analysis of this parameter is useful for understanding how to
meet nutritional needs, based on the quality and quantity of microbial protein produced. The purpose of this study was to evaluate the relationship between ruminal microbial protein production and dairy cattle productivity. For this purpose, the microbial pellet was extracted from 47 samples of ruminal fluid from lactating cattle, and its quantity was studied in relation to the quantitative and qualitative milk production of the same cattle. The study was conducted in collaboration with the company veterinarian on a herd characterized by periodic milk quality anomalies. In general, it was found that the microbial biomass produced positively influences quantitative milk production, lactose percentage, and production expressed as Energy Corrected
Milk (ECM). Furthermore, the quantity of microbial biomass was found to be correlated with the
content of Olotrichi protozoa and protozoa belonging to the Entodinium genus.
The measurement of ruminal microbial synthesis is therefore indicative of cattle productivity and
the functionality of the fore-stomachs
Since the 1950s, an intense sand and gravel quarrying from the river floodplains led to the formation of pit lakes (PLs) which now dot floodplains and have changed the morphology and the drainage system of several Italian river basins. In the last decade, a renewed interest in PLs has grown as tools for rehabilitating rivers and their floodplains, in order to provide a similar ecological service once offered by pristine riverine wetlands, which were lost due to anthropogenic pressures. Despite the potential importance of these environments, systematic data on their location, morphology, and water quality remains limited. The primary aim of this study was to quantify the number, distribution, and major morphometric characteristics of PLs in the Po River basin, in order to assess their relevance and suitability for ecological restoration purposes. To achieve these aims, a synergistic approach was used including remote sensing imagery (Landsat, Sentinel-2, and PRISMA), satellite archives, and regional databases. In addition, by exploiting the synoptic and multispectral view of these satellites, we evaluated the physical and optical properties of a large sample of PLs from 1990 to 2021. Specifically, we focused on two key parameters of water quality: the dominant wavelength (λdom, i.e., the water colour) and the concentration of Suspended Particulate Matter (SPM), splitting it into its inorganic (Suspended Particulate Inorganic Matter, SPIM) and organic (Suspended Particulate Organic Matter, SPOM) fractions. The results show that more than 12,650 small lentic water bodies are present in the Po River basin, with agricultural ponds the most represented category, followed by PLs. Specifically, 1580 PLs were identified, divided into three subcategories: active (338), ceased (1188), and doubtful (54). These artificial aquatic systems, when compared with pristine riverine wetlands, are more numerous, widely distributed, and on average more extensive, although more regularly shaped. The temporal analysis of satellite imagery showed that the number and the total area of PLs have increased from 1990 to 2021. PLs currently account for 63.5 km2 surface area, an average water volume of 378x106 m3, and for a removal of 26.6 mg N m-2 d-1 via denitrification. The results also show a very strong correlation between SPM concentrations obtained in situ and those obtained from satellite images, both for data derived from Landsat (R2 = 0.85) and Sentinel-2 images (R2 = 0.82). In general, it appeared that PLs with the highest mean SPM concentrations and the highest mean λdom (i.e., poor water quality) are located along the main Po River, and more generally near rivers. Active PLs exhibit a poor water quality status, especially those of small size (<5 ha) and directly connected to a river. Seasonal comparison shows the same trend for both SPM concentration and λdom: higher values in winter gradually decreasing until spring-summer, then increasing again. In addition, it emerged that the end of quarrying activity led to a reduction in SPM concentration from a minimum of 43% to a maximum of 72%. Finally, we demonstrated that the PRISMA hyperspectral satellite is a reliable tool for estimating the inorganic and organic fraction of SPM concentration. In this context, the use of multi-source satellite imagery enabled the quantification and analysis of PLs and other water bodies in a vast area such as the Po River basin, as well as the evaluation of the temporal evolution of the physical and optical properties of the PLs. In particular, the Sentinel-2 images consistently proved to be a reliable resource for capturing episodic and recurring quarrying events and portraying the ever-changing dynamics of these aquatic systems

African swine fever: evolution of the disease in Europe
African Swine Fever (ASF) is a viral disease caused by a DNA virus from the Asfaviridae family, ASFV.
It affects pigs and wild boars, proving highly contagious and often lethal, but it does not transmit to humans. In recent years, the pathology has emerged as a significant threat to the global swine industry, with a considerable impact on the economy and food security. This thesis investigates the spread of ASF in Europe, focusing on the complex interaction between the virus, host populations, and various ecological factors that facilitate its dissemination. Through a multidisciplinary approach encompassing epidemiology, molecular biology, and socio-economic considerations, this thesis aims to provide a comprehensive understanding of the disease's evolution in the European context.
The first chapter provides an overview of the virus from a genomic and morphological perspective, serving as a starting point to clarify the antigenic diversity that allows differentiation of ASFV into 24 genotypes, identifying genotype II as the main source of infection in European territory. It includes an analysis of the pathogenic mechanisms the virus manifests in the host to induce disease, describing current knowledge regarding the host's triggered immune response and the viral mechanisms enabling ASFV to evade it. Special attention is given to the disease transmission methods, involving pig and wild boar hosts, vectors, and the virus's resistance and persistence in the environment.
The second chapter begins with an assessment of the pathogenesis of symptoms, resulting in different clinical forms induced by ASFV: asymptomatic, peracute, acute, subacute, and chronic. It is followed by an evaluation of anatomopathological findings in infected subjects and a comparison of the identified lesions with those common to other diseases, allowing a differential diagnosis. These insights provide understanding tools for the diagnosis of African Swine Fever, detectable through direct and/or indirect research methods, without overlooking the latest developments in diagnostic techniques.
The third chapter offers a comprehensive overview of the historical context, tracing the introduction and subsequent spread of ASF from Africa to Europe, following its waves, with references to the current global situation. The fourth chapter is dedicated to describing the current situation in Italy, focusing on the management of detected outbreaks and the regulatory plans governing their handling, assessing encountered challenges due to various factors, including the characteristics of the territory and fauna, as well as the socio-economic aspects of affected areas.
The fifth chapter revolves around the overall evaluation of the effectiveness of existing control and management strategies, considering both biosecurity measures and the results obtained in the search for new safe and effective vaccines. It focuses on encouraging results that could lead to the development of new candidates. Future directions are provided regarding disease management and animal welfare

Several studies demonstrated that action observation and execution typically recruit inferior parietal and ventral premotor cortex plus the caudal part of inferior frontal gyrus. These areas have been described as the main nodes of the mirror neuron system (MNS). The MNS, by transforming the sensory information of observed motor acts into their corresponding motor representations, enables the immediate understating of others' actions. A wider network that also includes lateral occipito-temporal areas, dorsal premotor and parietal areas, as well as the lateral parts of the cerebellum, has been collectively termed as action observation network (AON). Although the main nodes of AON mostly encode the goal of the observed actions, previous research demonstrated that some areas of this network also process action kinematic features, by elaborating aspects of biological motion. In order to isolate the specific contribution of kinematic information to the decoding of observed actions, previous studies employed the point-light displays (PLDs) technique, in which biological motion is presented without pictorial aspects.
The studies described in the present thesis are aimed at assessing the neural representations of kinematic features of PLDs grasping actions during observation tasks performed by healthy human volunteers. In the first study, by comparing fully visible (FV) and PLDs grasping actions, while controlling for confounding effects due to low-level visual features, motion and context, we aimed at verifying whether: a) the observation of PLDs actions could elicit activations of the AON comparable to that produced by FV stimuli; b) it was possible to disentangle, by means of machine learning algorithms, neural pattern distributions of the activated areas in the two experimental conditions (FV and PLDs). Results showed a comparable bilateral activation of the AON during the observation of both FV and PLDs. Analyses conducted on the BOLD (blood oxygenation level dependent) signal, showed no significant differences in signal intensity between PLDs and FV. These data suggest the recruitment of a similar action coding mechanism. By using multivoxel pattern analysis (MVPA), it was possible to disentangle, with significant accuracy, the neural pattern elicited by observation of FV from that evoked by observation of PLDs grasping actions in occipital, parietal and premotor areas of the AON.
Taken together, these data reveal that kinematic features conveyed by PLDs stimuli are sufficient to elicit a complete action representation and that action kinematic features can be distinguished within the AON.
While previous neuroimaging studies using PLDs have explored the neural basis of action observation focusing on the distinction between biological and non-biological motion, in particular of whole-body movements, they have not examined the modulation of activity within the AON in response to differences in kinematic parameters of biological actions. In the second study, we investigated the neural substrates activated during the observation of PLDs grasping actions performed with different directions (right, left) and velocities (fast, slow). The main aim of this study was to assess which areas of the AON are mainly involved in encoding differences in direction and velocity of the observed action. By combining univariate and multivariate approaches, we were able to reveal differences in processing specific kinematic information within AON. Univariate results showed that coding differences in stimuli direction activated mainly posterior and lateral occipital areas, as well as occipito-parietal areas. Differences in velocity activated the lateral occipito-temporal cortex and both superior parietal and intraparietal areas. Multivariate results demonstrated that it is possible to decode velocity and direction at the network level, with several areas of the AON contributing to the decoding of either direction or velocity, or both. Taken together, the results of the two types of analyses indicate that posterior parietal nodes of the AON are mainly involved in coding grasping direction and that a wider set of areas, including premotor regions, are crucial for coding grasping velocity.
This evidence could be relevant for future studies aimed at investigating the neural correlates of action imitation based on PLDs stimuli. Futhermore, these insights could be exploited to personalize upper-limb observation-based rehabilitation programs, focusing on action kinematics. Finally, machine learning-based hand action recognition models could be enhanced by training them on the most informative kinematic features of observed actions
Parasite management in fish products: the genus Dibothriocephalus
Trypanorhyncha spp., Mixosporidi, Microsporidi e Gymnorynchus gigas) e zoonosici
(Anisakidae, Opisthorchis spp., Clonorchis sinensis, Eustrongylides spp. e
Dibothriocephalus spp.).
nota come difillobotriasi o botriocefalosi intestinale. La malattia in genere si manifesta con
dolori addominali, diarrea, malessere generale, mialgia, vertigini, anemia, colecistite,
europeo. The number of outbreaks associated with the consumption of products of animal origin
steadily increased in the EU in recent years a . In Italy, as many as 107 epidemic outbreaks
linked to the consumption of fish and fishery products were reported in the year period 2018-
2022. Despite that parasites of fish species, due to their intrinsic characteristics, do not
often cause epidemic outbreaks, but most often are responsible for sporadic cases, they are
of concern for human.
Parasites of fish include non-zoonotic (Pennella spp., Trypanorhyncha spp., Myxosporidae,
Microsporidae and Gymnorynchus gigas) and zoonotic (Anisakidae, Opisthorchis spp.,
Clonorchis sinensis, Eustrongylides spp. and Dibothriocephalus spp.) species. Nonzoonotic parasites do not cause illness in humans, but depreciate infested products and
exclude them from consumption as they render the food unfit for human consumption, as
set in Regulation (EC) 178/2002, article 14, subsection 2, letter b.
Zoonotic parasites, on the other hand, cause disease in humans and are therefore regulated
by EU regulations (Regulation (EC) 853/2004, Regulation (EC) 2074/2005 and Regulation
(EC) 1276/2011) and national guidelines which, when applied correctly, prevent infestations
in the consumer.
It should also be recalled that Article 5 of Italian Law 283/1962 establishes, under letter d,
the criminal offence of the food business operator (FBO) who failing to comply with the
aforementioned Regulations, uses in the preparation of food, sells, holds for sale, gives to
his employees or in any case distributes for consumption foodstuffs that are invaded by
parasites.
The parasites belonging to the genus Dibothriocephalus are cestodes of marine and
freshwater fish that are also present in Italy (Esox lucius, Gymnocephalus cernua, Lota lota,
Perca fluviatilis) and cause the human disease known as diphyllobotriasis or intestinal
botryocephalosis. The disease usually manifests itself with abdominal pain, diarrhoea,
general malaise, myalgia, dizziness, anaemia, cholecystitis, cholangitis and, in the case of
massive infestations, intestinal obstruction.
Humans become infested through the consumption of raw or undercooked parasitised fish
meat. The patient may become aware of the infestation by noticing the presence of
7
proglottids in faeces or by undergoing copro-microscopic examinations following the
symptoms to confirm the presence of parasite eggs.
The prognosis is generally favourable, and the therapy of choice is based on the use of
praziquantel and niclosamide.
In the light of the biological characteristics of the parasite, its endemic areas, the
particularities of the disease, the constant expansion of the world fish market and the
phenomenon of “culinary tourism”, the parasites belonging to the genus Dibothriocephalus
must be considered a real threat to public health in Italy and Europe
Trajectory generation strategies for safe autonomous driving in urban scenario
In this dissertation we develop novel strategies based on nonlinear optimal control techniques for trajectory generation of autonomous vehicles. These strategies are designed to enable the development of autonomous vehicles that navigate dynamic environments while enhancing safety and passenger comfort.
In the first part of the work, we introduce a family of reduced-order car models suited for trajectory generation strategies. We derive the equations of motion for both kinematic and dynamic bicycle models, the latter of which includes tire modeling for a more realistic representation of vehicle behavior. We re-write the kinematic model in terms of longitudinal and lateral coordinates, aligning them with the way humans perceive and control vehicle motion.
In the second part, we propose an optimization-based strategy to address merging maneuvers in busy intersections. We describe vehicle dynamics in terms of longitudinal and transverse coordinates and introduce a "virtual target vehicle" constrained to move within the target lane for merging. We formulate an optimal control problem in terms of longitudinal and lateral coordinates, including the kinematic position error between the autonomous vehicle and the virtual target vehicle. We also use obstacle predictions to enforce suitable kinematic constraints for generating collision-free trajectories. We show the efficacy of the proposed strategy through a set of numerical computations and highlight the main features of the generated trajectories.
In the third part, we present a real-time maneuver generation algorithm. Given a planar road geometry with static and moving obstacles along it, we are interested in finding collision-free maneuvers that satisfied the vehicle dynamics and subject to physical and comfort limits. Based on longitudinal and transverse coordinates, we propose a novel collision avoidance constraint and formulate a suitable optimal control problem. The optimization problem is solved by using a nonlinear optimal control technique that generates (local) optimal trajectories. We demonstrate the efficacy of the proposed algorithm by providing numerical computations on a simulated scenario. Experimental results are presented to demonstrate the efficiency
of the proposed algorithm both in terms of computational effort and dynamic features captured.
In the fourth part, we address the lane change maneuver using a parametric model predictive control approach. We recognize that successful lane changes involve both the decision of when to initiate these maneuvers and the generation of collision-free trajectories. Our approach combines decision-making and planning tasks, guiding the low-level policy through upper-level policy search. Additionally, we incorporate self-supervised learning techniques to adapt to dynamic, online scenarios, ensuring the vehicle can handle unexpected changes in its environment. We provide numerical results that highlight the effectiveness of this approach in improving vehicle maneuvering in dynamic environments
