    Fosforilazione del canale del sodio cardiaco anticipa la manifestazione della miopatia dell'invecchiamento

    La disfunzione diastolica e il prolungamento della ripolarizzazione ventricolare si osservano tipicamente negli anziani, tuttavia non e’ chiaro se questi fattori sono associati alla progressiva manifestazione della cardiomiopatia dell'invecchiamento. In modelli sperimentali, l’invecchiamento porta ad un aumento della corrente tardiva del sodio (INaL) nei cardiomiociti, e pertanto abbiamo ipotizzato che questa corrente e’ coinvolta nel prolungamento della ripolarizzazione con conseguenze sul rilassamento del miocardio dell’animale anziano. A tal proposito, abbiamo studiato topi wild-type (WT) e animali geneticamente modificati con mutazioni della proteina Nav1.5, che forma i canali ionici del sodio. In particolare, abbiamo utilizzato mutazioni a guadagno di funzione (Gain of Function, GoF) o a perdita di funzione (Loss of Function, LoF) del sito fosforilativo di Nav1.5 che causano, rispettivamente, un aumento e una stabilizzazione della corrente tardiva del sodio. I risultati sono stati ottenuti sia in topi maschi che femmine. Con l’invecchiamento, i topi WT hanno sviluppato un prolungamento della ripolarizzazione (intervallo QT dell'elettrocardiogramma) ed un’alterata funzione diastolica. La protratta ripolarizzazione e la disfunzione diastolica si sono manifestate prematuramente nei topi giovani GoF, e sono progressivamente deteriorate con l’eta’. Invece, gli animali LoF hanno mantenuto una buona funzione contrattile e di rirpolarizzazione durante l’invecchiamento. A livello cellulare, le fasi di contrazione/rilassamento erano prolungate nei miociti WT invecchiati rispetto alle cellule giovani. Invece, il prolungamento della meccanica di contrazione e’ comparso prematuramente allo stadio giovanile nei miociti GoF, ed e’ progressivamente deteriorato con l’età. Al contrario, miociti LoF hanno mantenuto una buona funzione meccanica durante l’invecchiamento. Collettivamente, questi risultati dimostrano che la fosforilazione della proteina Nav1.5 e la corrente tardiva di sodio modulano il rilassamento dei miociti, costituendo il meccanismo che collega la ripolarizzazione ventricolare ritardata con la disfunzione diastolica.Diastolic dysfunction and delayed ventricular repolarization are typically observed in the elderly, but whether these defects are intimately associated in the progressive manifestation of the aging myopathy remains to be determined. In this regard, aging in experimental animals is coupled with increased late Na+ current (INaL) in cardiomyocytes, raising the possibility that INaL conditions the modality of electrical recovery and myocardial relaxation of the aged heart. For this purpose, aging male and female wild-type (WT) C57Bl/6 mice were studied together with genetically engineered mice with phosphomimetic (gain-of-function, GoF) or ablated (loss-of-function, LoF) mutations of the sodium channel Nav1.5 at Ser571 associated with, respectively, increased and stabilized INaL. At ~18 months (m) of age, WT mice developed prolonged duration of the QT interval of the electrocardiogram and impaired diastolic left ventricular (LV) filling, defects that were reversed by INaL inhibition. Prolonged repolarization and impaired LV filling occurred prematurely in adult (~5 m) GoF mutant mice, whereas these alterations were largely attenuated in aging LoF mutant animals. Ca2+ transient decay and kinetics of myocyte shortening/relengthening were delayed in aged (~24 m) WT myocytes, with respect to adult cells. In contrast, delayed Ca2+ transients and contractile dynamics occurred at adult stage in GoF myocytes and further deteriorated at old age. Conversely, myocyte mechanics were minimally affected in aging LoF cells. Collectively, these results document that Nav1.5 phosphorylation at Ser571 and the late Na+ current modulates the modality of myocyte relaxation, constituting the mechanism linking delayed ventricular repolarization and diastolic dysfunction

    Il presente progetto nasce con l’obiettivo di migliorare l’esperienza del pasto per i pazienti affetti da disfagia, una condizione che comporta difficoltà nella deglutizione e impatta significativamente sulla qualità della vita. In particolare, il progetto si concentra sulla sperimentazione per un approccio sensoriale e personalizzato, mirato a rendere il pasto non solo sicuro dal punto di vista medico, ma anche piacevole ed appagante. La ricerca è stata condotta in collaborazione con il Centro Diurno Socio-Riabilitativo di Torino, che accoglie pazienti con Sclerosi Multipla e patologie neurologiche similari, e ha visto l’utilizzo dei pasti forniti dalla Mensa CAMST. Attraverso l’introduzione di consistenze modificate, spezie, e una maggiore attenzione alla presentazione, il progetto ha mostrato un impatto positivo sulla soddisfazione dei pazienti e sulla loro disponibilità al consumo del cibo. Questo studio sperimentale ha fornito risultati incoraggianti, suggerendo la possibilità di continuare la ricerca per migliorare ulteriormente l'efficacia operativa e integrare nuove tecnologie. Il progetto ha gettato le basi per una visione inclusiva del momento del pasto, con l’obiettivo di promuovere la dignità e la qualità della vita dei pazienti affetti da disfagia, aprendo la strada a futuri sviluppi nel trattamento della condizione.This project was created with the goal of improving the mealtime experience for patients with dysphagia, a condition that involves difficulty swallowing and significantly impacts quality of life. Specifically, the project focuses on experimentation for a sensory and personalized approach aimed at making mealtime not only medically safe, but also enjoyable and satisfying. The research was conducted in collaboration with the Centro Diurno Socio-Riabilitativo di Torino, which accommodates patients with Multiple Sclerosis and similar neurological disorders, and involved the use of meals provided by the CAMST Canteen. Through the introduction of modified textures, spices, and increased attention to presentation, the project showed a positive impact on patient satisfaction and willingness to eat. This experimental study provided encouraging results, suggesting the possibility of continuing research to further improve operational effectiveness and integrate new technologies. The project laid the foundation for an inclusive view of mealtime, with the goal of promoting the dignity and quality of life of patients with dysphagia, paving the way for future developments in the treatment of the condition

    ​Therapeutic potential of cannabidiol (CBD) in canine refractory epilepsy: an analysis of clinical evidence.

    Epilepsy is one of the most common neurological conditions in veterinary medicine. It is a brain disorder characterized by a persistent predisposition to develop epileptic seizures, which are episodes of abnormal electrical brain activity. These seizures manifests with symptoms such as muscle spasms, loss of consciousness, and involuntary movements. In this context, it is important to distinguish epilepsy from other conditions that cause seizures, such as those triggered by toxins or metabolic imbalances, which don’t fit classical definition of epilepsy. Seizures can develop in several forms, depending on the brain area involved, and can be classified as focal, focal processing to generalized or generalized. Diagnosing epilepsy in dogs is complex and is primarily based on the medical history provided by the owners, as it is rare for the veterinarian to be a direct witness of a seizure. It is crucial to gather detailed information on the frequency, duration, and circumstances of the seizures, as well as on the dog’s behavior between episodes. This is followed by physical and neurological examinations to identify the underlying cause of the seizures, which may be idiopathic (genetic origin), structural (related to brain lesions or diseases), or of unknown origin. The treatment of epilepsy relies on the use of antiepileptic drugs (AEDs), aimed to reduce frequency and severity of seizures. Complete seizure remission is often difficult to achieve, so the primary goal is to maintain an acceptable quality of animal life. The most commonly used drugs are Phenobarbital and Potassium Bromide, but in recent years, newer antiepileptic medications such as Imepitoin, Zonisamide, and Levetiracetam have been introduced, providing greater tolerability. Despite wide availability of AEDs, some dogs do not respond adequately to treatments. In such cases, this is referred to refractory epilepsy, where seizure control is difficult even with multiple combined drugs. This is a significant challenge in managing the condition, and research is ongoing to develop new solutions. Recently, Cannabis has gained increasing interest for its therapeutic potential, particularly cannabidiol (CBD), which has demonstrated anticonvulsant properties. A non-psychoactive derivative of Cannabis sativa, appears to be well-tolerated by animal patients, making it a promising option for the future. Although the mechanisms of action are not yet fully understood; the expectation is that it could become a new frontier in the treatment of canine epilepsy

    Advanced synchronization and estimation techniques for modern satellite communications

    Le comunicazioni satellitari sono diventate parte integrante del nostro mondo moderno, rivoluzionando il modo in cui ci connettiamo, condividiamo le informazioni e accediamo ai servizi su scala globale. L'ubiquità dei satelliti nella società contemporanea è evidente nelle loro molteplici applicazioni, tra cui telecomunicazioni, telerilevamento, localizzazione, navigazione e altro. Le reti satellitari sono diventate anche una componente integrale dell'infrastruttura 5G, offrendo un prezioso complemento ed estensione alle reti terrestri. L'introduzione delle reti non terrestri (non terrestrial networks, NTN), di cui le comunicazioni satellitari sono una parte essenziale, nel 5G, garantirà una maggiore connettività e fornirà l'accesso a Internet anche a parti del pianeta non servite o poco servite dal segmento terrestre dei sistemi di comunicazione globale, come le isole o le regioni polari, nonché a navi e aerei situati lontano dalle coste. Motivata dalla moltitudine di possibili applicazioni satellitari, l'analisi degli effetti di canale diventa cruciale nei sistemi di comunicazione moderni e futuri. In questa tesi, consideriamo la progettazione di nuovi algoritmi per la sincronizzazione e la stima dei parametri nelle comunicazioni satellitari. La nostra analisi si concentra sulle modulazioni di ordine elevato, in quanto sono le migliori candidate a soddisfare la crescente domanda di alta velocità di trasmissione dei dati e di efficienza spettrale nelle comunicazioni moderne, ma anche le più colpite dai comuni disturbi del canale. Consideriamo i tipici disturbi del canale che influenzano il segnale nelle comunicazioni digitali, ovvero gli offset di temporizzazione dei simboli, gli effetti del rumore di fase e le non linearità del canale. L'obiettivo di questo lavoro è proporre nuove soluzioni che possono essere adottate per contrastare questi effetti e confrontarle con le strategie esistenti in letteratura. Come prima cosa, affrontiamo il problema del rilevamento physical layer header (PLH) a lunghezza variabile nella seconda generazione della trasmissione video digitale via satellite (DVB-S2X), che è una fase cruciale dopo la sincronizzazione del frame. L'uso di un PLH di lunghezza variabile si inserisce perfettamente nello scenario dello standard DVB-S2X, in cui vengono impiegate la codifica e la modulazione adattive (adaptive coding and modulation, ACM). Lo standard DVB-S2X prevede l'uso di PLH a lunghezza costante. Tuttavia, una soluzione più flessibile potrebbe essere l'uso di un PLH di lunghezza variabile, a seconda dei ModCods adottati. Infatti, quando si utilizzano ModCod che operano con rapporti segnale-rumore più elevati, la lunghezza del PLH può essere ridotta rispetto a quella prevista dallo standard. Per questo, proponiamo diverse strategie di decodifica per stimare la lunghezza del campo PLH trasmesso, che è sconosciuta al ricevitore, insieme alla decodifica del codice PLH. Dimostriamo che è possibile ottenere un'eccellente prestazione di decodifica riducendo significativamente l'overhead dovuto alla trasmissione del campo PLH rispetto a un PLH standard a lunghezza fissa. Dopo la sincronizzazione del frame, possono ancora esistere alcuni disallineamenti di temporizzazione. L'errore di temporizzazione residuo è, in genere, inferiore al tempo di simbolo. Tuttavia, soprattutto per le frequenze di simbolo elevate, il tempo di simbolo è relativamente breve e quindi anche un piccolo errore di temporizzazione rappresenta una frazione significativa del tempo di simbolo. Pertanto, per compensare questo errore, è necessario ricorrere a schemi di recupero dei tempi dei simboli (symbol timing recovery, STR). Abbiamo affrontato l'implementazione parallela di un sincronizzatore di temporizzazione dei simboli ad anello chiuso, proponendo un'architettura parallela a bassa complessità, che è particolarmente adatta ai ricevitori ad alta velocità. Un altro disturbo del canale che caratterizza la maggior parte dei collegamenti di comunicazione è il rumore di fase. Esso può derivare da instabilità degli oscillatori locali, sia al trasmettitore che al ricevitore, e può degradare significativamente le prestazioni del sistema. Il rumore di fase causa rotazioni di fase casuali nel segnale portante trasmesso ed è particolarmente critico con modulazioni di ordine elevato, come l'amplitude phase-shift keying (APSK), in cui i punti sono distribuiti sia in modo circolare che radiale. Per questi motivi, concentriamo la nostra analisi sulle tecniche di recupero del rumore di fase per modulazioni di ordine elevato, come quelle adottate nel DVB-S2X. Confrontiamo le prestazioni ottenibili da diversi algoritmi, in diverse condizioni di canale. Come per il recupero della temporizzazione, proponiamo anche una semplice architettura parallela che può essere utilizzata in applicazioni ad alta velocità e confrontiamo le sue prestazioni con quelle ottenibili con schemi seriali. Infine, affrontiamo il problema degli effetti non lineari, tipici dei collegamenti di comunicazione in cui vengono utilizzati amplificatori ad alta potenza (HPA), come i collegamenti satellitari o ottici. Proponiamo nuove costellazioni appositamente progettate per i canali non lineari, dimostriando che rispetto ad altri formati di modulazione permettono di ottenere buone prestazioni, soprattutto se si considerano costellazioni di grandi dimensioni.Satellite communications have become an integral part of our modern world, revolutionizing the way we connect, share information, and access services on a global scale. The ubiquity of satellite in contemporary society is evident in their multifaceted applications, including telecommunications, remote sensing, localization, navigation and beyond. Satellite networks have also become an integral component of the 5G infrastructure, offering a valuable complement and extension to the terrestrial networks. The introduction of non-terrestrial networks (NTNs), of which satellite communications is an essential part, into 5G, will ensure higher connectivity and provide internet access also to parts of the planet that are unserved or underserved by the terrestrial segment of the global communication systems, such as islands or polar regions, as well as to ships and planes located far from the coastlines. Motivated by the multitude of possible satellite applications, the analysis of channel effects becomes crucial in modern and future communication systems. In this thesis, we consider the design of novel algorithms for synchronization and parameter estimation in satellite communications. We focus our analysis on high order modulations, as they are the best candidates to satisfy the growing demand of high data rates and spectral efficiency in modern communications, but also the most affected by common channel impairments. We consider typical channel impairments that affect the signal in digital communications, namely symbol timing offsets, phase noise effects and channel nonlinearities. The aim of this work is to propose novel solutions that can be adopted to counteract these effects, and compare them with the existing strategies in the literature. We first address the problem of variable length physical layer header (PLH) detection in the second generation of the digital video broadcasting for satellite (DVB-S2X), which is a crucial step after frame synchronization. The use of a variable length PLH perfectly fits into the scenario of the DVB-S2X standard, where adaptive coding and modulation (ACM) is employed. The DVB-S2X standard foresees the use of constant length PLH. However, a more flexible solution could be the use of a variable length PLH, depending on the adopted modulation and coding (ModCod) schemes. In fact, when using ModCods operating at higher signal-to-noise ratios, the PLH length may be reduced with respect to the length foreseen by the standard. We propose different decoding strategies to estimate the length of the transmitted PLH field, which is unknown at the receiver, jointly with the decoding of the PLH code. We demonstrate that it is possible to achieve an excellent decoding performance while significantly reducing the overhead due to the transmission of the PLH field with respect to a standard fixed-length PLH. After frame synchronization, some timing misalignments may still exist. The residual timing error is, typically, smaller than the symbol time. However, especially for high symbol rates, the symbol time is relatively short, therefore even a small timing error represents a significant fraction of the symbol time. Therefore, symbol timing recovery (STR) schemes should be employed to compensate this offset. We address the parallel implementation of a closed-loop symbol timing synchronizer, by proposing a low-complexity parallel architecture, which is practically suitable for high-speed receivers. Another channel impairment that characterizes most of the communication links is phase noise (PN). It can arise from instabilities of local oscillators, either at the transmitter or at the receiver, and it can significantly degrade the system performance. PN causes random phase rotations in the transmitted carrier signal, and it is especially critical with high order modulations, such as amplitude phase-shift keying (APSK), where the points are distributed both in a circular and radial pattern. For these reasons, we focus our analysis on phase noise recovery techniques for high order modulations, as the ones adopted in DVB-S2X. We compare the performance achievable by different algorithms, under different channel conditions. Then, as we did for the timing recovery, we also propose a simple parallel architecture that can be used in high-speed applications, and we compare its performance with that achievable by serial schemes. Finally, we address the problem of nonlinear effects, which is typical of communication links where high-power amplifiers (HPA) are used, such as satellite or optical links. We propose new constellations specifically designed for nonlinear channels. We show that, compared to conventional modulation formats, they achieve good performance, especially when considering large constellation size

    Integrated liquid biopsy for cancer diagnostics via genomic biomarkers detection

    Cancer is one of the leading causes of mortality worldwide, accounting for about 10 million deaths annually. The survival rates can be improved by an early-stage diagnosis achieved by regular population screening, but existing methods are invasive (tissue biopsy), expose patients to radiation (image-based techniques), or show limited accuracy (blood and stool tests). Liquid biopsy is a promising minimally invasive alternative method based on the analysis of body fluids, like blood, saliva, or urine, for the presence of cancer-related biomarkers. Genomic biomarkers based on DNA and RNA alterations are of particular interest because of the contribution of genetic changes to cancer onset and progression. The currently used gold standard for nucleic acids detection, fluorescence-based techniques, requires a multi-step assay and labeling with a fluorescent dye due to weak auto-fluorescence from biological samples. This thesis aims to explore alternative methods with more straightforward assays and better multiplexing abilities for genomic biomarker detection. Surface-enhanced Raman Spectroscopy (SERS) has a promising potential for liquid biopsy application due to its ability to provide the qualitative and potentially quantitative composition of a sample by analyzing the enhanced inelastic scattering of light. An amplification of Raman scattering is achieved by metal NPs arranged as a colloidal solution, as an array on a solid substrate, or as a uniform nanostructure. The selective detection of DNAs and RNAs from body fluids requires biological protocols based on the application of DNAs complementary to the target. Chapter 1 provides details about the working principle of SERS and a literature review of the biological protocols for target-specific capturing. Chapter 2 presents the fabrication and testing of SERS substrates based on NPs colloidal solution and NPs on the solid substrate. Chapter 3 studies the application of one of the biological protocols for the SERS-based detection of microRNAs. Chapter 4 explores the application of photonic crystal fibers as a microfluidic device for the SERS-based measurement of microRNAs. Chapter 5 discusses the possibility of achieving spatially resolved chemical measurement with a technique similar to SERS called tip-enhanced Raman Spectroscopy. This thesis has shown that there is significant research attention towards the SERS-based detection of genomic biomarkers, especially the ones based on the dysregulation of microRNA expression level. The literature review has shown that some one-to-two-step biological protocols with relatively straightforward assays have a detection ability comparable to fluorescence-based sensing and even better multiplexing abilities. The SERS substrates developed during this PhD have shown the limit of detection of down to 10-8 M with a promising potential for further improvements. The use of photonic crystal fibers was found to be an exciting research direction due to the longer interaction area between the light, NPs, and the analytes, and further optimization of the experimental parameters can be achieved with the COMSOL model presented in the thesis. The last chapter also has shown that there are other techniques to amplify Raman scattering that can provide even more advantages, like spatially resolved measurements, and that also have a potential for liquid biopsy application

    Psychological well-being of university students : the impact of Not Just Right Experiences, Fear of Guilt, Harm Avoidance and Obsessive Compulsive Personality Disorder symptomatology

    Nonostante la stima della prevalenza del Disturbo Ossessivo Compulsivo di Personalità (DOCP) tra la popolazione generale non sia trascurabile, la ricerca su tale disturbo risulta essere non particolarmente sviluppata e contrastante. Secondo la letteratura esistente, il DOCP risulta un disturbo collegato ad alcune sensazioni, sentimenti e tratti temperamentali, come il fenomeno della Not Just Right Experience, la paura del Senso di Colpa e l’Harm Avoidance; inoltre, il DOCP risulta un disturbo che può presentarsi in comorbilità con sintomi depressivi. Sulla base di tali evidenze, il presente lavoro di tesi si pone l’obiettivo di analizzare tali fattori e i collegamenti tra loro. I dati sono stati raccolti in un campione non clinico di studenti universitari tramite una batteria di questionari composta da vari test: la presenza dei criteri del DOCP, la Fear of Guilt Scale, il Not Just Right Experiences Questionnaire-Revised, il Temperament and Character Inventory per l’Harm Avoidance; è stato inoltre valutato il benessere psicologico soggettivo dei partecipanti tramite l’Oxford Happiness Questionnaire. L’ipotesi di questo progetto è quindi verificare se e in che modo i punteggi nei fattori elencati possano essere correlati ai punteggi di benessere psicologico percepito degli studenti universitari. Lo scopo di questo progetto riguarda l’individuazione e l’analisi di alcuni fattori che possono essere correlati a bassi punteggi nel benessere psicologico degli studenti universitari, così da identificare quelle che possono essere tecniche e terapie specifiche da poter implementare nel counseling psicologico diretto a tali studenti; inoltre, questa tesi si propone di spingere ed incoraggiare la ricerca verso lo studio e l’approfondimento del DOCP anche in popolazioni non cliniche.Although the estimated prevalence of the Obsessive-Compulsive Personality Disorder (OCPD) among the general population is not negligible, the research about this condition shows little development and conflicting results. According to current literature, the OCPD is a condition linked to certain sensations, feelings and temperament traits, as the phenomenon of Not Just Right Experience, Fear of Guilt and Harm Avoidance; furthermore, the OCPD is a condition which can show co-morbidity with depressive symptoms. On the basis of this evidence, the objective of this thesis work is to analyse the factors and connections occurring between the aforementioned conditions. Data have been collected through a non-clinical sample of University students, through a battery of questionnaires made up by various tests: the presence of criteria of OCPD, the Fear of Guilt Scale, the Not Just Right Experiences Questionnaire-Revised, the Harm Avoidance Temperament and Character Inventory; it has also been evaluated the personal psychological well-being among the participants through the Oxford Happiness Questionnaire. The Hypothesis of this project is so to verify if and in which way the scoring of the listed factors can be related to the psychological well-being scores perceived by University students. The aim of this work concerns both the identification and the analysis of some factors that can be related to low scoring of students’ psychological well-being, so as to identify those that can be used as specific techniques and therapies to implement psychological counselling meant for such students; moreover, this thesis aims to push and encourage research towards the study and the in-depth analysis of OCPD also in non-clinical population

    Il Symbolic Learning (SL) studia algoritmi di apprendimento per modelli computazionali che si basano sulla logica formale (o logica simbolica) e, come tale, fornisce modelli di intelligenza artificiale che eseguono ragionamenti logici trasparenti, espliciti, nonostante il suo ruolo centrale nell'intelligenza artificiale (AI) degli anni '80/'90, gli ultimi due decenni di ricerca sull'IA sono stati contrassegnati da grandi progressi nel deep learning, che hanno spostato l'attenzione su modelli a scatola nera altamente accurati. Negli ultimi anni la SL è tornata alla ribalta come soluzione promettente al cosiddetto problema della scatola nera, a causa della natura interpretabile della rappresentazione della conoscenza sottostante, ma allo stato attuale appare sia sottosviluppata che sottosviluppata. frammentato e generalmente non è considerato un serio concorrente del deep learning, probabilmente a causa di due fattori principali. L’obiettivo di questo lavoro è incoraggiare la ricerca in SL e, in definitiva, consentire interazioni virtuose tra intelligenza umana e computazionale, durante la negoziazione di compiti difficili.Symbolic Learning (SL) studies learning algorithms for computational models that rely on formal (or symbolic) logic and, as such, it provides AI models that perform transparent, explicit logical reasoning. Inspite of its central role in the Artificial Intelligence (AI) of the 80s/90s, the last two decades of research in AI had been marked by great advances in Deep Learning, which shifted the focus on highly accurate, black-box models. While this caused a slowdown in SL research, over the very last years, the field has been making a comeback as a promising solution to the so-called black box problem, due to the interpretable nature of its underlying knowledge representation. In its current state, SL appears fragmented, underdeveloped, and it is generally not regarded as a serious competitor to Deep Learning. This is likely due to two major factors. First, while deep learning methods specialized to time-series, images, graphs, etc., most of the widespread SL approaches are still based on propositional logic or predicate logic, which are only suitable for static, tabular data, and for purely relational data, respectively. Second, while several established programming frameworks exist for Deep Learning (e.g., tensorflow, Theano, Deeplearning4j, scikit-learn), implementations of SL algorithms (when available) are scattered across different programming languages and libraries, making it difficult to appreciate the true potential of explicit knowledge representations. This thesis addresses both of these gaps. First, with the aim of providing a modern, unifying view on the subject, we present SOLE, a comprehensive abstract framework for SL methodologies. The framework is based on a solid logical foundation that allow for the use of virtually any logical formalism, and covers (logic-specific and -agonistic) methods from Symbolic Data Analysis, Symbolic Learning and Symbolic Modelling. SOLE is also implemented and publicly available in the Julia programming language as Sole.jl, the first programming framework specifically tailored for symbolic methods. Second, we report recent, published experimental results showing how extensions of common SL methods to more expressive, modal logics can lead to interpretable and accurate models for tasks involving non-tabular data. The experimental part of this thesis considers four different classification tasks involving multivariate time-series and images. For each task, we provide an experimental evaluation of a novel decision tree learning algorithm based on temporal/spatial modal logics. The learned models display medium to high accuracies, and qualitative inspections of the extracted knowledge reveal insights into the underlying processes of the classification problems at hand. The objective of this work is to encourage research in SL, and to, ultimately, enable virtuous interactions between human and computational intelligence, when tackling hard tasks

    Design e sviluppo di biomateriali ecosostenibili per la salute

    Questo lavoro si è focalizzato sulla progettazione e sviluppo di biomateriali polimerici e ibridi applicabili nel settore sanitario seguendo i principi di ecosostenibilità ed economia circolare. Oggigiorno c’è una crescente necessità di sviluppare dispositivi sanitari composti da risorse rinnovabili che siano economici, ma soprattutto facili da smaltire e che non danneggino l’ambiente, oltre che sicuri ed efficaci per l’uomo. In questo contesto, lo scopo di questo lavoro è stato quello di sfruttare materiali biocompatibili e biodegradabili provenienti da fonti naturali e processi biomimetici green per sviluppare nuovi biomateriali multifunzionali in grado di rispondere a specifiche esigenze sanitarie. Nello specifico sono stati sviluppati due biomateriali ecosostenibili, uno completamente polimerico e l'altro di composizione ibrida, che hanno evidenziato notevoli potenzialità nei campi della ventilazione meccanica e della cosmesi a basso impatto ambientale. Il primo materiale è un aerogel polimerico composto da gelatina e chitosano, sviluppato attraverso un processo di chimica green e ottimizzato per ottenere un dispositivo medico ecosostenibile in grado di garantire, durante la terapia intensiva o l'anestesia, i normali livelli di umidità, calore e filtrazione dell'aria quando le vie aeree superiori del paziente vengono bypassate. Nel dettaglio, sono state selezionate materie prime derivate da scarti alimentari per realizzare un filtro monouso biodegradabile a ridotto impatto ambientale e a basso costo. Inoltre, i processi di liofilizzazione e reticolazione sono stati ottimizzati per sviluppare un materiale 3D stabile con un’elevata efficienza di filtrazione scalabile per l’industrializzazione. Il secondo materiale è un composto ibrido costituito da una fase minerale di idrossiapatite drogata con ioni titanio e nucleata su una matrice biopolimerica di alginato attraverso un processo di biomineralizzazione ispirato alla natura, con proprietà riflettenti adatte alla formulazione di filtri solari ecosostenibili. In particolare, oltre a proteggere adeguatamente la pelle dalle radiazioni UV ed eliminare gli effetti indesiderati causati dai componenti tipici dei filtri solari commerciali (effetto fotocatalitico, irritazione, penetrazione di nanoparticelle, ecc.), ci siamo concentrati sullo sviluppo di un filtro UV biocompatibile, che una volta formulato in crema e applicato sulla pelle, non rilascia sostanze dannose per l’ecosistema marino e per l'uomo. Infine, è stata effettuata una caratterizzazione a tutto tondo degli aspetti chiave di ciascun materiale, evidenziando i principali risultati e ostacoli, nonché i principali aspetti complessivi che potrebbero essere migliorati.This work has been focused on the design and development of polymeric and hybrid biomaterials applicable in the healthcare sector following the principles of eco-sustainability and circular economy. Nowadays there is a growing need to develop healthcare devices composed of renewable resources that are inexpensive, but above all easy to dispose of and which do not harm the environment, as well as being safe and effective for humans. In this context, the aim of this work was to exploit biocompatible and biodegradable materials from natural sources and green biomimetic processes to develop new multifunctional biomaterials capable of responding to specific healthcare needs. Specifically, two eco-sustainable biomaterials have been developed, one fully polymeric and the other of hybrid composition, which have highlighted considerable potential in the fields of mechanical ventilation and of cosmetics with low environmental impact. The first material is a polymeric aerogel made of gelatin and chitosan, developed through a green-chemistry process optimized to obtain an eco-sustainable medical device capable of guaranteeing, during intensive care or anesthesia, the normal levels of humidity, heat and filtration of the trachea, when the patient's upper airways are by-passed. In detail, raw materials derived from food waste were selected to create a biodegradable disposable filter with reduced environmental impact and low cost. Furthermore, the freeze-drying and cross linking processes were optimized to develop a stable 3D material with high filtration efficiency that is scalable for industrialization. The second material is a hybrid compound consisting of a mineral phase, a hydroxyapatite doped with titanium ions, nucleated on a biopolymeric matrix of alginate through a nature inspired biomineralization process, demonstrating reflecting properties adapt for the formulation of eco-sustainable sunscreens. In particular, in addition to adequately protecting the skin from UV radiation and eliminating the undesirable effects caused by the typical components of commercial sunscreens (photocatalytic effect, irritation, penetration of nanoparticles, etc.), we focused on the development of a biocompatible UV filter which once formulated into a cream and applied to the skin, does not release substances harmful for marine ecosystems or humans. Finally, an all-round characterization of the key aspects of each material was carried out, highlighting the main achievements and obstacles, as well as the main overall aspects that could be improved

    In the last decade, regenerative medicine has gained interest in the veterinary clinical practice, since it offers a promising therapeutic alternative, especially for the treatment of musculoskeletal disorders (MSDs), characterized by multimodal management and complex healing processes. Among MSDs, osteoarthritis affects a high percentage of canine, equine, and even human populations, thus representing a one-health challenge. Furthermore, there is still a lack of effective long-term treatment for osteoarthritis. In this scenario, Mesenchymal stromal cells (MSCs) have been shown to modulate inflammation and stimulate the repair of damaged tissue, and they can have a positive long-term impact on the animal’s quality of life. Despite promising results obtained in experimental and clinical studies, MSCs therapeutic applications are seriously influenced by the in-vivo implantation of an active and proliferating cell population. Recently, the new trend has been represented by MSCs-derived secretome, a cell-free product composed of soluble factors and extracellular vesicles, with similar biological activity but greater practical advantages when compared to the cells themselves. The present thesis work focused on a freeze-dried form of secretome, called Lyo-secretome, prepared from adipose-derived MSCs from canine and equine species. The first aim was the validation of the technological process for veterinary clinical-use medicinal production of different canine and equine batches of Lyo-secretome, and its characterization in terms of quantitative and qualitative analysis. Furthermore, in vitro metabolic tests were performed on target cells, such as tenocytes, chondrocytes, and synovial fluid derived-MSCs, demonstrating positive proliferative effects. Additionally, proteomic analysis demonstrated the presence of proteins involved in regulating the inflammatory response. The next step was a double-blinded randomized controlled clinical trial, enrolling dogs and horses suffering from naturally occurring osteoarthritis to assess the in vivo safety and efficacy of Lyo-secretome. The protocol included a double intra-articular injection of Lyo-secretome resuspended in hyaluronic acid, at 40 days interval. Clinical and orthopedic examinations, as well as owner questionnaires, were used to evaluate the patients during the 180-day follow-up. Results indicated that the double intra-articular injection of allogeneic Lyosecretome is safe and does not induce systemic adverse response for both species. A mild to moderate joint flare was observed in some patients after the injection, although they were self-limiting and not correlated to the treatment, since no differences were found with the control group (hyaluronic acid). Regarding the efficacy in canine patients, the study revealed mild clinical improvement following the two injections in both treatment and control groups, without highlighting one treatment to be significantly superior, even if a positive tendency in terms of lameness and pain score, as well as an improvement in the Helsinki Chronic Pain Index score was observed in the Lyo-secretome group
