Cardiovascular Complications of COVID-19: Pharmacotherapy Perspective
Coronavirus disease of 2019 (COVID-19), which is caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), is spreading rapidly the world over. The disease was declared �pandemic� by the World Health Organization. An approved therapy for patients with COVID-19 has yet to emerge; however, there are some medications used in the treatment of SARS-CoV-2 infection globally including hydroxychloroquine, remdesivir, dexamethasone, protease inhibitors, and anti-inflammatory agents. Patients with underlying cardiovascular disease are at increased risk of mortality and morbidity from COVID-19. Moreover, patients with chronic stable states and even otherwise healthy individuals might sustain acute cardiovascular problems due to COVID-19 infection. This article seeks to review the latest evidence with a view to explaining possible pharmacotherapies for the cardiovascular complications of COVID-19 including acute coronary syndrome, heart failure, myocarditis, arrhythmias, and venous thromboembolism, as well as possible interactions between these medications and those currently administered (or under evaluation) in the treatment of COVID-19. © 2020, Springer Science+Business Media, LLC, part of Springer Nature
The predictive value of the epicardial adipose thickness in the rate of expansion of the aortic root Dicke des epikardialen Fettgewebes als Prädiktor für die Ausdehnung der Aortenwurzel
Background: Epicardial adipose tissue (ECAT) is metabolically active and is involved in the development of atherosclerosis. The thickness of ECAT has been positively correlated with the dimensions of the ascending aorta. We aimed to examine whether ECAT thickness predicted the expansion of the aortic dimensions. Methods: The imaging results of patients who had undergone transthoracic echocardiographic (TTE) examinations more than twice during the period 2005�2015 were surveyed. We included adult patients who had undergone TTE examinations at least 1 year apart. The ECAT was measured in the parasternal long-axis view from the index TTE study. End-diastolic dimensions in three consecutive beats were averaged for all measurements. The annulus, root, and sinotubular junction (STJ) were also measured. The amount of increase (if any) in aortic dimensions per year was calculated and the correlation of this increase with the initial thickness of the ECAT was analyzed. Results: In total, 429 examinations were performed with 197 patients (17 females), from which 394 examinations were analyzed. The ECAT thickness was 8.6�± 3.6�mm. In the initial examinations, the annulus, STJ, and root measured 23�± 4, 28�± 4, and 34�± 4�mm, respectively. In univariate analysis, for every 1�mm of ECAT thickness, the STJ expanded 0.056 (95 CI: 0.001�0.112�mm/year; p�= 0.030) and the aortic root expanded 0.088�mm/year (p�< 0.001). In multivariate analysis, ECAT thickness remained an independent predictor of the aortic root expansion. For every 1�mm increase in ECAT thickness, the aortic root expanded by 0.036�mm (95 CI: 0.010�0.062) per year (p�= 0.007). Conclusion: The thickness of the ECAT is a predictor of more rapid increases in the dimensions of the aortic root. Clinicopathological significance of tumor stem cell markers aldh1 and CD133 in colorectal carcinoma
Background & Objective: Colorectal cancer (CRC) is the third most common cancer worldwide with a high mortality rate. The main causes of death in patients are recurrence and metastasis which are mainly attributed to the small subpopulation of cells within tumors called cancer stem cells (CSCs). This study aimed to evaluate the correlation between the expression of ALDH1 and CD133 as CSC associated markers and clinicopathological characteristics in CRC. Methods: In this cross-sectional study, a total of 483 CRC tumor samples were immunohistochemically stained for detection of CD133 and ALDH1 markers. Correlations of marker expression with clinicopathological factors were also evaluated. Results: There was a significant correlation between the luminal intensity of CD133 and neural invasion (P=0.05) and between the cytoplasmic intensity of CD133 and metastasis (P=0.05). In terms of H-score, a positive significant relation was observed between cytoplasmic expression of CD133 and lymph node (P=0.02), neural (P=0.04) and vascular invasion (P=0.02). The ALDH1 cytoplasmic expression showed a significant correlation with tumor size (P=0.001). Conclusion: Our findings showed that increased expression of CD133 and ALDH1 is associated with tumor progression and worse outcomes in CRC patients. These markers can be good candidates for localized targeting of CSCs using antibodies. Future researches need to be improved approaches for early detection of CRC, and treatment monitoring for CRC and other cancers. © 2021, Iranian Society of Pathology. All rights reserved
Psychological well-being of infertile women and its relationship with demographic factors and fertility history: a cross-sectional study
Background: Infertility leads to a wide range of psychological injuries that may reduce psychological well-being. This study aimed to determine the psychological well-being of infertile women and its relation with demographic factors and fertility history. Methods: This cross-sectional study was conducted on 300 infertile women referred to three infertility centres, Tehran, Iran. The sampling was continuous. We collected data from a self-generated demographic and fertility questionnaire and Ryff's Psychological Well-being Scale (PWB). Data analysis was done by independent t-test, one way ANOVA. The significance level was set at P < 0.05. Results: The results showed that there was no significant relationship between demographic variables including age, occupation of each couple, spousal�s education, economic status and place of residence with PWB, but the mean score of PWB was significantly different in women's educational levels (P = 0.03). There was also a significant difference between the mean score of PWB among different groups in the duration of marriage (P = 0.01). Fertility characteristics variables include the duration of infertility, duration of treatment of infertility, and current treatment were not the relation with PWB. However, the mean score of PWB in the number of IVF (P = 0.003) and the failed IVF pregnancies (P = 0.01) had a significant statistical difference. Conclusion: The results showed that PWB related to several variables. Paying attention to these variables can help in the preparation and development of counseling or educational programs. © 2021, The Author(s)
The pro-apoptosis effects of Echinacea purpurea and Cannabis sativa extracts in human lung cancer cells through caspase-dependent pathway
Background: Considering the advantages of using medicinal herbs as supplementary treatments to sensitize conventional anti-cancer drugs, studying functional mechanisms and regulatory effects of Echinacea purpurea (as a non-cannabinoid plant) and Cannabis sativa (as a cannabinoid plant) are timely and required. The potential effects of such herbs on lung cancer cell growth, apoptosis, cell cycle distribution, cellular reactive oxygen species (ROS) level, caspase activity and their cannabinomimetic properties on the CB2 receptor are addressed in the current study. Methods: The cytotoxic effect of both herb extracts on the growth of lung cancer cells (A549) was assessed using the MTT assay. The annexin-V-FITC staining and propidium iodide (PI) staining methods were applied for the detection of apoptosis and cell cycle distribution using flow cytometry. The cellular level of ROS was measured using 7�-dichlorofluorescin diacetate (DCFH-DA) as a fluorescent probe in flow cytometry. The caspase 3 activity was assessed using a colorimetric assay Kit. Results: Echinacea purpurea (EP) root extract induced a considerable decrease in A549 viable cells, showing a time and dose-dependent response. The cell toxicity of EP was accompanied by induction of early apoptosis and cell accumulation at the sub G1 phase of the cell cycle. The elevation of cellular ROS level and caspase 3 activity indicate ROS-induced caspase-dependent apoptosis following the treatment of A549 cells by EP extract. The observed effects of EP extract on A549 growth and death were abrogated following blockage of CB2 using AM630, a specific antagonist of the CB2 receptor. Increasing concentrations of Cannabis sativa (CS) induced A549 cell death in a time-dependent manner, followed by induction of early apoptosis, cell cycle arrest at sub G1 phase, elevation of ROS level, and activation of caspase 3. The CB2 blockage caused attenuation of CS effects on A549 cell death which revealed consistency with the effects of EP extract on A549 cells. Conclusions: The pro-apoptotic effects of EP and CS extracts on A549 cells and their possible regulatory role of CB2 activity might be attributed to metabolites of both herbs. Deep COVID DeteCT: an international experience on COVID-19 lung detection and prognosis using chest CT
The Coronavirus disease 2019 (COVID-19) presents open questions in how we clinically diagnose and assess disease course. Recently, chest computed tomography (CT) has shown utility for COVID-19 diagnosis. In this study, we developed Deep COVID DeteCT (DCD), a deep learning convolutional neural network (CNN) that uses the entire chest CT volume to automatically predict COVID-19 (COVID+) from non-COVID-19 (COVID�) pneumonia and normal controls. We discuss training strategies and differences in performance across 13 international institutions and 8 countries. The inclusion of non-China sites in training significantly improved classification performance with area under the curve (AUCs) and accuracies above 0.8 on most test sites. Furthermore, using available follow-up scans, we investigate methods to track patient disease course and predict prognosis. © 2021, The Author(s)
Nanocomposite scaffolds for accelerating chronic wound healing by enhancing angiogenesis
Skin is the body�s first barrier against external pathogens that maintains the homeostasis of the body. Any serious damage to the skin could have an impact on human health and quality of life. Tissue engineering aims to improve the quality of damaged tissue regeneration. One of the most effective treatments for skin tissue regeneration is to improve angiogenesis during the healing period. Over the last decade, there has been an impressive growth of new potential applications for nanobiomaterials in tissue engineering. Various approaches have been developed to improve the rate and quality of the healing process using angiogenic nanomaterials. In this review, we focused on molecular mechanisms and key factors in angiogenesis, the role of nanobiomaterials in angiogenesis, and scaffold-based tissue engineering approaches for accelerated wound healing based on improved angiogenesis.Figure not available: see fulltext. © 2021, The Author(s)
A combination of herbal compound (SPTC) along with exercise or metformin more efficiently alleviated diabetic complications through down-regulation of stress oxidative pathway upon activating Nrf2-Keap1 axis in AGE rich diet-induced type 2 diabetic mice
Background: SPTC is a mix of four herbal components (Salvia officinalis, Panax ginseng, Trigonella foenum-graeceum, and Cinnamomum zeylanicum) which might be prevented the development of AGE rich diet-induced diabetic complication and liver injury through activated the nuclear factor erythroid-2-related-factor-2 (Nrf2) pathway. Nrf2, as a master regulator of antioxidant response elements by activating cytoprotective genes expression, is decreased oxidative stress that associated with hyperglycemia and increases insulin sensitivity. the aim of this study was to assess whether the combination therapy of SPTC along with exercise or metformin moderate oxidative stress related liver injurie with more favorable effects in the treatment of AGE rich diet-induced type 2 diabetic mice. Methods: We induced diabetes in C57BL/6 mice by AGE using a diet supplementation and limitation of physical activity. After 16 weeks of intervention, AGE fed mice were compared to control mice. Diabetic mice were assigned into seven experimental groups (each group; n = 5): diabetic mice, diabetic mice treated with SPTC (130 mg/kg), diabetic mice treated with Salvia Officinalis (65 mg/kg), diabetic mice treated with metformin (300 mg/kg), diabetic mice with endurance exercise training, diabetic mice treated with SPTC + metformin (130/300 mg/kg), diabetic mice treated with SPTC + exercise training. Results: SPTC + exercise and SPTC + metformin reduced diabetic complications like gain weight, water and calorie intake, blood glucose, insulin, and GLUT4 content more efficiently than each treatment. These combinations improved oxidative stress hemostasis by activating the Nrf2 signaling pathway and attenuating keap1 protein more significantly. Conclusion: Eventually, combined treatment of SPTC with exercise or metformin as a novel approach had more beneficial effects to prevent the development of diabetes and oxidative stress associated with hyperglycemia. Chitosan hydrogel loaded with Aloe vera gel and tetrasodium ethylenediaminetetraacetic acid (EDTA) as the wound healing material: in vitro and in vivo study
The main aim of the current study was to develop a chitosan hydrogel containing Aloe vera gel and Ethylenediaminetetraacetic acid (EDTA) as the wound healing materials. Chitosan with the concentration of (2 w/v) was prepared in AA (0.5, v/v) and Tetrasodium EDTA (0.01 w/w) and AV (0.5 v/v) were added to the prepared polymer solution. As prepared solution was cross-linked by β-GP with the weight ratio of 1/6 w/w (1 chitosan and 6 β-GP). The characterization of the hydrogels showed that the hydrogels have porous structures and interconnected pores with the pores size range from 41.5 ± 14 to 48.3 ± 11 μm. The swelling and weight loss measurements of the hydrogels showed that the hydrogels could swell up to 240 of their initial weight during 8 h and loss 79.7 ± 3.5 of the initial weight during 14 days. The antibacterial studies depicted that the prepared Cs/tEDTA/AV hydrogel inhibited the growth of Staphylococcus aureus (the minimum inhibition concentration, MIC of 73 ± 4.8) and Pseudomonas aeruginosa (the MIC of 40 ± 7.9). Moreover, the prepared hydrogels were hemocompatible (Cs/tEDTA/AV: OD of 0.24 ± 0.30) and biocompatible (Cs/tEDTA/AV: OD of 0.38 ± 0.01). At the final stage, the wound healing assessments in the animal model revealed that the application of the prepared hydrogels effectively enhanced the wound healing process. In conclusion, the results confirmed the efficacy of the prepared hydrogels as the wound healing materials. © 2020 Wiley Periodicals LLC
Ultrasound�assisted synthesis of FeTiO3/GO nanocomposite for photocatalytic degradation of phenol under visible light irradiation
Phenolic compounds are dangerous pollutants in industrial effluents that have caused many human and environmental problems. These compounds are resistant to decomposition conventional biological systems. Photocatalytic treatment is amongst the most efficient technique for phenol removal from aqueous media. In this study, FeTiO3/GO nanocomposite was synthesized through the ultrasound�assisted method. Structural, morphological, and optical properties of the synthesized nanocomposite were characterized by TEM, XRD, EDS, XPS, BET, TGA, FTIR, and UV�visible techniques. Optimum conditions for phenol photocatalytic degradation were obtained in 3 of GO content, 0.75 g/L nanocomposite dosage, and pH = 8. The degradation efficiency decreased by increasing phenol concentrations and complete mineralization was observed after 240 min of irradiation in optimum conditions. The effect of scavengers indicated that superoxide and hydroxyl radicals had the main role in the photodegradation process. The hindrance effect of inorganic ions was obtained asHCO3- > Cl- > SO42� > NO3-. Our experimental results put forward the FeTiO3/GO nanocomposite as a highly stable catalyst that can successfully remove phenolic compounds from aqueous solutions. Only a 10.8 decrease was observed in phenol degradation efficiency after five cycles, showing high recyclability of the FeTiO3/GO nanocomposite. © 2020 Elsevier B.V
