    Peptidoglycan synthesis drives a single population of septal cell wall synthases during division in Bacillus subtilis

    Bacterial cell division requires septal peptidoglycan (sPG) synthesis by the divisome complex. Treadmilling of the essential tubulin homologue FtsZ has been implicated in septal constriction, though its precise role remains unclear. Here we used live-cell single-molecule imaging of the divisome transpeptidase PBP2B to investigate sPG synthesis dynamics in Bacillus subtilis. In contrast to previous models, we observed a single population of processively moving PBP2B molecules whose motion is driven by peptidoglycan synthesis and is not associated with FtsZ treadmilling. However, despite the asynchronous motions of PBP2B and FtsZ, a partial dependence of PBP2B processivity on FtsZ treadmilling was observed. Additionally, through single-molecule counting experiments we provide evidence that the divisome synthesis complex is multimeric. Our results support a model for B. subtilis division where a multimeric synthesis complex follows a single track dependent on sPG synthesis whose activity and dynamics are asynchronous with FtsZ treadmilling

    Self- and informant-reported personality traits and vaccination against COVID-19

    As COVID-19 vaccines’ accessibility has grown, so has the role of personal choice in vaccination, and not everybody is willing to vaccinate. Exploring personality traits’ associations with vaccination could highlight some person-level drivers of, and barriers to, vaccination. We used self- and informant-ratings of the Five-Factor Model domains and their subtraits (a) measured approximately at the time of vaccination with the 100 Nuances of Personality (100NP) item pool (N = 56,575) and (b) measured on average ten years before the pandemic with the NEO Personality Inventory-3 (NEO-PI-3; N = 3,168). We tested individual domains’ and either items’ (in the 100NP sample) or facets’ (in the NEO-PI-3 sample) associations with vaccination, as well as their collective ability to predict vaccination using elastic net models trained and tested in independent sample partitions. Although the NEO-PI-3 domains and facets did not predict vaccination ten years later, the domains correlated with vaccination in the 100NP sample, with vaccinated people scoring slightly higher on neuroticism and agreeableness and lower on openness, controlling for age, sex, and education. Collectively, the five domains predicted vaccination with an accuracy of r = .08. Associations were stronger at the item level. Vaccinated people were, on average, more science-minded, politically liberal, respectful of rules and authority, and anxious but less spiritual, religious, and self-assured. The 100NP items collectively predicted vaccination with r = .31 accuracy. We conclude that unvaccinated people may be a psychologically heterogeneous group and highlight some potential areas for action in vaccination campaigns

    Mixed method evaluation of a learning from excellence programme for community health workers in Neno, Malawi

    Background: Community Health Workers (CHWs) play an essential role in linking communities to facility-based healthcare. However, CHW programmes have often been hampered by low levels of staff motivation, and new tools aimed at improving staff motivation and work environment are needed. One such intervention is the “Learning from Excellence” (LfE) programme. We aimed to assess feasibility, outputs, and impact of a co-designed LfE programme on CHW motivation, in Neno District. Methods: We conducted a convergent mixed-method evaluation of the LfE programme. Co-design of the programme and forms took place between October 2019 and January 2020. LfE forms submitted between September and November 2020 were analysed using descriptive statistics and memos summarising answers to the open-ended question. To investigate experiences with LfE we conducted in-depth semi-structured interviews with key stakeholders, CHWs, and site supervisors, which were analysed thematically. A pre-post intervention questionnaire was developed to assess the impact of the co-designed LfE intervention on CHW motivation and perceived supervision. Outcomes were triangulated into a logic model. Results: In total 555 LfE forms were submitted, with 34.4% of CHWs in Neno District submitting at least one LfE report. Four themes were identified in the interviews: LfE implementation processes, experience, consequences, and recommendations. A total of 50 CHWs participated in the questionnaire in January 2020 and 46 of them completed the questionnaire in December 2020. No statistically significant differences were identified between pre-and post-LfE measurements for both motivation (Site F: p = 0.86; Site G: p = 0.31) and perceived supervision (Site F: p = 0.95; Site G: p = 0.45). A logic model, explaining how the LfE programme could impact CHWs was developed. Conclusions: Many CHWs participated in the LfE intervention between September 2020 and November 2020. LfE was welcomed by CHWs and stakeholders as it allowed them to appreciate excellent work in absence of other opportunities to do so. However, no statistically significant differences in CHW motivation and perceived supervision were identified. While the intervention was feasible in Neno District, we identified several barriers and facilitators for implementation. We developed a logic model to explain contextual factors, and mechanisms that could lead to LfE outcomes for CHWs in Neno District. The developed logic model can be used by those designing and implementing interventions like LfE for health workers

    Agricultural improvements that reduce conventional pesticide use and support environmental aims are a priority. Current approaches develop promising alternative products but meet significant challenges in bringing them to market. This article reports findings of an Association of Applied Biologists event at which delegates from relevant industry sectors discussed the establishment of an effective integrated pest management innovation system linking multiple stakeholders. Interrelated recommendations were agreed upon, focused on structured gap analysis, co‐design processes reflecting the complete innovation system, the approval process, application equipment, enhancing grower confidence, integrating knowledge exchange activities, promulgation of public good information and the need for an overarching national action plan and supporting legislation

    Science for all? School science education policy and STEM skills shortages

    Whether enough highly qualified STEM workers are being educated and trained in the UK is an important question. The answer has implications not only for educators, employers and policymakers but also for individuals who are currently engaged in, or are considering entering, education or training in this area. Set against a policy backdrop that prioritises students studying more science for longer, this paper considers long-term patterns of participation in STEM education – from school science through to graduate entry into the highly skilled STEM labour market. Using a unique dataset that extends across seven decades and comprises many hundreds of thousands of students, the paper finds that patterns of participation in most STEM subjects have varied little over the period considered; suggesting that efforts to increase the numbers of students studying science in school has had limited impact on the throughput of students who study STEM, including the pure sciences, at university level and, subsequently, on the number of graduates who would be available to undertake highly skilled work in areas for which degree-level skills are a pre-requisite

    The impact of reduced routine community mental healthcare on people from minority ethnic groups during the COVID-19 pandemic : qualitative study of stakeholder perspectives

    Background: Enduring ethnic inequalities exist in mental healthcare. The COVID-19 pandemic has widened these. Aims: To explore stakeholder perspectives on how the COVID-19 pandemic has increased ethnic inequalities in mental healthcare. Method: A qualitative interview study of four areas in England with 34 patients, 15 carers and 39 mental health professionals from National Health Service (NHS) and community organisations (July 2021 to July 2022). Framework analysis was used to develop a logic model of inter-relationships between pre-pandemic barriers and COVID-19 impacts. Results: Impacts were largely similar across sites, with some small variations (e.g. positive service impacts of higher ethnic diversity in area 2). Pre-pandemic barriers at individual level included mistrust and thus avoidance of services and at a service level included the dominance of a monocultural model, leading to poor communication, disengagement and alienation. During the pandemic remote service delivery, closure of community organisations and media scapegoating exacerbated existing barriers by worsening alienation and communication barriers, fuelling prejudice and division, and increasing mistrust in services. Some minority ethnic patients reported positive developments, experiencing empowerment through self-determination and creative activities. Conclusions: During the COVID-19 pandemic some patients showed resilience and developed adaptations that could be nurtured by services. However, there has been a reduction in the availability of group-specific NHS and third-sector services in the community, exacerbating pre-existing barriers. As these developments are likely to have long-term consequences for minority ethnic groups’ engagement with mental healthcare, they need to be addressed as a priority by the NHS and its partners

    Robust NAS benchmark under adversarial training : benchmark, theory, and beyond

    Recent developments in neural architecture search (NAS) emphasize the significance of considering robust architectures against malicious data. However, there is a notable absence of benchmark evaluations and theoretical guarantees for searching these robust architectures, especially when adversarial training is considered. In this work, we aim to address these two challenges, making twofold contributions. First, we release a comprehensive data set that encompasses both clean accuracy and robust accuracy for a vast array of adversarially trained networks from the NAS-Bench-201 search space on image datasets. Then, leveraging the neural tangent kernel (NTK) tool from deep learning theory, we establish a generalization theory for searching architecture in terms of clean accuracy and robust accuracy under multi-objective adversarial training. We firmly believe that our benchmark and theoretical insights will significantly benefit the NAS community through reliable reproducibility, efficient assessment, and theoretical foundation, particularly in the pursuit of robust architectures

    Improved prediction of wave overtopping rates at vertical seawalls with recurve retrofitting

    This study investigates the reduction in overtopping discharge along a vertical seawall through the implementation of a recurve retrofitting. A comprehensive set of physical modelling experiments were undertaken in a laboratory-scale wave flume at the University of Warwick, to investigate the wave overtopping processes under both swell and storm wave conditions. The tests measured overtopping discharges for impulsive and non-impulsive wave conditions. The effects of geometrical design of recurve retrofitting on overtopping reduction are examined by four configurations with varying overhang length and recurve hight. The study revealed that the reduction in overtopping is primarily determined by the length of the overhang in the recurve wall, while the influence of the recurve height is limited. A longer overhang length results in a more substantial decrease in overtopping discharges on the seawall crest. The results also highlight the role of incident wave steepness and the crest freeboard on the overtopping mitigation performance of the recurve walls. A new enhanced methodology is proposed to predict the wave overtopping from vertical seawalls with recurve retrofitting., considering the effects of freeboard and wave steepness. The findings of this study provide new important insight in the role of retrofitting as a robust intervention to improve the wave overtopping mitigation performance of seawalls. The predictive empirical formulae proposed by this study facilitate readily and accurate estimation of overtopping rates as a function of retrofitting geometrical design, allowing for wider application of retrofitting solutions
