Diagnosis of quarantine organisms at the JKI in the National Reference Laboratory for organisms harmful to plants
Dem JKI wurde im April 2019 durch das Bundesministerium für Ernährung und Landwirtschaft (BMEL) die Funktion des nationalen Referenzlaboratoriums (NRL) für Schadorganismen der Pflanzen zugewiesen. Mit dieser Funktion des NRL für Deutschland sind bestimmte Zuständigkeiten und Aufgaben verbunden, die in der EU-Verordnung 2017/625 (EU, 2017) geregelt sind. Dazu gehören auch Referenzuntersuchungen bzw. die Diagnose von Quarantäneschadorganismen (QSO). Das NRL stellt eine übergeordnete Einheit innerhalb des JKI dar. Durch insgesamt 14 Prüflabore der JKI-Institute für Pflanzenschutz in Ackerbau und Grünland (A), nationale und internationale Angelegenheiten der Pflanzengesundheit (AG), Epidemiologie und Pathogendiagnostik (EP), Pflanzenschutz in Gartenbau und Forst (GF), Pflanzenschutz in Obst- und Weinbau (OW) wird die Referenzfunktion bei der Diagnose zu verschiedensten (Quarantäne)-Schadorganismen der Pathogengruppen Bakterien, Insekten, Nematoden, Pilze (einschließlich Oomyceten), Phytoplasmen und Viren wahrgenommen.In April 2019, the JKI was officially designated as the National Reference Laboratory (NRL) for organisms harmful to plants by the Federal Ministry of Food and Agriculture (BMEL). This function as NRL for Germany is associated with certain responsibilities and tasks, which are specified in the EU Regulation 2017/625 (EU, 2017). This also includes reference tests and the diagnosis of quarantine pests, respectively. The NRL represents a superordinate unit inside JKI. A total of 14 test laboratories from different JKI institutes, namely for Plant Protection in Field Crops and Grassland (A), for National and International Plant Health (AG), for Epidemiology and Pathogen Diagnostics (EP), Plant Protection in Horticulture and Forests (GF), and for Plant Protection in Fruit Crops and Viticulture (OW) are in charge to carry out a reference function in the diagnosis of (quarantine) pests in the pathogen groups of bacteria, fungi (including oomycetes), insects, nematodes, phytoplasma und viruses
The Japanese beetle (Popillia japonica) – a pest with major phytosanitary risk potential for Germany and Europe
Das hohe Schadpotential eines Pflanzenschädlings rechtzeitig zu erkennen und dem Einschleppen vorzubeugen bzw. ein Ausbreiten zu verhindern, ist ein zentrales Anliegen der Pflanzengesundheit. Daher gelten für den Japankäfer (Popillia japonica) als Unionsquarantäneschädling besondere Regelungen und Maßnahmen. Der polyphage Käfer entwickelt sich in der Regel innerhalb eines Jahres und verursacht in nichtendemischen Gebieten gravierende wirtschaftliche Schäden. Eine Bestimmung der Käfer ist bis zur Art möglich. Erste Funde von Japankäfern in Deutschland konnten nicht bestätigt werden.The timely recognition of the high damage potential of a plant pest and the prevention of its introduction or spread is a central concern of plant health. For this reason, special regulations and measures apply to the Japanese beetle (Popillia japonica) as a union quarantine pest. The polyphagous beetle usually develops within one year and causes severe economic damage in non-endemic areas. A determination of the beetles is possible up to the species. First findings of Japanese beetles in Germany have not been confirmed
Nickel toxicity in Brassica rapa seedlings: Impact on sulfur metabolism and mineral nutrient content
Weltweit haben anthropogene Aktivitäten zu erhöhten Nickelgehalten im Boden (Ni2+) geführt, was sich negativ auf die Pflanzenproduktivität auswirken kann. Der physiologische Hintergrund der Ni2+ Phytotoxizität ist noch weitgehend unklar. Eine zehntägige Exposition von Brassica rapa Sämlingen mit 1, 2 und 5 μM NiCl2 führte zu stark erhöhten Ni Gehalten im Gewebe, einer verringerten Biomasseproduktion und zu Blattchlorosen bei Konzentrationen von ≥ 2 μM Ni2+. Bei einer Konzentration von 5 μM Ni2+ war kein Pflanzenwachstum mehr zu beobachten. Eine Ni Toxizität trat auf, wenn der Ni Gehalt im Sprosses 1,0 μmol g–1 Trockengewicht und der in der Wurzel 23 μmol g–1 Trockengewicht überschritt. Eine Ni2+ Exposition von 2 μM beeinflusste den Mineralstoffgehalt in Spross und Wurzel nur geringfügig. Daher beeinflusste eine Ni2+ Exposition die Gehalte an Schwefelmetaboliten in der Pflanze kaum. Bei ≥ 1 μM Ni2+ war der Gesamtschwefelgehalt der Wurzel nur geringfügig erniedrigt, was vollständig auf einen verminderten Sulfatgehalt zurückzuführen war. Darüber hinaus war der Gehalt an wasserlöslichen Nicht-Protein-Thiolen sowohl im Spross als auch in der Wurzel nur bei 5 μM Ni2+ erhöht. Aus diesen Ergebnissen geht hervor, dass der Schwefelstoffwechsel wahrscheinlich nicht direkt an den Ni2+ Toleranzmechanismen von B. rapa beteiligt ist.Throughout the world anthropogenic activity has resulted in enhanced soil nickel (Ni2+) levels, which may negatively affect plant productivity. The physiological background of Ni2+ phytotoxicity is still largely unclear. Ten-day exposures of Brassica rapa seedlings to 1, 2 and 5 μM NiCl2 resulted in strongly enhanced tissue Ni levels, a decreased biomass production and leaf chlorosis at ≥ 2 μM Ni2+. At 5 μM Ni2+ plant growth was completely halted. Ni toxicity occurred when the content of the shoot exceeded 1.0 μmol g–1 dry weight and that of the root, 23 μmol g–1 dry weight. Ni2+ exposure at ≤ 2 μM only slightly affected the mineral nutrient content of both shoot and root. Hence, Ni2+ exposure hardly affected the sulfur metabolite content of the plant. At ≥ 1 μM Ni2+ the total sulfur content of the root was only slightly lowered, which could fully be ascribed to a decreased sulfate content. Moreover, the water-soluble non-protein thiol content of both shoot and root was only enhanced at 5 μM Ni2+. From these results it was clear that sulfur metabolism was unlikely to be directly involved in the Ni2+ tolerance mechanisms of B. rapa

Directed Inoculum Production of arbuscular mycorrhizal fungi – the state of the art
Since the mutualistic nature of arbuscular mycorrhiza (AM) had been discovered, an enormous amount of experiments elucidated the huge potential of mycorrhizal technology. Nevertheless, the predict-ability of mycorrhizal effectiveness (i.e. the quantitative measure of a pronounced effect) in agro-ecosystems and plant production systems remained low. On this background, Directed Inoculum Production (DIP) had been developed based on quantitative genetics. During the inoculum production process the ecological niche of an arbuscular mycorrhizal fungus (AMF) is considered, the effective ecological niche widened and population engineering included in large-scale inoculum production. This is done in order to place effective mycor-rhiza products on the market. This article reviews briefly the histori-cal development and the idea of DIP and its performance in the last 25 years. Recent and current scientific explanations for the variabili-ty of mycorrhizal effectiveness and future demands of research are outlined
Ripening effect on the concentration of polyfunctional thiol precursors in \u27Gewürztraminer\u27
The effect of ripeness on the concentration of polyfunctional thiol precursors was investigated in \u27Gewürztraminer\u27 juices in two vintages characterised by very different climate conditions. An incremental trend of glutathionyl-3-mercaptohexan-1-ol and cysteinyl-3-mercaptohexan-1-ol during ripening was observed. The increase in the last phase of maturation was noticeable in both vintages. The concentration of 4-S-glutathionyl-4-methylpentan-2-one and 4-S-cysteinyl-4-methylpentan-2-one was confirmed to be low in \u27Gewürztraminer\u27, although the latter was found at quantifiable levels in a couple of samples. The management of the harvest date appears to be highly important in order to exploit the potential grapefruit-like note related to polyfunctional thiols of little aromatic GWT wines, particularly in case of challenging vintage years
The involvement of phenolic metabolism in superficial scald development in ‘Wujiuxiang’ pear
Superficial scald often occurs after a long term of cold storage in apples and pears. In this study, the superficial scald index, the contents of major phenolic compounds, polyphenol oxidase (PPO) activity and its related genes expression in peel was investigated during cold storage period and at shelf life in ‘Wujiuxiang’ pear (Pyrus communis L. cv. Wujiuxiang) with or without 1-MCP treatment. It showed that arbutin, chlorogenic acid, catechin and epi-catechin were the main phenolic compounds in the peel, and 1-MCP treatment significantly inhibited scald development while altering the composition of phenolic compounds, inhibited PPO activity and the expression of phenylalanine ammonia ligase (PAL1, PAL2), cinnamate 4-hydroxylase (C4H1, C4H2) and PPO (PPO1, PPO5) and up-regulated the expression of hydroxycinnamoyl-CoA shikimate/quinate hydroycinnamoyltransferase (HCT1), p-coumarate-3-hydro-xylase (C3H) and PPO (PPO4 and PPO6) in the peel. These results suggested that the phenolic metabolism is closely related to the scald development, and several genes related to phenolic metabolism were involved in scald development
Control of Umbelliferae in Grassland and Pasture, a new herbicide containing ArylexTM active
Anthriscus sylvestris ist eine mehr- oder zweijährige, krautige Pflanze im Dauergrünland. Mit seiner halben Rosette verdrängt es wertvolle Futtergräser und hat nur einen mittleren Futterwert. Gegen dieses und andere schwer bekämpfbare Unkräuter aus der Familie der Doldenblütler gibt es derzeit keine wirksamen und nachhaltigen Lösungen mit selektiv wirkenden Herbiziden.
ArylexTM active (Halauxifen-Methyl) ist ein neuer Wirkstoff aus der zu den synthetischen Auxinen gehörenden Untergruppe der Arylpicolinate. Herbizide mit diesem Wirkstoff werden in Deutschland seit 2018 zur Unkrautbekämpfung in Getreide vermarktet.
Für den Einsatz in Wiesen und Weiden wird GF-3635 derzeit von Corteva Agriscience in Deutschland und anderen europäischen Ländern entwickelt. Es ist eine Mischung aus Arylex, Fluroxypyr und Triclopyr und enthält bei einer maximalen Aufwandmenge von 1,5 l/ ha 10 g/ha Arylex, 300 g/ha Fluroxypyr und 300 g/ha Triclopyr. GF-3635 ist eine Weiterentwicklung des seit 2005 verfügbaren Produktes Starane Ranger®, bzw. Ranger® ab 2012 mit gleichen Mengen an Fluroxypyr und Triclopyr pro Hektar. Es kann sicher während der gesamten Vegetationsperiode angewendet werden, schädigt keine wertvollen Grasarten und hat keinen negativen Einfluss auf den Trockenmasseertrag oder die Futterqualität. GF-3635 bekämpft A. sylvestris, Heracleum sphondylium, Plantago lanceolata, Ranunculus acris, Ranunculus repens, Rumex crispus, Rumex obtusifolius, Taraxacum officinale und Urtica dioica.Anthriscus sylvestris is a perennial or biennial herbaceous plant in permanent grassland and pasture. With its half rosette it is able to displace valuable forage grasses and has only a medium feed value. At present, no effective and long-lasting solutions with selective acting herbicides is available against it and other hard-to-control weeds of the Umbelliferae family.
ArylexTM active (halauxifen-methyl) is a new active ingredient in the subgroup of aryl picolinates belonging to synthetic auxins. Herbicides containing this active ingredient have been marketed in Germany since 2018 for weed control in cereals.
For the use in grassland and pasture GF-3635 is now under development by Corteva Agriscience in Germany and other European countries. It is a premix of Arylex, fluroxypyr and triclopyr and contains at the 1.5 L/ha maximum use rate 10 g/ha Arylex, 300 g/ha Fluroxypyr and 300 g/ha triclopyr. GF-3635 is thus a further development of the Product Starane Ranger® available since 2005, Ranger® since 2012 respectively, with equal amounts of fluroxypyr and triclopyr per hectare. It can be safely applied during the whole vegetation period, does not harm valuable grass species and has no negative impact on yield quantity and Forage quality. GF-3635 controls A. sylvestris, Heracleum sphondylium, Plantago lanceolata, Ranunculus acris, Ranunculus repens, Rumex crispus, Rumex obtusifolius, Taraxacum officinale and Urtica dioica
Weeds in maize - variation in species trait assembly through soil cultivation and crop rotation
Mais ist die zweitwichtigste Kulturpflanze in Deutschland. Die genutzten Anbauverfahren im Mais variieren zunehmend, sodass neben der klassisch wendenden Bodenbearbeitung, vermehrt die nicht-wendende Bodenbearbeitung genutzt wird. Mais wird zudem als Teil einer Fruchtfrequenz oder in Selbstfolge angebaut. Die Folgen des veränderten Managements auf die Eigenschaften der Unkrautarten sind unklar. Es wird jedoch angenommen, dass ein entsprechendes Management mit bestimmten Unkrauteigenschaften assoziiert ist. Die wendende Bodenbearbeitung begünstigt scheinbar konkurrenzstarke Unkrauteigenschaften als die nicht-wendenden Bodenbearbeitung. Weiterhin wird vermutet, dass konkurrenzstarke Eigenschaften eher durch den Anbau von Mais in Selbstfolge als durch den Anbau in Rotation gefördert werden. Dies wurde mit Hilfe einer modernen RLQ-Analyse getestet. Als Datengrundlage für diese Analyse wurden Feld- und Unkrautdaten aus einem deutschlandweiten Unkrautmonitoring im Mais 2017 und 2018 sowie ein Datensatz zu Unkrauteigenschaften (Raunkiear Lebensformen, Lebenszyklus, Leaf-Height-Seed Strategien, Phenology, Klassische Verwandschaftsgruppen der Unkräuter sowie deren Neigung Herbizidresistenzen zu bilden) aus anderen Studien zusammengestellt. Die Ergebnisse zeigen, das unabhängig vom Management im Mais, konkurrenzstarke Unkrauteigenschaften, wie Therophyten, Arten mit hoher Affinität zu nährstoffreichen Böden, große Blattflächen, hohe Pflanzenhöhe und ein schweres Samengewicht vorkommen können. Managementfaktoren können jedoch die Gruppierung bestimmter Unkrauteigenschaften fördern. So werden, entgegen der Annahme, konkurrenzstarke Unkrauteigenschaften eher der nicht-wendenden als der wendenden Bodenbearbeitung zugeordnet. Wie erwartet, kann der Anbau von Mais in Selbstfolge eine Anpassung der Unkrautarten an die Bedingungen im Mais fördern.Maize is the second most important weed in Germany. Maize cultivation varies between conventional tillage and reduced tillage and cultivation of maize in crop sequences or continuous maize. The effect of the management on the weed traits is unknown. Assoziation of weed traits to the management, however, are likely. We expect that conventional tillage is more associated to competitive weed traits than to reduced tillage. Furthermore, cultivation of continuous maize in contrast to crop rotation leads to a specialization of weeds to maize growing conditions. We tested these expectations by using the modern RLQ-analysis. Field and weed data were obtained from a germanwide weed monitoring in maize in 2017 and 2018 and trait data (life form, life history, L-H-S strategy, phenology, division and weeds possibility to develop herbicide resistance) from various datasets. The results showed that traits linked with competitive weed species, such as Therophytes, weed species with a high affinity to soil nutrients, large leaf area, plant height and seed weight were independent from the tested management factors. Management factors, however, can enhance the selection of weed traits. Thus, competitive traits are more associated to reduced than to conventional tillage. Conservation tillage systems in sugarbeet without use of glyphosate
Mit drohenden Indikationseinschränkungen von Glyphosat im Ackerbau und dem eventuellen mittelfristigen Wegfall des Wirkstoffes stellt sich die Frage, wie und ob Mulchsaatsysteme von Sommerungen aufrechterhalten werden können. Am Beispiel von Zuckerrüben werden Ergebnisse aus drei Feldversuchsjahren präsentiert, in denen die Wechselwirkungen von Zwischenfrüchten und Bodenbearbeitungssystemen auf die Unkrautpopulationen und die Zuckerrübenetablierung aufgezeigt werden. Es wurden je nach Standort 12 bis 35 Systeme in 2 bzw. 4 Wiederholungen geprüft.
Die Ergebnisse fallen sehr unterschiedlich aus. Je nach Jahreswitterung, Bodenbearbeitungssystem, Zwischenfrucht und Herbizideinsatz kann eine Zuckerrübenmulchsaat ohne Glyphosateinsatz sehr gut funktionieren aber auch scheitern. In vielen Fällen bedeutet ein Glyphosatverzicht einen deutlich höheren Aufwand an Bodenbearbeitung und teilweise auch einen zusätzlichen Einsatz von Herbiziden.With the threat of restrictions on glyphosate use and the possible omission of the active substance in the medium term, the question arises as to whether and how soil conservation tillage systems of annual spring crops can be maintained. Using sugar beet, results from three years of field trials are presented in which the interactions between catch crops and soil tillage systems on weed populations and sugar beet establishment are demonstrated. Depending on the location, 12 to 35 systems were tested in 2 or 4 replications. The results are very different. Depending on the weather conditions, tillage system, catch crop and herbicide use, sugar beet seed without glyphosate can work very well, but it can also fail. In many cases, a waiver of glyphosate means a significantly higher effort for tillage and, in some cases, also an increase in the use of herbicides
