    A pathogen complex between the root knot nematode Meloidogyne incognita and Fusarium verticillioides results in extreme mortality of the inka nut (Plukenetia volubilis)

    The combined infection of the root knot nematode Meloidogyne incognita and two Fusarium species led to high plant mortality of inka nut (Plukenetia volubilis) seedlings in pots after 80 days growth in two independent inoculation experiments. Inoculation of M. incognita juveniles and conidia of F. solani or F. verticillioides, simultaneously, increased plant mortality by 25-30%, compared with M. incognita alone (5-10% mortality). When inoculated with F. verticillioides at 20 days post nematode inoculation, plant mortality increased to 55%. Either of the Fusarium spp. alone caused some plant mortality, but less than when combined with M. incognita. The synergistic interaction of M. incognita and especially F. verticillioides demonstrates a lethal outcome for inka nuts. It appears that M. incognita creates favorable conditions that lead to enhanced effect and damage by the Fusarium species, especially F. verticillioides, leading to devastating levels of plant death

    Correlation analysis of chlorogenic acid and luteoloside biosyntheses with transcription levels of HQTs and FNSs in Lonicera species

    Organic acids and flavonoids are the main active components in Lonicera species. Chlorogenic acid and luteoloside are important components, and their synthesis is regulated in plants by the phenyl-propanoid synthesis pathway. Downstream of the phenylpropanoid synthesis pathway, hydroxycinnamoyl CoA quinate hydroxycinnamoyl transferase (HQT) and flavone synthase (FNS) are critical enzymes that are involved in chlorogenic acid and luteoloside biosynthesis, respectively.In this study, we first determined the dynamic accumulations of chlorogenic acid, luteoloside and other active components in different growth stages of the flower buds of Lonicera fulvotomentosa through HPLC-DAD and then investigated the expressions of the LJHQT and LJFNS gene families by q-RT-PCR. In addition, we also compared the expression levels of HQT and FNS orthologous genes in vari-ous tissues of Lonicera japonica, L. fulvotomentosa, and Lonicera hypoglauca.The results indicated that the chlorogenic acid contents exhibit leaf accumulation that is preferential in L. fulvotomentosa but exhibit bud accumulation that is preferential in L. japonica and L. hypoglauca. The luteoloside contents show preferential leaf accumulation in these three species. Our results suggest that the leaves and buds of these three species are rich in medicinal ingredients, including chlorogenic acid (CGA) and luteoloside, and therefore can be used as a material to extract CGA and luteoloside rather than being wasted. Furthermore, combined with the transcript expression levels of HQTs and FNSs, we explained the species-specific and tissue-specific occurrence of CGA and luteoloside. We analyzed dynamic changes of components and gene expression and demonstrated that the expressions of HQTs and FNSs in these three species are closely related to the synthesis of chlorogenic acid and luteoloside

    Site-specific weed control as a component of smart farming

    Unkräuter treten auf Ackerflächen selten gleichmäßig verteilt auf. Heterogene Unkrautdichten eröffnen der Landwirtschaft die Möglichkeit, Herbizidmaßnahmen teilflächenspezifisch durchzuführen. Hieraus ergibt sich ein erhebliches Einsparpotential für den Einsatz von Pflanzenschutzmitteln. Moderne Technologien wie Direkt­einspeisungssysteme und verschiedene Verfahren der automatischen Unkrauterkennung und -kartierung sind die technischen Grundlagen für dieses Verfahren. Es zeigt sich, dass die fortlaufende Forschung und Entwicklung seit den 1980er Jahren das Verfahren der teilflächenspezifischen Unkrautbekämpfung weit vorangebracht hat und dadurch hohe Pflanzenschutzmitteleinsparungen rea­lisiert werden können. Dennoch bleibt die Nutzung des Verfahrens in der landwirtschaftlichen Praxis hinter den technischen Möglichkeiten zurück, obwohl automatische Unkrautdetektion und der Einsatz unbemannter Flugsysteme die Erstellung von Unkrautverteilungskarten stark vereinfacht haben. Zukünftig kann es jedoch dazu beitragen, Pflanzenschutzmittel einzusparen und Biodiversität auf Ackerflächen zu fördern, wenn sich die Akzeptanz in der landwirtschaftlichen Praxis weiter erhöht.Weeds are rarely evenly distributed on arable land. Hete­rogeneous weed densities and distributions open up the possibility for farmers to apply herbicides on a site-specific basis. This results in considerable saving potential for the consumption of plant protection products. Modern technologies such as direct injection systems and various weed detection and mapping methods are the technical basis for this procedure. It can be seen that ongoing develop­ment and research since the 1980 s has advanced the process a long way. In the future, site-specific weed control can help to save pesticides and create biodiversity on arable land

    Assessment of sensory profile and instrumental analyzed attributes influenced by different potassium fertilization levels in three tomato cultivars

    Sensory properties are an essential quality aspect when the consumption of fresh tomato is under consideration. The flavor of tomato is defined as a combination of taste sensations (sweetness, sourness), aroma (volatile compounds), and texture (firmness, mealiness), some of which are proven to be affected by insufficient nutrient supply − especially potassium (K). This study intends to undertake a holistic assessment of the K fertilization effect on the flavor of tomato by connecting the use of sensorial and instrumental methods. An optimal K supply significantly increased the sensory descriptors sweetness, sourness, and aroma as well as the instrumental estimated color, firmness, total soluble solids (TSS), titratable acids (TA), and dry matter (DM) in a cultivar-specific manner. The volatile organic compounds (VOCs) were not significantly affected by K fertilization. The evaluation by the panelists confirmed the results of the instru-mental analyses, by which an increment in the fruit quality with rising K supply could be detected. An optimal K supply of 3.66 g/plant could be suggested to increase tomato flavor in the cocktail cultivars studied: Primavera and Yellow Submarine. Cultivar effects should, therefore, be considered for defining the optimal K fertilizer dose that favors high tomato fruit quality and, hence, better flavor

    Weed detection and mapping for automatic application map generation in crop protection

    Für eine künftig umweltverträgliche und teilflächen­spezifische Anwendung von Herbiziden ist eine genaue Kenntnis über die Zusammensetzung und die räumliche Verteilung von Nutzpflanzen und Unkräutern auf Ackerflächen erforderlich. Sind diese Informationen bekannt, können sie in Unkrautverteilungskarten abgebildet werden und dienen dann als Basis für die Erstellung von Appli­kationskarten. Eine bislang fehlende Prozesskette beginnend beim Pflanzenmonitoring auf dem Acker, über Methoden zur automatischen Identifizierung von Einzelpflanzen und der Generierung von Verteilungskarten wurden erstmals durchlaufen. Hierzu wurden im Projekt „Assistenzsystem zur teilflächenspezifischen Applikation von Pflanzenschutzmitteln (AssSys)“ manuelle Kamera-gestützte Feld- und Halbfreilandbonituren typischer Verunkrautungssituationen und Feldbefliegungen mit einem Multicopter genutzt, um die Position und die Bestandsdichten von Unkräutern und Kulturpflanzen zu bestimmen und zu kartieren. Die Segmentierung aller Aufnahmen erfolgte manuell mit einer eigenständig entwickelten Software zur Annotation von Bilddaten und nach dem Training von umfangreichen Datensätzen auto­matisch mit Methoden des maschinellen (Bag-of-visual Words) und tiefen Lernens (Convolutional Neural Networks). Es zeigte sich, dass die getesteten Algorithmen für eine Vorhersage von ein- und zweikeimblättrigen Pflanzenarten geeignet sind. Die aus den Feldbonituren gewonnenen Daten wurden in Applikationskarten überführt und von den Partnern des Projektes genutzt, um in praxisnahen Feldversuchen teilflächenspezifische, selektive Unkrautbekämpfungsmaßnahmen mit einem Direkteinspeisungssystem durchzuführen.For environmental and site-specific application of herbicides in the coming future, precise knowledge of the structure and spatial distribution of crops and weeds on arable land is required. If this information is known, it can be mapped in weed distribution maps and then serve as a basis for the generation of application maps. A process chain that has been missing so far, starting with plant monitoring in the field, through methods for the automatic identification of individual plants and the generation of distribution maps, was run through for the first time in a first approach as part of the work package “Weed Identification and Mapping”. For this purpose, the AssSys project used manual camera-based field and semi-field sampling of typical weed situations and automatic field aerial photographs with a multicopter to deter­mine and map the position and population densities of weeds and crop plants. All images were segmented manually using an own developed software for annotation of image data and after training of large datasets comparatively using methods of machine learning (Bag-of-visual Words) and deep learning (Convolutional Neural Networks). It was shown that the tested algorithms are suitable for predicting mono- and dicotyledonous plant species. The data obtained from the field sampling were converted into application maps and used by the project partners as part of their work packages to carry out site-specific, selective weed control treatments with a direct-injection system in practical field trials

    Methodology for site-specific determination of slope inclination using digital height information in the Plant Protection Application Manager (PAM)

    In der Landwirtschaft werden viele Pflanzenschutzmittel im Zuge des Zulassungsverfahrens mit Hangneigungsauflagen belegt. Ziel ist es, den durch Oberflächenabfluss und Bodenerosion bedingten Eintrag von Pflanzenschutzmitteln in angrenzende Gewässer zu vermeiden. Aktuell existieren keine praktikablen, deutschlandweit verfügbaren Instrumente, die den Landwirt in einer zuverlässigen und nachvollziehbaren Weise bei der objektiven Ermittlung der Hangneigung unterstützten. Mit dem entwickelten PAM3D-Dienst wird dem Nutzer ein internetbasiertes Entscheidungshilfesystem zur Einhaltung von Hangneigungsauflagen zur Verfügung stehen. Gleichzeit wird diese Methode für die zuständigen Kon­troll­behörden der Länder zur Diskussion gestellt. Die Ermittlung der Hangneigung eines Feldes erfolgt automatisiert auf Grundlage von digitalen Höheninformationen mittels eines neuartigen Algorithmus, der Randpixelmethode. Im Gegensatz zu anderen Services gibt diese nicht nur einen Durchschnittswert pro Feld aus, sondern erlaubt eine teilflächenspezifische Beurteilung der Hangneigung. Die Methode ist gegenüber gängigen rasterbasierten Verfahren flexibel genug, Höheninformation aus verschiedenen Quellen (Laserscan-, Traktor,- Drohnen- und Satellitendaten) zu verarbeiten und ist zudem robust gegen­über unterschiedlichen räumlichen Auflösungen dieser Datensätze.In agriculture, many plant protection products (PPP) are subject to slope restrictions as part of their approval process. The aim is to prevent the discharge of pesticides into adjacent water bodies caused by surface runoff and soil erosion. There are currently no practicable instruments available throughout Germany, supporting farmers to deter­mine the slope inclination in a reliable and verifiable manner. The PAM3D-service will provide an internet-based decision support system for complying with slope requirements in the context of PPP risk management. The slope inclination of a field is determined automatically based on digital elevation information using a novel algorithm. Contrary to other services providing an unreliable average field value, this new methodology allows for a site-specific slope assessment. The service supports various sources of elevation information including official laser, tractor, satellite, and drone data, and it is robust to the spatial resolution of input elevation data
