    Phosphine against arthropods in stored product protection with special consideration of the resistance problem worldwide and in Germany: A review article

    Die Begasung mit Phosphorwasserstoff (PH3) ist eine der wirksamsten Methoden zur Bekämpfung von schädlichen Insekten und Milben im Vorratsschutz. Die übermäßige und fehlerhafte Anwendung dieses Wirkstoffes hat jedoch zur Resistenzentwicklung bei vorratsschädlichen Insekten in vielen Ländern geführt, was die Bekämpfung eines Befalls deutlich erschwert und im ungünstigsten Fall zum Totalverlust der Vorratsgüter pflanzlicher Herkunft führen kann. Falls die Begasungsmaßnahme nicht hinreichend wirkt, wird normalerweise weiter bekämpft, ehe die Ware aufgegeben wird. Die Strategien zur Vermeidung von Resistenzentwicklung gegen PH3 und das Management vorhandener PH3-Resistenzen sind in vielen Ländern bereits ein Teil der guten Praxisanwendung bei Begasungstätigkeiten. In Deutschland sind derzeit keine resistenten vorratsschädlichen Insekten gegen PH3 bekannt. Damit das Risiko der Resistenzentwicklung und Ausbreitung bei den vorratsschädlichen Insekten in Deutschland weiterhin gering bleibt, müssen die Anwendungsregeln und -vorschriften bei der Begasung und Lagerung pflanzlicher Vorratsgüter eingehalten werden. Im vorliegenden Übersichtsartikel werden die weltweite aktuelle Situation und die möglichen Faktoren für eine Resistenzentwicklung gegen PH3 und deren Vermeidung in Deutschland diskutiert.Fumigation with phosphine (PH3) is one of the most effective ways of control insect and mite pests in stored agricultural products. However, the excessive and incorrect use of this active ingredient has led to the development of resistant insects towards phosphine fumigation, which makes it much more difficult to control an infestation and, in the worst case, can lead to the total loss of the stored products. In case of an insufficiency of the fumigation, the treatment usually continues until the controlling of entire insect pests in the stored commodities. The strategies to avoid development of resistance to phosphine are already part of the good practice in fumigation activities in many countries. In Germany, no insects are currently known that are resistant to phosphine in stored products. To ensure that the risk of development and spread of resistance among insects remains low in Germany, the application rules and regulations for fumigation and storage of agricultural products must be fulfilled. In this review article, the current world-wide situation and the possible factors for a development of resistance to phosphine and its prevention in Germany are discussed

    Molecular and ampelographic characterization of genotypes used for Pajarete wine, an old Denomination of Origin from Huasco and Elqui Valleys in northern Chile

    The characterization of 30 grape accessions used to produce Pajarete wine is presented, one of the first American "Denomination of Origin" (D.O.). Accessions were collected from small vineyards from the Huasco Valley in the Atacama Region, northern Chile, which are very heterogeneous for varietal assortment, as was demonstrated by microsatellite marker analysis. Eighteen different genotypes were identified, including the two varieties considered the founders of American viticulture, \u27Listán Prieto\u27 (syn. \u27País\u27) and \u27Muscat of Alexandria\u27. A series of criolla varieties was also identified, some of them already described plus others not yet included in international databases. Based on nine SSR markers, allelic matchings suggest that four of these new criolla varieties (NN-311, NN-313, NN-368 and NN-370) derives from \u27Listán Prieto\u27 x \u27Muscat of Alexandria\u27, but three (NN-314, NN-276 and NN-369) are derived from \u27M. of Alexandria\u27 crossed with a yet undetermined parent. Seven criolla accessions were described by ampelography, including some not yet recorded in the European Vitis International Variety Catalogue (VIVC) or in the Instituto de Investigaciones Agropecuarias (INIA) database. Three accessions were considered as not appropriate for the production of Pajarete wine since two of them are table grape varieties and the other is the American variety \u27Isabella\u27 (Vitis labrusca x V. vinifera), all considered of poor quality for winemaking. All these findings are very useful to direct the conservation of this unique germplasm, as well as for small producers of Pajarete wine, since based on this information on the overall variety assortment of the area, they can now make informed decisions to improve their vineyard management and wine production

    Biological control of wireworms (Agriotes spp.) in potato cultivation using the entomopathogenic fungus Metarhizium brunneum: Factors that influence the effectiveness of mycoinsecticide formulations

    Die landwirtschaftliche Produktion von Ackerbaukulturen ist eine der wichtigsten Aufgaben, um die wachsende Weltbevölkerung zu ernähren. Da die für den Ackerbau geeigneten Flächen begrenzt sind, spielt der Pflanzenschutz durch die Bekämpfung von Unkräutern, mikrobiellen Krankheitserregern, Viren und tierischen Schädlingen eine Schlüsselrolle bei der effizienten Nutzung der vorhandenen Felder. Unter den tierischen Schädlingen verursachen Insekten bei weitem die größten wirtschaftlichen Verluste, indem sie durch den Befall ober- oder unterirdischer Pflanzenteile den Ertrag oder die Ertragsqualität mindern. Ein Beispiel für bodenbewohnende Schadinsekten sind Drahtwürmer, die Larven von Schnellkäfern (Coleoptera: Elateridae), die weltweit als wichtige Schädlinge im Kartoffelanbau gelten. In Deutschland stellen Drahtwürmer der Gattung Agriotes, mit den am weitesten verbreiteten Arten A. obscurus, A. lineatus und A. sputator, eine ernsthafte Bedrohung für Landwirte dar, die Speisekartoffeln erzeugen. Da die Europäische Kommission wirksame Agrochemikalien für den Pflanzenschutz aufgrund ihrer schädlichen Auswirkungen auf die Umwelt verboten hat, ist die Drahtwurmbekämpfung im Kartoffelanbau nur begrenzt möglich. Auf der Suche nach nachhaltigen Alternativen im Rahmen des integrierten Pflanzenschutzes ist die biologische Bekämpfung von Drahtwürmern mit entomopathogenen Pilzen in der aktuellen Forschung sehr populär. Obwohl die Pathogenität von Metarhizium brunneum-Isolaten gegen Drahtwürmer der Gattung Agriotes bestätigt und verschiedene Mykoinsektizid-Formulierungen entwickelt wurden, ist die Wirksamkeit im Feldeinsatz sehr unterschiedlich und die genauen Ursachen sind kaum bekannt. In der vorliegenden Dissertation wurde das Drahtwurmbekämpfungspotenzial von zwei neuen Formulierungen auf der Grundlage des entomopathogenen Pilzes M. brunneum eingehender untersucht. Ein Granulat (AgriMet-Granulat) und ein lösbares Pulver (AgriMet-Trockenprodukt) wurden für die Ausbringung in den Boden während der Kartoffelpflanzung getestet, um die jeweilige Drahtwurmpopulation als Teil einer inokulativen Biokontrollstrategie zu reduzieren. Vor der Feldtestung zeigten Laborversuche mit dem AgriMet-Granulat, dass der erforderliche Kontakt zwischen den infektiösen Konidien und den Larven wahrscheinlich zustande kommt, da mit Hilfe eines Y-Olfactometer eine anziehende Wirkung der Formulierung auf A. obscurus, A. sputator und A. lineatus beobachtet wurde. Neben der olfaktorischen Präferenz für das AgriMet-Granulat akzeptierten die Larven den Träger als Nahrungsquelle, was das Bekämpfungspotenzial der Granulat-Formulierung unterstrich. Das AgriMet-Trockenprodukt enthielt keine zusätzlichen Lockstoffe und sollte seine Wirkung über die gleichmäßige Verteilung in die Furche des Kartoffeldaches mit der Spritzung der Kartoffelbeize entfalten. Die Anwendung der AgriMet-Formulierungen während der Kartoffelpflanzung wurde jedoch durch die Kompatibilität des formulierten M. brunneum-Isolats mit nur einem Wirkstoff der im Kartoffelanbau üblichen Fungizid-Saatgutbeizmittel eingeschränkt. Ein weiteres Defizit in Bezug auf die Formulierungseigenschaften der getesteten Mykoinsektizide war ein Verlust der Lebensfähigkeit der Pilze nach der Lagerung oder dem Versand, der bei Qualitätskontrolltests festgestellt wurde. Trotz dieser technischen Hindernisse wurde die Wirksamkeit des AgriMet-Granulats und des AgriMet-Trockenprodukt in einem mehrjährigen Feldversuch (2018-2020) getestet, und es wurde eine signifikante Reduzierung der Drahtwurmknollenschäden beobachtet. Die Wirksamkeit war jedoch relativ gering und variierte zwischen den Versuchsjahren. Als Teil der angestrebten inokulativen Biokontrollstrategie war eine Pilzvermehrung am Zielort erforderlich, um das letale Potenzial für eine angemessene Wirksamkeit aufzubauen. Die Pilzvermehrung wurde durch die Bestimmung der koloniebildenden Einheiten von Metarhizium spp. nach der Anwendung der AgriMet-Formulierungen im Kartoffeldamm untersucht, jedoch nicht im ausreichendne Maß beobachte. Ein wichtiger Faktor, der die Pilzvermehrung und damit die Wirksamkeit beeinflusste, war die niedrige Bodentemperatur (< 17 °C) während der Ausbringung im Frühjahr. Die unzureichende Rhizosphären-Kompetenz von M. brunneum bei niedrigen Bodentemperaturen stellt eine große Herausforderung für den beabsichtigten Einsatz der Mykoinsektizide bei der Kartoffelpflanzung im April oder Mai dar. Zusätzlich zu diesem ökologischen Problem wurde bei der Prüfung der geeigneten Aufwandmengen der Formulierungen in standardisierten Gewächshaus- und Labor-Bioassays ein intrinsischer Faktor identifiziert, der die Wirksamkeit beeinflusst. Neben den hohen Aufwandmengen, die zum Schutz der Kartoffelknollen erforderlich sind, wurde festgestellt, dass das verwendete M. brunneum-Isolat Unterschiede in der Virulenz gegenüber A. obscurus, A. sputator und A. lineatus aufwies, wobei A. obscurus am empfindlichsten war. In Anbetracht gemischter Drahtwurmpopulationen auf landwirtschaftlich genutzten Flächen scheint die Anwendung von Mykoinsektiziden auf der Grundlage eines einzigen M. brunneum-Isolats mit Unterschieden in der Virulenz gegen die wichtigsten Drahtwurmarten ungeeignet zu sein. Daher wurde die kombinierte Behandlung von zwei Metarhizium spp.-Isolaten, die jeweils ein spezifisches Virulenzprofil aufwiesen, getestet, um die Wirksamkeit im Feldeinsatz zu erhöhen, indem mehrere Drahtwurmarten in gleichem Maße abgetötet werden. Obwohl eine additive Wirksamkeit gegen A. obscurus und A. sputator mit nahezu gleichen Überlebenswahrscheinlichkeiten nach der kombinierten Behandlung beobachtet wurde, stellen Unterschiede in der Vermehrungsleistung der getesteten Isolate eine Herausforderung für die nachfolgende Formulierung dar. Die identifizierten technischen, ökologischen und intrinsischen Faktoren zeigen die Komplexität des Einsatzes des entomopathogenen Pilzes M. brunneum gegen Drahtwürmer im Kartoffelanbau und müssen bei der künftigen Entwicklung marktfähiger Mykoinsektizide berücksichtigt werden. Der kommerzielle Erfolg von Mykoinsektiziden hängt von der Schaffung eines letalen Potenzials am Zielort gegen die wichtigsten Drahtwurmarten ab, weshalb die Anwendung in das ganzheitliche agronomische System eingebettet sein sollte, um geeignete Bedingungen für die pilzliche Wirkungsweise und damit eine effektive Drahtwurmbekämpfung zu schaffen.Agricultural production of field crops is one of the most important tasks to feed the growing human population. Since suitable cultivation areas are limited, crop protection plays a key role in the efficient use of existing fields by controlling weeds, microbial pathogens, viruses and animal pests. Among animal pests, insects cause by far the greatest economic losses by crop damage through infestation of above- or belowground plant parts. An example of soil-dwelling pest insects are wireworms, the larvae of click beetles (Coleoptera: Elateridae), which are considered important pests in potato cultivation worldwide. In Germany, wireworms of genus Agriotes, with the most widespread species A. obscurus, A. lineatus and A. sputator, pose a serious threat to farmers producing fresh potatoes. Since the European Commission has banned effective agrochemicals for plant protection due to their harmful effects on the environment, wireworm control in potato cultivation is limited. In search for sustainable alternatives in the integrated pest management, biological control of wireworms using entomopathogenic fungi seems promising. However, although pathogenicity of Metarhizium brunneum isolates against wireworms of the genus Agriotes has been confirmed and various mycoinsecticide formulations have been developed, the effectiveness in field use varied considerably and the exact causes are poorly understood. In the present dissertation, the wireworm control potential of two new formulations based on the entomopathogenic fungus M. brunneum was investigated in more detail. A granule (AgriMet-Granule) and a wettable powder (AgriMet-Dry Product) were tested for soil application during potato planting to reduce the respective wireworm population as part of an inoculative biocontrol strategy. Prior to the field trials, laboratory assays with the AgriMet-Granule showed that the required contact between infectious conidia and the larvae was likely to occur, since an attracting effect of the formulation on A. obscurus, A. sputator and A. lineatus was observed using a Y-Olfactometer. In addition to the olfactory preference to the AgriMet-Granule, the larvae accepted the carrier as food source, which emphasised the control potential of the granule formulation. The AgriMet-Dry Product did not contain any additional attractants and was expected to be effective by being evenly sprayed in the furrow of the potato ridge with the potato seed dressing. However, application of the AgriMet formulations during potato planting was limited by the compatibility of the formulated M. brunneum isolate with only one active ingredient of the fungicide seed dressings commonly used in potato cultivation. Another shortcoming of the formulation properties of the mycoinsecticides tested was the loss of fungal viability after storage and shipment, which was observed in quality control tests. Despite these technical obstacles, the effectiveness of the AgriMet-Granule and the AgriMet-Dry Product was tested in a multi-year field trial (2018-2020) and a significant reduction of wireworm tuber damage was observed. However, the effectiveness was relatively low and varied between years. Adequate effectiveness of the desired inoculative biocontrol strategy required fungal proliferation at the target site, but this was not observed by determining the colony-forming units of Metarhizium spp. after application of the AgriMet formulations in the potato ridge. An important factor affecting fungal proliferation was the low soil temperature (< 17 °C) during application in spring. The insufficient rhizosphere competence of M. brunneum at low soil temperatures poses great challenges to the intended use of the mycoinsecticides at potato planting in April or May. In addition to this ecological problem, an intrinsic factor influencing the effectiveness was identified when testing the appropriate application rates of the formulations in standardised greenhouse and laboratory bioassays. Besides the high application rates required to protect potato tubers, it was found that the used M. brunneum isolate exhibited differences in virulence against A. obscurus, A. sputator and A. lineatus, with A. obscurus being the most sensitive. Considering mixed wireworm populations on a given infested area, the application of mycoinsecticides based on a single M. brunneum isolate with differences in virulence against the most important wireworm species seems unsuitable. Therefore, the combined treatment of two Metarhizium spp. isolates, each with a specific virulence profile, were tested to enhance the effectiveness in field use by targeting multiple wireworm species to the same extent. Although additive effectiveness against A. obscurus and A. sputator was observed with almost equal survival probabilities after the combined treatment, differences in the proliferation performance of the tested isolates pose challenges for subsequent formulations. The identified technical, ecological and intrinsic factors demonstrate the complexity of using the entomopathogenic fungus M. brunneum against wireworms in potato cultivation and must be considered in the future development of marketable mycoinsecticides. The commercial success of mycoinsecticides depends on the creation of a lethal potential at the target site against the most important wireworm species, which is why the application should be embedded in the holistic agronomic system to create suitable conditions for the fungal mode of action and thus effective wireworm control

    The positive response in grape secondary metabolites under controlled stresses: a review

    Grapevine is cultivated worldwide with great economic importance. In recent years, our knowledge of the physiological and molecular basis of berry quality regulation has substantially increased. Abiotic and biotic stresses, such as deficit irrigation, low temperature, light/UV and microbes, to a certain extent, could improve grape berry quality by enhancing flavor metabolites, colorization or aroma compounds. This review summarizes recent data related to the stress of grape berry development, with special emphasis on secondary metabolism and its response to stresses. A full understanding of how grape berry metabolism responds to different stresses is important to improve the biochemical qualities of grapes and resultant products, such as wine in practice

    A possibility of influence factors on winter physical damage of grapevines in a snowy vineyard

    Hokkaido in northern Japan is a typical subpolar region and wine production using traditional cultivars can be done recently. However, there is an issue that the grapevines have short life spans in snowy central Hokkaido. This study investigates the effect of snow itself and its interaction with vineyard slope and grapevine stiffness on physical stress. In comparison of the flatness of the vineyard, the distortion of the basal trunk in the slope zone was greater than in the flat zones in the snowy mid-season. In comparison between cultivars, the distortion of the basal trunk in the flexible \u27Kerner\u27 was greater than in rigid \u27Gewürztraminer\u27, even in the flat zones. The magnitude of the distortion coincides with the mortality of grapevines. Although the changes in distortion showed a transition in just two winter durations, these results suggested a possibility that the snow cover might be linked to one of the physical damages of grapevines in vineyards in heavy snowy regions

    Enhancement of biomass production, salinity tolerance and nutraceutical content of spinach (Spinacia oleracea L.) with the cuticular wax constituent triacontanol

    The present study investigates the effect of foliar application of triacontanol (TRIA) on various physiological parameters with regard to crop yield and quality attributes of spinach (Spinacia oleracea L.) under normal growth conditions and salinity stress. Plantlets were grown for 21 days in perlite-containing pots supplemented with Hoagland’s nutrient solution, then they were subjected to 0 (control) or 150 mM NaCl. Two concentrations of TRIA (25 nM and 1 μM) were applied during seed germination and as foliar spray treatment, in itself or simultaneously with salt stress at 4-day intervals for three weeks. Exogenous application of TRIA enhanced germination energy and capacity, as well as shoot and root biomass of young spinach plants. Inhibition of net CO2 assimilation (Pn) and of potential quantum yield of photosystem II (Fv/Fm), caused by salt stress, was significantly reduced by treatment with triacontanol. Increment of carotenoid pigment and ascorbate (vitamin C) content, as well as reduction of membrane lipid peroxidation due to triacontanol treatment improves the health-promoting quality of spinach leaves developed under high salinity conditions. The presented results offer a novel solution for optimization of spinach cultivation on soils affected by high salinity, as well as for an increased content of health-promoting metabolites of spinach leaves upon human consumption

    How harvest, cleaning and conservation good practices affect the quality of saffron: results of a research conducted in Italy

    Saffron (Crocus sativus L.) spice making requires time-spending manual operations: stigmas are separated from the flower picked in bud early in the morning, and once dried they are preserved protected from light. This study verified how the correct pursuing of these good practices affects saffron quality. Few hours of exposure of the flower to the sun determined a significant decrease in the colouring strength (239.66 ± 10.33 versus 255.35 ± 11.87). The correct cleaning of stigmas determined a very significant increase of colouring strength (247.12 ± 13.32 instead of 224.35 ± 14.88) and a significant increase of flavour strength (99.72 ±7.48 against 90.31 ± 6.32, p <0.05). In 24 months, all the samples kept in the dark were still of first category of quality while the ones kept in the light dropped in second category. For all samples there was an increase of aroma strength and a decrease of the flavour and colouring strength, but only the colouring strength loss followed a significantly more sloping trendline. A detectable difference in the content of trans-crocin 4 correlated to the ageing or the way of conservation was not found, nevertheless, it was confirmed that the isomers tend towards a photostationary state where the trans isomer is more present

    Toxicity and neurophysiological impacts of three plant-derived essential oils against the vineyard mealybug Planococcus ficus

    Many natural products are able to control pests and can be used as alternatives for chemical treatments. Plant essential oils (EOs) have been found to exhibit some biological activity against many insects including mealybugs. This study aimed at studying the insecticidal activity and behavioral and neurophysiological impacts of three plant essential oils against the vine mealybug Planococcus ficus. The topical and fumigant toxicity of Cymbopogon citratus, Mentha piperita, and Pelargonium graveolens essential oils was evaluated against P. ficus adults. The chemical composition analysis of EOs by gas chromatographic-mass spectrometry (GC-MS) revealed citronellal (31.69 %), menthol (73.78 %), and geraniol (39.6%), as major components, respectively. Bioassays of EOs against vine mealybug adults through fumigation toxicity method revealed lethal concentrations LC50 values of 17.01, 26.27 and 24.52 µL·L-1 air for C. citratus, M. piperita, and P. graveolens, respectively. In both topical and fumigant bioassays, essential oil from C. citratus was the most active altering the behavioral response of treated mealybugs which becomes hyperactive and disoriented. EOs induced general stress in P. ficus adults, as evidenced by oxidative stress biomarker analyses. Biochemical analyses showed that the EOs exposure reduced the activity of acetylcholinesterase and significantly induced the glutathione S-transferases and Malondialdehydes accumulation in the vine mealybug tissues. Mortality caused by lemongrass EO positively correlated with the significant decrease in the AChE activity indicating lethal neurological effects. These toxicity bioassays and neurological impact findings provide new informations for formulating effective essential oil based-insecticides to control P. ficus in the framework of integrated pest management programs

    Evaluation of Sensor-based Early Detection Methods for Grapevine Diseases like Palatinate grapevine yellows, Bois noir, Grapevine leafroll disease and Esca

    Während ihrer Standzeit über mehrere Jahrzehnte kann die Weinrebe (Vitis vinifera ssp. vinifera) von einer Vielzahl unterschiedlicher Pathogene infiziert werden, wovon einige in der Rebe verbleiben und sich so im Laufe der Zeit ansammeln. Derzeit stehen nur prophylaktische Ansätze zum Schutz vor vielen dieser endogenen Krankheiten zur Verfügung. Diese Maßnahmen beinhalten beispielsweise visuelle Bonituren im Weinberg und anschließendes Roden infizierter Reben oder Pathogennachweise in Rebvermehrungsanlagen, die dazu beitragen sollen gesundes Pflanzmaterial zu produzieren. Der Einsatz von Sensor-basierten Verfahren zur Krankheitsdetektion kann hierzu einen entscheidenden Beitrag leisten. Hyperspektrale Sensoren erfassen objektiv und nicht-invasiv die Reflektion von Pflanzen im visuellen Bereich des Lichts (400 – 700 nm) sowie im Nahinfrarot- (700 – 1000 nm) und kurzwelligen Infrarot-Bereich (1000 – 2500 nm). Biochemische und biophysikalische Änderungen, die durch Pathogenbefall induziert werden, führen zu Abweichungen in den Reflektionsspektren, welche mit Hilfe verschiedener Machine und Deep Learning Modelle analysiert und Infektionen dadurch frühzeitig erkannt werden können. Im Rahmen dieser Arbeit wurde die Eignung der Hyperspektralanalyse zur Erkennung der Rebkrankheiten FD Pfalz (PGY) und Schwarzholzkrankheit (BN), welche durch Phytoplasmen hervorgerufen werden, sowie der Viruserkrankung Blattrollkrankheit (GLD) und der durch Pilze induzierten Esca Krankheit evaluiert. Anhand von Gewächshauspflanzen konnten unter kontrollierten Bedingungen Detektionsmodelle für BN und PGY erstellt werden, die bis zu 96% der Pflanzen korrekt als gesund oder infiziert klassifizierten. Die Erkennung von infizierten, aber nicht symptomatischen Pflanzen bedarf allerdings noch weiterer Versuche. Da die Symptomentwicklung beider Krankheiten stark von Umgebungsfaktoren abhängig ist, wurden zusätzlich Triebe verschiedener Rebsorten aus dem Freiland analysiert. Auch hier konnten sehr gute Klassifikationsgenauigkeiten von bis zu 100% erzielt werden, was die Vermutung nahelegt, dass beide Krankheiten auch direkt im Feld detektiert werden können. Die Erkennung von GLD erfolgte zunächst ebenfalls an Gewächshauspflanzen. Dabei konnten 83 – 100% der symptomatischen Reben und 85 – 100% der infizierten, aber nicht symptomatischen Reben von Kontrollpflanzen unterschieden werden. Darüber hinaus wurden im Zeitraum von 2016 bis 2018 rund 500 Reben im Freiland näher untersucht. Auch hier wurden ähnlich hohe Klassifikationsgenauigkeiten erzielt. Außerdem konnte das Potential der Hyperspektralanalyse zur Erkennung infizierter, aber nicht symptomatischer Reben im Freiland gezeigt werden. Da die Ergebnisse jedoch sehr stark zwischen den Versuchsjahren schwankten, bedarf es weiterer Untersuchungen zur abschließenden Bewertung dieses Aspekts. Darüber hinaus konnten Symptome der beiden GLD-Krankheitserreger Grapevine leafroll-associated virus-1 und Grapevine leafroll-associated virus-3 erfolgreich voneinander unterschieden werden. Auch die Erkennung von Esca wurde über drei Jahre hinweg im Feld durchgeführt. Hyperspektrale Detektionsmodelle konnten erfolgreich sowohl für originale Felddaten als auch für manuell annotierte Daten erstellt werden. Erste Ergebnisse zeigten zudem deutlich das Potential der präsymptomatischen Detektion. Des Weiteren wurde die Übertragbarkeit der Modelle auf unbekannte Daten simuliert. Dabei zeigte sich, dass die Modelle zwar von einem Jahr auf ein anderes Jahr übertragen werden können, die Modellperformanz dabei jedoch deutlich nachlässt. Basierend auf den Hyperspektraldaten wurden für alle Krankheiten und alle Analyseansätze die wichtigsten Wellenlängen ermittelt, um eine Vereinfachung des komplexen Systems zu ermöglichen. Anhand dieser Wellenlängen könnten schließlich multispektrale Sensoren entwickelt werden, die schneller, günstiger und flexibler einsetzbar sind als hyperspektrale Anwendungen. Im Fall von Esca wurden bereits im Rahmen dieser Arbeit zusätzlich Multispektraldaten mittels Drohne erhoben und mit dem simulierten Ansatz aus den Hyperspektraldaten verglichen. Obwohl die simulierten Multispektraldaten sehr gute Ergebnisse erzielten und somit das Potential dieser Methode aufzeigen, bleibt die Luft-gestützte Krankheitserkennung derzeit noch eine Herausforderung.Grapevine (Vitis vinifera ssp. vinifera) as a perennial crop is typically grown over several decades. During their lifespan, vines may be infected by a number of different pathogens, some of which remain inside the vine and thus accumulate over time. At the moment, only prophylactic measures are available to reduce the spread of many endogenic diseases, which include visual ratings and subsequent uprooting of infected grapevines in the field or mandatory pathogen tests in nurseries to provide healthy planting material. Sensor-based approaches could significantly contribute to early disease diagnosis. Hyperspectral sensors detect plants’ reflection objectively and non-invasively in the visible range of light (400 – 700 nm) but also in the near infrared (700 – 1000 nm) and short-wave infrared region (1000 – 2500 nm). Biochemical and biophysical changes induced by pathogen infestation cause deviations in the reflectance spectra, which can be analyzed by different machine and deep learning models enabling disease detection at early infection stages. In this study, the suitability of ground-based hyperspectral analyses for the detection of the grapevine phytoplasma diseases Palatinate grapevine yellows (PGY) and Bois noir (BN), the virus infection grapevine leafroll disease (GLD), and the fungal Esca disease has been evaluated. Disease detection models for both BN and PGY could be developed under controlled conditions using greenhouse plants. These models were able to classify plants correctly as either healthy or infected with an accuracy of up to 96%. However, identification of infected but symptomless plants needs further improvements. Since symptoms of both diseases may vary strongly depending on environmental factors, shoots collected in the field from different cultivars were also analyzed. Again, high classification accuracies of up to 100% could be achieved leading to the assumption that both diseases might also be detectable directly in the field. GLD detection was at first tested using different greenhouse plants. Thereby, 83 – 100% of symptomatic and 85 – 100% of infected but symptomless vines could be correctly identified. Moreover, approximately 500 grapevines were analyzed directly in the field during the years 2016 – 2018 leading to similar classification accuracies. Furthermore, the potential of hyperspectral analyses for the in-field detection of infected but symptomless vines could be shown. However, results strongly differed between experimental years, therefore, further analyses are necessary for a final evaluation of this aspect. Moreover, symptoms caused by the two main GLD agents Grapevine leafroll-associated virus-1 and Grapevine leafroll-associated virus-3 could successfully be discriminated. Esca disease detection was also performed directly in the field during three consecutive years. Thereby, hyperspectral detection models could successfully be established for original field data as well as for manually annotated data. In addition, first results clearly showed the potential of pre-symptomatic disease detection. Moreover, model transferability to unknown data was tested but remains challenging and will require to include further experimental years. Based on hyperspectral data, most important wavelengths were determined for every disease and every analysis approach in order to simplify this complex system. Multispectral sensors could eventually be developed using these wavelengths being faster, cheaper and more flexible than a hyperspectral application. In the case of Esca, additional airborne multispectral data were acquired during this study and compared to a multispectral simulation based on hyperspectral approaches. Although, the simulated multispectral data achieved good results, thus, showing the potential of this method, airborne disease detection needs to be improved
