    Clonal heterogeneity of the MDCK cell population: Effect on cell-to-cell variability in influenza A virus replication

    Madin-Darby canine kidney (MDCK) cells are a continuous epithelial cell line and comprise a heterogeneous non-clonal cell population with clonal subpopulations displaying differences in various characteristics. Furthermore, a large cell-to-cell variability in influenza A virus (IAV) replication was revealed among individual MDCK cells by single-cell analysis (Heldt and Kupke et al., 2015, Nat Commun 6, 8938). The present thesis is the first study that aims to reveal if the clonal heterogeneity of the MDCK cell population contributes to the large cell-to-cell variability in IAV replication. 31 clonal MDCK cell lines were derived from a parental MDCK cell population by limiting dilution cloning. The individual clones reflected heterogeneous characteristics in terms of morphology, cell size and cell density. Strikingly, two distinct groups of clones became apparent: 1) clones of small cells growing to high cell densities and 2) clones of big cells growing to low cell densities. All clones were infected with IAV and investigated for the virus titer they produce at the population level. However, no significant differences in volumetric virus titers were observed between the clones. Yet, further analysis showed that 1) clones of small cells were associated to lower and 2) clones of big cells were associated to higher cell-specific virus yields. It was therefore assumed that the combination of cell size, cell density and cell-specific virus yield, in turn, resulted in similar volumetric virus titers produced by similar cell volumes. Finally, the IAV replication was investigated at the single-cell level comparing two clones, one representing small cells and one representing big cells, with the parental MDCK cells. Single-cell analysis demonstrated that the cell-to-cell variability in extracellular virus yields and intracellular vRNA content was similarly high in all three populations. (...

    In der folgenden Ausarbeitung wird die „Projektentwicklung eines unbebauten Grundstücks an der Fördestraße 20-48 (Bebauungsplan Nr. 258) in Flensburg“ entwickelt. Eine Projektentwicklung ist das „Zusammenführen von Projektidee, Grundstück, Kapital und Nutzer mit dem Ziel, eine rentable Investition zu tätigen“. Sie beinhaltet die Projektidee, Projektkonzeption, Bauvorbereitung, Vermietung, Projektrealisierung und den Verkauf des geplanten Objekts. Betrachtet werden in dieser Arbeit die Prozesse von der Projektidee bis zur Projektkonzeption und der wirtschaftlichen Rentabilität. Es gibt mehrere unterschiedliche Ausgangssitua tionen einer Projektentwicklung. Zusammenfassend sind hier der Nutzer, das Kapital, das Grundstück und die Projektidee von zentraler Bedeutung. Diese verschiedenen Aspekte können in unterschiedlicher Art miteinander fungieren. Entscheidend ist, ob der Entwickler selbst ein bestimmtes Grundstück, das benötigte Kapital oder eine bestimmte Projektidee besitzt. In diesem Fall ist von einem Grundstück an der Fördestraße 20-48 in Flensburg auszugehen, das von der Stadt Flensburg angeboten wird. Der Projektentwickler versucht daraufhin auf dieser Fläche ein geeignetes Nutzungskonzept zu entwickeln

Indonesia is a vast country comprising many islands with a lot of business opportunity that can be explored, especially towards its consumer buying behavior. With growing of the middle-income class and its young population, given the country’s good economic growth by increased of domestic consumption. In this era of Globalization, many foreign brands have been doing their business and established their brands there. Indonesia has become a good profit market for multinational company. Therefore, until now there are still new companies invest in Indonesia for strengthen their position and expand their market to Asian market. Focusing on expanding market, it also can be applied to German Business, TITANIA Fabrik GmbH, a German medium size family company with its headquarter in Wülfrath, Germany. They have been actively on international body care market during the last 60 years and now have managed their business over 120 countries. By seeing this opportunity that they have from the product, it’s match with the behavior that still applied in Indonesian consumer perception about the German product. The product it’s have special place on Indonesian consumer mind, which is high quality and a first-class exclusive product. With this research, it will discover how consumer perception towards German cosmetic and body care product which can gain useful information for their future research and development to entering Indonesian market especially knowing as well that there is already existing foreign competitors

By the nodal tensor method the value of packet delay along a traffic transmission route for the network with bypass routes of traffic transmission and without them is received and the results comparison of traffic balancing is conducted. It is shown, that in the network with the bypass routes of traffic transmission less time of packets delay is received.     Die Herausforderung als Chance nutzen : Arbeitsmarktintegration und Beschäftigung von Flüchtlingen in Sachsen-Anhalt

    The purpose of the study is to investigate the effects of refugees on the German Labor market. The first section gives an overview on the labor conditions. The second part includes an analysis of the demographic structure of Saxony-Anhalt, a possible skill shortage resulting from the aging population and a comparison of several projects and integration processes, which support refugees in their job searches. It must be pointed out that the study will only consider refugees from the wave of refugees in 2014 to 2016. To overcome obstacles such as language barriers and bureaucracy processes, it is necessary to develop an effective change concept, which includes aspects of diversity management and integration measures, to support companies with the placement of foreign employees. Two companies, that are located in the south of Saxony-Anhalt, were selected. A questionnaire was sent to the two companies that involved an electronic engineering firm and a construction firm. An interview was recorded on an audio tape and conducted with the manager of the engineering company. The results of the study show that refugees cannot fully reduce the skill shortage in several branches in Saxony-Anhalt. On the basis of the study it can be concluded that the integration will require a long way of attending integration and language courses until refugees are able to apply for a job
