    Effect of Nickel and Titanium on DNA Methylation in Human Gingival fibroblast cells In Vitro

    Metallic dental materials are frequently used in clinics. Cytotoxicity and allergy tests in vitro and in vivo have primarily been used to evaluate their safety. However, whether these dental materials cause genetic modifications leading to pathological conditions in oral tissues has not been shown. Gene mutations and epigenetic alterations are the initial steps of genetic modification. Environmental factors, including chemical components and mechanical stimulations, can cause epigenetic modifications that are often involved in pathogenic changes in humans. Therefore, we hypothesize that dental materials induce specific alterations in DNA methylation. This study aims to demonstrate specific alterations in DNA methylations in human gingival fibroblast cells (HGnF) induced by metal components (Ni and Ti ) of dental materials. The mRNA expression level of IL−6 for 2 weeks was significantly higher in the HGnF cells treated with 50 μM of Ni and 10 μM of Ti compared to the controls. Quantitative methylation − specific PCR ( qMSP) revealed that the DNA methylation percentage levels of IL−6 and IFN−γ in HGnF cells treated with 10 μM of Ti for 2 weeks were significantly lower compared to the controls. In conclusion, this study found hypomethylation of IL−6 and IFN−γ, followed by their upregulated expressions in HGnF cells stimulated with Ti. Methylation levels may serve as a new cytotoxic assessment tool, even for biocompatible materials.departmental bulletin pape

    Relationship between head position tilt on the examination of masticatory function test.

    This study aimed to investigate whether head position influences the results of masticatory function tests. Masticatory function tests were conducted using the Glucosensor GS−II with bitescan in participants classified as Eichner A and without temporomandibular joint symptoms. A total of 126 data points were collected from 63 participants who were divided into two groups based on head position. Of the data points, 50 were obtained from the group with no head tilt during mastication, whereas 76 were obtained from the group with a head tilt to the right or left during mastication. The group with a tilted head position during mastication demonstrated a smaller decline in masticatory function compared to the group with no head tilt. This result may be due to increased mandibular displacement associated with the head tilt during mastication. These findings suggest that head tilt during mastication may be one of the factors influencing the results of masticatory function tests in individuals classified as Eichner A.departmental bulletin pape

    Change of the attitude of students taking geriatric dentistry toward older adults

    Due to the high ratio of elderly adults, knowledge, skills and attitudes regarding geriatric dentistry are considered a critical subject for students at the faculty of dentistry. The aim of this study is to observe changes in students’ attitudes of students before and after taking geriatric dentistry course using the GAS−14. Thirty−nine students were surveyed using the GAS−14 three times during their 4th, 5th and 6th years at the university. Students answered 14 questions in the GAS−14 using a 5−point rating scale ; strongly agree (5 points), agree (4 points), neutral (3 points), disagree (2 points), strongly disagree (1 point). Points to negative questions were reversed. Each point was summed to calculate the GAS score. Average GAS scores were analyzed using the Friedman test with Bonferroni correction, and the ratios of answers were analyzed using the chi−square test with Bonferroni correction. The GAS scores were 3.18±1.09 at 4th year students, 3.14±1.01 at 5th year students and 3.17±1.07 at 6th year students. There was no statistical difference among the years and the situation had not changed significantly. The response ratio for Q2 “The federal government should reallocate money from Medicare to research on AIDS or pediatric diseases.” showed a statistical difference between 4th year students and 5th year students, as well as between 4th year students and 6 th year students. Additionally, Q11 “Treatment of chronically ill old patients is hopeless.” and Q14 “It is interesting listening to old people’s accounts of their past experiences. ” also showed statistical difference between 4th year students and 5th year students, respectively. This study suggests that although the attitudes of students over a span of 3 years did not change significantly, the lectures and practices in geriatric dentistry encouraged students to contemplate the needs of older adults.departmental bulletin pape
