    Embryogenesis in Oak species. A review

    Aim of study: A review on the propagation methods of four Quercus species, namely Q. suber, Q. robur, Q. ilex and Q. canariensis, through somatic embryogenesis and anther embryogenesis are presented.Area of study: The study comprises both Mediterranean and Atlantic oak species located in Spain.Material and Methods: Somatic embryogenesis was induced on immature zygotic embryos of diverse oak species, permitting the multiplication of half-sib families. Induction of haploid embryos and doubled haploids was assayed in both Q. suber and Q. ilex by temperature stress treatments of anthers containing late vacuolated microspores. The haploid origin of the anther embryos has been evaluated by quantitative nuclear DNA analysis through flow cytometry and by DNA microsatellite markers. Genetic transformation of cork oak has also been performed by means of Agrobacterium tumefaciens vectors. Proteomic analysis has been conducted to screen the diverse protein profiles followed by in vitro derived embryos during their development.Research highlights: Successful plant regeneration from both somatic and haploid embryos has been achieved. In the particular case of cork oak, doubled-haploid plants were obtained. Plantlets regenerated from selected parent trees through somatic embryogenesis were acclimated in the greenhouse and in the nursery, and were planted in an experimental plot in the field. Preliminary evaluation of the cork quality of the plants showed a good heritability correlation with the parent trees.     Detection and molecular characterization of Polymyxa betae, transmitting agent of sugar beet rhizomania disease, in Iran

    The plasmodiophorid Polymyxa betae is considered as the only natural transmitting agent of Beet necrotic yellow vein virus (BNYVV), the most devastating agent of sugar beet fields throughout the world. To evaluate for the first time the genetic diversity of P. betae isolated from different autumn and spring sugar beet fields, and also to detect the presence of virus in these isolates, susceptible sugar beet plants (cv. Regina) were grown in soil samples collected from 10 different regions of Iran. P. betae detection was carried out using root microscopic observations, DAS-ELISA, and PCR-based methods. Results showed the presence of plasmodiophorids in all soil samples. Complementary assays also revealed the presence of viruses in soil samples collected from Khorasan Razavi, Fars, Hamadan and Kermanshah regions. The genetic diversity was evaluated through comparing glutathione-S-transferase nucleotide sequences amplified by PCR with the Internal Transcribed Spacers region. Results showed no significant differences in nucleotide sequences between virus-bearing and virus-free isolates of P. betae

    Grain quality of drought tolerant accessions within the MRI Zemun Polje maize germplasm collection

    Maize Research Institute Zemun Polje (MRI) gene bank created an elite drought tolerant core collection of 40 accessions, based on field trials and general combining ability with inbred lines from the main heterotic groups (Lancaster, Iowa Stiff Stalk Synthetic - BSSS and Iodent). A total of seven genetic groups were identified. Seven accessions showed good combining abilities with three testers from chosen heterotic groups, thus forming a dinstinctive genetic group (Unknown). In the present research, accessions with drought tolerance were also analyzed for grain quality, as these two traits are becoming highly important due to global warming and population growth. Kernel macronutrients contents (oil, protein and starch) were determined using Near Infrared Spectroscopy (NIR). Oil, protein and starch contents were significantly higher in introduced populations than in landraces for 0.43%, 0.12% and 0.85%, respectively (p<0.01). The greatest progress from the selection based on the expected genetic gain (ΔG) for 5% selection intensity would be obtained for oil (14.74%) followed by protein (10.14%). Landraces showed the least potential for the grain quality improvement due to the lowest expected ΔG for the three macronutrients. The differences between macronutrient content among genetic groups defined them as potentially favourable sources for a specific trait. According to ΔG values, the greatest progress in breeding would be accomplished for increased oil content with accessions from the Unknown group. Identification of the accessions with several favorable traits is valuable for simultaneous breeding for drought tolerance and grain quality

    Salinity contamination response to changes in irrigation management. Application of geochemical codes

    Salinity contamination caused by irrigation has been widely studied but the analysis of geochemical processes regarding agronomic variables has not adequately been considered yet. The research presented here analyzes the influence of changes in irrigation management on salinity contamination, through the use of geochemical modeling techniques, in an agricultural basin during the hydrological year of 2001 and within the period 2005-2008. The results indicate that the changes implemented in irrigation management reduced the masses of salts exported in 72%, although water salinity increased by 25% (this salinity level does not restrict its use for irrigation). The different ionic ratios in drainage water, the results of the salinity balances, and the results of geochemical calculations (mass balances and speciation-solubility) indicate, mainly, precipitation of calcite, dissolution of gypsum and halite and cation exchange. The salt contamination index decreased from approximately 70% to levels close to those presented in modern irrigation areas, indicating that the changes in irrigation management were effective. Petrocalcic genesis and punctual sodification of soils can constitute an agroenvironmental problem that requires adequate management of irrigation and drainage considering future modernization of irrigation areas

    Effects of in ovo injection of conjugated linoleic acid on immune status and blood biochemical factors of broiler chickens

    This experiment evaluated the effects of an in ovo injection of conjugated linoleic acid (CLA) on immune status and blood metabolites of broiler chicks at 21 and 42 days of age. One hundred and sixty fertilized eggs were selected from a young broiler breeder flock and allocated to4 treatments with 4 replicates each. CLA was injected into the air sack of the selected eggs (150 and 300 mg, dissolved in 100 mL of commercial diluents, CLA150 and CLA300) on Day18 of incubation. Two control groups (injected with or without diluent) were also included. In ovo injection of 150 or 300 ppm CLA increased (p<0.05) feed intake of chickens as compared to control groups up to 42 days of age. Weight gain and feed conversion ratio of chickens from CLA300, were improved (p<0.05) as compared to chicks hatched from eggs injected diluent or control group during total period. Cutaneous basophil hypersensitivity response to injection of phytohaemagglutinin-P significantly (p<0.05) increased in chickens from CLA300 comparing to chicks from CLA150 or control ones. In ovo injection of 300 ppm CLA, decreased blood serum cholesterol and low density lipoprotein (LDL) of chickens as compared to control chicks or those from CLA150 (p<0.05). Central lymphoid tissues such as bursa of Fabricius and thymus relative weights were increased (p<0.05) in chickens from CLA300 as compared to chicks injected diluent or control groups. It is concluded that feeding 300 ppm CLA through in ovo injection may be effective to subsequent growth rate, immune response and decrease blood serum cholesterol and LDL of broiler chickens during whole experimental period

    Honeybee populations around the world are experiencing a decrease in colony numbers probably due to a combination of different causes, such as diseases, poor nutrition and frequent applications of insecticides to control pests. Previous studies about the effect of pesticide Bacillus thuringiensis subsp. kurstaki (Btk) on Apis mellifera L. report different results. The aim of this study was to analyze the effect of field aerial applications of Btk on bee colony performance, specifically on the brood cell percentage evolution, which can be used as an indicator of queen health and brood development breeding rates. To achieve it, the brood cell surface was photographed in every sampling, and data were analyzed using a method based on image treatment software. A total of 480 pictures were examined from two groups of four nucleus hives in two areas, one receiving aerial spraying with Btk and the other without treatment. A mixed factorial design was realized to analyse the data showing no differences in colony performance between the two groups of colonies either before the treatment, during and at the end of the assay. Furthermore, the brood surface ratio of Btk-treated/untreated increased along the experiment. Therefore, the results of the present study suggest that Btk aerial applications did not affect the brood development of honeybees under natural conditions. Nevertheless further field studies are required to ascertain a safe use of Btk in forest pest management

    Revealing additional preference heterogeneity with an extended random parameter logit model: the case of extra virgin olive oil

    Methods that account for preference heterogeneity have received a significant amount of attention in recent literature. Most of them have focused on preference heterogeneity around the mean of the random parameters, which has been specified as a function of socio-demographic characteristics. This paper aims at analyzing consumers’ preferences towards extra-virgin olive oil in Catalonia using a methodological framework with two novelties over past studies: 1) it accounts for both preference heterogeneity around the mean and the variance; and 2) it considers both socio-demographic characteristics of consumers as well as their attitudinal factors. Estimated coefficients and moments of willingness to pay (WTP) distributions are compared with those obtained from alternative Random Parameter Logit (RPL) models. Results suggest that the proposed framework increases the goodness-of-fit and provides more useful insights for policy analysis. The most important attributes affecting consumers’ preferences towards extra virgin olive oil are the price and the product’s origin. The consumers perceive the organic olive oil attribute negatively, as they think that it is not worth paying a premium for a product that is healthy in nature

    Simulating water distribution patterns for fixed spray plate sprinkler using the ballistic theory

    Ballistic simulation of the spray sprinkler for self-propelled irrigation machines requires the incorporation of the effect of the jet impact with the deflecting plate. The kinetic energy losses produced by the jet impact with the spray plate were experimentally characterized for different nozzle sizes and two working pressures for fixed spray plate sprinklers (FSPS). A technique of low speed photography was used to determine drop velocity at the point where the jet is broken into droplets. The water distribution pattern of FSPS for different nozzle sizes, working at two pressures and under different wind conditions were characterized in field experiments. The ballistic model was calibrated to simulate water distribution in different technical and meteorological conditions. Field experiments and the ballistic model were used to obtain the model parameters (D50, n, K1and K2). The results show that kinetic energy losses decrease with nozzle diameter increments; from 80% for the smallest nozzle diameter (2 mm) to 45% for nozzle diameters larger than 5.1 mm, and from 80% for the smallest nozzle diameter (2 mm) to 34.7% for nozzle diameters larger than 6.8 mm, at 138 kPa and 69 kPa working pressures, respectively. The results from the model compared well with field observations. The calibrated model has reproduced accurately the water distribution pattern in calm (r=0.98) and high windy conditions (r=0.76). A new relationship was found between the corrector parameters (K1’ and K2’) and the wind speed. As a consequence, model simulation will be possible for untested meteorological conditions

    Evaluation of different mechanical fruit harvesting systems and oil quality in very large size olive trees

    In 2006 and 2009, trials were carried out in the Apulia region in Southern Italy to evalu-ate the possibility of mechanizing olive harvesting in groves of old and very large trees. The trees belonged to the cultivars ‘Cellina di Nardò’ and ‘Ogliarola Salentina’. They were 60-100 years old and 7-9 m tall with a canopy volume of 140-360 m3. In the first half of November 2006, with a mechanical beater mounted on a tractor plus hand-held pneumatic combs, the harvesting yield was close to 90% of the total olives present in the canopy, and the harvesting working productivity was around 60 kg of harvested olives h-1 worker-1. With a self-propelled shaker attached to the main branches the harvesting yield was about 73% in ‘Cellina di Nardò’, and 40% in ‘Ogliarola Salentina’, while the harvesting working productivities were around 103 and 85 kg of harvested olives h-1 worker-1, respectively. In the second half of No-vember 2009, in ‘Cellina di Nardò’, with a mechanical beater mounted on a tractor plus nets on the ground or a catching frame (reversed umbrella) mounted on another tractor, the harvesting yield was about 97%. The working productivity was about 98 kg of harvested olives h-1 worker-1 with the mechanical beater plus nets and around 133 kg of harvested olives h-1 worker-1 when the mechanical beater was combined with a reversed umbrella. The oil obtained from the mechanically harvested olives was always of high quality. A basic economic evaluation of the harvesting costs is also reported

    Effects of wheat cultivar, metabolizable energy level, and xylanase supplementation to laying hens diet on performance, egg quality traits, and selected blood parameters

    A 2 x 2 x 2 factorial arrangement of treatments was conducted to evaluate the effects of two dietary apparent metabolizable energy (AME) levels (2,720 and 2,580 kcal kg-1 diet) and enzyme (0 and 0.3 g kg-1 diet, Grindazym® GP 15,000 with mostly xylanase activity) supplementation on the performance of laying hens fed diets based on two wheat cultivars (Marvdasht and Sardari). Experimental diets were formulated to have a constant energy to protein ratio and were fed to 65-wk-old Lohmann LSL-Lite laying hens for 7 wk. The lower level of AME reduced egg production and egg mass (p<0.05) and increased feed conversion ratio (p<0.05). Enzyme addition increased feed intake of the birds fed a diet with Sardari cultivar (p<0.05) but had no effect on feed intake of the birds fed a diet with Marvdasht cultivar (p>0.05). Nevertheless, birds receiving diets based on Marvdasht cultivar had higher feed intake and egg mass than that of those receiving diets based on Sardari cultivar (p<0.05). The birds fed diets based on Marvdasht cultivar produced less undesired eggs and had better yolk color as compared with the birds fed diets based on Sardari cultivar (p<0.05). The serum concentration of glucose increased by enzyme supplementation when birds receiving lower AME level (p<0.05). These results indicate that enzyme supplementation may have a positive effect on the feed intake of laying hens when fed on wheat-based diets; however, this effect is cultivar dependent and does not necessarily mean that enzyme supplementation always benefit production
