Factors and Issues Related to the Environmental Impact Caused by the Life Cycle of Timber Building Construction Projects
Timber construction offers a number of advantages in terms of environmental sustainability particularly in comparison to other construction technologies such as concrete construction. Although the calculation of the environmental impact caused by the life cycle of a timber construction project provides results that display some of these advantages, it is also necessary to examine the manner in which timber construction should move forward in order to maximize its sustainability potential. The aim of the current research is to determine the key factors and issues that are related to the environmental impact caused by timber construction and thus provide the basis for future considerations regarding the optimal delivery of such projects. A timber building is used as the basis for the calculations that are used to quantify the influence of the issues examined. Furthermore, the conclusions that are derived highlight recommendations intended for application in similar projects, while also providing suggestions for the way forward regarding sustainable practice within the timber building sector

Comparative genomics among dairy strains of Streptococcus thermophilus
Microorganisms like lactic acid bacteria are employed for the biotransformation of raw materials into fermented foods. Fermented foods have increased nutritional value and shelf life as well as improved organoleptic characteristics compared to the raw materials. Interestingly, there are several genera within lactic acid bacteria that are considered to be important for food fermentations including the Streptococcus genus. However, only Streptococcus thermophilus is used as a starter culture. Streptococcus thermophilus has been adapted to milk and dairy products through a reductive evolution process that has led to the loss of typical streptococcal pathogenictraits. In this work we present the comparative genomic analysis among the recently sequenced genome of S. thermophilus ACA-DC 29 is olated from yogurt and the existing seven complete genome sequences of S. thermophilus. Full chromosome alignments revealed a high degree of synteny among the different strains although strain specific differences could also be observed. The pangenome of the eight strains was comprised of approximately 2,300 genes. Concerning the ACA-DC 29 strain, the majority of genes was distributed in the core and the accessory genomes. We also identified a significant percentage of unique genes, i.e. approximately 250, involved in various biological processes. Further analysis of these unique genes revealed that several of them may have been acquired through horizontal gene transfer. We also predicted five potential antimicrobial peptides and two CRISPR systems, which may confer resistance against phages. Overall, our analysis provides useful insights into the technological potential of the ACA-DC 29 strain

Catalytic features and crystal structure of a tau class glutathione transferase from Glycine max specifically upregulated in response to soybean mosaic virus infections
The plant tau class glutathione transferases (GSTs) play important roles in biotic and abiotic stress tolerance in crops and weeds. In this study, we systematically examined the catalytic and structural features of a GST isoenzyme from Glycine max (GmGSTU10-10). GmGSTU10-10 is a unique isoenzyme in soybean that is specifically expressed in response to biotic stress caused by soybean mosaic virus (SMV) infections. GmGSTU10-10 was cloned, expressed in Escherichia coli, purified and characterized. The results showed that GmGSTU10-10 catalyzes several different reactions and exhibits wide substrate specificity. Of particular importance is the finding that the enzyme shows high antioxidant catalytic function and acts as hydroperoxidase. In addition, its Km for GSH is significantly lower, compared to other plant GSTs, suggesting that GmGSTU10-10 is able to perform efficient catalysis under conditions where the concentration of reduced glutathione is low (e.g. oxidative stress). The crystal structure of GmGSTU10-10 was solved by molecular replacement at 1.6 Å resolution in complex with glutathione sulfenic acid (GSOH). Structural analysis showed that GmGSTU10-10 shares the same overall fold and domain organization as other plant cytosolic GSTs; however, major variations were identified in helix H9 and the upper part of helix H4 that affect the size of the active site pockets, substrate recognition and the catalytic mechanism. The results of the present study provide new information into GST diversity and give further insights into the complex regulation and enzymatic functions of this plant gene superfamily
