Isoenzyme- and Allozyme-Specific Inhibitors: 2,20-Dihydroxybenzophenones and Their Carbonyl N-Analogues that Discriminate between Human Glutathione Transferase A1-1 and P1-1 Allozymes
The selectivity of certain benzophenones and their carbonyl N-analogues was investigated towards the human GSTP1-1 allozymes A, B and C involved in MDR. The allozymes were purified from extracts derived from E. coli harbouring the plasmids pEXP5-CT/TOPO-TAhGSTP1* A, pOXO4-hGSTP1*B or pOXO4-hGSTP1*C. Compound screening with each allozyme activity indicated three compounds with appreciable inhibitory potencies, 12 and 13 with P1-1A 62% and 67%, 11 and 12 with P1-1C 51% and 70%, whereas that of 15 fell behind with P1-1B (41%). These findings were confirmed by IC50 values (74–125 lM). Enzyme inhibition kinetics, aided by molecular modelling and docking, revealed that there is competition with the substrate CDNB for the same binding site on the allozyme (Ki(13/ A) = 63.6 +- 3.0 lM, K (15/B) = 198.6 +- 14.3 lM, and Ki(11/ C) = 16.5 +- 2.7 lM). These data were brought into context by an in silico structural comparative analysis of the targeted proteins. Although the screened compounds showed moderate inhibitory potency against hGSTP1-1, remarkably, some of them demonstrated absolute isoenzyme and/or allozyme selectivity
Acquired acid adaptation of Listeria monocytogenes during its planktonic growth enhances subsequent survival of its sessile population to disinfection with natural organic compounds
In this study, the possible influence of acid adaptation of L. (monocytogenes cells during their planktonic growth on their subsequent resistance against some such compounds (i.e. lactic acid, essential oil or hydrosol of Mediterranean spice Satureja( thymbra) nupon their attachment to SS, was evaluated by simultaneously using the bead vortexing technique and a conductance method
Multichromophores Onto Graphene: Supramolecular Non-Covalent Approaches for Efficient Light Harvesting.
The idea of attaching multiple porphyrins to graphene is explored. A charged porphyrin salt is stabilized onto exfoliated graphene by taking advantage of π-π* interactions and a second porphyrin light harvester is anchored through electrostatic interactions with the former. The interactions are capable of allowing electronic communication of the second, electrostatically attached, porphyrin with graphene, effectively quenching its emission. The graphene-porphyrin-porphyrin triad is examined through optical (UV-Vis, steady state and time resolved photoluminescence) techniques, while electrochemistry is employed to study the thermodynamically favored pathways through which the interaction occurs. The porphyrin that is electrostatically stabilized onto the graphene nanoensemble shows lifetimes one order of magnitude faster than its π-π* stacked analogue suggesting a more efficient pathwa
New hybrid materials with porphyrin-ferrocene and porphyrin-pyrene covalently linked to single-walled carbon nanotubes.
Novel porphyrin derivatives bearing additional pyrene or ferrocene units as light harvesting antenna systems were synthesized and fully characterized. Following a covalent functionalization approach for single-walled carbon nanotubes (SWCNTs), stable SWCNT suspensions in common organic solvents 10 were produced. Subsequently, the resulting porphyrin-pyrene and porphyrin-ferrocene dyads were incorporated onto the nanotubes' backbone yielding donor-donor-acceptor hybrids. The resulting hybrid materials were soluble in common organic solvents and were characterized using micro-Raman, ATR-IR,
UV-Vis and photoluminescence spectroscopy, transmission electron microscopy, thermogravimetric analysis and εlectrochemistry. Photoluminescence quenching of the porphyrin emission in both hybrid 15 materials was detected thus suggesting the potentiality of these materials in photoelectrochemical cells
Insulin/Poly(ethylene glycol)-block-poly(L-lysine) Complexes: Physicochemical Properties and Protein Encapsulation
Insulin (INS) was encapsulated into complexes with poly(ethylene glycol)-block poly(L-lysine) (PEG-b-PLys), which is a polypeptide-based block copolymer (a neutral-cationic block polyelectrolyte). These macromolecules can encapsulate INS molecules in aqueous conditions via electrostatic interactions. Light scattering techniques are used in order to examine the complexation process of the hybrid nanoparticles in a gamut of buffers, as a function of protein concnetration. The physicochemical and structural characteristics of the complexes depend on the ionic strength of the aqueous medium, while the concentration of PEG-b-PLys was constant through the series of solutions. As INS concentration increased each polyelectrolyte chain interacts with an increasing number of INS molecules, the degree of charge neutralization becomes higher and the size distribution of the complexes decreased also, especially at the highest ionic strength. The size/structure of complexes diluted in biological medium indicated that the copolymer imparts stealth properties and colloidal and biological stability to the complexes, which could in turn affect the clearance properties in vivo. Therefore, these studies could be a rational roadmap for designing the optimum complexes/effective nanocarriers for proteins and peptides
