    In vivo evaluation of local theranostic approach to high-grade glioma with hypoxia-targeting radiopharmaceutical 64Cu-ATSM

    Introduction: High-grade gliomas (HGGs) are highly malignant brain tumors with a very poor prognosis. Due to their diffuse infiltrative features, small residual tumors remain around the resection cavity even after extensive resection, leading to local recurrence and indicating the need of a novel therapeutic approach. Our clinical PET study has demonstrated that Cu-diacetyl-bis (N4-methylthiosemicarbazone) (Cu-ATSM) highly accumulated in the HGGs generally expressing hypoxia marker HIF-1α. Herein, we investigated the feasibility and efficacy of local treatment and coupled PET monitoring with 64Cu-diacetyl-bis (N4-methylthiosemicarbazone) (64Cu-ATSM), a hypoxia-targeting radiopharmaceutical, to develop theranostic approaches for the local control of HGGs.Methods: The maximum tolerated dose (MTD) of 64Cu-ATSM after localadministration was determined in mice using biodistribution, dosimetry, and toxicity studies. Multiple HGG patient-derived xenograft (PDX) models recapitulating patient tumor characteristics were used for imaging and treatment studies. Imaging with PET/CT and MRI and autoradiography analyses were conducted to examine the distribution of 64Cu-ATSM after local administration. The efficacy and cytotoxic effect of 64Cu-ATSM local treatment were then investigated.Results: The MTD of 64Cu-ATSM local treatment in mice was 3.7 MBq. After the local treatment with MTD,Imaging and autoradiography analyses demonstrated high tumor penetration, distribution, and retention of64Cu-ATSM within HGG PDX tumors, with highly expressed hypoxic markers of HIF-1α and BNIP3. A single 64Cu-ATSM local treatment significantly prolonged overall survival as well as potently induced DNA double-strand breaks and apoptosis in tumors.Conclusions: Local treatment and PET monitoring with 64Cu-ATSM may be a novel theranostic approach for treating HGGs.SNMMI 2025 Annual Meetingconference poste

    Uterine serous carcinoma (USC) is a rare aggressive type 2 endometrial carcinoma with dismal prognosis. Peritoneal metastasis (PM) is often observed even at early stage in USC. HER2 is a therapeutic target of USC and trastuzumab, the anti-HER2 antibody, has shown benefit in the treatment of HER2 positive USC. However, the acquired resistance of trastuzumab also has been challenges to overcome for HER2 positive USC. Targeted alpha therapy (TAT) is useful not only for local tumor but also metastasis. Having the target specificity of antibody and high cell cytotoxicity and short range of alpha-particle, TAT can effectively control targeted tumor with minimal toxicity to untargeted normal tissues. Astatine 211 (211 At) is one of the attractive alpha-emitter. We have performed experimental TAT using 211At-trastuzumab to preclinical mouse model with trastuzumab resistant PM of HER2 positive USC in order to investigate the therapeutic efficacy and toxicity.     心の中に想像するイメージを脳から読み出し、映像として可視化することはできるだろうか?脳の信号から、見ている画像や運動の意図などを読み出す技術は「脳情報デコーディング」と呼ばれ、AI・機械学習の力を借りて大きく発展してきました。しかし、多くの研究で見ている画像を脳信号から復元できることは2008年ごろから示されてきましたが、心の中に想像しているイメージの外在化は難しく、課題として残されてきました。成功例を報告する先行研究は2,3件あったものの、対象をアルファベット文字や単純な図形に絞ったものに限られていました。2024年、我々は制限なしでの想起イメージの可視化に挑み、ある程度の精度で成功しました[1]。今回、これについて紹介します。また、近年行っている量子アルゴリズムや量子計算から着想を得たアルゴリズムを脳データ解析に用いる取り組みも紹介します[2,3,4]。どちらの取り組み・試みもまだまだ課題が多く、多種多様なバックグラウンドをもつBeyond AIのメンバーとその点を議論できればと思います。[1] Mental image reconstruction from human brain activity.     Metabotropic glutamate receptor 1 (mGluR1), a key mediator of glutamatergic signaling, is frequently overexpressedin tumor cells and is an attractive drug target for most cancers. Here, we present a targeted radiopharmaceuticaltherapy strategy that antagonistically recognizes mGluR1 and eradicates mGluR1+ humantumors by harnessing a small-molecule alpha (a)-emitting radiopharmaceutical, 211At-AITM. A single doseof 211At-AITM (2.96 MBq) in mGluR1+ cancers exhibits long-lasting in vivo antitumor efficacy across sevensubtypes of four of the most common tumors, namely, breast cancer, pancreatic cancer, melanoma, and coloncancers, with little toxicity. Moreover, complete regression of mGluR1+ breast cancer and pancreatic canceris observed in approximate 50% of tumor-bearing mice. Mechanistically, the functions of 211At-AITM areuncovered in downregulating mGluR1 oncoprotein and inducing senescence of tumor cells with a reprogrammedsenescence-associated secretory phenotype.     High temperature operating characteristics of 4H-SiC Active Pixel Sensors

    Silicon carbide (SiC) photo diodes (PDs) and active pixel sensors (APSs) were fabricated and their characteristics were investigated raising temperature from 50 to 500 degrees. Quantum efficiency (QE) of PDs was measured in the wavelength range from 200 nm to 550 nm. Output voltage of the APS was measured under dark and under UV irradiation at 250 nm. Successful operation of SiC PD and APS even at 500 degrees was demonstrated, additionally, output waveforms of the APS increased with increasing temperature. It indicates increase in the gain of SiC MOSFET as the source follower amplifier.
