MetaLP: An integrative linear programming method for protein inference in metaproteomics
Metaproteomics based on high-throughput tandem mass spectrometry (MS/MS) plays a crucial role in characterizing microbiome functions. The acquired MS/MS data is searched against a protein sequence database to identify peptides, which are then used to infer a list of proteins present in a metaproteome sample. While the problem of protein inference has been well-studied for proteomics of single organisms, it remains a major challenge for metaproteomics of complex microbial communities because of the large number of degenerate peptides shared among homologous proteins in different organisms. This challenge calls for improved discrimination of true protein identifications from false protein identifications given a set of unique and degenerate peptides identified in metaproteomics. MetaLP was developed here for protein inference in metaproteomics using an integrative linear programming method. Taxonomic abundance information extracted from metagenomics shotgun sequencing or 16s rRNA gene amplicon sequencing, was incorporated as prior information in MetaLP. Benchmarking with mock, human gut, soil, and marine microbial communities demonstrated significantly higher numbers of protein identifications by MetaLP than ProteinLP, PeptideProphet, DeepPep, PIPQ, and Sipros Ensemble. In conclusion, MetaLP could substantially improve protein inference for complex metaproteomes by incorporating taxonomic abundance information in a linear programming model

Although preventable, pressure injuries are an overwhelming problem in the healthcare system and can lead to poor quality of life, increased length of hospitalization, and unsatisfactory patient outcomes. Many cardiac surgeries take anywhere from six to twelve hours, and patients are completely immobilized for the entire length of time. Alternating pressure overlays placed on the operating room table provide tissue offloading with inflating and deflating nodes, leading to micro-movements on high pressure bony prominences. In modern times it is an expectation of students that technology be incorporated into how they receive their education. The COVID-19 pandemic brought to light how pure reliance on traditional face-to-face (F2F) learning can be detrimental to student learning. Many institutes of higher education had already implemented hybrid/blended learning prior to the pandemic and saw no difference in their NCLEX-RN pass rates. This evidence-based practice project gathered, reviewed, and synthesized evidence in order to determine if implementing different teaching strategies would maintain or improve student performance on both unit exams and end of course exit exams
A Mixed Methods Study of Cultural Competence Among Nursing Students in Kenya
Significance and Background: Kenya has been experiencing an increase in cultural and ethnic diversity. As a result of cultural differences, health disparities among ethnic groups in Kenya are increasing. A component of patient centered care involves the incorporation of cultural aspects of health and illness in the delivery of nursing care. Understanding cultural competence of nursing students will provide guidance on educational needs related to cultural competence.
Purpose:. The purpose of this study was to assess nursing students’ cultural competence (CC) in the areas of cultural awareness, knowledge, sensitivity, skills, encounters and desire. Nursing students’ views on caring for culturally diverse patients and how their views relate to CC were also explored.
Design: A convergent mixed methods design was used to answer the following research questions: a) What are the levels of CC in nursing students in Kenya? b) what demographic and cultural characteristics impact the students’ level of CC? c) what are the nursing students’ views on caring for diverse patients? and d) how do the nursing students’ views on caring for diverse patients compare to or explain the nursing students’ level of CC?
Methods: A convenient sample of nursing students (N = 190) from a university in Kenya were recruited. Cultural competence was assessed using a questionnaire and interviews. Descriptive, inferential statistics, multiple regression and content analyses were used to analyze responses. Common concepts from both quantitative and qualitative data were identified.
Results: Quantitative results showed that most nursing students had a high level of CC. The highest scores were in the awareness and knowledge subscales. Skills and sensitivity were ranked lower in the subscales. Working in rural areas, private healthcare settings, having taken more continuous education units (CEUs) and lower age were associated with greater CC. Qualitative and quantitative data converged for most variables in the study. Areas of divergence explained why the scores were not at the highest possible level.
Conclusion: These findings inform stakeholders in education and healthcare about nursing students\u27 needs for CC. Strategies to support nursing students’ ongoing competency are recommended
ROLE OF CMYC IN THE PROLIFERATION AND FUNCTION OF CYTOKINE INDUCED HUMAN MEMORY LIKE NK CELLS
Natural killer cells protect against virus infection and tumors. Although initially recognized as a part of the innate immunity, several recent studies have revealed NK cells exhibit immune memory to pathogens and haptens. NK cell memory has also been observed after exposure to combination of cytokines (IL12, IL15, IL18). The cytokine induced memory like NK cells exhibited potent antitumor immunity. To understand the role of cMyc in effector function of cytokine induced memory like NK cells, the effects of cMyc chemical inhibitor and Myc gene deletion by Crispr/Cas9 were studied. We found that treatment with cMyc inhibitor (cMYCi975) reduced NK cell proliferation, IFN-γ production and partially inhibited NK cell mediated tumor cell killing. Moreover, CRISPR/Cas9 mediated deletion of Myc gene was validated using real time PCR and Myc deletion compromised NK cell proliferation. Overall, our results suggested involvement of cMyc in the proliferation and function of cytokine induced memory like NK cells
