    Co-infection of Trichuris vulpis and Toxocara canis in different aged dogs: Influence on the haematological indices

    Trichuris vulpis and Toxocara canis are worldwide parasitic nematodes affecting dogs and mammals of the Canine family. Due to the special structure of the shells, the eggs of these geohelminths can maintain their viability in soil, sand, water and the environment for a long time. The study involved young (6–12 months old) and adult (1.5–8.0 years old) dogs affected by co-infection of T. vulpis + T. canis; the control group consisted of dewormed healthy animals of the same age. Parasitological examination of dogs' faeces was performed using a "Counting Chamber for Ovoscopic Researches"; morphological parameters, indicators of nutrient metabolism, mineral metabolism and activity of enzymatic systems were determined in blood and serum. According to the results of parasitological research on the dogs, it was found that young animals are more prone to toxocarosis, and adults – trichurosis. The co-infection of nematodes T. vulpis + T. canis in dogs develops several changes in haematological parameters: a significant decrease in erythrocytes, haemoglobin, MCV, MCH, and haematocrit, leukocytosis with basophils and eosinophilia in young infected animals; and eosinophilia and basophilia (15.9 times) in adults, compared with healthy dogs of the same age. Among the changes in serum biochemical parameters, young infected dogs showed a decrease in the concentration of total protein and albumin content, an increase in the content of "acute phase" proteins (α-1, α-2 and β-globulins), an increase in cholesterol and total bilirubin; in adult infected dogs, a decrease in albumin content, an increase in the content of α-1, α-2, and β-globulins, an increase in the concentration of cholesterol and total bilirubin, a decrease in the concentration of urea in comparison with healthy animals were determined. Also, among the indicators of mineral metabolism, a decrease in the concentration of calcium and magnesium in the serum was found in young infected dogs. Among the changes in the enzyme metabolism in the serum of infected dogs, there was an increase in the activity of all studied enzymes in animals of both research groups: ALT, AST, α-amylase, GGT and ALP. Thus, co-infection with nematodes T. vulpis + T. canis has a complex pathogenic effect on the body of dogs of all ages, which manifested itself in multiple changes in haematological parameters. In the future, the authors’ team plan to develop comprehensive measures to combat nematode infections in different living conditions of dogs, taking into account the results of the current research

    Impact of lead on the amount of chlorophyll and carotenoids in the leaves of Triticum durum and T. aestivum, Hordeum vulgare and Avena sativa

    Lead is one of the most dangerous pollutants to both the environment and humans. It causes structural changes in photosynthetic apparatus and reduced biosynthesis of chlorophyll pigments inhibits carbon metabolism. The aim of our study was to determine the dynamics of photosynthetic pigments in leaves of wheat (Triticum durum and T. aestivum), barley (Hordeum vulgare) and oats (Avena sativa) at different lead acetate, Pb(CH3COO)2 levels: 0, 0.15, 0.30 and 0.60 g/L. The results of this research indicate that these concentrations significantly affected chlorophyll content of H. vulgare and A. sativa as compared to T. durum and T. aestivum. Analysis of variance showed that lead concentration and interaction between cereal species had a significant effect on all chlorophyll characteristics at 0.1% probability and on carotenoids contents at 1% significance. Lead acetate in 0.3 and 0.6 g/L concentrations had a highly significant effect on chlorophyll a, b and carotenoids in H. vulgare seedlings, its carotenoid contents increased from 0.002 mg/g FW at 0 g/L to 0.107 mg/g FW at 0.6 g/L, whereas its chlorophyll content decreased under heavy metal stress, corresponding to the concentration of the metal ion. Carotenoids of A. sativa were not affected compared to Chl a and Chl b, while higher concentrations significantly increased chlorophyll contents of the seedlings from 1.384 mg/g FW of total chlorophyll at 0 g/L to 1.883 mg/g FW at 0.6 g/L. The increased amount of carotenoids was indicative of the formation of free radicals in plants under heavy metal stress, while decreased levels of chlorophyll content were an indication of reduction in the growth of the plants leading to decrease in the yield. It is suggested that chlorophyll content can be adopted as a very useful in vivo indicator of heavy metal toxicity

    Changes in the structure and dominance of the zooplankton community of the Kremenchuk Reservoir under the effect of climate changes

    Zooplankton plays an important role in aquatic food webs and changes in its abundance and diversity under changing climatic conditions can significantly affect the abundance and diversity of other aquatic organisms. The aim of the study was to assess quantitative and qualitative parameters of zooplankton development, dynamics of development of its dominant groups and the effect of water temperature. Species composition and abundance of zooplankton of the Kremenchuk Reservoir were studied in 2006, 2010–2012 and 2020 in relation to water temperature and pollution. Zooplankton species diversity fluctuated significantly over the years of the study. A total of 46 taxa were recorded in the reservoir during the study period. The number of recorded zooplankton taxa ranged 26 to 32 depending on the year. The most abundant zooplankters were Chydorus sphaericus, Copepoda nauplii, Brachionus diversicornis. The abundance of zooplankters in the reservoir during the study period ranged 23·103 to 256·103 ind./m3, and biomass 0.14 to 0.89 g/m3. A significant positive relationship was observed between the abundance of cladocerans, including some individual species (Ch. sphaericus and Ceriodaphnia sp.), and water temperature. Indicator species of water pollution in different years and in different parts of the Kremenchuk Reservoir differed significantly, which probably depended on the presence or absence (in a certain period of time) of polluting discharges. The total saprobity index in different years in different parts of the reservoir was in the range of 1.5–1.9. Structural indicators of zooplankton and its dominant complexes characterize the water in the Kremenchuk Reservoir as β-mesosaprobic. It is probably too early to assess the impact of climate change zooplankton as these changes are still unstable and short in time, but it is necessary to constantly monitor the biota of aquatic ecosystems to further study and summarize the data, which could later allow an identification of such changes

    Fouling communities of microscopic fungi on various substrates of the Black Sea

    Fungi are the most active biodeteriorators of natural and man-made materials. The article presents generalizations of the studies (2001–2019) of communities of microscopic fungi within biofilms on various substrates: shells of live Mytilus (Mytilus galloprovincialis, 670 specimens) and Ostreidae (Crassostrea gigas, 90 specimens), fragments of driftwood (over 7,000), stones (40), concrete of hydrotechnical constructions along the shoreline (80) and wood between concrete blocks in constructions on the shores (80). The studies were carried out in Odessa Oblast, the coastal zone of Sevastopol and open area of the Black Sea. There were identified 123 species of micromycetes, belonging to 65 genera, 33 families, 21 orders, 10 classes, 4 divisions, 2 kingdoms: Fungi and Chromista (fungi-like organisms). The Chromista kingdom was represented by 1 species – Ostracoblabe implexa, on shells of C. gigas. The number of species of micromycetes on various substrates varied 23 (wood between concrete blocks of hydrotechnical constructions) to 74 (shells of M. galloprovincialis at the depths of 3 and 6 m). On all the substrates, the following species were found; Alternaria alternata, Botryotrichum murorum. The communities were found to contain pathogenic fungi Aspergillus fumigatus (shells of mollusks, stones, concrete), A. terreus (concrete), Fusarium oxysporum, Pseudallescheria boydii (shells of mollusks). The best representation was seen for the Pleosporales order – from 12.9% (shells of M. galloprovincialis, 0.3 m depth) to 33.3% (shells of C. gigas) of the species composition. Toxin-producing species of Microascales in mycological communities accounted for 1.6% (driftwood) to 40.0% (concrete), and were also observed on shells of Bivalvia – 11.1–32.3%. Similarity of species composition of mycological communities according to Bray-Curtis coefficient varied 21.1% (driftwood and concrete, 10 shared species) to 72.7% (shells of M. galloprovincialis, the depths of 3 and 7 m and shells of C. gigas, 45 shared species). Using graphs of indices of mean taxonomic distinctness (AvTD, Δ+) and variation (Variation in Taxonomic Distinctness index, VarTD, Λ+), we determined deviations of taxonomic structure of the studied mycological communities from the level of mean expected values, calculated based on the list of species, taking into account their systematic positions. The lowest values of index Δ+ were determined for communities on shells of M. galloprovincialis, 0.3 m depth, driftwood, stones and concrete. These communities had uneven distribution of species according to higher taxonomic ranks and minimum number of the highest taxa: 4–6 classes, 1–2 divisions, Fungi kingdom. Disproportion in species composition with decrease in the number of the highest taxa occurred in extreme environmental conditions. Using index Λ+, we found that the most complex taxonomic structure of fungi communities has developed on concrete and shells of C. gigas. In mycological communities on those substrates, the number of species was low (25 and 46), but they belonged to 4–7 classes, 2–3 divisions, 1–2 kingdoms. To compare the structures of mycological communities that have developed in such substrates in biotopes sea, sea-land-air, land-air, we compiled a list of fungi based on the literature data, which, taking into account our data, comprised 445 species of 240 genera, 103 families, 51 orders, 15 classes, 5 divisions, 2 kingdoms. The analysis revealed that on substrates with similar chemical composition, in all the biotopes, the species of the same divisions dominated (genus and family may vary). Therefore, in the biotope land-air – Hypocreales, Pleosporales, Eurotiales (genera Acremonium, Fusarium, Alternaria, Aspergillus, Penicillium); sea – Pleosporales, Eurotiales, Microascales (Alternaria, Aspergillus, Penicillium, Corollospora); sea-land-air – Pleosporales, Microascales (Alternaria, Leptosphaeria, Aspergillus, Penicillium, Corollospora, Halosarpheia). Monitoring of species composition of myxomycetes is needed in farms that cultivate industrial objects, recreation sites, various buildings for prevention of mycotoxin intoxication and infestation by mycodermatoses and other diseases caused by opportunistic and pathogenic fungi

    Water vole (Arvicola amphibius) as an object of long-term biomonitoring in a floodplain area (Western Siberia, Russia)

    The paper is devoted to the dynamics of the water vole population in the conditions of total deforestation of valley forests and their subsequent restoration. We analyzed the relative population of small mammals in the typical biotopes in the Tom River basin (Western Siberia) on the border of the forest-steppe and taiga zones. From 1978 to 2019, 1,139 water voles Arvicola amphibius (Linnaeus, 1758) (synonym of A. terrestris) were caught with 50-meter trapping grooves; for 788 individuals we assessed the condition of non-metric features (phenes) of the skull. It was found that changes in the population level are non-cyclical, while against the background of a generally low occurrence of the species in the region, the population level increased tenfold in some years. It was shown that such episodic population surges can significantly impact the structure of the community of small mammals. In the valley of the Tom River, the water vole prefers inhabit wet meadows and ecotone areas between the meadow and the dark coniferous taiga forest. During the years of population surges, the water vole intensely occupies new nesting sites due to the dispersal activity of young animals but at the same time retains the original biotopic preferences. Dispersal of the species takes place in waterlogged wetlands. Most of the animals caught during the peak of their numbers were young animals of late broods born from overwintered individuals. The conducted phenetic analysis revealed the heterogeneity of young animals during the population surge, which allowed us to assume the participation of several populations in the formation of the peak. In the final surge year, the surge was characterized by an extremely low percentage of participation in the breeding of young females and the appearance of a large number of weakened animals, which led to crisis in the species population and the disappearance of the water vole from the captures. The research shows that one cannot predict the success of this process at the current stage; therefore, after flooding, it is necessary to continue monitoring

    Allelopathic potential and cytostatic activity of Antarctic moss Sanionia georgicouncinata (Amblystegiaceae)

    From the first half of the XX century, the maritime Antarctic was a region with high regional warming rates. At the beginning of the XXI century, regional warming strongly slowed down, but during the next twenty years the near-surface temperatures and mean annual precipitation at the Antarctic Peninsula will most probably increase. Higher temperatures and precipitation make the conditions in the maritime Antarctic more favourable for plants, including invasive species. Moreover, the increasing number of Antarctic tourists unintentionally transport thousands of seeds to the Antarctic on their clothes and equipment. In such circumstances, it is important to evaluate possible defensive mechanisms of native plant species, including their allelopathic potential. The objective of this study was to evaluate the allelopathic potential and cytostatic activity of Sanionia georgicouncinata. This species is widespread in the maritime Antarctic and is one of the most abundant moss species on the Argentine Islands (Wilhelm Archipelago). Seeds of cucumber Cucumis sativus, radish Raphanus raphanistrum subsp. sativus, wheat Triticum vulgare, and amaranth Amaranthus paniculatus were used as bioassay species. Fresh green biomass of S. georgicouncinata was used to prepare aqueous extracts with 1 : 10, 1 : 50, and 1 : 100 plant to water ratios. The average root length of wheat sprouts and cucumber sprouts in distilled water did not differ significantly from those in plant extracts. The root length of radish sprouts in 1 : 50 plant extract was significantly shorter than in distilled water. Meanwhile, in 1 : 50 and 1 : 100 water extracts, roots of amaranth plants were significantly longer than in water. The number of lateral roots of cucumber sprouts in water and studied aqueous extracts varied from 1 to 6 with medians from 3 to 4. The medians of control (distilled water) and sample groups (aqueous extracts) did not differ significantly, but only 38.1 ± 6.1% of cucumber sprouts developed lateral roots in the 1 : 10 aqueous extract of S. georgicouncinata, and even less – 28.3 ± 7.5% in the 1 : 50 aqueous extract. The results of this study show that the allelopathic potential of S. georgicouncinata is low. However, a significant cytostatic activity of the studied extracts shows that this species may still affect other plants and animals. Further research is needed to evaluate the impact of the studied extracts on root growth and germination of the invasive plants, particularly Poa annua

    Phytoplankton succession in the anthropogenic and climate ecological transformation of freshwater ecosystems

    The study is focused on the influence of mechanisms of anthropogenic activity and climate changes of different types and levels on the autotrophic link of freshwater ecosystems. The studies of the river, lake, water reservoir and quarry phytoplankton in the territory of the Ukrainian Polissia were carried out during 2005–2019 according to the standard methods. At the same time, we determined water hydrophysical and hydrochemical characteristics. Exactly 812 algal species represented by 877 infraspecific taxa were identified, including 75 new ones for the Ukrainian Polissia, with a significant share of thermophiles and halophiles. The dominance of Chlorophyta, Bacillariophyta, Euglenozoa and Cyanobacteria was observed in all types of water bodies. With the increase in the nutrient content in the composition of dominant complexes of river ecosystems, the role of centric diatoms Cyclotella meneghiniana, C. stelligera, Stephanodiscus hantzschii increased. The vegetation intensity of Aphanizomenon flos-aquae increased in the water courses with partially regulated flow, while vegetation of Chrysococcus rufescens, Pseudokephyrion pillidium, Kephyrion ovum intensified in rivers with increased colour of water. Water reservoirs are dominated by representatives of Bacillariophyta, Cyanobacteria, Euglenozoa, Miozoa, mostly by Aphanizomenon flos-aquae and Peridinium cinctum. The lowest number of dominant species was identified in the lakes, and belonged to the genera Trachelomonas, Peridinium, Chlamydomonas, Crucigeniella, Cyclotella, Aulacoseira. The dominant species in quarries were Cyclotella meneghiniana, Chlamydomonas monadina, Ch. globosa, Coelastrum microporum, Gomphosphaeria aponina, Trachelomonas volvocina and Microcystis aeruginosa. The obtained data can be used to predict scenarios of changes in the autotrophic link under the influence of natural and anthropogenic factors, to develop methods for monitoring and managing ecosystems at different levels of anthropogenic transformation necessary to ensure an appropriate level of their functions and ecological services

    Long-term dynamics of photosynthetic pigments in plankton of a large plains reservoir

    Long-term observations are necessary to analyze and predict changes occurring in biological communities under global and regional climatic changes. The data on seasonal and long-term dynamics of chlorophyll in plankton of the Rybinsk Reservoir (Upper Volga, Russia) obtained in May – October 2009–2019 at six standard stations using the fluorescence method are presented. In the years with contrasting hydroclimatic conditions and water regime, the chlorophyll content varied from 1–3 to >100 μg/L. The significant variability of mean seasonal values (7.9 ± 0.5 μg/L in 2009 to 27.6 ± 1.7 μg/L in 2013 with variation coefficients of 52–134%) indicates the low resistance of the community. The total chlorophyll content is associated with the development of three main phytoplankton divisions i.e., diatoms, cyanoprokaryots, and green algae. The trophic status of the reservoir was characterized as mesotrophic in 2009 and 2017, eutrophic in 2011–2014, and moderately eutrophic in other years. In the long-term seasonal cycle of phytoplankton, there are five periods with stable temperature conditions and transparency, but variable chlorophyll content. A moderate positive relationship was found between the seasonal dynamics of chlorophyll and water temperature, but a moderate negative relationship with transparency. The priority factors regulating the long-term dynamics of chlorophyll include the NAO indices, Wolf numbers, temperature, and underwater light conditions, as well as the inflow volume and water level.     Regulation of superoxide dismutase activity in soybean plants by inoculating seeds with rhizobia containing nanoparticles of metal carboxylates under conditions of different water supply

    Soybean is one of the most profitable advanced crops in agricultural production in Ukraine and the world as a whole. Therefore, studies of means of regulation and increase in the adaptive capacity of soybeans in symbiosis with nodule bacteria under the action of unfavourable environmental factors are relevant and should be aimed at the use of complex bacterial compositions involving modern nanotechnological approaches. Nanocarboxylates of ferrum, molybdenum and germanium metals were used as components of rhizobia inoculation suspension for soybean seed treatment to study the effectiveness of their complex effect on the regulation of the activity of the key antioxidant enzyme superoxide dismutase in plants under drought. Various symbiotic systems were used, which included soybean plants and inoculation suspensions based on the active, virulent Tn5-mutant Bradyrhizobium japonicum B1-20 by adding nanoparticles of ferrum, germanium and molybdenum carboxylates to the culture medium in a ratio of 1: 1000. Citric acid was the chelator. A model drought lasting 14 days was created during the period of active fixation of atmospheric molecular nitrogen by root nodules of soybeans in the budding and flowering stages, by means of controlled watering of plants to 30% of the total moisture content. In the stage of bean formation, watering of plants was resumed to the optimal level – 60% of the total moisture content. The control was soybean plants, the seeds of which were inoculated with a suspension of rhizobia without the addition of chelated metals. The following research methods were used in the work – microbiological, physiological and biochemical. According to the results, it was found that when nanoparticles of carboxylates of ferrum, molybdenum and germanium were added to the inoculation suspension of rhizobia, there was an increase in superoxide dismutase activity in root nodules and a decrease in soybean leaves under optimal water supply conditions of plants. This indicates the initial changes in the activity of the antioxidant enzyme in these symbiotic systems, induced by the influence of chelated metals in combination with the rhizobia of the active Tn5-mutant B. japonicum B1-20. Prolonged drought induced an increase in the overall level of superoxide dismutase activity in soybean nodules and leaves, compared to plants grown under optimal watering conditions. The symbiotic system formed by soybeans and B. japonicum with molybdenum carboxylate nanoparticles was the most sensitive to long-term drought exposure, compared to two other soybean-rhizobial symbioses using ferrum and germanium nanocarboxylates. This was manifested in the unstable reaction of the enzyme to the action of drought – suppression or intensification of the level of its activity in the root nodules and leaves of soybeans inoculated with rhizobia containing molybdenum carboxylate nanoparticles. In symbiotic systems with the participation of germanium and ferrum nanocarboxylates, slight changes were revealed in superoxide dismutase activity in root nodules and leaves of plants during drought and restoration of enzyme activity to the level of plants with optimal watering after water stress. It is concluded that the addition to the culture medium of rhizobia Tn5-mutant B1-20 of nanocarboxylates of germanium or ferrum is an effective means of regulating the activity of the antioxidant enzyme superoxide dismutase in soybean root nodules and leaves, which can contribute to an increase in the protective properties and adaptation of plants to the action of dehydration

    Variability of the structure of winter microbial communities in Chelyabinsk lakes

    Microorganisms form complex and dynamic communities that play a key role in the biogeochemical cycles of lakes. A high level of urbanization is currently a serious threat to bacterial communities and the ecosystem of freshwater bodies. To assess the contribution of anthropogenic load to variations in the structure of winter microbial communities in lakes, microorganisms of four water bodies of Chelyabinsk region were studied for the first time. We used cultural, chromatography-mass spectrometric, and modern methods of statistical data processing (particularly, multivariate exploratory analysis and canonical analysis of correspondences). The research showed that the composition of winter microbial communities in lakes Chebarkul’, Smolino, Pervoye, and Shershenevskoye Reservoir did not differ significantly between the main phyla of microorganisms. The dominant microorganisms were found to be of the Firmicutes phylum and Actinobacteria phylum. The structure of bacterial communities had special features depending on the characteristics of the water body and the sampling depths. Thus, in the lakes Smolino, Pervoye, and Shershenevskoye Reservoir, an important role was played by associations between microorganisms – indicators of fecal contamination: coliform bacteria and Enterococcus. On the contrary, in Chebarkul’ Lake, members of the genus Bacillus, which are natural bioremediators, formed stable winter associations. However, the differences between water bodies and sampling depths reflected 28.1% and 9.8% of the variability of the winter microbial communities, respectively. The largest contribution (about 60%) to the variability of the structure was made by intra-water processes, which determined the high heterogeneity of samples from different water areas. We assume that an important role in this variability was played by the high anthropogenic impact in a large industrial metropolis. In our opinion, this line of research is very promising for addressing key environmental issues
